Effects of Aqueous Chlorine Dioxide and UV-C Irradiation on Decontamination and Growth of Microbes during Chilled Storage of Celery and Cherries (이산화염소수, Ultraviolet-C 또는 병합처리가 샐러리와 체리에서의 살균 및 냉장저장 중 미생물 성장에 미치는 효과)

  • The effects of a combined treatment of aqueous chlorine dioxide ($ClO_2$) and ultraviolet-C (UV-C) irradiation on microbial growth in celery and cherries were investigated. Celery and cherry samples were treated with 50 ppm $ClO_2$, UV-C at dose of 10 kJ/$m^2$, and a combination of $ClO_2$ and UV-C. The changes in the counts of Escherichia coli O157:H7 inoculated in the celery and cherries as well as those of total aerobic bacteria, yeast and molds in the celery and cherries were investigated after each treatment. After the combined treatment of aqueous $ClO_2$ and UV-C irradiation, the populations of E. coli O157:H7 in the inoculated celery and cherries were reduced by 2.8 and 3.0 log CFU/g, respectively, compared to those of the control. For the un-inoculated celery and cherries, the populations of total aerobic bacteria were reduced by 2.9 and 1.8 log CFU/g, respectively, compared to the control. In addition, the populations of yeast and molds were decreased by 1.8 and 1.2 log CFU/g, respectively. These results suggest that the combined treatment of 50 ppm $ClO_2$ and UV-C at a dose of 10 kJ/$m^2$ would be an effective technology for decontamination and improving the microbiological safety in celery and cherries during chilled storage.

Selection and Physico-Chemical Characteristics of Lactic Acid Bacteria which had Cholesterol Lowering Activities (콜레스테롤 저하 유산균의 분리 및 이들 균주의 이화학적 특성)

  • Four strains of lactic acid bacteria which had cholesterol lowering activities were selected from foreign fermented milk. The strains were identified as Lactobacillus(L.) rhamnosus 2084, L. casei 0781, Lactococcus (Lacto.) lactis spp. 204, and Enterococcus(E.) faecium 402. We observed that the L. rhamnosus 2084 was the most tolerant against pH 1.5, L. casei 0781 against pH 2.0, but not significantly different in the tolerance against pH 3.0. The L. rhamnosus 2084 was the most tolerant against bile acid and prominent in the degree of lowering cholesterol level. All four strains were used as starters in producing yogurt, and then investigated physico-chemical characteristics, such as pH, titratable acidity, and viable cell counts of yogurt base. L. casei 0781, L. rhamnosus 2084, Lacto. lactis 204, and E. faecium 402 were incubated for 6 hours at $40^{\circ}C$, 4 hours at $40^{\circ}C$, 6 hours at $37^{\circ}C$, and 12 hours at $37^{\circ}C$ and $40^{\circ}C$, respectively, for the optimum conditions of fermented milk.

Seasonal Monitoring of Airborne Microbial Concentrations in Kindergartens (유치원의 실내환경에서 공기중 미생물 수의 계절적 변화)

  • To assess microbiological indoor air quality in kindergartens, concentrations of viable airborne microorganisms were seasonally determined at three kindergartens in Ulsan from April, 2002 to January, 2003. Sampling was performed with an impaction-type air sampler and three different media. The numbers of bacteria grown on Staphylococcus medium were between 84 and 4,150 MPN/m3 with an average of 827 MPN/m3, and those on standard method agar ranged from 50 to 2,636 MPN/m3 with an average of 580 MPN/m3. The bacterial concentrations were highest in summer, followed by fall, spring, and winter, and were significantly correlated with indoor temperature. Among the colonies, 45.6~61.0% were observed as Gram-positive cocci and 8.5~20.6% were Gramnegative rods. Micrococcus species were the dominant organisms. The numbers of fungi ranged from 0 to 1,888 MPN/m3(661 MPN/m3 average) based on colony counts with dichloran rose bengal chloramphenicol agar. On average, the fungal concentrations were highest in summer and lowest in winter. Penicillium species and Aspergillus species were identified from the colonies. The obtained data can be utilized as a step to set a guideline for bioaerosols in indoor environment of schools.

