지류 및 섬유질 문화재의 미생물에 관한 연구 (紙類 및 纖維質 文化財의 微生物에 關한 硏究) ­경남지방(慶南地方)을 중심(中心)으로-

  • The investigation of air fungal population in the storages to keep papers and textiles that are designated as important folk life materials or treasures was carried out from Dec. 17 to. 23, 1980. Isolation media was used for malt extract agar with chloramphnicol to prevent bacterial contamination. Isolation and identification of air fungi from the four preserved rooms were Cladosporium cladosporioides, Alternaria chlamydospora, Aspergillus fumigatus, A. versicolor, Eurotium chevalieri, Penicillium charlesii var. rapidum, P. oxalicum. P. viridicatum, Trichoderma viride, Acremomium sp., Mucor sp. and Yeast. It was found that nine species in eight genera was isolated. Among them, underscribed species in Korea was two species ; Eurotium chevalieri and Penicillium visidicatum. The fungal population of four storages was showed to be dominant species such as Cladosporium cladosporioides and the order was Acremonium, Penicillium, Aspergillus, Trichoderma, Alternaria and Eurotium. Eurotium chevalieri was ascomycetous fungi including distinctive ascospores in cleistothecia, the filamentous fungi was directly isolated from the papers and cellulose materials showing to be fourteen species in eight genera. The most species of the fungi isolated was also Cladosporium cladosporioides and the other fungi were found as Acremonium, Penicillium, Aspergillus and Trichoderma. It was confirmed that underscribed fungi were two species ; Mucor racemosus and Penicillium spinulosvm. The effect of four antifungal agents, benzoic acid, sorbic acid, dehydroacetic acid and thymol was also examined on eight species of Aspergillus, Penicillium, Cladosporium. and Tricoderma. this results were shown that more than 0.5% concentration of thymol inhibited the grow of all fungalspecies and other three chemicals appeared various inhibition zones of fungal growth depending in their different concentrations.

Development and Evaluation of the Supplementary Foods for Korean Infants and Children (지역 식품을 이용한 이유 보충식의 개발과 이의 영양학적 검토 및 저장성에 관한 연구)

  • The purpose of this study was to develop supplementary foods for infants and young children in order to improve their nutritional status. Three formulas composed of rice, soybeans, fish, dry skim milk and sesame in varying proportions were studied. The three formulas, $RS_{1}S_{2}$, $RFS_{1}S_{2}$, and $RMS_{1}S_{2}$, were consisted of Rice(R), Soybean$(S_{1})$, Sesame$(S_{2})$ (60 : 35 : 5) , Rice, Fish(F), Soybean, Sesame (60 : 10 : 25 : 5) , and Rice, Dry Skin Milk (M), Soybean, Sesame (60 : 10 : 25 : 5), respectively. A proximate analysis and amino acid determination were made on the developed formulas. In the animal assay, growth rate, PER and FER were evaluated and biochemical analyses were also carried out. A storage test and the cost evaluation were also conducted. The summarized results are as follows : 1) The proximate composition of the three formulas were 7.3-7.4% of moisture, 15.9-21.5% of crude protein, 7.8-9.6% of crude fat and 2.5-2.8% ash. 2) The result of amino acid analysis showed that the 1st limiting amino acids of $RS_{1}S_{2}$ and $RFS_{1}S_{2}$ were lysine (amino acid score, 76.6) and threonine (amino acid score, 93.3), and that of $RMS_{1}S_{2}$ and the commercially prepared formula were sulfur containing amino acids (amino acid score, 82.0 and 54.4). When the contents of the amino acids of the three formulas were compared with mother's milk and cow's milk, the balance of the amino acid of each formula was superior to mother's milk but inferior to cow's milk. 3) In the animal assay, the growth rate of all groups increased gradually during the experimental period. 4) The C- PER, which was corrected on the basis of the casein PER of 2.5 was 2.99, 3.38 and 3.10 in the $RS_{1}S_{2}$, $RFS_{1}S_{2}$ and $RMS_{1}S_{2}$ respectively. The C- PER of $RFS_{1}S_{2}$ and $RMS_{1}S_{2}$ were Significantly (P<.05) higher than that of the casein. 5) The FER of the casein, $RS_{1}S_{2}$, $RFS_{1}S_{2}$, and $RMS_{1}S_{2}$ were 0.37, 0.39, 0.43 and 0.39, respectively. The FER of $RFS_{1}S_{2}$ and $RMS_{1}S_{2}$ were also significantly (P<.05) higher than that of the casein. 6) The concentrations of hematocrit, hemoglobin, total protein and albumin in the serum of the rats of all groups were not significantly different among groups. 7) The storage stability test showed that the total plate count (TPC), the coliforms count and the bacterial spore count in the ingredients were quiet low. However, after 30 and 60 days storage, the count in $RFS_{1}S_{2}$ increased and were higher at room temperature than refrigerated temperature. 8) In the cost evaluation, the cost of the developed formulas was \1,826-2,626 / kg. This was less than that of the commercially prepared formula (\3,300-4,073 / kg) and that of the imported formula (\4,250-8,720 / kg).

