簡易上水道의 水質管理實態와 給水利用住民의 滿足度 (Water Quality Management of Simple Piped Water Supply System and Its Satisfaction of the Residents)

  • For the purpose of inquiring into the actual conditions of water quality management of Simple Piped Water Supply(SPWS) and the satisfaction and attitudes of inhabitants about utilizing water-supply, this research was done through the questionnaires, the investigation of surrounding environment and water quality analysis in the Sangju city, Kyungsang-pookdo during 4 months from March to June, 1998. The fountainhead of SPES using ground water accounted for 65.3%, which was the highest rate in the group. Most of them was at least 10 years in the number of utilization year. 79.6% of them were exposed to many sorts of surrounding pollution origins. The examination of water was performed only through a test about water purification and 24.5% of them disinfection by chlorine also no residual chlorine was detected at all. All the waterworks did not have any education to the managers of water quality and 81.5% of them held physical examination. As a result of the water examination about SPWS, 65.3% of them were found incongruity and the valley and springing water accounted for higher than the underground water in the rate of incongruity. Looking into the details of the result in the water examination, a category of colon bacilli was ranked in the highest rate and the next one was general bacilli, nitric acid nitrogen and turbidity in order. In the satisfaction degree of the water quality, the satisfied accounted for 44.6%, and 29.2% each. Over the state of satisfaction about the ways of water examination, satisfaction accounted for 44.6%, and unsatisfaction 28.3%. Summarizing the result of the above-stated, the state of Water Quality Management of the SPWS was in a poor condition. Thus for the water Quality Management, systematic and scientific water Quality Management mainly by the administrative organization other than voluntary management by the village should be done beyond doubt. Additionally the opinions and demands of inhabitants utilizing the water supply have to be positively reflected in the affairs of water Quality Management so that the distrust of inhabitants to the SPWS should be settled.

일부 종합병원 내 영역별 공기 중 미생물 평가 (Assessment of airborne bioaerosols among different areas in the hospitals)

  • Three major hospitals with over 500 beds located in and near Seoul were surveyed for airbone microorganisms from February 1, 1998 to February 18, 1998. The purpose of the study was to identify and quantify microbiological organisms circulating in the air of three different areas in the hospitals. For the study, a RCS air sampler was utilized equipped with two different collection media, the agar strip GK-A for bacteria and the agar strip HS for fungi. The areas investigated were the intensive care unit (ICU) in the Department of internal medicine, the Newborns room in the Department of Obstetrics, and the microbiology laboratory. The results were as follows; 1. The average numbers of general microbiological particles collected on the agar strip GK-A media were $205CFU/m^3$, $232CFU/m^3$, and $128CFU/m^3$ in each hospitals. The highest concentration of $387CFU/m^3$ was found in the ICU of A hospital at 15:00 during the day. Further analysis of the collected bioaerosols by gram staining, revealed that there were gram positive cocci (89.5%), gram positive bacilli (7.2%), gram negative bacilli (2.8%), and fungi (0.5%), in descending order of frequency. 2. Ten different genes were identified from the agar strip GK-A. The most frequently identified organisms were: the Coagulase negative staphylococcus (55.0%), Micrococcus (21.4%), Enterococcus species(10.4%), and Bacillus species (7.2%). A series of antibiotics susceptibility test were conducted against the aforementioned four(4) organisms. Ninety percent of coagulase negative stapylococcus were sensitive to Penicillins. Pathogenic microbes isolated include: Staphylococcus aureus, Acinetobacter species, Klebsiella pneumonia, Klebsiella oxytoca, and Pseudomonas aeroginosa. 3 Although 56.8% of the microorganisms grown on the strip HS media for fungi could not be identified, some of them were successfully identified. The most frequently found fungi were Aspergillus (35.3%), Yeast or Molds (6.2%), and Penicillium (0.7%). Based on the results obtained from the study, it was concluded that some areas in the hospitals had abnormally high bioaerosol concentrations which could be attributed to human activity. Therefore, it is recommended that periodic assessments of indoor bioaerosols aiming to identify the possible sources should be conducted in order to maintain clean indoor environment in the hospitals.

