Motion of Microbeads Propelled by Bacterial Chemotaxis (박테리아의 주화성에 의한 미세입자의 운동)

  • Recently, several research groups have been investigating the motion of flagellated bacteria, with the aim of examining the feasibility of using bacterial chemotaxis as an efficient power source for microactuators. In this study, microparticle-tracking velocimetry ($\mu$-PTV) is used for investigating the motion of fluorescent microbeads propelled by bacterial chemotaxis. Flagellated bacteria, Serratia marcescens, are spontaneously attached to the surface of the fluorescent polystyrene (PS) microbeads in an aqueous culture. The microbeads thus treated are injected into the test medium, which contains the solidified chemoattractant L-aspartate. With time, the particles slowly move toward the zone in which the L-aspartate concentration is high. This study shows that chemotaxis of flagellated bacteria can be applied as an efficient power source for microactuators.

Study of Oral Microbial Prevalence and Oral Health in Adults

  • Objective: This study performed a quantitative analysis using the real-time polymerase chain reaction technique to examine the oral microbial prevalence in adults and intended to examine the correlations between risk factors of periodontal disease and oral bacteria and correlation between oral test scores and oral microorganisms. Methods: We examined papillary marginal attached (PMA) index, modified patient hygiene performance (M-PHP) index, probing depth (PD), modified gingival index, and oral bacteria counts and surveyed 117, 20 years or older adult males and females who visited dental clinics in the Daejeon region to analyze the prevalence and oral health. Results: The prevalence was 100% for Fusobacterium nucleatum, meaning it was observed in all examined subject, 85.5% for Parvimonas micra, 76.1% for Prevotella intermedia, and 72.6% for Tannerella forsythia. The averages of P. gingivalis and T. forsythia increased as the examined subjects were older, and there was a statistically significant difference between T. forsythia and E. nodatum in relation to medical history, between P. intermedia and P. micra in relation to gender, and between P. intermedia and E. corrodens in relation to smoking (p<0.05). For a correlation between the oral test scores and oral microorganisms, P. gingivalis and F. nucleatum was highly correlated with PD (correlation coefficient of 0.51 and 0.41) (p<0.01) while P. gingivalis, P. micra, C. rectus, and E. nodatum were significantly correlated with M-PHP index, gingival index, PD, and PMA index (p<0.01, p<0.05). Conclusion: For oral health management of adults, the age, systemic disease, and smoking are closely related to oral bacteria, and P. gingivalis, T. forsythia, F. nucleatum, P. intermedia, P. micra, C. rectus, E. corrodens, and E. nodatum are considered to be the oral microorganisms that indicate periodontal health.

Hydrolysis of ${\beta}-glycosidic$ Bonds of Isoflavone Conjugates in the Lactic Acid Fermentation of Soy Milk (대두 요구르트 제조에서 이소플라본 배당체의 가수분해)

  • Isoflavones of soy milk were mainly present as sugar conjugates such as genistin and daidzin which a glucosyl residue was attached to their aglycones, genistein and daidzein through ${\beta}-glycosidic$ bond, respectively. When soy milk containing sucrose as a sugar source was fermented with lactic acid bacteria, small amount of lactic acid $(0.16{\sim}0.29%)$ was produced but isoflavone conjugates were fully hydrolyzed. Supplementation of glucose or lactose was required for normal lactic acid production and affected the hydrolysis of isoflavone conjugates in some lactic acid bacteria. In the case of Lactobacillus delbrueckii subsp. delbrueckii KCTC 1047, glycosidic bond of isoflavone was fully hydrolyzed regardless of glucose supplementation. But only $25{\sim}40%$ of daidzin and $65{\sim}80%$ of genistin was hydrolyzed when glucose was added into soy milk in the other lactic acid bacteria, Lactobacillus bulgaricus KCTC 3188, Lactobacillus casei KCTC 3109, Lactobacillus delbrueckii subsp lactis KCTC 1058, Lactobacillus lactis KCTC 2181. The hydrolyzing enzyme, ${\beta}-glucosidase$ produced by lactic acid bacteria except Lactobacillus delbrueckii subsp. delbrueckii KCTC 1047 could be considered as inducible in the fermentation of soy milk and its production was decreased when glucose was added.

Microflora of Manufacturing Process and Final Products of Saengshik (시판생식의 제조공정 및 최종제품의 미생물분포)

  • Microflora and contamination process of Saengshik products were investigated to ensure microbial safety of Saengshik. Food-borne pathogenic bacteria, such as Staphylococcus aureus, Bacillus cereus, and Clostridium perfringens detected mainly from grains were not removed by washing with tap water and freeze-drying. Contaminations of food-borne pathogenic bacteria occurred through raw material powder processed at other factories and during actual product manufacturing process, because detection rates of final products were higher than those of raw materials. Concentration of food-borne pathogenic bacteria increased with advancing of process after first pulverization. Dusts of powder and powder attached to machine were good media for air-borne microorganisms and caused to increase of food-borne pathogenic bacteria during process. Improvement of manufacturing process and sanitary control of machines arc necessary to ensure microbial safety of Saengshik.

