Identification of bird species and their prey using DNA barcode on feces from Korean traditional village groves and forests (maeulsoop)

  • A DNA barcode based on 648 bp of cytochrome c oxidase I (COI) gene aims to build species-specific libraries for animal groups. However, it is hard to recover full-length (648 bp) barcode gene from environmental fecal samples due to DNA degradation. In this study, we designed a new primer set (K_Bird), which amplifies a 226 bp fragment targeted an inner position of full-length COI barcode based on 102 species of Korean birds to improve amplification success, and we attempted to identify bird species from 39 avian fecal samples collected during 4 months from Jinan, South Korea. Simultaneously, we conducted a dietary analysis using a universal DNA mini-barcode (Uni_Minibar) from same fecal samples. In silico analysis on newly designed mini-barcode represented that genetic distances were 0.5% in species and 9.1% in genera. Intraspecific variations of 149 species out of 174 species (86%) between Korea and North America were within the threshold (5.3% threshold in this study). From environmental fecal samples collected in Jinan, we identified seven avian species, which have high similarity (99-100%) with registered COI sequences in GenBank. Eight kinds of prey species, such as moth, spider, fly, and dragonfly, were identified in dietary analysis. We suppose that our strategy applying mini-barcode for environmental fecal samples, might be a useful and convenient tool for species identification and dietary analysis for birds.

Infection by a Filarial Nematode from the Family Onchocercidae in the Wild Bird Anas falcata

  • A filarial nematode was found in a blood sample of an Anas falcata individual collected in South Korea in 2018. Phylogenetic analysis based on partial cytochrome C oxidase subunit I (COI) sequences placed the nematode as a novel genus of the family Onchocercidae and as closely related to Mansonella species, Chandlerella quiscali, and filarial nematodes recently reported in avian species. However, different phylogenetic relationship was observed in the NADH dehydrogenase subunit 5 and 12S rRNA-based phylogenetic trees, which might indicate the filarial nematode found in this study was not defined to belong to the known specific genera of the family Onchocercidae. The screening of 105 additional avian blood samples retrieved only one 12S rRNA-targeting polymerase chain reaction (PCR)-positive sample, which indicates that filarial nematode infection is rare in wild birds or that it occurs below the detection limit of PCR in blood samples. Nevertheless, considering the recent findings about ancient interactions between birds and human pathogenic filarial nematodes and their pathogenic potential in several avian species, additional exploration of novel filarial nematodes in wild birds remains necessary.

시설원예단지와 논습지의 조류서식 및 다양성 비교분석 연구 (The Comparative Studies on the Avian Diversity in Protected Horticulture Complex and Paddy Wetland)

  • We examined the impact of paddy wetland's avian-diversity on the development of the protected horticulture complex. The results of this study were made to propose ecological protected horticulture complex and development direction suitable for avian habitat. The total number of species surveyed was analyzed as 1168 individuals in 11 orders 26 families and 36 generic 48 species. Type average was analyzed as paddy $17.25{\pm}4.83$ species and $74.50{\pm}38.08$ individuals, glass $10.00{\pm}0.82$ species and $46.75{\pm}2.06$ individuals, single $13.75{\pm}7.27$ species and $59.50{\pm}35.34$ individuals, multi $8.75{\pm}0.96$ species and $36.75{\pm}9.29$ individuals. Paddy showed higher species diversity than protected horticulture complex. The average number of species surveyed for one year was glass $14{\pm}2.83$, multi $12{\pm}1.41$, single $18{\pm}11.31$, and paddy $26{\pm}11.31$. The diversity difference was identified as paddy> single> glass and multi. Statistical analysis showed statistical difference of Ciconiiformes. Development of protected horticulture complex causes loss of avian habitat. The diversity index was glass $1.73{\pm}0.07$, multi $1.68{\pm}0.14$, single $1.91{\pm}0.47$, and paddy $2.29{\pm}0.17$. Paddy has a higher species diversity than the protected horticulture complex. For the purpose of ecological protected horticulture complex, detention ponds, artificial wetlands and habitats should be reflected in the design. This should be applied to reclamation areas or large-scale protected horticulture complexes.

Avian Fauna of Con Dao and Xuan Thuy Ramsar Sites in Vietnam

  • Xuan Thuy and Con Dao are natural Ramsar sites in Vietnam with different ecosystems, geography and topography. Avian fauna in these two Ramsar sites were recorded over a 2-year period from 2018 to 2019. A total of 234 bird species belonging to 57 families and 16 orders were confirmed in Xuan Thuy, while only 71 species involving 32 families and 12 orders were found in Con Dao. In total, 25 endangered species have been found in these two national parks based on the IUCN Red List. The Spoon-billed Sandpiper and Baer's Pochard are the most endangered species ranked as Critically Endangered, whereas Nicobar pigeon, which is endemic to Con Dao, is the only endangered bird species here. This study provides the list of bird species of international importance identified in these two wetlands.