빨강불가사리(Certonardoa semiregularis)에서 분리된 세균의 군집구조 분석 (Microbial Community Analysis Isolated from Red Starfish (Certonardoa semiregularis) Gut)

  • 불가사리를 이용하여 다양한 생리활성에 대한 연구가 많이 진행되었지만, 다른 천연물 연구에 비해 불가사리로 부터 분리한 미생물에 대한 연구는 부족하다. 이에 본 연구에서는 제주도에서 채집한 극피동물인 빨강불가사리(Certonardoa semiregularis)의 내장으로부터 Marine Agar와 R2A를 이용하여 총 103개의 균주를 분리하여 세균군집에 대해 조사하였다. 분리된 균주들은 16S rRNA유전자를 이용하여 염기서열을 분석 및 동정하였다. 그 결과 주요 계통군은 Proteobacteria (Alpha-proteobacteria 24%, Beta-proteobacteria 4%, Gamma-proteobacteria 65%) 93%, Bacteroidetes 4%, Actinobacteria 2%, Firmicutes 1%로 4개의 문이 확인되었고, 7개의 강(Actinobacteria, Flavobacteria, Bacilli, Sphingobacteria, Alpha-proteobacteria, Beta-proteobacteria, Gamma-proteobacteria), 15개의 목, 19개의 과, 24속이 관찰되었다. 또한 계통 분석 결과 2개의 균주(Lysobacter sp., Pedobacter sp.)가 각각 97.55%, 97.58%로 상동성이 98% 이하로 나타나 새로운 속 또는 종으로 분류될 가능성이 있다고 여겨지며, 표준 균주와 함께 신종 후보 균주에 대한 생화학적, 형태학적 등의 추가적인 신종실험을 향후 진행해야 할 것으로 사료 된다.

객담을 이용한 Mycobacteria의 검출과 중합효소 연쇄반응의 민감성 비교 (Identification of Mycobacteria Using Polymerase Chain Reaction and Sputum Sample)

  • 결핵(Mycobacterium tuberculosis, MTB) 감염은 아직까지 전 세계에서 높은 유병률과 사망의 주요 원인이 되고 있으며 비정형 결핵(nontuberculous mycobacteria, NTM)은 최근 후천성 면역결핍증(AIDS)이나, 종양, 이식 등으로 면역력이 저하된 환자들의 임상 검체에서 분리빈도가 증가함에 따라 분리 균주의 임상적 의의가 중요시 되고 있다. 이에 항산균 염색을 이용한 객담 도말검사는 결핵균과 비정형 결핵균을 구별할 수 없다는 제한점이 있고, 균 분리배양검사는 시간이 오래 걸린다는 단점이 있어, 본 연구에서는 이러한 제한점을 가진 기존의 검사법을 대신하여 분자생물학적 방법인 중합효소 연쇄반응(polymerase chain reaction, PCR)을 이용하여 균 분리배양 검사와 비교하여 보았다. Mycobacteria를 동정하는데 항산균 염색과 3% ogawa 배지를 이용하였고, 균 분리배양 후 M. tuberculosis를 확인하기 위해서 niacin test를 실시한 결과 집락의 DNA를 추출하여 PCR후 동정된 M. tuberculosis와 niacin 양성이 일치함을 확인할 수 있었다. 또, 이 실험에서 항산성균 염색이 2+ 이상인 객담검체와 집락검체 각각에서 DNA를 추출하여 결핵균을 동정하는 방법으로 M. tuberculosis complex에 특징적으로 존재하는 insertion sequence (IS) 6110의 특정부위인 547 bp와 285 bp 부분을 증폭한 two-tube nested polymerase chain reaction을 시행하였고, 비정형 결핵균 동정법으로는 mycobacteria에 공통적으로 존재하는 rpoB 유전자 중 일부인 360 bp 부분을 증폭한 후, 제한효소 Msp I을 첨가 PCR-restriction fragment length polymorphism (RFLP)을 시행하였으며, 결핵균과 비정형 결핵균 모두 동정율에 거의 차이가 없었다. 1+인 경우는 객담검체에서 PCR한 결과가 31.2%에서 집락검체의 PCR한 결과 93.7%까지 결핵균과 비정형결핵균의 동정율이 높아졌고, trace인 경우 객담검체에서 PCR 결과가 2%에서 집락검체에서 PCR한 결과가 97.9%까지 결핵균과 비정형 결핵균의 동정율을 높일 수 있었다. 이 실험에서 항산성균 염색 1+이하 일때 객담검체와 집락검체로 PCR을 실시하면 결핵균과 비정형 결핵균의 동정율에 차이가 있고, 배양만으로는 결핵균의 동정은 가능하지만 비정형 결핵균의 동정이 가능하지 않으므로 배양검사와 PCR 검사 모두를 병행하므로써 보다 신속하고 정확한 검사결과를 내는데 도움 될 것이다.

