Enzyme의 처리에 의한 Cheese Base의 숙성중 Casein의 변화 (The Changes of Casein of Cheese Base Treated with Enzyme during Ripening)

  • 본 연구는 UF에 의한 cheese base제조과정 중 calf rennet 과 대용효소인 porcine pepsin을 각각 단독, 또는 1:1로 혼합첨가하여 cheese base를 제조한 후, HPLC와 전기영동을 이 용하여 casein의 변화를 관찰하기 위여 실시하였으며 얻어진 결과는 다음과 같다. HPLC를 이용, Superose 12 column에 의하여cheese casein을 gel filtration한 결과, Cheddar cheese는 제조직후 5개의 분획이었던 것이 6개월 숙성후 8개의 분획으로 분별되었으며, 효소처리하지 않은 대조구 cheese base는 8주 숙성중 4개의 분획으로 분별되어 분획수의 큰 변동은 없었고, calf rennet 단독처리구는 4개에서 5개로, 1:1혼합효소처리구(calf rennet : porcine pepsin)는 4개에서 7개로, porcine pepsin 단독처리구는 4개에서 8개로 각각 분별되어 porcine pepsin이 혼합됨에 따라 casein의 분해 정도가 높았다. 효소처리하여 8주간 숙성시킨 cheese base의 casein을 전기영동한 결과, ${\alpha}$$_{s1}$-casein의 경우, calf rennet 단독처리구는 3개의 band로 분리되었고, 1:1혼합효소처리구(calf rennet :porcine pepsin)와 Porcine pepsin 단독처리구는 5개의 band로 분리되었으며, 대조구는 1개의 major band와 2개의 minor band를 나타내었다. ${\beta}$-casein은 porcine pepsin의 첨가비율이 증가할수록 그 농도는 감소하였고, ${\gamma}_1$ ${\gamma}_2$-casein으로 추정되는 band는 상대적으로 증가함을 나타내었다. 또한 para-${\kappa}$-casein은 대조구를 제외하고 calf rennet, porcine pepsin단독처리구, 1:1혼합효소처리구에서 분리되었다.

정제봉독의 MAC-T 세포에서 유단백 합성 촉진효과 (Stimulation of the milk protein production in MAC-T cells by purified bee venom)

  • Purified bee venom was collected from colonies of honeybees (Apis mellifera L.) using a bee venom collector under sterile conditions and then purified under strict laboratory conditions. Purified bee venom contained $63.9{\pm}5.4%$ melittin, $10.9{\pm}1.6%$ phospholipase A2, and $2.3{\pm}0.3%$ apamin. Purified bee venom has various anti-bacterial, anti-inflammatory and immunostimulating effects. In this study, we evaluated purified bee venom which are mammary gland cells, MAC-T cells are used to increase the synthesis of milk protein. Purified bee venom promoted the proliferation of MAC-T cells at concentrations below $1{\mu}g/mL$, but cytotoxicity at $10{\mu}g/mL$ and above. As a result of the increase in the synthesis of ${\beta}-casein$, a milk protein after treatment with MAC-T cells at a concentration of the bee venom without cytotoxicity, the ${\beta}-casein$ content in the cell culture was increased when treated at a concentration of 1 ng/mL or more. In addition, it was confirmed that purified bee venom significantly increased the expression of bovine ${\beta}-casein$ (bCSNB) mRNA, a ${\beta}-casein$ synthesis gene, at a concentration of 1 ng/mL or more. These results suggest that purified bee venom can be used to increase the production of livestock by ultimately increasing the expression of milk protein.

