Study on Genotoxicity of Crocin, a Component of Gardenia Fruit, in Bacterial and Mammalian Cell Systems

  • Crocin is one of the major components of gardenia fruit and saffron which are widely used as natural food colorants and as traditional Chinese medicines. However, the genotoxicity data on crocin are not sufficient for safety evaluation. The purpose of this study was the examination of the genotoxicity on crocin from gardenia yellow in bacterial and mammalian cells, using various genotoxic battery testing assays and the influence of crocin on methyl methanesulfonate (MMS) and ${H_2}{O_2}$-induced DNA damage in vitro, using single cell gel electrophoresis (comet) assay. From results, no considerable mutagenicity and clastogenicity were seen in bacteria and mammalian cells treated with crocin, by Ames test, chromosomal aberration assay, ${tk}^{+/-}$ gene forward mutation assay and comet assay. And, post-treatment with crocin significantly suppressed ${H_2}{O_2}$-induced DNA damage in a dose-dependent manner. In conclusion, the findings of the present study and other previous observations indicate that crocin has no genotoxic potential. And it showed that crocin clearly repressed the genotoxic potency of ${H_2}{O_2}$. These results suggest that anti-oxidative effects of crocin may be involved in the protective effects of DNA damage.

Anti-biofilm Activity of Xanthorrhizol Isolated from Curcuma xanthorrhiza Roxb. against Bacterial Biofilms Formed by Saliva and Artificial Multi-species Oral Strains

  • Xanthorrhizol, a sesquiterpene isolated from Curcuma xanthorrhiza Roxb., was used to investigate its effect on reducing the saliva and multi-species oral biofilms consisting of Streptococcus mutans, Streptococcus sanguis, and Actinomyces viscosus by anti-biofilm and confocal laser scanning microscopy (CLSM) assays. Xanthorrhizol exhibited significant antibiofilm activity in the dose- and time-dependent manners. Exposure to 2 and $5{\mu}g/mL$ xanthorrhizol for 30 min remained <50% of saliva and multi-species biofilms formed for 24 hr. In addition, exposure to $10{\mu}g/mL$ xanthorrhizol for 30 min reduced 65 and 77% of 24 hr saliva and multi-species oral biofilms, respectively. CLSM results visually demonstrated that xanthorrhizol reduced bacterial viability in the saliva and multi-species oral biofilms. These results suggest that C. xanthorrhiza Roxb. containing xanthorrhizol with strong anti-biofilm activity can be employed as a plant source for oral care functional foods.

Inhibitory Effects of Artemisia asiatica on Osteoclast Formation Induced by Periodontopathogens

  • Bone resorption surrounding tooth root causes tooth loss in periodontitis patients. Osteoclast has bone resorption activity. Effects of Artemisia asiatica on bone resorption induced by periodontopathogens, Porphyromonas gingivalis and Treponema denticola, were examined using co-culture systems of mouse osteoblasts and bone marrow cells. Addition of A. asiatica ethanol extract to bacterial sonicate abolished bacteria-induced osteoclastogenesis. To determine inhibitory mechanism of A. asiatica against osteoclastogenesis, effects of A. asiatica on expressions of osteoclastogenesis-inducing factors such as receptor activator of NF-${\kappa}B$ ligand (RANKL), prostaglandin $E_2\;(PGE_2)$, interleukin (IL)-1, and tumor necrosis factor (TNF)-${\alpha}$, in osteoblasts were examined. A. asiatica suppressed expressions of RANKL, $PGE_2$, IL-$1{\beta}$, and TNF-${\alpha}$ increased by each bacterial sonicate. These results suggest inhibitory action of A. asiatica against osteoclastogenesis is associated with down-regulations of RANKL, $PGE_2$ IL-$1{\beta}$, and TNF-${\alpha}$ expressions.

