In previously study, we evaluated seed germination for longevity derived from a cross between 'Ilmi' and 'Dharial', a weed rice collected in Bangladesh. The strong germination trait originated from 'Dharial' was incorporated into 'Ilmi', through backcross method. The germination ratio was evaluated after two years of room temperature storage conditions. A high germination ratio of 80.5% in donor plant of 'Dharial' and 77.3% in an introgression lines was observed based on the two years of storage while the recurrent japonica cultivars, 'Ilmi' was failed in germination. In this study, we investigate changes in physicochemical properties of 'Ilmi' and introgression lines (ILs) stored at room temperature. We analyzed germination rate, texture of cooked rice, toyo glossiness value, pasting properties, amylose content, protein content and ${\alpha}-amylase$ content of 'Ilmi' and 5 introgressions every 4 months on the room temperature condition. Seed germinations were decreased by storage periods. Three ILs germination rate was slowly decreased more than 'Ilmi' and 2 another ILs after 4 months. Toyo glossiness value of 'Ilmi' and three ILs were no difference, but, 2 ILs gradually decreased every 4 months at storage periods. Pasting properties were affected by storage temperatures and periods of 'Ilmi' and ILs. The increase at breakdown was observed but setback was decreased by storage periods. Amylose content and protein content were no significant difference at storage periods, respectively. The ${\alpha}-amylase$ content was gradually increased during the storage periods. The introgression line could be useful to increase longevity and maintain quality during storage of japonica rice seed.
Powdery mildew of Indian mustard (Brassica juncea), caused by Erysiphe cruciferarum, is emerging as major problem in India. All the Indian mustard cultivars presently grown in India are highly susceptible to powdery mildew and so far no resistance source has been reported. In this study, with an aim to identify resistant source, 1,020 Indian mustard accessions were evaluated against E. cruciferarum PMN isolate, at Wellington, The Nilgiris, Tamil Nadu, India under natural hot spot conditions. The study identified one accession (RDV 29) with complete resistance against E. cruciferarum PMN isolate for the first time, which was consistent in five independent evaluations. Genetic analysis of F1, F2 and backcross populations obtained from the cross RSEJ 775 (highly susceptible) × RDV 29 (highly resistant) for two season revealed that the resistance is governed by two genes with semi-dominant and gene dosage effect. Further, a new disease rating system using six scales (0, 1, 2, 3, 4, and 5) has also been proposed in this study to score powdery mildew based on progress of fungal growth in different plant parts of the F2 population. The outcome of this study viz. newly identified powdery mildew-resistant Indian mustard accession (RDV 29), information on inheritance of resistance and the newly developed disease rating scale will provide the base for development of powdery mildew-resistant cultivars of Indian mustard.
Beta-carotene producing transformants were produced in the background of 'Nagdongbyeo', a Japonica rice cultivar. Introgression of the carotenoid locus in the transformant, PAC4-2 into the elite cultivar 'Ilpumbyeo' was started. To initiate a backcrossing program, we surveyed 220 SSR markers and found that 38% of them were polymorphic between 'Ilpumbyeo' as a recurrent parent and the PAC4-2 as a recipient parent. The selection strategy comprising foreground and background selection was employed. First, foreground selection was practiced in $BC_1$, $BC_2$, and $BC_3$ generations using the transgene specific PCR-based marker in addition to visual scoring of the seed color. Marker-based background selection combined with phenotypic selection was employed from $BC_3F_2$ to $BC_3F_4$ generations. Blast search indicated that the transgene PAC4-2 was located between SSR markers, RM6 and RM482. 240 $BC_3F_3$ and 63 $BC_3F_4$ lines were evaluated for four agronomic traits including days to heading. Most of the lines were similar to Ilpumbyeo in agronomic traits evaluated. The percentage of PAC4-2 genome ranged from 4% to 21% with a mean of 12.5%, which was higher than the expected for an unselected $BC_3$ backcross population. This could be explained by the fact that two genes for beta-carotene and the stripe virus resistance were targeted in this study. We selected 10 representative $BC_3F_5$ lines from 63 $BC_3F_4$ lines based on agronomic traits and carotenoids content. The selection strategy would be appropriate for the introgression of beta-carotene gene in a breeding program.
This experiment was conducted to characterize the inheritance of petal color, flower-eye color, flower shape, leaf variegation pattern, and leaf-end shape in Cyclamen persicum. The segregation of $F_2$ and backcross populations from crosses between inbred lines was tested for Mendelian inheritance mode. From four different combinations of crosses, it was found that the petal color was controlled by a single incomplete dominant gene. The other characters, flower-eye color, flower shape, leaf variegation pattern and leaf-end shape were confirmed to be controlled by single complete dominant genes.