신생아 집중 치료실에 입원한 신생아에서 장내 세균에 의한 장관 집락화와 관련된 위험 인자 (Risk Factors Associated with Gastrointestinal Tract Colonization by Enterobacteriaceae in Neonatal Intensive Care Unit Patients)

  • 목적: 최근 신생아 집중 치료실(neonatal intensive care unit, NICU)에 입원한 신생아들의 병원내 감염 중 그람 음성 간균(Gram negative bacilli, GNB)에 의한 패혈증이 증가하고 있다. NICU에서 정기적인 대변의 감시 배양을 통해 GNB에 의한 장관 집락화가 같은 균에 의한 패혈증으로 이행되는 지를 확인하고, 집락화된 GNB 병원균의 종류를 확인하며 집락화에 관여하는 위험인자를 조사하고자 하였다. 방법: 2007년 10월부터 2008년 5월까지 고신대학교 복음병원 신생아 집중치료실에 입원한 86명의 환아를 대상으로 하였다. 모든 환아에게 총 3회의 대변 채집을 계획하였는데, 출생 후 첫 대변과 2주 간격으로 2회 채집을 계획하였다. 장관의 GNB 집락화와 관련된 위험 인자들은 대상 환아들의 의무기록을 열람하거나 연관된 논문의 고찰을 통해 설정하였다. 결과: 86명의 환아 중 22명(25.6%)이 GNB에 의한 장관 집락화가 있었으나 이 환아들을 퇴원까지 추적하였을 때 GNB로 인한 패혈증은 없었다. Pseudomonas aeruginosa, Enterobcter cloacae, Citrobacter freundii가 가장 많이 분리되었다. 분리된 GNB 중 약 89% (32/36)는 amikacin에 감수성이 있었다. GNB 에 의한 장관 집락화의 가능성은 집락화 당시 수유량이 적을 때 증가한 반면, 첫 수유 시기가 지연될 때 그 가능성은 감소하였다. 결론: 저자들은 NICU에 입원한 신생아에서 장관 감시배양을 통해 장관에 집락화된 GNB를 확인하였으나 연구에 포함된 환아에서 GNB에 의한 패혈증이 발생하지 않았으므로 집락화가 패혈증으로 이행되는 지를 확인할 수는 없었다. 집락화시 적은 수유량(feeding volume at colonization day)이 집락화의 위험요인이었으므로 장관 영양의 진행이 원활하지 않을 경우 장관의 GNB 집락화 가능성을 고려해야 한다.

Bacillus spp. 엽면살포에 의한 가로수 및 고추의 병 방제 (Disease Management in Road Trees and Pepper Plants by Foliar Application of Bacillus spp.)

  • 식물생장촉진세균은 식물의 생장과 수확량을 촉진하고, 식물병에 대한 유도저항성을 유도하는 것으로 보고되었다. 본 논문에서 연구의 목적은 가로수와 고추의 엽면에 엽권정착 식물생장촉진세균을 처리하여, 식물생장촉진세균의 적용 범위를 확장하였다. 수목의 엽권에서 내생포자 형성 세균 1,056개 균주를 분리하여, protease, chitinase, lipase를 포함한 효소활성과 진균병인 C. graminicola와 B. cinerea에 대한 길항작용을 측정하였다. 1차 선발된 bacilli 14개 균주를 고추의 잎에 살포하여 엽권정착능을 시험하였다. 5B6, 8D4, 8G12 단독처리와 그 혼합처리군을 고추 엽면에 살포하여 생장촉진, 수확량증진, 병방제 효과를 고추 포장에서 관찰하였다. 대량배양을 통하여 선발된 균주를 대한민국 대전광역시 유성구 일대의 가로수에 살포하였을 때, 대조군과 비교하여 엽록소함량과 잎 두께가 증가하였다. 선발된 3개 균주를 수목에 엽면살포했을 때, 벚나무 진균성갈색무늬구멍병을 저해하였고 은행나무의 낙엽생성을 촉진하였다. 종합적으로 본 연구는 엽권정착세균의 엽면살포를 통하여 가로수와 고추의 생장을 촉진시키고, 식물병을 방제하는 엽권정착세균의 적용 가능성을 제시한다.