Hyper-CMCase-Producing Mutants of Bacillus sp. 79-23 Induced by Gamma- Radiation

  • Bacillus sp. 79-23 spores were irradiated with $^{60}Co$ gamma-rays at doses ranging from 0.5 to 5 kGy. Following gamma-irradiation, seven mutant strains were isolated by scoring the halo sizes formed around the colonies grown on LB agar plates containing 4% carboxymethylcellulose (CMC) and trypan blue. The mutant strains showed a 1.5 to 2-fold increase in carboxymethylcellulase (CMCase) activity over the parent strain. Wheat bran acted as an effective inducer for CMCase production in the parent and mutant strains. Mutant strains 68 and 70 were identified as exhibiting higher CMCase activities than those of other mutants in LB media both with and without 3% wheat bran. In addition, these strains seem to produce substantially lower amounts of capsular materials, whereas the parent strain produced large amounts of them in both liquid and solid LB media. In flask cultures, the CMCase production by mutants 68 and 70 reached maximum levels of 17.5 unit/ml and 15.7 unit/ml, respectively, in an LB medium containing 3% wheat bran.

REVIEW: Footpad dermatitis (FPD) in chickens

  • Footpad dermatitis (FPD) can be considered as a threat for poultry production due to it causes losses due to condemnation in slaughterhouse. The prevalence and the severity of FPD in broiler breeders increase over time with development of poultry industry. The condition is produced by multifactorial including include drinker design and management, diet composition, house (temperature and humidity levels), litter (type, quality, and quantity) and gut health. Bacterial infections can be associated. FPD lesions can develop in less than a week. FPD is a variable size of inflammatory lesion of the footpad in commercial poultry characterized by necrotic lesions on the plantar surface of the footpad of poultry. These lesions can be exposed after removal of fecal mass and litter stuck on it. There are considerable and numerous interactions between the environment and the genetic traits. Prevention depends mainly on improvement of litter condition. Genetic selection against footpad dermatitis must be contributed to reduce pain and suffering for particular bird experiencing contact dermatitis as well as to avoid economic losses. The objective of this review is to collect different literature written about FPD to be available to students, researchers and veterinarian in poultry practical.

Antimicrobial Properties of Turmeric (Curcuma longa L.) Rhizome-Derived ar-Turmerone and Curcumin

  • The growth responses of six bacterial strains exposed to materials extracted from turmeric (Curcuma longa) rhizomes were examined using impregnated paper disk agar diffusion. Methanol extracts of turmeric rhizomes exhibited strong inhibitory activity against Clostridium perfringens and weak inhibitory activity toward Escherichia coli at 5 mg/disk. However, in tests conducted with Bifidobacterium adolescentis, B. bifidum, B. longum, and Lactobacillus casei, the methanol extract showed no inhibitory response. The biologically active constituent isolated from the turmeric rhizomes extracts was characterized as ar-turmerone using various spectroscopic analyses including EI-MS and NMR. The responses varied according to the dosage, chemicals, and bacterial strain tested. At 2 and 1 mg/disk, ar-turmerone strongly inhibited the growth of C. perfringens and moderately inhibited the growth of E. coli without any adverse effects on the growth of four lactic acid-bacteria. Of the commercially available compounds originating from turmeric rhizomes, curcumin exhibited strong and moderate growth inhibition against C. perfringens at 2 and 1 mg/disk, respectively, and weak growth inhibition against E. coli at 1 mg/disk. However, little or no activity was observed for borneol, 1,8-cineole, and sabinene against all six bacteria strains tested. The observed inhibitory activity of the turmeric rhizome-derived curcumin and ar-turmerone against C. perfringens and E. coli demonstrate one of the important pharmacological activities of turmeric rhizomes.

진주담치(Mytilus edulis) 추출물의 항균활성 및 단백질 분해효소에 대한 안정성 탐색 (Screening of Antimicrobial Activity and Proteolytic Enzyme Stability of Extract of the Blue Mussel Mytilus edulis)