Evaluation of Bacillus subtilis Native Strains for Plant Growth Promotion and Induced Systemic Resistance in Tomato and Red-pepper (토마토, 고추의 생육촉진 및 병 저항성 의 농업적 활용을 위한 토착 Bacillus subtilis의 생물활성 평가)

  • Bacillus subtilis strains isolated from different regions of Korea were screened for their plant growth promotion and induced systemic resistance (ISR) in tomato and red-pepper. The plant growth promotion on red-pepper and tomato revealed maximum plant height (22.73 cm) on red pepper treated with B. subtilis strain JE 21-1 and 30.18cm in case of tomato treated with B. subtilis strain JE 8-1. There was also significant improvement in root and shoot dry weight in both the plants. The strain JE 21-1 showed better promise for all growth parameters in red-pepper and tomato when compared to other strains and positive check BTH. Different strains screened in square plate method also revealed maximum plant height and leaf width, and suppressed anthracnose on red pepper in case of strain JE 21-1 at $10^6$ and $10^7$ cells/ml when compared to other strains. In all the bacterial inoculations the population was significantly high when compared to untreated check. In plant growth promotion with respect to fruit length and weight, fruit length was maximal in treating with JE 9-4 and ES 2-2, while fruit weight was maximal in treating with JE 3-6, ES4-2, ES2-2 and JE 21-2 on red pepper. In case of tomato, comparatively better fruit weight was in JE 21-1, ES 3-3 and JE 10-2 when compared to BTH and untreated control. The soft rot disease caused by Pectobacterium carotovorum SCCI was completely suppressed in case of transgenic tobacco harboring GUS gene related to PR1a and increased the level of salicylic acid significantly in combined application of JE 9-4 on par with BTH. Thus, this study clarified some potential Bacillus subtilis strains for plant growth promotion and ISR in red-pepper and tomato.

Effects on Surgical Repair of VSD by TATV (막성주위형 심실중격결손중의 봉합시 경삼첨판륜 절개방법의 외과적 치료효과)