Bioluminescence 반응에 의한 ATP측정을 이용한 젖소 유방염 치료에 관한 연구 (Treatment test for bovine mastitis by the determination of ATP based on firefly bioluminescence)

  • This study was carried out to treatment test for bovine mastitis by the determination of adenosine triphosphate (ATP) based on firefly bioluminescence. The results obtained are followed; 1. In the susceptibility test, cephalothin which looks the most effective were sensitive to Staphylococcus sp. (72.3%), Micrococcus sp. (84.2%), Streptococcus sp. (72.7%) and Gram positive bacilli (72.7%), Gram negative bacilli were sensitive to gentamicin (92.3%) and Yeast-like-fungi was the most sensitive to clotrimazole, and nystatin in order. 2. When the number of bacteria, such as Staphylococcus aureus, Candida tropicalis isolated from the mastitis milk were counted by conventional agar plating technique, and compared with the concentration of bacterial ATP, it gave a good linear relationship. The content of ATP per Staphylococcus aureus, cell was 3.1fM and Candida tropicalis showed the high level of A TP (90fM). 3. The ATP assay was applied to the determination of minimal inhibitory concentration (MIC) of various antibiotics. When Staphylococcus aureus was incubated in the presence of different concentration of tetracycline, erythromycin, kanamycin and streptomycin sulfate and the growth was monitored by the conventional agar plating technique and ATP assay, both methods shown the same results that they were 1mcg/ml, 2mcg/ml, 6.25mcg/ml and 8mcg/ml, respectively. 4. For the determination of susceptibility of sensitive and resistant Staphylococcus au reus isolated for the milk with mastitis to tetracycline, erythromycin kanamycin and streptomycin sulfate, the minimum time required for the test was determined by the assay of ATP every 30 minutes during incubation of 3 hours at $37^{\circ}C$. ATP concentration time curve calculated on both resistant and sensitive strains incubated 3 hours as the optimum time for the determination of susceptibilities of various antibiotics exemed. The ATP concentration of each test broth (antibiotic containing), expressed as a percentage of its own control brith (antibioticfree) indicated values of 30% to be indicative of each antibiotic sensitivity. Single time point ATP assay carried out on the various sensitive and resistant of Staphylococcus aureus to antibiotics examined after 3 hours at $37^{\circ}C$ correlated exactly with disc diffusion and MIC. 5. In the cure of intramammary treatment of bovine mastitis in lactating quarters, the cure rate of Staphylococcal mastitis showed to cephalexin (80%), cloxacillin and gentamicin (70%), ampicillin and oxytetracycline (60%), and Streptococcal mastitis showed to cephalexin (85%), penicillin (80%), cloxacillin and oxytetracycline (75%), and ampicillin (70%), but intramammary antimycotic drug (clotrimazol) were only a little effect about fungal mastitis.

Diversity of Root-Associated Paenibacillus spp. in Winter Crops from the Southern Part of Korea

  • The genus Paenibacillus is a new group of bacilli separated from the genus Bacillus, and most of species have been isolated from soil. In the present study, we collected 450 spore-forming bacilli from the roots of winter crops, such as barley, wheat, onion, green onion, and Chinese cabbage, which were cultivated in the southern part of Korea. Among these 450 isolates, 104 Paenibacillus-like isolates were selected, based on their colony shape, odor, color, and endospore morphology, and 41 isolates were then finally identified as Paenibacillus spp. by 16S rDNA sequencing. Among the 41 Paenibacillus isolates, 23 were classified as P. polymyxa, a type species of the genus Paenibacillus, based on comparison of the 16S rDNA sequences with those of 32 type strains of the genus Paenibacillus from the GenBank database. Thirty-five isolates among the 41 Paenibacillus isolates exhibited antagonistic activity towards plant fungal and bacterial pathogens, whereas 24 isolates had a significant growth-enhancing effect on cucumber seedlings, when applied to the seeds. An assessment of the root-colonization capacity under gnotobiotic conditions revealed that all 41 isolates were able to colonize cucumber roots without any significant difference. Twenty-one of the Paenibacillus isolates were shown to contain the nifH gene, which is an indicator of $N_{2}$ fixation. However, the other 20 isolates, including the reference strain E681, did not incorporate the nifH gene. To investigate the diversity of the isolates, a BOX-PCR was performed, and the resulting electrophoresis patterns allowed the 41 Paenibacillus isolates to be divided into three groups (Groups A, B, and C). One group included Paenibacillus strains isolated mainly from barley or wheat, whereas the other two groups contained strains isolated from diverse plant samples. Accordingly, the present results showed that the Paenibacillus isolates collected from the rhizosphere of winter crops were diverse in their biological and genetic characteristics, and they are good candidates for further application studies.

