급식소에서 제공되는 비가열조리 음식의 위해요인 분석과 HACCP 적용 후 위생개선효과 (Microbiological Hazard Analysis of Non-Heating Process Menus Served at Foodservice Operations and Hygienic Improvements by Implementing HACCP)

  • The purpose of this study was to evaluate the microbiological quality of non-heat-processed foods and the effects of HACCP implementation. Here, cabbage salad and cucumber&onion salad were selected and we investigated HA(Hazard Analysis) by checking microbiological quality, time and temperature, pH, and water activity at each processing stage. Thus, the receiving of spices and dressings, washing and sterilizing, cutting, cooking, and serving stages were all considered CCPs. Before implementing HACCP, microbial analysis showed that standard plate counts and coliform counts were higher than standard levels in most of the raw ingredients of each menu, as well as during the production process. The microbiological quality of the utensils and employee's hands used during cooking indicated levels requiring direct management. Evaluations of falling bacteria-in the foodservice establishment work areas ranged from $2{\sim}12CFU/plate$. However, after HACCP implementation, microbiological levels improved to standard levels fly sanitation education. Also, the number of falling bacteria were lower than before implementing HACCP. Therefore, it is essential the foodservice operations make efforts to implement HACCP, so that microbiological hazard levels are lowered and hygienic status improved.

Changes in the Physico-chemical and Microbial Quality during the Production of Pastırma Cured with Different Levels of Sodium Nitrite

  • Pastırma, a dry-cured meat product, is produced from the whole muscle and/or muscles obtained from certain parts of beef and water buffalo carcasses. The purpose of this study was to determine the effects of different levels of sodium nitrite on changes in the physicochemical and microbial quality during the production of pastırma. The changes in residual nitrite, salt, pH, moisture, thiobarbutiric acid reactive substances (TBARS), colour (L*, a*, b*), total aerobic mesophilic bacteria (TAMB), lactic acid bacteria (LAB), Micrococcus/Staphylococcus (M/S), mould-yeast (M-Y), and Enterobacteriaceae counts of pastırma with 0, 50, 100 and 150 ppm sodium nitrite were determined during the production. The nitrite levels and the production stages had significant effects (p<0.01) on residual nitrite, TBARS, pH, salt, and colour values. The TBARS values of the pastırma with nitrite were significantly lower (p<0.05) than the control. The final TAMB, LAB, M/S, and M-Y counts of pastırma with 150 ppm nitrite were significantly (p<0.05) lower than the control. Also, the a* (relative redness) values of control pastırma were significantly lower (p<0.05) than the pastırma with nitrite. The production stages had a significant effect (p<0.01) on the moisture.

김치찌개용 양념의 저장 및 품질특성의 변화 (Changes in Physicochemical Properties and Microorganisms during the Storage of Kimchi Stew저s Sauce)

  • Physicochemical properties and microflora of kimchi stew's sauce were investigated during 6 months at 13oC, 27oC and 37oC to study the changes of its quality. The final pH values in the sauce showed a slight decrease to 4.46, 4.38 and 4.21 during storage at 13oC, 27oC and 37oC, respectively. Color(L, a, b values), salinity, moisture and Aw in the sauce were decreased during storage. However, the values of brix and browning, and the amounts of total sugar, crude protein and crude fat in the sauce were increased during storage. All the reactions occurred more rapidly in the samples stored at 37oC than those stored at 27oC and 13oC. The viable cell counts of aerobic bacteria in the sauce were changed remarkably during storage, but viable cell counts of lactic acid bacteria were decreased. Yeast, mold and E. coli were not found in the above temperatures during storage. Regression analysis between sensory scores and quality characteristics showed that the ΔE value was a major quality index for the deterioration of kimchi stew's sauce. The shelf lives of sauces at 13oC, 27oC, 37oC were estimated to be 24 months, 6 months and 6 months, respectively.

Effectiveness of the Verif $EYE^{TM}$ machine -vision technology for complying with reducing microbial indicator counts on beef carcasses

  • The slaughter process for cattle will inevitably transfer some bacteria onto the carcasses. The goal of food safety programs is to minimize and effectively remove this contamination. This study was attempted by the Verif $EYE^{TM}$ machine-vision technology that might be useful for reducing microbial indicator counts and could reduce the contamination chance of E coli O157:H7 and Salmonella spp on beef carcasses. For the evaluation of the effectiveness of the Verif $EYE^{TM}$ technology, 80 samples were examined by the inspection device over 15 days. On an examination of FDS-positive samples compared to negative controls from the same carcasses, aerobic plate counts were bigger than the negative control samples (5.26 vs 4.60 log). Enterobacteriaceae counts were greater on the positive samples than the corresponding negative control samples (2.07 vs 1.17log). There was a consistent correlation between samples detected by the Verif $EYE^{TM}$ system with detectable counts. For example, 100% of positive samples had detectable APC and 91.2% of positive samples had detectable TCC. Therefore, if areas detected as positive for contamination by the Verif $EYE^{TM}$ system were removed from the carcasses, significant sources of microbial contamination will be reduced for objective compliance with HACCP. This results suggest that the use of Verif $EYE^{TM}$ machine-vision technology might be useful for reducing microbial indicator counts (APC, TCC) and could help reduce the risk of presence of E coJi O157:H7 and Salmonella spp on Beef carcasses.