Combination Effect of Modified Atmosphere Packaging and Electron Beam Irradiation on the Oxidative and Microbiological Stability of Ground Pork during Storage (공기 조절 포장과 전자선 조사의 병용이 분쇄돈육의 저장 중 산화와 미생물적 안정성에 미치는 영향)

  • Ground pork was packaged(purged) with modified atmosphere (n$_2$ and CO$_2$) and irradiated with the electron beam in order to find out whether modified atmosphere packaging (MAP) inhibit the microbial growth and lipid oxidation development caused by electron beam irradiation. After packaging and irradiation, ground pork was stored at 4$^{\circ}C$ for 6 days and -15$^{\circ}C$ for 3 months, and periodically the microbial counts and the thiobarbituric acid reactive substances (TBARS) for the determination of lipid oxidation were measured. The inhibition of growth of total aerobic bacteria and mesophiles was confirmed when the ground pork was irradiated with the electron beam dose of 1.5 and 3.0 kGy. The N$_2$ or CO$_2$ purging alone was also effective in reducing the development of lipid oxidation of ground pork during storage at 4 and -15$^{\circ}C$. The combination of electron beam irradiation(1.5 and 3.0 kGy) with MAP (N$_2$ or CO$_2$) was effective to inhibit the growth of total aerobic bacteria and mesophiles, and retard the lipid oxidation of ground pork during storage at 4$^{\circ}C$ for 6 days and -15$^{\circ}C$ for 3 months.

Effect of Chlorella Extract on Quality Characteristics of Yoghurt (Chlorella 추출물 첨가가 요구르트의 품질 특성에 미치는 영향)

  • Yoghurt base was prepared from skim milk added with 0.25∼2.0%(w/v) of chlorella extract powder and fermented with lactic acid bacteria(Streptococcus thermophilus : Lactobacillus casei = 1:1) at 37$^{\circ}C$ for 24 hours. Quality characteristics of the yoghurts were evaluated in terms of acid production, number of viable cell, color, texture property, and sensory property during lactic acid fermentation. The addition of 0.25% chlorella extract powder stimulated the growth of lactic acid bacteria which showed the highest number of viable cell counts(l.46${\times}$10$\^$8/ CFU/mL) after 12 hours incubation, and also enhanced the acid production which was pH 4.33 after 12 hours incubation. The L values(brightness) of the yoghurts containing chlorella extract powder were lower than control group, whereas the b values(yellowness) were higher. The hardness and gumminess of the yoghurt added with 0.25% chlorella extract powder were higher than others. Sensory scores of the yoghurt added with 0.25% chlorella extract powder were significantly higher than other groups in color, chlorella odor, sweet taste, chlorella taste, mouth feel, aftertaste and overall acceptability.

A survey for Pb, Cd and microbiological contamination from by-products of cattle in Incheon city (인천지역 소 부산물의 미생물 및 Pb, Cd 오염도 조사)

  • This study was carried out to investigate the heavy metal and microbiological hazards on by-products (liver, omasum, small intestines) of cattle. From April to October in 2011, one hundred and twenty samples were equally collected from slaughterhouse and meat by-product markets in Incheon city. The total bacteria counts and E. coli count were applied to assess the microbiological quality. Food borne bacteria including Salmonella spp., Listeria monocytogenes, Staphylococcus aureus, Clostridium perfringens and E. coli O157:H7 were also determined. The results were obtained as follows: The undesirable grade (more than $10^5\;CFU/cm^2$) was detected in the by-product from 18.3% (slaughterhouse) and 23.3% (by-product markets). The frequency of generic E. coli (more than $10^2\;CFU/cm^2$) from the slaughterhouse was 20.0%, whereas that of the meat by-product markets was 26.7%. Of the samples from slaughterhouse, 3 (5.0%), 5 (8.3%), and 12 (20.0%) samples were contaminated with Salmonella spp, S. aureus, and C. perfringens, respectively. S. aureus and C. perfringens were also detected in 6 (10.0%) and 25 (41.7%) samples in the meat by-product markets, respectively. Nine of 11 S. aureus isolates harbored toxin gene. However, the cpe gene of C. perfringens was not detected among the 37 isolates. The detection rate was higher in August than in February, April and June. The levels of Cd and Pb in all the samples tended to be low (<0.2 mg/kg). This preliminary data could be used for legislation on the regulation and control of microorganism and heavy metal in by-products of cattle.

Change of Quality Characteristics of Commercial and Prepared Kimchi Depending on Fermentation (상업용 김치와 담금 배추김치의 발효에 따른 품질특성 변화)