Holstein 종(種) 유우(乳牛)의 유단백질(乳蛋白質)의 유전적다형(遺傳的多型)과 유조성분간(乳組成分間)의 연관성 (Association between Genetic Polymorphisms of Milk Proteins and Milk Compositions in Holstein Cows)

  • 본(本) 연구(硏究)는 유단백질(乳蛋白質)의 유전적표식(遺傳的標識)(genetic marker)를 유우(乳牛)의 유전적(遺傳的) 개량(改良)을 위한 유우등록 및 선발(選拔) 자료로 활용하고자 1992년 국립 종축원에서 사육증인 Holstein종 159두에서 각각 유즙 시료를 채취하여 Polyacrylarnide gel(PAGE)에 의한 전기영동방법에 의하여 ${\alpha}S1$-casein(${\alpha}S1$-CN) 및 ${\beta}$-casein(${\beta}$-CN), ${\kappa}$-casein(${\kappa}$-CN) 및 ${\beta}$-lactoglobulin(${\beta}$-LG)의 유전적(遺傳的) 다형(多型)을 분석(分析)하여 유전적(遺傳的) 변이체의 표현형(表現型), 유전자형(遺傳子型) 및 유전자빈도(遺傳子頻度)를 추정하고 이들 단백질의 유전적(遺傳的) 다형(genetic polyrnorphisms)과 유조성분과의 연관성을 분석(分析) 검토(檢討)하였다. 유단백질(乳蛋白質)인 ${\alpha}S1$-CN, ${\beta}$-CN, ${\kappa}$-CN 및 ${\beta}$-LG 유전좌위(遺傳座位)의 유전적(遺傳的) 변이체의 표현형(表現型) 분포의 관찰치와 기대치간에 유의적(有意的)인 차이가 인정 되지않아 이들 유전자(遺傳子)들이 유전적(遺傳的) 평형을 이루고 있는 것으로 사료되었다. 유단백질(乳蛋白質) 좌위(座位)의 유전자형(遺傳子型)을 분석(分析)한 결과(結果) ${\alpha}S1$-CN BB유전자형(遺傳子型), ${\beta}$-CN AA유전자형(遺傳子型), ${\kappa}$-CN AA유전자형(遺傳子型) 및 ${\beta}$-LG AB 유전자형(遺傳子型)의 출현빈도(出現頻度)가 각각 79.87%, 84.28%, 71.70% 및 49.10%로 높게 나타났다. 유단백질(乳蛋白質)의 유전자빈도(遺傳子頻度)에 있어서는 ${\alpha}S1-CN^B$${\alpha}S1-CN^C$가 각각 0.899 및 0.101이었고, ${\beta}-CN^A$${\beta}-CN^B$은 각각 0.921 및 0.079이었으며, ${\kappa}-CN^A$${\kappa}-CN^B$은 각각 0.837 및 0.163이었고, ${\beta}-LG^A$${\beta}-LG^B$은 각각 0.378 및 0.622이었다. 분산분석(分散分析) 결과(結果) 유단백질(乳蛋白質)의 유전자형(遺傳子型)들은 유조성분인 유지율, 단백질 함량 및 총 고형분 함량에서 유의적(有意的)인 연관성이 인정되었다. 유조성분 함량에 있어서는 ${\kappa}$-CN좌위(座位)의 BB유전자형(遺傳子型)은 유지율 및 유단백질(乳蛋白質) 함량에서 ${\kappa}$-CN좌위(座位)의 AA 및 AB 유전자형(遺傳子型)보다 유의적(有意的)으로 높게 나타났으며, ${\beta}$-LG좌위(座位)의 AA유전자형(遺傳子型)은 유지율에서 ${\beta}$-LG좌위(座位)의 AB 및 BB 유전자형(遺傳子型)보다 유의적(有意的)으로 높게 추정되었다. 이상의 연구결과(硏究結果)를 종합하여 볼때 유조 성분인 유지율 및 단백질함량을 높히기 위하여서는 ${\kappa}$-CN좌위(座位)의 BB 유전자형(遺傳子型)과 ${\beta}$-LG좌위(座位)의 AA 유전자형(遺傳子型)을 선발(選拔)하는것이 보다 유리할 것으로 생각되었다.

Comparison of Alpha-Factor Preprosequence and a Classical Mammalian Signal Peptide for Secretion of Recombinant Xylanase xynB from Yeast Pichia pastoris

  • The secretory efficiency of recombinant xylanase xynB from yeast Pichia pastoris between the ${\alpha}$-factor preprosequence and a classical mammalian signal peptide derived from bovine ${\beta}$-casein was compared. The results showed that although the bovine ${\beta}$-casein signal peptide could direct high-level secretion of recombinant xylanase, it was relatively less efficient than the ${\alpha}$-factor preprosequence. In contrast, the bovine ${\beta}$-casein signal peptide caused remarkably more recombinant xylanase trapped intracellularly. Real-time RT-PCR analysis indicated that the difference in the secretory level between the two signal sequences was not due to the difference in the transcriptional efficiency.