Sodium polyanethol sulfonate의 혈액배양에서의 Salmonella typhi 분리에 대한 영향 (Effect of sodium polyanethol sulfonate on the isolation of Salmonella typhi from blood culture)

  • Blood is one of the most important clinical specimens for the isolation of bacteria. A rapid isolation and a high isolation rate of bacteria are very important in blood culture because bacteremic patients are mostly in grave condition. Various blood culture media which support growth of most fastidious bacteria are available commercially. However, growth of bacteria are frequently delayed because of antibacterial activity of blood. Sodium polyanethol sulfonate(Liquoid) has been reported to inactivate the antibacterial substance and disrupt phagocytic cells. The beneficial effect of SPS is well recognized in the isolation of gram-positive bacteria. However, the effect does not seem to be prominent for gram-negative bacilli isolation mainly due to the rapidity of their growth. It has been experienced with Sal. typhi that the growth is much slower than that of other gram-negative bacilli. For the rapid growth of the organism, use of bile broth has been recommended. Although Sal. typhi is the most frequently isolated organism at present, about one half of total isolates are other organisms and, in case bile broth is used, other media which support growth of these organisms should be used together. Fluid thioglycollate medium(FTM) which is always used in blood culture to isolate anaerobes is inferior to brain heart infusion(BHI) for the isolation of aerobes. This study was done to determine the effect of SPS on the isolation of Sal. typhi from blood. During the Sep. 1973 to Sep. 1974 study period, 2460 blood cultures were made from the Severance hospital patients: BHI and FTM sets 1431 specimens, BHI with SPS(0.05%) and FTM sets 396 specimens, BHI and FTM with SPS sets 359 specimens, BHI and BHI with SPS sets 274 specimens. Mean incubation time required for the macroscopic detection of growth of Sal. typhi were 3.5 days on BHI and 2.7 days on BHI with SPS. The 0.8 day difference was statistically significant. On FTM the mean incubation time was 3.8 days while it was 2.9 days on FTM with SPS. The 0.9 day difference was statistically significant. The result on BHI with and without SPS sets showed faster growth on BRI with SPS in 7 specimens and slower growth in one specimen and the remaining 12 showed growth at the same time. These specimens had mean incubation time of 3.2 days on BHI and 2.3 days on BHI with SPS. The 0.9 day difference was statistically significant. This study indicates beneficial effect of SPS for the rapid isolation of Sal. typhi from clinical blood specimens.

싱싱회류 생산업체의 HACCP 시스템 구축 전 후의 미생물학적 평가 (Microbiological Evaluation of Chilled Freshes Raw-fish Manufacturers before and after HACCP System Establishment)

  • Raw-fish food contains a lot of moisture and is a high-protein food. It is a first-stage processed food taking a lot of manual work. Therefore, it is classified as a PHF food, very liable to cause a bacterial food-poisoning. But its manufacturers are usually small-sized and a systematic sanitation management is difficult to expect. But the manufacturer participating in this study produces chilled fresh raw-fish food. Fish are sliced into two fillets, which are packaged under vacuum, kept and distributed in refrigerators, and sold within a day. It is a newly-developed kind of raw-fish food, and a more improved kind of raw-fish food making possible a systematic sanitation management. The HACCP (Hazard Analysis and Critical Control Point) is a systematic and continuous process-control method which is very efficient for controling food sanitation and reducing the expenses. A new HACCP model has been developed to be applied to a large-sized chilled fresh raw-fish food manufacturer. To ascertain its efficiency, the baterial examination was done to its workplace and products. The significance test was done on its data by "SPSS 12.0 for Window" and "Mann-Whitney U Test". The numbers of bacteria on its final products were significantly different in flatfish and porgy. The number of bacteria tended to decrease in each time-differential sampling (P<.00l). The final food products showed no food-poisoning bacteria in all the time-differential tests and in all the samplings, which proves that the CCP of the HACCP system is under control. After the SSOP program was applied, no pathogenic bacteria were found in the work-place, and the kinds and numbers of bacteria decreased. The numbers of general bacteria and colon bacilli also showed a significant difference from those before the SSOP program in the filleting board (P<.05), in the skinning board (P<.0l), in the neck-removing knife (P<.05), and in the filleting knife (P<.01). The working equipments, periodically disinfected, also showed a significant difference in sanitary conditions (in the dehydrator, P<.05). The number of bacteria found on the food-touching surface was within the standard (below 500/l00 cm$^2$) After the SSOP program was applied, the general bacteria and colon bacilli were not found. The quality of water used in the food processing was also within the standard. The numbers of bacteria falling from the air in the work-place were negligible in all the samplings (<30CFU/l000ι). The staphylococci and fungi were not found.
