Eicosapentaenoic acid (EPA) produced from marine organisms is widely used in nutraceuticals. Monodus subterraneus and Nannochloropsis oceanica, which are representative freshwater and marine Eustigmatophyceae, respectively, are known to have a high content of protein and lipid, particularly, EPA. In this study, to compare the growth and nutritional composition of M. subterraneus and N. oceanica, they were cultured in autotrophic and mixotrophic conditions with JM and f/2 medium, respectively, at $25^{\circ}C$. In addition, $80{\mu}mol\;photons\;m^{-2}s^{-1}$ with 24-hour and 12-hour light was provided, with the addition of 2% glucose to the medium for the mixotrophic culture. With regard to growth, M. subterraneus showed 10 times higher biomass in a mixotrophic culture than in an autotrophic one. However, no significant difference was observed for N. oceanica between the two culture methods. With respect to nutritional composition, M. subterraneus cultured autotrophically had a higher protein and lipid content, particularly EPA, than that cultured mixotrophically, but no significant difference was found in the two cultures of N. oceanica. Furthermore, M. subterraneus cultured autotrophically with continuous light showed higher nutritional composition, particularly EPA, than N. oceanica. In conclusion, the mass culture of freshwater M. subterraneus is much easier and more economical than marine N. oceanica. In addition, production of EPA will be economically improved if mixotrophic culturing of M. subterraneus is first conducted to maximize the biomass, and then secondary autotrophic culturing is performed.
Generally speaking, there are two widely used methods of Nitrogen removal from waste water: 1) nitrification using autotrophic microorganisms, and 2) denitrification using heterotrophic microorganisms. The C/N ratio is an important factor of the denitrification process. In this case, if methanol is added to increase the lacking organic matter, a high economic cost is incurred and methanol is left in the processed water. In an effort to fix these issues, autotrophic denitrification through the use of Hydrogen, Iron and Sulfur is being studied, and among those Sulfur is cheaper and carries out denitrification effectively, and therefore is being studied the most. In this study, after cultivating T. denitrificans, the presence of T. denitrificans was determined and the effectiveness of denitirification via T. denitrificans was studied. In order to find out about the inhibition of T. denitrificans from the loading of organic matter, this shows that the greater the loading of organic matter, the more the denitrification ability of T. denitrificans is hindered. In order to research the hindrance of T. denitrificans resulting from the loading of $NO_3{^-}-N$, these results show that concentrations less than 100mg/L per 100mL of gel volume do not hinder T. denitrificans. In order to research the optimization of denitrification resulting from T. denitrificans, three 500mL samples of Sulfur granules were prepared: 1) one with only T. denitrificans attached (Mode I), 2) one with both T. denitrificans and active sludge attached (Mode II), and 3) one with only active sludge attached (Mode III). The results showed that autotrophic denitrification using S from Mode I was the most active.
An autotrophic denitrification reactor (ADR-l) and a heterotrophic denitrification reactor (HDR-2) were operated to remove nitrate and nitrite in an anoxic environment in raw sewage. The $NO_3$-N removal rate of ADR-l was shown to range from 52.8% to 78.7%, which was higher than the $NO_3$-N removal rate of HDR-2. Specific denitrification rates (SDNR) of ADR-l and HDR-2 were 3.0 to 4.0 and 1.1 to $1.2\;mgNO_3$-N/gVSS/h, respectively. From results of restriction fragment length polymorphism (RFLP) of the 16S rRNA gene, Aquaspirillum metamorphum, Alcaligenes defragrans, and Azoarcus sp. were $\beta$-Proteobacteria that are affiliated with denitritying bacteria in the ADR-l. Specifically, Thiobacillus denitrificans was detected as an autotrophic denitrification bacteria. In HDR-2, the $\beta$-Proteobacteria such as Denitritying-Fe-oxidizing bacteria, Alcaligenes defragrans, Acidovorax sp., Azoarcus denitrificans, and Aquaspirillum metamorphum were the main bacteria related to denitrifying bacteria. The $\beta$-and $\alpha$-Proteobacteria were the important bacterial groups in ADR-l, whereas the $\beta$-Proteobacteria were the main bacterial group in HDR-2 based on results of fluorescent in situ hybridization (FISH). The number of Thiobacillus denitrificans increased in ADR-l during the operation period but not in HRD-2. Overall, the data presented here demonstrate that many heterotrophic denitritying bacteria coexisted with autotrophic denitrifying bacteria such as Thiobacillus denitrificans for nitrate removal in ADR-l. On the other hand, only heterotrophic denitritying bacteria were identified as dominant bacterial groups in HDR-2. Our research may provide a foundation for the complete nitrate removal in raw sewage of low-COD concentration under anoxic condition without any external organic carbon or the requirement of post-treatment.