투과도 조절 폴리에틸렌 필름의 절임배추 보존기간 연장효과 (Effect of Permeability-Controlled Polyethylene Film on Extension of Shelf-life of Brined Baechu Cabbage)

  • 절임배추를 각각 다른 5종류의 필름에 sealing 한 다음 4주간 4${^{\circ}C}$로 저장하면서 품질 특성을 평가하였다. 기체투과도 조절 필름인 MF필름(US patent No. 5972815)으로 포장한 절임배추의 경우에는 미생물의 성장수가 가장 적었고 HDPE필름으로 포장한 절임배추의 경우가 미생물수가 가장 많았다. MF필름으로 포장한 절임배추의 경우 다른 필름으로 포장한 절임배추보다 pH가 천천히 감소하였고 산도의 경우에는 모든 필름에서 포장한 절임배추에서 4주 동안 점점 증가하지만 MF필름으로 포장한 절임배추의 산도가 가장 낮았다. MF필름으로 포장한 절임배추의 경우 탄력성의 감소율이 가장 낮았고 HDPE필름으로 포장한 절임배추의 경우에 탄력성의 감소율이 가장 높았다. MF필름에 절임배추를 포장한 경우 포장 필름의 투과성과 절임배추의 호흡에 의해 4${^{\circ}C}$에서 1주 후 필름 안에 $O_2$ 조성이 0.66%로 나타났는데, 이러한 필름 안의 $O_2$ 조성 결과와 미생물수, pH, 산도의 결과를 비교해보면 포장필름 안에 $O_2$ 조성이 1%에 가까울 때 절임배추의 품질특성이 가장 우수하게 유지되었다. 본 연구 결과로 절임배추의 저장성 증진과 품질특성의 유지를 위한 필름으로 MF필름이 가장 적합하였다. 그리고 LDPE필름 역시 MF필름보다는 저장성 증진과 품질특성 유지 면에서 성능은 떨어졌지만 가격 면이나 대중적인 면에서 고려해 보았을 때 LDPE필름의 사용도 가능하다고 하겠다.

영남지역 지하수에서 대장균군의 분포 및 분리한 세균의 특성 (Distribution and Characteristics of Coliform Bacteria in Groundwater of Yeungnam Province)

  • 생활용수로 사용하는 영남지역 지하수의 세균학적 수질을 평가하기 위해, 여름 123개 지점, 겨울 117개 지점에서 지하수 시료를 채수하였다. 일반세균 수의 중앙간은 여름 30 CFU/ml, 겨울 40 CFU/ml이었고, 25% 이상의 조사지점에서 일반세균 수는 먹는물 수질기준 100 CFU/ml을 초과하였다. 전체 조사지점 중 여름 46%, 겨울 30%의 지점에서는 대장균군이 검출되었으며, 이들 지점에서 대장균군 수의 중앙간은 여름 20 CFU/ml, 겨울 4 CFU/ml로 여름이 겨울보다 높았다. 분리한 대장균군 세균은 Citrobacter속, Enterobacter속, Eschericitia속, Klebsiella속, Pantoea속, Rahnella속, 그리고 Serratia속으로 동정되었으며, Enterobacter속이 48%를 차지하여 가장 많았다. 종별로는 E. cloacae, E. amnigenus, K. pneumoniae등이 빈도가 높게 분리되었으며, 반면 E. coli는 1개 조사지점에서만 분리되었다. 대장균군 수는 일반세균 수와 양의 상관관계를 보였으며, 일반세균 수는 수온과도 양의 상관관계가 있었다. 본 연구의 결과는 지하수에서 대장균군의 오염에 대한 보다 자세한 정보를 제공해 주며, 영남지역의 지하수에서 병원성 미생물의 오염 가능성이 여전히 높음을 시사한다.

Effects of Gaseous Ozone Exposure on Bacterial Counts and Oxidative Properties in Chicken and Duck Breast Meat

  • The effects of gaseous ozone exposure on the bacterial counts and oxidative properties were evaluated in duck and chicken breast fillets, which were stored under a continuous flux of gaseous ozone (10×10−6 kg O3/m3/h) at 4±1℃ for 4 d. The ozone generator was set to on for 15 min and off for 105 min, and this cyclic timer was set during storage. Ozone effectively reduced the growth of coliform, aerobic and anaerobic bacteria in both chicken and duck breast. However, lipid oxidation occurred faster in duck breast than chicken breast with higher degree of discoloration, TBARS value, and antioxidant enzyme (glutathione peroxidase and catalase) activity decline rates. It is concluded that ozone effectively controlled the growth of bacteria in both chicken and duck breast with less effects on oxidative deterioration in chicken breast.