  • Perimembranous ventri ular septal defects(PMVSDS) are the most common type of ventricular septal defects(VSDs) and consist morphologically of deficiency of the membranous septum and variable portions of the adjacent muscular septum. Repair of VSD has begun via a right ventriculotomy. Even with this exposure, however, it mght lead to ventricular dysfunction. Transatrial exposure of VSDs is luiown to a versatile approach to PMVSDS and even malaligunent defects can be repaired by this method. Although transatrial exposure can be improved by taking down'the atrioventricular valve at the annulus, surgeons have been hesitant to do so because of concern for valvular competence. Therefore, this study was undertaken to clarity the effects of transamlular approach of tricuspid valve (TATV) at operation of PMVSD. During last 5 years, twenty eight cases from 96 patients of PMVSD were closed by TATV and follow up study was done from 3 months to 33 months and results were obtained as follows. 1. Age at operation was fr m 4 months to 38 years and most patients(17, 62%) were above 5 years. 2. Preoperative pulmonary-systemic flow ratio(QPIQS) was ranged from 1 to 2.8 and 22 patients(79%) were less than 2. 3. Peak systolic pulmonary artery pressure was below 30mmHg in 8, 30-50mmHg in 17, above 50mmHg in 3 patients and 25 patients(89%) were less than 50mmHg. 4. Preoperative tricuspid regurgitation(TR) is none in 12, trivial in 6, mild in 3, moderate in 5, severe in 2 patients but postoperative TR was none in 18, trivial in 6, mild in 4 patients, so TR in most patients had decreased or not. 5. Indications for operation were based on the presence of a significant shunt. However, in patients with small shunts, indications for operation were included additional factors, tricuspid valve pouch, RVOT obstruction(right ventricular outflow tract obstruction), subacute bacterial endocarditis and associated anomalies. 6. There were no hospital deaths and residual shunts in postoperative echocardiography. Therefore TATV is especially a good method in PMVSn where patients have trcuspid valve pouch. And it is a safe and effective technique that improves exposure for PMVSD repair and does not adversely affect tricuspid valvular competence.

Biological Control of the Mushroom Fly, Lycoriella mali, Using Bacillus thuringiensis subsp. israelensis (Bacillus thuringiensis subsp. israelensis를 이용한 느타리 재배에서 버섯파리의 생물학적 방제)

  • Biological control against mushroom fly, Lycoriella mali, was performed by using Bacillus thuringiensis subsp. israelensis Bti-D and Bti-U, isolated from dead mushroom fly in oyster mushroom houses. Control values of the bacterial strains Bti-D and Bti-U against L. mali in bottle culture of oyster mushroom were 74.4% and 64.2%, respectively, and the value in small tray culture were 75.8% and 56.8%, respectively. In the experiment to develop the mass, cheap media for Bti-D and Bti-U isolates, the Biji broth (bean curd residue, called Biji in Korean language) was selected as a culture medium for an inexpensive and mass cultivation by the measurement of optical density of the two bacteria grown in the different media tested. Insecticidal effect of the formulation contained different ingredients that were prepared by using the Bti-D strain cultured in the Biji broth was tested in tray and bottle culture of oyster mushroom. The WCS formulation that contained corn starch as bio-gel (86.4%) was more effective to control the mushroom fly than living cells (69.1%) in bottle culture of oyster mushroom. Moreover, insecticidal effect of the WCS formulation was improved when water of pH 8 was used for dilution of the formulation. Effect of the WCS formulation using water of pH 8 and chemicals, Zuron (dimillin) W.P. on the control of mushroom fly and the productivity of oyster mushroom was investigated in tray culture of oyster mushroom. The Zuron W.P. was more effective to control the mushroom fly than the WCS formulation. However, compared with no treatment, the productivity of the mushroom treated with the WCS formulation was improved than that of the mushroom with Zuron W.P.

Effect of Preparation method and Fermentation Conditions on Microbiological Characteristics of Sikhae (어류를 이용한 식해의 제조 방법에 따른 미생물의 특성변화)

  • This study investigated microbial populations and the sensory quality of sikhae including globefish (GLS), flounder (FLS), gastropods (GAS) and whelks (WHS) during storage at 4C for 16 d following fermentation at 10C for 4 d. General bacterial numbers increased to 102 in GLS and FLS, and to 1045 in WHS and GAS after the 20 d fermentation/storage period. Lactic acid bacteria increased to 108 log cycle in GLS and FLS after 10 d ripening time, and reached this level in GAS and WHS after 15 d and 20 d, respectively. After 20 days the number of lactic acid bacteria in each of the four samples was 108. There were 104105 yeast cells/g in each of the four samples after 20 d. The number of Leuconostoc increased to over 108 log cycle after 10 d in GLS and FLS, and 15 days in GAS for WHS the increase was to 107 log cycle. The pH values of GLS, FLS, GAS and WHS 4.42, 4.56, 4.31 and pH 4.26, respectively. The Sikhae acidity for all four samples ranged from 1.551.85%. From the sensory evaluation the overall acceptability was in the order of FLS > GLS > GAS > WHS.