Principal Milk Components in Buffalo, Holstein Cross, Indigenous Cattle and Red Chittagong Cattle from Bangladesh

  • The aim of the present study was to get a total physical and chemical characterization and comparison of the principal components in Bangladeshi buffalo (B), Holstein cross (HX), Indigenous cattle (IC) and Red Chittagong Cattle (RCC) milk. Protein and casein (CN) composition and type, casein micellar size (CMS), naturally occurring peptides, free amino acids, fat, milk fat globule size (MFGS), fatty acid composition, carbohydrates, total and individual minerals were analyzed. These components are related to technological and nutritional properties of milk. Consequently, they are important for the dairy industry and in the animal feeding and breeding strategies. Considerable variation in most of the principal components of milk were observed among the animals. The milk of RCC and IC contained higher protein, CN, ${\beta}$-CN, whey protein, lactose, total mineral and P. They were more or less similar in most of the all other components. The B milk was found higher in CN number, in the content of ${\alpha}_{s2}-$, ${\kappa}$-CN and ${\beta}$-lactalbumin, free amino acids, unsaturated fatty acids, Ca and Ca:P. The B milk was also lower in ${\beta}$-lactoglobulin content and had the largest CMS and MFGS. Proportion of CN to whey protein was lower in HX milk and this milk was found higher in ${\beta}$-lactoglobulin and naturally occuring peptides. Considering the results obtained including the ratio of ${\alpha}_{s1}-$, ${\alpha}_{s2}-$, ${\beta}$- and ${\kappa}$-CN, B and RCC milk showed best data both from nutritional and technological aspects.

DNA Polymorphisms of κ-Casein, β-Lactoglobulin, Growth Hormone and Prolactin Genes in Korean Cattle

  • The gene and genotypic frequencies of ${\kappa}$-casein (${\kappa}$-CN), ${\beta}$-lactoglobulin (${\beta}$-LG), growth hormone (bGH) and prolactin (bPRL) loci in Korean cattle were investigated using PCR-RFLP analyses. Genomic DNA samples were obtained from 290 cows and 30 AI bulls. In both cows and bulls, the most predominant genotypes of ${\kappa}$-CN, ${\beta}$-LG, bGH and bPRL loci were AB, BB, AA and AA, respecitively. The frequencies of A and B alleles for ${\kappa}$-CN locus were .612 and .388 for cows and .567 and .433 for bulls. The respective frequencies of A and B alleles for ${\beta}$-LG locus were .153 and .847 in cows and .217 and .783 in bulls. The frequencies of A and B alleles for bGH locus were .769 and .231 in cows and .784 and .216 in bulls, respectively. The frequencies of A and B alleles for bPRL locus were .678 and .322 for cows and .767 and .233 for bulls. Differences in frequencies of these alleles were not significant between cows and bulls at all loci examined. If the DNA polymorphisms of these candidate genes are associated with economically important traits, they could serve as genetic markers for genetic improvement in future marker-assisted selection programs in Korean cattle.

Camembert Cheese 숙성 중 Casein의 변화에 관한 연구 (Changes in Caseins during the Ripening of Camembert Cheese)

  • P. caseicolum V.B.와 복합유산균을 사용하여 제조한 Camembert cheese를 45일간 숙성시키면서 카제인 변화와 그에 따른 기호성을 조사하였다. 숙성과정 중 수용성 질소, 비카제인태 질소, 비단백태 질소량은 증가하였다. pH 4.6-insoluble casein의 전기영동 결과 숙성 10일에는 4개의 band에서 45일에 12개의 band로 분리되었는데 ${\alpha}_{s1}-casein$은 숙성 17일에 완전히 분해되었으며, ${\beta}-casein$도 숙성 45일 동안 거의 대부분이 분해되었다. 또한 pH 4.6-soluble casein fragments의 gel filtration에서 숙성 10일에는 3개의 fraction, 24일까지는 4개의 fraction이 나타났고 31일부터는 5개의 fraction으로 분별되었다. 관능검사 결과 숙성 31일된 Camembert cheese의 기호성이 제일 높았으며, 숙성이 진행됨에 따라 hardness는 감소하였고 관능적 기호성과는 0.1%의 유의수준으로 높은 상관관계를 나타냈다.