  • Purpose: The purpose of this study was to evaluate the folate contents and quality characteristics of commercial and fermented Kimchi and compare the correlation between folic acid and quality characteristics. Methods: The contents of total folate and quality characteristics were evaluated in 10 kinds of industrial and local Kimchi and nine kinds of other commonly consumed Kimchi. Changes in folate content and quality characteristics of Kimchi during 14 days of fermentation at $15^{\circ}C$ were compared. Results: Chungnam (L4) Kimchi had the lowest pH of 4.62, and acidity (0.57%) and salinity (3.26%) were highest compared to other areas. The content of total folate of D company (A) was the highest at $73.66{\mu}g/100g$. The content of total folate was significantly higher than those of Sesame leaf Kimchi (O9) and Young leafy radish Kimchi (O5) containing 65.77 and $62.82{\mu}g/100g$, respectively. The pH of fermented Kimchi decreased from 6.09 to 4.11 (p<0.05), and the acidity significantly increased (p<0.001) to 0.32-0.66%. Reducing sugar content decreased rapidly by 4 days and then slowly decreased (p<0.001). Total bacteria and lactic acid bacteria counts significantly increased (p<0.05) with fermentation period. Content of total folate was highest at $69.82{\mu}g/100g$ and $68.16{\mu}g/100g$ on days 0 and 2 of fermentation, after which it decreased to 77.6% at $15.61{\mu}g/100g$ on day 10 of fermentation. Conclusion: As a result, there was no definite trend regarding folate content in commercial Kimchi and other Kimchi. Young leafy radish, Chives, and Welsh onion Kimchi, which are rich in green leaves, are excellent folate source foods. The highest folate content of fermented Kimchi was identified on day 2 of fermentation. Therefore, it is recommended to use raw or immature Kimchi when using Kimchi as a folate source food.

Fermentation and Quality Evaluation of makgeolli, Korean Rice Wine Supplemented with Alcohol-tolerant Pediococcus acidilactici K3 (알코올 내성 젖산균 P. acidilactici K3와 혼합 발효한 막걸리의 품질 연구)

  • This study's purpose was to investigate the characteristics of a traditional Korean rice wine containing lactic acid bacteria (LAB), called makgeolli. The makgeolli was brewed with the alcohol-tolerant Pediococcus acidilactici strain K3, and was analyzed for LAB cell counts, alcoholic content, turbidity, pH, total acidity, amino nitrogen, total sugars, reducing sugars, solid contents, and organic acids. The physicochemical properties of the makgeolli were mostly maintained during fermentation (9 d) and storage (15 d). We also monitored the properties of LAB-supplemented commercial makgeollies, after adding P. acidilactici K3 at a concentration of $10^7CFU/ml$ makgeolli, for one month. Most of their properties, such as alcoholic content, turbidity, pH, total acidity, amino nitrogen, total sugars, reducing sugars, solid contents, and organic acids, were preserved during storage at $10^{\circ}C$, suggesting that makgeolli supplemented with live LAB can be produced. These results suggest that alcohol-tolerant P. acidilactici K3 can be used for makgeolli brewing either as a starter or as a supplement.

Determination of Microbial Community as an Indicator of Kimchi Fermentation (김치발효의 지표로서 미생물군집의 측정)

  • Attempts were made to define the characteristics of microbial community as an indicator of Kimchi fermentation. Determination of communities was carried out by simple Gram-stain, followed by direct microcopic counts. In room-temperature $(15^{\circ}C)$ fermentation, microbial succession was occurred in the order of communities of Gram-positive bacteria, yeasts and Gram-negative bacteria. It was characteristic that Gram-positive bacterial community was developed during the production of lactic acid, yeasts community was developed to cause rancidity, and Gram-negative bacterial community was relevant to maceration (or softening) as well as rancidity. The fluctuation of apparent Gram-negative reaction group might be used as a criterion of death or aging of Gram-positive bacterial populations. In low-temperature fermentation $(5^{\circ}C)$, however, it was found that yeasts and Gram-negative bacterial communities did not developed but only Gram-positive bacterial community did. It follows from these results mentioned above that maturity of Kimchi depends on the development of Cram-positive bacterial community. Thus, the size and occurrence of microbial community are avaiable for an indicator of Kimchi fermentation, and also determination of community could be a useful method to predict the maturity.

Effects of Wasabi (Wasabia japonica Matsum) on the Microbiological and Sensory Characteristics of Dongchimi (고추냉이 첨가가 동치미의 미생물학적 및 관능적 특성에 미치는 영향)

  • The Dongchimi added with Wasabi (Wasabia japonica Matsum) for the improvement of quality was scientifically explored by reviewing the optimum level and its effect on the microbiological and sensory characteristics of product fermentation. The final weight percentage of Wasabi in Dongchimi was adjusted to 0, 3, 5, 7 and 9%, per radish, respectively. Microbiological and sensory characteristics were determined during fermentation at $10^{\circ}C$ for 55 days. The viable cell counts of total and lactic acid bacteria had increased to their maximum values during fermentation until they began to drop at the later stage. Here, the 7% treatment showed a distinctive low and high numbers of total cells and lactic acid bacteria, respectively, during the later stage of fermentation. As for the sensory evaluation results of the fermented Donchimi with Wasabi, the 7% treatment was most favored for the color, the smell, the sour taste, the hot taste, the carbonated taste, the texture, and the overall acceptability of the products. The optimum levels of Wasabi in Dongchimi were estimated between 5 and 7% per added radish weight, and the 7% level was more preferred when the fermentation-retarding effect of the product was considered.