Psittacine Beak and Feather Disease, Budgerigar Fledgling Disease and Aspergillosis in an African Grey Parrot (Psittacus erithacus)

  • A five-month-old African grey parrot was presented with alopecia, yellowish diarrhea, depression, and paralysis in the veterinary medical center, Chungbuk National University. The patient died 3 h later after hospitalization. For the accurate diagnosis, necropsy was performed and fungi were detected in the air sac. PCR was done for the viral detection which caused the alopecia, and for the species identification of fungi. Final diagnosis was a multi infection with avian circoviruses that caused psittacine beak and feather disease (PBFD), avian polyomavirus cause budgerigar fledgling disease (BFD), and Aspergillus fumigatus. This is the first report of a multi infection in South Korea.

도심하천과 자연하천의 식생형에 따른 조류 서식지 유형분석 (Classification of Avian Habitats Based on Vegetation Types in Urban and Natural Streams)

  • 우리나라 중부지역에 입지하는 3개 하천(안양천, 갑천, 섬강)을 대상으로 식생형에 따른 조류의 서식 유형을 파악하였다. 식생 유형은 교목형, 관목형, 장경-습생초지형, 단경-건생초지형, 기타 경작지형 등 5가지 유형이었다. 식생형과 출현 조류는 상관성이 높았는데, 교목형에서는 멧비둘기(Streptopelia orientalis) 및 직박구리(Hypsipetes amaurotis) 등의 종이, 관목형에서는 붉은머리오목눈이(Paradoxornis webbiana), 딱새(Phoenicurus auroreus) 등이, 장경 및 습생초지에서는 덤불해오라기(Ixobrychus sinensis), 개개비(Acrocephalus arundinaceus)등이, 단경초지에서는 꿩(Phasianus colchicus), 황조롱이(Falco tinnunculus) 등의 종이, 경작지에서는 중대백로(Egretta alba modesta), 흰뺨검둥오리(Anas poecilorhyncha) 및 밭종다리(Anthus spinoletta) 등의 종이 각각 번식 및 채식지로 이용하였다. 현존식생 및 입지유형에 따라서 조류의 종별 분포유형이 뚜렷이 구분되었다.

강원도 야생동물 조류의 조난.구조 실태분석 (Analysis of the wildlife distress and rescue of wild avian animals in Gangwon province)

  • This survey aims to provide information on the pattern of distress in wild avian animals in Gangwon province. This survey is based on 315 wild avian which were rescued by the Kangwon University Wildlife Rescue Center. The data was collected at the Wildlife Animal Center from September 2007 to August 2010, and by analyzing the shape of the wild animals killed or injured, the center seeks an efficient and systematic way to manage such distress. There are 55 species found, in this study including 40 common kestrel, 34 eurasian eagles, 20 common buzzards, 20 collared scops owls, 19 oriental scops owls, 16 brown hawk owls and 160 others. These are the results of the survey: 129 (40.3%) of 315 avian birds were found between June and August. Many cases in that term were observed falling from the nest or losing their mother because of unskilled flight during the breeding season's flight practicing process. Between November and December, 64 individual (20.3%) were found, and it's the second largest number. The main reasons for these results were starvation and exhaustion from the winter migratory season. The results indicated that the main reason for avian distress came from the following: collision with architecture windows in 73 cases (23.2%), starvation and exhaustion in 63 cases (20.0%), collision with vehicles in 51 cases (16.2%), becoming orphaned in 69 cases (21.9%), trespassing into buildings occurred in 19 cases (6.0%), collision with electric wires in 18 cases (5.7%), poaching materials in 13 cases (4.1%), becoming entangled by fishing wire and other miscellaneous materials in 4 cases (1.3%), infection from parasites in 4 cases (1.3%), toxicosis from such toxins like agricultural chemicals in 1 case (0.3%). Overall, the purpose of this investigation was to analyze cases of avian distress in Korea's wildlife. This study used areas, seasons and causes to understand origins of these animals distress. So that it can be utilized for organized wild avian rescue and treatment.