SEQUENCE ANALYSIS AND COMPARISON OF BOVINE αS1-CASEIN GENOMIC DNA

  • A phage clone containing the partial ${\alpha}_{S1}$-casein gene was isolated from a bovine genomic library by using mixed probes of ovine ${\alpha}_{S1}$-, ${\beta}$- and ${\kappa}$-casein cDNAs. Restriction enzyme mapping analysis for 14.6 kb revealed that the map was in conflict with the report of Meade et al. (1990), especially in the 3'-end fragment. Sequence analysis of 12.6 kb revealed a high AT/GC ratio (1.64); we have identified eight exon sequences according to the bovine ${\alpha}_{S1}$-casein cDNA sequence. The same exon/intron splice junction sequence was observed between these exons. We suggest that the bovine ${\alpha}_{S1}$-casein gene night contain a minimum of 18 exons and the full length is approximately 18-19 kb.

The Relation between Genetic Polymorphism Markers and Milk Yield in Brown Swiss Cattle Imported to Slovakia

  • The aim of this study was to determine genotypes of four genetic markers and to investigate their association with milk production traits in Brown Swiss cattle imported to Slovakia. The bovine $\kappa$-casein, $\beta$-lactoglobulin, growth hormone and prolactin genotypes of 107 cows were identified by polymerase chain reaction. Effects all four genetic markers on milk, fat, protein and lactose yields and fat, protein and lactose percentage were estimated from a data set of 249 lactations. The frequency of desirable B allele of $\kappa$-casein gene to milk production was 0.46, alleles A of $\beta$-lactoglobulin gene was 0.55, allele and L of growth hormone gene was 0.45 and allele A and B of bovine prolactin gene were 0.61 and 0.39. The results of milk production obtained in our work showed that BB genotypes of $\kappa$-CN gene, AA genotypes of $\beta$-LG gene, LL genotypes of bGH gene were significantly associated with better milk production traits, mainly about the fat content. Association of a bovine prolactin genotypes with milk production were not found.

상업적 응유효소의 탈지유에 대한 단백질 분해 작용 (Comparative Study of Proteolytic Activities of Some Commercial Milk Clotting Enzymes on Bovine Skim Milk)

  • 상업적 단백질 분해 효소에 0.02% $CaCl_2$를 첨가하여 응유 활성화를 시킨 탈지유에 대한 분해 작용의 결과를 요약하면 다음과 같다. 다양한 효소별 가수분해 시간에 따른 가수분해도는 미생물 유래 효소와 trypsin은 pepsin과 papain W-40보다 높은 분해도를 나타냈다. 12% TCA 용액에 가용성인 NPN의 양은 trypsin이 가장 높은 분해도를 나타내었고 rennet과 pepsin이 가장 낮은 분해도를 보였다. 전기영동에 있어서 trypsin과 protease S는 $\alpha$- lactalbumin을 분해하였고 papain w-40은 $\beta$- lactoglobulin을 미약하게 분해하였으며 neutrase 1.5는 90분 이후부터 $\alpha$-lactalbumin과 $\beta$-lactoglobulin을 분해하였다. Rennet과 비교한 전기영동상에서는 rennet에 의해 분해 되지 않은 ${\alpha}_s$- casein과 $\beta$-casein을 trypsin과 protease S가 다량 분해하였고 $\kappa$-casein은 rennet에 비해 papain W-40이 상당 수준의 분해상을 나타내었다. 이상의 결과 가수분해도 및 NPN 양은 trypsin, neutrase 1.5 및 protease S가 다른 효소에 비해 높게 나타났으며, 전기영동상에서는 pepsin과 neutrase 1.5가 rennet과 유사한 경향을 나타내었다.