The Change of Attached Bacterial Community on Aggregates in Lake Paldang (팔당호에서 Aggregates에 부착한 세균군집구조의 변화)

  • For scrutinizing the changes of aggregated bacteria in Lake Paldang, the FISH method was applied by using the rhodamine labeled probes, and total bacteria, chloropyll a concentrations and nutrients concentrations were measured. The aggregates were collected with sediment traps. The T-N, T-P, chlorophyll a concentrations of aggregates were higher 5-15 times, 81-140 and 49-66 times than water samples, respectively. Also, the bacterial numbers of aggregates were 200 times higher than those of water smaples. The ratios of each groups of water sample were 2.1-7.4% for $\alpha$-group, 4.5-8.3% for $\beta$-group, 2.1-7.4% for $\gamma$-group, 2.1-6.1% for Cytophaga-Flavobacterium group and 0.1-2.5% for 'other'group, respectively. While, in aggregates, the ratios of $\alpha$-, $\beta$-, $\gamma$- and Cytophaga-Flavobacterium groups were very small and most abundant group was 'other' bacteria. With these results, the aggregated bacteria in Lake Paldang had a particular group composition of bacteria.

The Bioleaching of Sphalerite by Moderately Thermophilic Bacteria (고온성 박테리아를 이용한 섬아연석의 용출 특성)

  • Bioleaching experiments were effectively carried out at $42^{\circ}C$, $52^{\circ}C$ and $62^{\circ}C$ to leach the more valuable metal ions from sphalerite using bacteria. The pH values of the bioleaching solution were constantly maintained for 10 days in the range of 2.40 to 2.55. In these bioleaching experiments, rod-shaped bacteria attached to the sphalerite surface were continuously observed in the sample. Along with the increase in the leaching temperatures, the concentration of Zn and Pb increased in the control sample of leachates, whereas the concentration of Fe increased in the sample containing bacteria. At $42^{\circ}C$, $52^{\circ}C$ and $62^{\circ}C$ the biological leaching content of Zn was found to be 9.5, 2.8 and 2.9 times higher than that in the chemical leaching content, respectively. At these temperatures, the content of Pb in the bacterial sample of the leachate was detected to be 14.8, 7.4 and 3.8 times higher than that of the control sample of the leachate, respectively.

  • GL, a glycolipid type produced from yeast, inhibited the growth of bacteria and fungi. Algicidal effect by GL against dinoflagellate was motility stop and lysis. When over 10 ${\mu}g/ml$ showed strong algicidal effect. GL attached to the algal body directly.

  • The objective of this study is to estimate the effect of the bed expansion and the characteristics of attached biomass in the start-up in the anaerobic fluidized bed reactor(AFBR). The fluidized bed reactor was operated with bacteria supported on the bed of granular activated carbon(GAC). The reactor was operated at 35$\circ$C, 5 kg $COD/m^3\cdot day$ at bed expansion varying from 0 to 100% with soluble glucose wastewater(5,000 mg/l). When the effluent reached a steady state at 100% of bed expansion, maximum COD removal efficiency of 87.3% and 0.031 $m^3CH_4/kg COD_{removed}$ were obtained. At higher bed expansion, COD removal efficiency, methane production rate and biogas production rate increased. Especially, at 50% of bed expansion, the efficiency of the treatment increasedg rapidly in the AFBR. The biomass colonized in the pits and crevices of the GAC particle and no complete biofilm was established in the bioreactor during the experiment.

광합성세균에 의한 미생물막의 형성

  • The formation of microbial films(biofilm) by a non-sulfur phototrophic bacteria, Rhodopseudomonas capsulata, on inorganic media was studied. Porous ceramic beads(PCB) were superior to other immobilizing media for the biofilm formation in a packed-bed reactor. It was found that the formation of microbial films favored a lower hydraulic retention time, showing a higher ratio of cells attatched to the media to those suspended in the solution. The cell concentration in the biofilm reactor was as high as 11,400mg/l, which is 8-folds of the cell concentration in an ordinary suspended treatment. It was observed that the formation of micribial film by R. capsulata followed a general serial process of cell attachment, microcolony formation, and biofilm formation. The microbial films thus formed was very stable even for an extremely high volumetric BOD loading rate of 15gBOD/l day. The scanning electron micrographs of the microbial films showed that the cells were attached to both the surface and pores of the media.

Steel Pile Corrosion in Potential Acid Sulfate Soil (잠재성 특이산성토중 강관말뚝의 부식)

  • The results and discussions of surveyed case site at constructed steel pile in potential acid sulfate soil were as follows. Topography at surveyed site was local alluvial valley and that site soils was classified as BanGog and YuGye series as detailed soil surveyed results in RDA and soil texture was Clay/Clay Loam. Soils pH was neutral, which was average 7.5 but much decreased to average 4.2 after $H_2O_2$ treatment. Organic matter and sulfate ions contents were very rich. The corrosion was severe at ground water fluctuation depth. Deposits colored black were attached to steel pile surface, which because of violent reaction in treatment HCI solution, were guessed as corrosion products (FeS) reduced by sulfate reducing bacteria(SRB). Consequently, main cause was thought microbiologically induced corrosion at this site where there is ground water fluctuation occurring oxidation and reduction reactions in turn and the soil is potential acid sulfate soil.