A traditional wheat breeding program needs more than 12-13 years to develop a new cultivar. In recent years, 'Speed breeding (SB)' system, which uses extremely extended photoperiod (22 h), enabled up to 4-6 generations of spring wheat per year. However, since almost 70% of wheat cultivars are winter type, and over 95% of total cultivation area is for winter wheat in Korea, optimized vernalization treatment was essential for improving the SB system. Several vernalization temperatures and durations were tested with various genotypes, and the 4 weeks of 8-10 ℃ vernalization treatment was the most effective to develop 4 generations per year, for both spring and winter type wheat cultivars. This 'Speed vernalization (SV)' system followed by SB, allowed developing a new F6 recombinant inbred lines (RILs) within 2 years. Among the 184 RILs, which derived from a cross between Jokoyung and Joongmo2008, two outstanding lines were selected for yield trial test, and then named Milyang52 and Milyang53. Compared to the traditional wheat breeding program, over 60% of the time was saved to develop these two lines. Marker-assisted selection and backcross were also combined with the SV system. YW3215-2B-1 (Jokoyung*3/Gamet), which has similar agronomic traits with Jokyoung and the same Glu-B1 allele with Garnet, was developed within 2.5 years. Thus, the SV system combined with molecular breeding technology would help breeders to make a new cultivar with less time and high efficiency.
We studies seed set in the interspecific F1 /backcross hybrids of Lilium species. In the interspecfic hybrid of L. longiflorum cv. Gelria with L.${\times}$ fomolongi cv. Raizan,93% fruit set was obtained by stigmatic pollination in comparison to 53% from cut-style pollination. Accordingly, the number of seed set resulting from stigmatic and cut-style pollination was 147 and 53, respectively. Pollination o( both stigmatic and cut-style pollination resulted in 47% fruit set in the hybrid of L. longiflorum cv. Lorina with L.${\times}$ fomolongi cv. Raizan. However, stigmatic pollination formed 413 seeds, whereas only 24 seeds were obtained by cut-style pollination in this cross. The hybrid of L.${\times}$ fomolongi cv. Raizan with L. longiflorum cv. Come set 40% fruit with a total of 43 seeds by stigmatic pollination. However, no fruit set was observed in cut-style pollination in this hybrid. Backcrossing the F1 hybrid by cut-style pollination of L. longiflorum cv. Lorina ${\times}$ Asiatic hybrid cv. Chicago with the latter parent led to 53% fruit set, and 109 embryos were obtained. Likewise, backcrossing following cut-style pollination of L. longiflorum cv. Lorina ${\times}$ Asiatic hybrid cv. Corsia with the latter parent formed 67% fruits and 107 embryos. However, in the remaining interspecific hybrids, cut-style pollination set no fruit.
It is reported that the absence of lipoxygenase-3 (LOX-3) may contribute to a reduction in stale flavor after the storage of rice. To improve the quality of stored rice of the Korean japonica rice cultivar, we conducted a breeding program to develop near-isogenic rice without LOX-3 in the genetic background of Saenuri, a mega variety of Korea. In the first step of the breeding program, we used a donor parent of LOX-3 null, Daw Dam, and a recurrent japonica parent, Sindongjin, to develop HR27873-AC12 by backcross (BC1), color test for introgression of lox-3, and anther culture for rapid fixation. In the second step, we used the donor parent, HR27873-AC12, and the recurrent parent, Saenuri, to develop HR28896-31-3-1-1 by backcross (BC1), marker-assisted selection (MAS) for lox-3, and phenotypic selection (PS) for agronomic traits. Finally, in the third step, we developed HR30960-186-2-1-2-1 (Jeonju624), derived from a cross between Saenuri and HR28896-31-3-1-1, by MAS for lox-3 and PS with high selection pressure for agronomic characteristics. Jeonju624 was confirmed with the introgression of lox-3 by molecular marker. Jeonju624 was a mid-late maturing rice with similar agronomic characteristics to Saenuri, lodging tolerance with short culm, erect plant architecture, and resistance to bacterial blight and rice stripe virus. The yield components of Jeonju624 were mostly similar to Saenuri, except for the 1,000-grain weight of brown rice. The appearance of the grain of Jeonju624 was better than that of Saenuri, and the characteristics of cooked rice were similar to those of Saenuri. In the genetic background analysis using 406 KASP (Kompetitive Allele-Specific PCR) markers, Jeonju624 was confirmed to be the near-isogenic line (NIL) of Saenuri with a 95.8% recovery rate. Jeonju624 is the NIL of Saenuri without LOX-3, and overcomes the linkage drag of Daw Dam with similar agronomic characteristics and genetic background to Saenuri. Jeonju624 can be utilized as a practical cultivar to improve the quality of stored rice, breeding material for the introgression of lox-3, and genetic material to elucidate the effect of introgressed genes.