Dispensable role of wild rodents in avian influenza A virus transmission in Gyeonggi province, Korea

  • Avian influenza A viruses (IAVs) present significant threats to both animal and human health through their potential for cross-species transmission and global spread. Clade 2.3.4.4 H5Nx highly pathogenic avian IAVs initially emerged in East Asia between 2013 and 2014. Since then, they have spread to Europe, Africa, and America via migratory bird flyways. However, beyond viral transmission primarily facilitated by migratory birds, the potential involvement of other intermediate factors for virus transmission remains poorly investigated. This study aimed to investigate the role of wild rodents as intermediary hosts in the ecology of avian IAVs in Gyeonggi province, South Korea. By capturing and analyzing 189 wild rodents near poultry farms and migratory bird habitats in 2013 and 2014 and employing serological assays and virus isolation techniques, we found no evidence of IAV infection among these populations. Our results suggest that wild rodents may not significantly contribute to the transmission dynamics of IAVs within these regions.

조류 서식지로서 지뢰지대 삼림습지의 경관생태학적 평가 (Landscape Ecological Evaluation for Avian Fauna Habitats at the Forest Swamp Minefields of Civilian Control Zone(CCZ) Close to the Demilitarized Zone(DMZ) of Korea)

  • 본 연구는 선택되어진 지뢰지역과 습지조각의 산림성조류의 군집에 대하여 경관생태학적 가치를 평가하도록 하였다. 각각 면적이 다른 세 곳(8.5ha, 17.4ha, 40.6ha)의 지뢰지역과 산지지역의 세 개 비교지역의 조류상분석을 이용하여 평가하였다. 평균 산림성조류의 종수, 개체수, 종다양성, 종풍부도와 종균등도를 조각의 크기와 습지의 존재와 연결성과 같은 환경적 특성에 의하여 분석하였다. 군집 유사도지수는 실험군과 대조군 조사구사이를 비교하였다. 조류군집은 다음과 같이 요약되었다. 첫째, 63종의 조류가 조사되었으며, 최대개체수 합계는 828개체였다. 종다양성지수는 3.8이었으며, 종풍부도는 9.2, 종균등도는 93.3 이었다. 둘째, 작고 숲과 고립된 숲은 낮은 평균 종의 수와 개체수를 가졌지만, 소하천을 가진 작은 숲 조각은 개활수면이 없는 숲 조각에 비하여 높은 조류 밀도를 보였다. 셋째, 갈대습지를 가진 숲 조각은 단위면적당 조류 종의 수(/10ha), 단위면적당 개체수, 종다양도지수, 종풍부도와 종균등도에서 낮은 가치를 가진 것으로 평가되었다. 마지막으로, 군집 유사도지수는 소하천을 가진 작은 숲 조각은 다른 숲과 연결되어 있는 큰 산지 숲과 유사성이 높게 나타났다. 결과적으로, 저지대 숲 조각은 작은 면적에도 불구하고 다른 서식지와 연결되고 서식지가 넓은 산지의 숲 조각과 군집유사도가 비슷하였다. 따라서, 하천숲의 복원은 산림성조류의 군집에서 생물다양성을 향상시킬 것으로 제안한다.

조류 유래 재조합 H7N1 인플루엔자 바이러스의 분자적 특성 규명 (Molecular Characterization of an Avian-origin Reassortant H7N1 Influenza Virus)

  • 최근에 재조합 H7Nx 인플루엔자 바이러스가 산발적으로 인체 감염 사례가 보고되고 있으며 이러한 바이러스는 조류 종으로부터 지속적으로 분리되고있다. 본 연구에서는 조류에서 유래된 H7N1 인플루엔자 바이러스를 분리하여 A/wild bird/South Korea/sw-anu/2023로 명명하였고, 전장유전체 분석과 분자적 특성을 분석하였다. 계통발생학적 분석 결과 A/wild bird/South Korea/sw-anu/2023는 유라시아 혈통에 속하는 H7N1 인플루엔자 바이러스로 확인되었다. A/wild bird/South Korea/sw-anu/2023 바이러스의 polymerase basic 1(PB)2, PB1, polymerase acidic (PA), nucleoprotein (NP) 유전자는 야생 조류에서 분리되었던 조류 인플루엔자 바이러스유전자와 밀접한 관련이 있는 것으로 밝혀졌으며, hemagglutinin (HA), neuraminidase (NA), matrix (M), nonstructural (NS) 유전자는 집오리에서 분리되었던 조류 인플루엔자 바이러스와 유사하였다. 이러한 결과는 동아시아-호주 이동 경로를 따라 이동하는 야생 조류들 사이에서 새롭게 유전자가 재배열된 재조합 H7N1 조류 인플루엔자 바이러스가 순환되고 있음을 시사하고 있다. 따라서, H7Nx 인플루엔자 바이러스는 전 세계적으로 순환하며, 돌연변이된 H7N1 조류 인플루엔자 바이러스는 인간을 감염시킬 수 있으므로 야생 조류 및 가금류에서 H7N1 조류 인플루엔자 바이러스의 지속적인 감시가 필요할 것이다.