This paper deals with the influence of chemical oxygen demand to nitrogen ratio ((COD/N) ratio) on the performance of an membrane bioreactor. We aim at establishing relations between COD/N ratio, organisms' distribution and sludge properties (specific resistance to filtration (SRF) and membrane fouling). It is also essential to define new criteria to characterize the autotrophic microorganisms, as the measurements of apparent removal rates of ammonium seem irrelevant to characterize their specific activity. Two experiments (A and B) have been carried on a 30 L lab scale membrane bioreactor with low COD/N ratio (2.3 and 1.5). The obtained results clearly indicate the role of the COD/N ratio on the biomass distribution and performance of the membrane bioreactor. New specific criteria for characterising the autotrophic microorganisms activity, is also defined as the ratio of maximum ammonium rate to the specific oxygen uptake rate in the endogenous state for autotrophic bacteria which seem to be constant whatever the operating conditions are. They are about 24.5 to 23.8 $gN-NH_4{^+}/gO_2$, for run A and B, respectively. Moreover, the filterability of the biological suspension appear significantly lower, specific resistance to filtration and membrane fouling rate are less than $10^{14}m^{-2}$ and $0.07\;10^{12}m^{-1}.d^{-1}$ respectively, than in conventional MBR confirming the adv < antage of the membrane bioreactor functioning under low COD/N ratio.
Soil autotrophic bacterial communities play a significant role in the soil carbon (C) cycle in paddy fields, but little is known about how rhizosphere soil microorganisms respond to different long-term (35 years) fertilization practices under double rice cropping ecosystems in southern China. Here, we investigated the variation characteristics of rhizosphere soil RubisCO gene cbbL in the double rice ecosystems of in southern China where such fertilization practices are used. For this experiment we set up the following fertilizer regime: without any fertilizer input as a control (CK), inorganic fertilizer (MF), straw returning (RF), and organic and inorganic fertilizer (OM). We found that abundances of cbbL, 16S rRNA genes and RubisCO activity in rhizosphere soil with OM, RF and MF treatments were significantly higher than that of CK treatment. The abundances of cbbL and 16S rRNA genes in rhizosphere soil with OM treatment were 5.46 and 3.64 times higher than that of CK treatment, respectively. Rhizosphere soil RubisCO activity with OM and RF treatments increased by 50.56 and 45.22%, compared to CK treatment. Shannon and Chao1 indices for rhizosphere soil cbbL libraries with RF and OM treatments increased by 44.28, 28.56, 29.60, and 23.13% compared to CK treatment. Rhizosphere soil cbbL sequences with MF, RF and OM treatments mainly belonged to Variovorax paradoxus, uncultured proteobacterium, Ralstonia pickettii, Thermononospora curvata, and Azoarcus sp.KH33C. Meanwhile, cbbL-carrying bacterial composition was obviously influenced by soil bulk density, rhizosphere soil dissolved organic C, soil organic C, and microbial biomass C contents. Fertilizer practices were the principal factor influencing rhizosphere soil cbbL-carrying bacterial communities. These results showed that rhizosphere soil autotrophic bacterial communities were significantly changed under conditions of different long-term fertilization practices Therefore, increasing rhizosphere soil autotrophic bacteria community with crop residue and organic manure practices was found to be beneficial for management of double rice ecosystems in southern China.