활성슬러지공정 하수종말처리장의 단위공정별 세균 제거효율 (Bacterial Removal Efficiencies by Unit Processes in a Sewage Treatment Plant using Activated Sludge Process)

  • To figure out the removal efficiency of indicator and pathogenic bacteria by unit processes of a sewage treatment plant using activated sludge process, analyses were done for incoming sewage, influent and effluent of primary clarifier, aeration tank, secondary clarifier and final discharge conduit of the plant. A matrix of bacterial items (average of bacterial reduction [log/ml], p value of paired t-test, number of decreased cases of twenty analyses, removal percentage only for decreased cases) between incoming sewage and final effluent of the plant were heterotrophic plate counts (1.54, 0.000, 20, 95.01), total coliforms (1.38, 0.000, 19, 83.94), fecal coliforms (0.90, 0.000, 20, 94.84), fecal streptococci (0.90, 0.000, 20, 98.08), presumptive Salmonella (0.23, 0.561, 7, 99.09), and presumptive Shigella (1.02, 0.002, 15, 92.98). Total coliforms, fecal coliforms, heterotrophic plate counts, and fecal streptococci showed highest decrease through secondary clarifier about 1-log (p<0.001) between 88% and 96%, and primary clarifier represented the significant (p<0.05) decrease. However, final effluent through discharge conduit showed higher total coliforms and fecal streptococci than effluent of secondary clarifier (p<0.05). In addition, final effluent once violated the water quality standard while effluent of secondary clarifier satisfied the standard. Hence some control measures including elimination of deposits in discharge conduit or disinfection of final effluent are necessary.

A Comparative Study on the Rumen Microbial Population of Cattle and Swamp Buffalo Raised under Traditional Village Conditions in the Northeast of Thailand

  • A comparative study on rumen bacterial and protozoal population and fungal zoospores in cattle (Brahman$\times$Native) and swamp buffalo (Bubalus bubalis) was conducted. Forty animals, twenty of each, with same sex and similar age which were raised under similar condition in the Northeast of Thailand, were used. Rumen digesta were sampled immediately post slaughtering for total microscopic counts of bacteria, protozoa and fungal zoospores. It was found that total bacterial population were higher in swamp buffalo that those in cattle (1.6 vs $1.36{\times}10^{8}cells/ml$) having more population of cocci, rods and ovals. Lower rumen protozoal pupulation in swamp buffalo with lower numbers of Holotrichs and Entodiniomorphs were found as compared to those in cattle. Significant higher fungal zoospore counts were in swamp buffalo than those in cattle being 7.30 and $3.78{\times}10^6$, respectively. Study under electron microscope, revealed Anaeromyces sp. with acuminate apex were more predominant in the rumen of swamp buffalo. With these findings, cattle and swamp buffaloes showing differences in rumen bacterial, protozoal population and fungal zoospore counts, offer new additional information as why swamp buffaloes exhibit conditionally better than cattle especially during long dry season without green grass.

Effects of Xylanase on Growth and Gut Development of Broiler Chickens Given a Wheat-based Diet

  • To study the working mechanisms for non-starch polysaccharidases to improve the growth performance of broiler chickens, a 21-day feeding trial was conducted. Two dietary treatments were included: 1) wheat diet (the control); 2) wheat+xylanase diet (xylanase, Allzyme PT, Alltech, Kentucky, USA). There were 8 replicates with 8 birds each for each treatment and the experimental diets were given to birds from hatch. Feed intake and body weight were measured on days 7 and 21. At the same ages, samples were taken for the determination of selected groups of luminal and mucosa-associated bacteria, mucosal morphology, brush-border membrane (BBM) bound enzyme activity and ileal nutrient digestibility. The xylanase supplement increased (p<0.05) body weight gain (BWG) and improved feed conversion ratio (FCR) at the end of the experiment but protein and starch digestibilities were not affected (p>0.05) by xylanase. Up to day 7, xylanase increased the counts of C. perfringens in the ileum and total anaerobic bacteria (TAB) in the caeca (p<0.05, p=0.07, respectively). By day 21, the counts of ileal lactobacilli (p<0.05) and TAB (p=0.07) were lower in birds given the xylanase-supplemented diet than in those on the control diet. No significant differences were observed in the counts of mucosa-associated lactobacilli and coliforms between xylanase treatment and the control at both ages. Villus height at the jejunum was not affected (p>0.05) by the supplement but crypt depth at the same site was reduced at day 7. Also, xylanase tended to increase the concentration of BBM protein (p = 0.09) and the specific activity of sucrase (p = 0.07) at day 21.