Effects of Baicalin on Gene Expression Profiles during Adipogenesis of 3T3-L1 Cells (3T3-L1 세포의 지방세포형성과정에서 Baicalin에 의한 유전자 발현 프로파일 분석)

  • Baicalin, a flavonoid, was shown to have diverse effects such as anti-inflammatory, anti-cancer, anti-viral, anti-bacterial and others. Recently, we found that the baicalin inhibits adipogenesis through the modulations of anti-adipogenic and pro-adipogenic factors of the adipogenesis pathway. In the present study, we further characterized the molecular mechanism of the anti-adipogenic effect of baicalin using microarray technology. Microarray analyses were conducted to analyze the gene expression profiles during the differentiation time course (0 day, 2 day, 4 day and 7 day) in 3T3-L1 cells with or without baicalin treatment. We identified a total of 3972 genes of which expressions were changed more than 2 fold. These 3972 genes were further analyzed using hierarchical clustering analysis, resulting in 20 clusters. Four clusters among 20 showed clearly up-regulated expression patterns (cluster 8 and cluster 10) or clearly down-regulated expression patterns (cluster 12 and cluster 14) by baicalin treatment for over-all differentiation period. The cluster 8 and cluster 10 included many genes which enhance cell proliferation or inhibit adipogenesis. On the other hand, the cluster 12 and cluster 14 included many genes which are related with proliferation inhibition, cell cycle arrest, cell growth suppression or adipogenesis induction. In conclusion, these data provide detailed information on the molecular mechanism of baicalin-induced inhibition of adipogenesis.

Antibacterial Effect of Cinnamon (Cinnamomum cassia) Bark Extract against Fish Pathogenic Bacteria (계피 추출물의 어류 질병 세균에 대한 항균 효과)

  • To develop a natural antibacterial agent for fish bacterial diseases, antibacterial activity, minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and bactericidal effect of cinnamon bark extract were examined against fish pathogenic bacteria. The antimicrobial effect of the extract to the fish diet was also estimated, Cinnamon bark extract showed the broad spectrum of antibacterial activity against fish pathogenic bacteria, especially, it had strong activity against Streptococcus iniae, Edwardsiella tarda and Listonella anguillarum. Its MIC was $75.8\sim189.6{\mu}g/mL$ against Cram positive bacteria, and $75.8\sim113.8{\mu}g/mL$ against Gram negative bacteria in liquid medium, It was found to show stronger bactericidal action against Gram negative bacteria than Cram positive bacteria. According to increasing concentrations of the extract, it resulted in a proportional reduction of viable cell counts of both S. iniae and L. anguillarum. The former was not detected by addition of $189.6{\mu}g/mL$ after 12 hours incubation and the latter by addition of $151.6{\mu}g/mL$ after 24 hours incubation, respectively. It was reasonable that fish diet was soaked in cinnamon bark extract for ten minutes. The relationship formula between the weight of fish diet and the extract absorbed to fish diet was Y=7.2726X+4.5083 ($R^2=0.9998$). The fish diet soaked in the extract inhibited the growth of all strains used in this study. Its antibacterial activity was stable at the range from $10^{\circ}C\;to\;35^{\circ}C$ during the storage period of 28 days. When the diet soaked in the extract was incubated in liquid medium at $35^{\circ}C$, it inhibited the growth of microorganisms inhabited in the diet.