Mutant mouse which show dwarfism has been developed by N-ethyl-N-nitrosourea (ENU) mutagenesis using BALB/c mice. The mutant mouse was inherited as autosomal recessive trait and named Small Body Size (SBS) mouse. The phenotype of SBS mouse was not apparent at birth, but it was possible to distinguish mutant phenotype from normal mice 1 week after birth. In this study, we examined body weight changes and bone mineral density (BMD), and we also carried out genetic linkage analysis to map the causative gene(s) of SBS mouse. Body weight changes were observed from birth to 14 weeks of age in both affected (n = 30) and normal mice (n = 24). BMD was examined in each five SBS and normal mice between 3 and 6 weeks of age, respectively. For the linkage analysis, we produced backcross progeny [(SBS${\times}$C57BL/6J) $F_1{\times}$ SBS] $N_2$ mice (n = 142), and seventy-four microsatellite markers were used for primary linkage analysis. Body weight of affected mice was consistently lower than that of the normal mice, and was 43.7% less than that of normal mice at 3 weeks of age (P < 0.001). As compared with normal mice at 3 and 6 weeks of age, BMD of the SBS mice was significantly low. The results showed 15.5% and 14.1 % lower in total body BMD, 15.3% and 8.7% lower in forearm BMD, and 29.7% and 20.1% lower in femur BMD, respectively. The causative gene was mapped on chromosome 10. The map order and the distance between markers were D10Mit248 - 2.1 cM - D10Mit51 - 4.2 cM - sbs - 0.7 cM - D10Mit283 - 1.4cM - D10Mit106 - 11.2cM - D10Mit170.

Circling mice were recorded to display profound deafness and a head-tossing and bidirectional circling behavior, showing an autosomal recessive mode of inheritance. In addition, the histological examination of inner ears revealed that the region around organ of Corti, spiral ganglion neurons and outer hair cells showed definite abnormality. On the other hand, a genetic linkage map was constructed in an intraspecific backcross between cir and C57BL/6J mice. The cir gene was mapped to a region between D9Mitl16/D9Mit15 and D9Mit38 on the mouse chromosome 9. Estimated distances between cir and D9Mitl16, and between cir and D9Mit38 are 0.70 $\pm$ 0.40 and 0.23 $\pm$ 0.23 cM, respectively. The markers in order was defined as follows: centromere-D9Mit182- D9Mit51/ D9Mit79/ D9Mit310- D9Mit212/ D9Mit184- D9Mit116/ D9Mit15- cir- D9Mit38- D9Mit20- D9Mit243- D9Mit16- D9Mit55/ D9Mit125- D9Mit281 Based on genetic mapping, we constructed for a YAC contig across cir region. They covered the entire region or cir and cir gene was located on between the lactotransferrin (ltf) and the macrotubule-associated protein (map4). It is known that sr gene is localized in 64cM of mouse chromosome 9. The two mouse were found to be allelic by complementation test. Recently the spinner mouse has been mapped to our cir region, and tmie gene were elucidated. And further study will be needed in circling mouse to prove tmie gene mutaiton.
A leading sweet pepper cultivar, Keystone Resistant Giant #3, was crossed with a line with resistance to Xanthomonas campestris pv. vesicatoria, PI271322, for incorporation of the resistance and to study the inheritance of resistance to the disease. Seedlings of parents, $F_1$, $F_2$ and backcross populations of a cross between Keystone Resistant Giant #3 and PI271322 were inoculated with race 1 and race 3 of Xanthamonas campestris pv. vesicatoria by infiltrating bacterial suspension into abaxial side of leaves. PI271322 was carrying a gene ($Bs_3$) for hypersensitive resistance to race 1 of X. c. pv. vesicatoria and also a component of non-hypersensitive resistance to race 3 inheriting in a quantitative mode. Correlation coefficient between disease indices to race 1 and race 3 of non-hypersensitive plants in $F_2$ population was highly significant, thus indicating that a component of non-hypersensitive general resistance in PI271322 acted consistently to both races. Individual plants with resistance to both race 1 and race 3 of X. c. pv. vesicatoria were selected and a breeding program for incorporation of the resistance is continued.