  • This study was performed to screen the antimicrobial activities and proteolytic enzyme stability of the acidified extract of the Blue mussel Mytilus edulis. The acidified extract showed potent antimicrobial activities against Gram-positive bacteria, Bacillus subtilis, and Gram-negative bacteria, Escherichia coli D31, but had no activity against Candida albicans. Treatment of extract with trypsin completely abolished all or significant antibacterial activity against the tested bacteria, but slightly decreased antimicrobial activity against B. subtilis, and treatment of extract with chymotrypsin retained almost antibacterial activity against the tested bacteria except for E. coli D31. To confirm the additional enzyme stability of the extract, antimicrobial activity of the extract was tested after treated with several enzymes. Enzymes treated extract showed potent antimicrobial activity against B. subtilis and its activity was also retained for 5 h after trypsin treatments. Non-proteinaceous materials in the acidified extract also showed strong DNA-binding ability but did not show bacterial membrane permeabilizing ability. All our results indicate that mussel extract might contain the proteinaceous or non-proteinaceous antibacterial materials target not bacterial membrane but intracellular components. These results could be used to develop mussel extract as an additive for the improvement of stability or antimicrobial activity of antibiotics against specific bacteria.

Characterization of Prophages in Leuconostoc Derived from Kimchi and Genomic Analysis of the Induced Prophage in Leuconostoc lactis

  • Leuconostoc has been used as a principal starter in natural kimchi fermentation, but limited research has been conducted on its phages. In this study, prophage distribution and characterization in kimchi-derived Leuconostoc strains were investigated, and phage induction was performed. Except for one strain, 16 Leuconostoc strains had at least one prophage region with questionable and incomplete regions, which comprised 0.5-6.0% of the bacterial genome. Based on major capsid protein analysis, ten intact prophages and an induced incomplete prophage of Leu. lactis CBA3626 belonged to the Siphoviridae family and were similar to Lc-Nu-like, sha1-like, phiMH1-like, and TPA_asm groups. Bacterial immunology genes, such as superinfection exclusion proteins and methylase, were found on several prophages. One prophage of Leu. lactis CBA3626 was induced using mitomycin C and was confirmed as belonging to the Siphoviridae family. Homology of the induced prophage with 21 reported prophages was not high (< 4%), and 47% identity was confirmed only with TPA_asm from Siphoviridae sp. isolate ct3pk4. Therefore, it is suggested that Leuconostoc from kimchi had diverse prophages with less than 6% genome proportion and some immunological genes. Interestingly, the induced prophage was very different from the reported prophages of other Leuconostoc species.

  • Rice is an important food crop for most of the population in the world and it is largely cultivated in Pakistan. It not only fulfills food demand in the country but also contributes to the wealth of Pakistan. But its production can be affected by climate change. The irregularities in the climate can cause several diseases such as brown spots, bacterial blight, tungro and leaf blasts, etc. Detection of these diseases is necessary for suitable treatment. These diseases can be effectively detected using deep learning such as Convolution Neural networks. Due to the small dataset, transfer learning models such as vgg16 model can effectively detect the diseases. In this paper, vgg16, inception and xception models are used. Vgg16, inception and xception models have achieved 99.22%, 88.48% and 93.92% validation accuracies when the epoch value is set to 10. Evaluation of models has also been done using accuracy, recall, precision, and confusion matrix.

Effect of Chaff Vinegar on the Growth of Food-Borne Pathogenic Bacteria

  • The growth inhibitiory effect of chaff vinegar against various food-borne pathogenic bacteria was evaluated. Bacterial growth was evaluated in chaff vinegar at concentrations of 15, 30, 50, 65, 80, and $100\%$ using the paper disc diffusion method and 0.5, 1.0, 1.5, 1.8, 2.0, 2.2 and $2.5\%$ in broth. In the paper disc diffusion assay, chaff vinegar showed a clear zone on both the Gram-positive bacteria; Listeria monocytogenes, Staphylococcus aureus and Gram-negative bacteria; Escherichia coli O157:H7, Salmonella Typhimurium, and Vibrio parahaemolyticus. Chaff vinegar exhibited the greatest growth inhibition for V. parahaemolyticus. The bactericidal effect of chaff vinegar on the E. coli O157:H7 was tested at concentrations ranging from 0.5 to $2.5\%$ (v/v) in the LB broth media. Chaff vinegar retarded the lag phase time of the growth curve in proportion in a concentration-dependent manner. Chaff vinegar at $2.5\%$ completely inhibited the growth of E. coli O157:H7.