An autotrophic sulfur oxidizer, Thiobacillus sp. ASWW-2, was isolated from activated sludge, and its sulfur oxidation activity was characterized. Thiobacillus sp. ASWW-2 could oxidize elemental sulfur on the broad range from pH 2 to 8. When 5-50 g/L of elemental sulfur was supplemented as a substrate, the growth and sulfur oxidation activity of Thiobacillus sp. ASWW-2 was not inhibited. The specific sulfur oxidation rate of strain ASWW-2 decreased gradually until sulfate was accumulated in medium up to 10 g/L. In the range of sulfate concentration from 10 g/L to 50 g/L, the sulfur oxidation rate could keep over $2.0g-S/g-DCW{\cdot}d$. It indicated that Thiobacillus sp. ASWW-2 has tolerance to high concentration of sulfate.
As a bench-scale study, transformation of nitrate to nitrogen gas under anoxic condition was determined by using autotrophic denitrifiers containing Thiobacillus denitrificans and elemental sulfur as an electron donor. The research objective is to measure the basic kinetic parameters of autotrophic denitrification reaction on the removal efficiency of nitrate. The results showed that nitrate was almost completely transformed to nitrite in the first 4 days of column operation. After 2 days of accumulation of nitrite, its concentration slowly decreased and the compound was detected less than 0.5 mg/L in 14 days. In the experiment, sulfate concentration in the effluent was the 70~90 mg-S/L and the pH was maintained around pH 7.5. When nitrate concentration of bank filtrate in the real field is considered, this sulfate concentration seems to be acceptable. At 17 cm from the bottom of the column, the effluent showed the highest nitrite concentration, and nitrate concentration decreased rapidly to the Point of 33 cm from the bottom. The results suggest that an appropriate thickness of permeable reactive barriers is about 30 cm.
Plants are able to recognize even small changes in surrounding temperatures to optimize their growth and development. At warm temperatures, plants exhibit diverse architectural adjustments, including hypocotyl and petiole elongation, leaf hyponasty, and reduced stomatal density. However, it was previously unknown how such warm temperatures affected the early stages of seedling development. In our recent study, we demonstrated that the RNA-binding protein, FCA, is critical for sustaining chlorophyll biosynthesis during early seedling development, which is a prerequisite for autotrophic transition at warm temperatures. FCA plays a dual role in this thermal response. It inhibits the rapid degradation of protochlorophyllide oxidoreductases (PORs) that mediate chlorophyll biosynthesis. In addition, it induces the expression of POR genes at the chromatin level, which contributes to maintaining functional enzyme levels. Our findings provide molecular basis for the thermal adaptation of chlorophyll biosynthesis during the early stages of seedling development in nature.
Two hollow fiber membrane biofilm reactors (HF-MBfRs) were operated for autotrophic nitrification and hydrogenotrophic denitrification for over 300 days. Oxygen and hydrogen were supplied through the hollow fiber membrane for nitrification and denitrification, respectively. During the period, the nitrogen was removed with the efficiency of 82-97% for ammonium and 87-97% for nitrate and with the nitrogen removal load of 0.09-0.26 kg NH4+-N/m3/d and 0.10-0.21 kg NO3--N/m3/d, depending on hydraulic retention time variation by the two HF-MBfRs for autotrophic nitrification and hydrogenotrophic denitrification, respectively. Biofilms were collected from diverse topological positions in the reactors, each at different nitrogen loading rates, and the microbial communities were analyzed with partial 16S rRNA gene sequences in denaturing gradient gel electrophoresis (DGGE). Detected DGGE band sequences in the reactors were correlated with nitrification or denitrification. The profile of the DGGE bands depended on the NH4+ or NO3- loading rate, but it was hard to find a major strain affecting the nitrogen removal efficiency. Nitrospira-related phylum was detected in all biofilm samples from the nitrification reactors. Paracoccus sp. and Aquaspirillum sp., which are an autohydrogenotrophic bacterium and an oligotrophic denitrifier, respectively, were observed in the denitrification reactors. The distribution of microbial communities was relatively stable at different nitrogen loading rates, and DGGE analysis based on 16S rRNA (341f /534r) could successfully detect nitrate-oxidizing and hydrogen-oxidizing bacteria but not ammonium-oxidizing bacteria in the HF-MBfRs.