Serum and Cerebrospinal Fluid(CSF) Nitric Oxide, Macrophage Inflammatory Protein-1 α and Lactoferrin Levels in Aseptic Meningitis (무균성 뇌수막염 환자의 뇌척수액과 혈청에서 Nitric Oxide, Macrophage Inflammatory Protein(MIP)-1α, Lactoferrin 값의 비교)

  • Purpose : The pathologic mechanisms of central nervous system(CNS) injuries in human meningitis are not yet completely understood. Recent studies indicate that the host inflammatory responses are as important in brain damage as the infecting organisms and toxins. There have been some reports on the relationship of nitric oxide(NO), macrophage inflammatory protein-$1{\alpha}$(MIP-$1{\alpha}$), and lactoferrin in bacterial meningitis, but few reports in aseptic meningitis. Thus, we investigated the concentrations of NO, MIP-$1{\alpha}$ and lactoferrin in cerebrospinal fluid(CSF) and serum of patients with aseptic meningitis and control subjects and evaluated their relationship with other parameters of meningitis. Methods : CSF and blood were obtained from 25 subjects with aseptic meningitis and 15 control subjects. After centrifugation, supernatants were stored at $-70^{\circ}C$ and we assayed the concentrations of NO, MIP-$1{\alpha}$ and lactoferrin with the ELISA method. There were no patients with neurologic sequelae after being recovered from aseptic meningitis. Results : Concentrations of CSF and serum NO, MIP-$1{\alpha}$ were not increased in aseptic meningitis subjects compared to control subjects. Concentration of CSF lactoferrin was significantly elevated in patients with aseptic meningitis and concentration of serum lactoferrin was significantly decreased in patients with aseptic meningitis compared with those in control subjects(P<0.05). CSF lactoferrin level was positively correlated with CSF WBC counts($r_s=0.449$, P=0.007), especially with neutrophil counts($r_s=0.574$, P<0.001) and CSF protein level($r_s=0.508$, P=0.002). Conclusion : Lactoferrin plays an important role in aseptic meningitis and may be released from neutrophils recruited from blood to the CSF through breakdown of blood-brain barrier. NO and MIP-$1{\alpha}$ may not be important factors in the pathogenesis of aseptic meningitis without neurologic sequelae.

Effect of the Concentration of Humic Acid on Growth and Yield of Organically Cultivated Hot-Pepper (휴믹산 농도가 유기농 고추의 생육 및 수량에 미치는 영향)

  • The purpose of this study was to investigate the effect of humic acid on the germination, the growth and the yield of hot pepper when treated with organic hot pepper seedlings and growing season. The germination rate of 0.05% and 0.1% humic acid was higher than that of untreated, but the germination rates of 0.4% and 1.0% humic acid were 90.0% and 86.7%, respectively, compared with the control treatment (96.7%). At 30 days after transplanting, hot pepper treated with low (0.05%) or high (1.0%) concentration of humic acid decreased the growth of hot pepper seedlings, whereas 0.2% humic acid treatment significantly increased a average height (97.6 cm), leaf number (84.7) and fresh weight ($128.1g\;plant^{-1}$) of hot pepper. After 60 days of treatment with humic acid, the height of hot pepper was significantly longer in 0.2% humic acid. The mean green fruit number of 0.2%, 0.1% and 0.05% humic acid were not significantly different among the treatments, but the mean green pepper number of 0.4% and 1.0% humic acid treatments were the higher with 35.2% and 29.1%, respectively than other treatments. However, the fresh weight of green pepper was found to be $111.5g\;plant^{-1}$ more heavier than the untreated in 0.2% humic acid. The total ($5.8kg\;plant^{-1}$) and average ($1.4kg\;plant^{-1}$) fresh weight of pepper were higher than that of untreated control, except for the 1.0% humic acid treatment after 60 days of soil irrigation. The total weight of hot pepper treated with 0.2% and 0.1% humic acid treatment was $9.3kg\;plant^{-1}$ and $8.6kg\;plant^{-1}$, respectively, which were heavier than the other treatments. The effect of humic acid concentrations on soil microbial populations, pH and EC was investigated. The soil bacterial population density of 0.2% humic acid treatment was 3.5 times higher than that of untreated control soil. As the concentration of humic acid increased from 0.05% to 1.0%, pH and EC of hot pepper grown soil also increased.
