Effects of Amyda sinensis on Allergic Inflammation Mechanism related Atopy Dermatitis (별갑이 아토피 피부염에서의 알러지성 염증 반응에 미치는 영향)

  • Objectives: This study aimed to investigate the effects of Amyda sinensis (AS) on allergic inflammation mechanism related atopy dermatitis. Methods: To investigate the effects of AS, We study inhibitory effect of AS on the levels of pro-inflammatory cytokines released from Raw264.7 cell stimulated with LPS (lipopolysaccaride), and EoL-1, THP-1, Jutkat cell stimulated with DP (Dermatophagoides pteronyssinus), and LPS indused acute inflammatory BALB/c mouse model. Result: AS reduced the levels of IL-$1{\beta}$ released from Raw264.7 cell stimulated with LPS at 20 ug/ml, 5 ug/ml concentration, and reduced the levels of IL-6 in a dose-dependent. AS significantly reduced the levels of MCP-1 released from EoL-1 cell stimulated with DP (Dermatophagoides pteronyssinus) at all the concentration, and significantly reduced the level of IL-8 at 0.1 ug/ml concentration. AS significantly reduced the levels of MCP-1 released from THP-1 cell stimulated with DP (Dermatophagoides pteronyssinus) at 1 ug/ml concentration, and reduced the level of IL-6 in a dose-dependent. AS significantly reduced the levels of IL-4 released from Jutkat cell stimulated with DP at all the concentration, and significantly reduced the level of IL-5 at 0.1 ug/ml. 1 ug/ml concentration,. and reduced the level of TNF-${\alpha}$ in a dose-dependent. AS significantly reduced the levels of TNF-${\alpha}$, IL-6, IL-$1{\beta}$, in LPS indused acute inflammatory BALB/c mouse model, in a dose-dependent. Conclusion: These result suggested that AS has suppressive effect on pro-inflammatory cytokines in various cell lines through the regulation of immune system. AS could be applied on the medicinal sources for treatment of immune abnormal diseases such as atopy dermatitis afterward.

Nano Capsulization of Ceramide and the Efficacy of Atopy Skin (나노세라마이드의 캡슐화와 아토피 피부의 치료)

  • The nano-ceramide capsulation is a technique that capsulates ceramide III and tocopheryl linoleate at the mono-vesicle to act on the horny layer in skin. In this technique, $0.5{\~}5.0\;wt\%$ of hydrogenated lecithin and $0.01{\~}2.00\;wt\%$ of lysolecithin are used as the membrane-strengthen agents of the mono-vesicle and $5.0{\~}10.0\;wt\%$ of propylene glycol and $5.0{\~}10.0\;wt\%$ of ethyl alcohol are used as solvents. Active ingredients such ceramide III and tocopheryl linoleate are utilized to enhance the moisturizing efficacy and treat atopy skin. These materials do not contain synthetic emulsifiers. The optimal conditions or nano-ceramide capsulation are such that particles pass Microfludizdizer 3 times at 1,000 bar and $60{\~}70^{\circ}C$ and pH of nano capsules is $5.8{\pm}0.5.$ The average size of particles is $63.1{\pm}7.34\;nm$ showing lucid state like water by the laser light scattering. A zeta potential value is $-55.1\pm0.84\;mV.$ Through clinical tests, the moisturizing effect (in-vivo, n=8, p-value<0.05) showed $21.15\%$ of improvement comparison to comparison-samples and $36.31\%$ of improvement compared to the state before treatment. Moreover, the effectiveness of atopy skin showed positive reaction from 10 volunteers.

Acceleration of DNCB-induced Early-apoptosis via Activation of Corticotropin Releasing Factor in the Hair Root of NC/Nga Mice (DNCB로 유도된 NC/Nga 아토피피부염마우스에서 부신피질자극호르몬방출인자 활성에 따른 모발세포의 초기세포사멸 연구)

  • Stress in skin plays a significant role in both the direct/indirect regulation of cellular processes occurring in hair, which in turn affect the hair cycle. However, experimental data regarding the effects of stress-related corticotropin releasing factor (CRF) released by stress on the apoptotic process involved in hair is limited. Therefore, we investigated the acceleration of early-stage apoptosis induced by atopy-related stress using a 2,4-dinitrochlorobenzene NC/Nga mice model. Expression of CRF, its related proteins, annexin V, and mitochondrial dysfunction were measured by immunohistochemical analyses. Atopic stress strongly stimulated stress hormones response, such as CRF and adrenocorticotropic hormone, in outer epithelial sheath of the hair. Moreover, its stress induced mitochondrial damage and early-stage apoptosis of cells in hair root. These findings suggest that hair damage due to apoptosis in atopy model is accelerated in a high CRF environment. Importantly, the effect of stress-related CRF on apoptosis processes involved in atopy dermatitis-related hair loss, suggests that the CRF-regulating development or maintenance materials may provide effective therapeutic strategies for hair health.

Effect of Coptidis Rhizoma extract on Atopic Dermatitis-like Skin Lesions in NC/Nga Mice (황련 추출물의 아토피피부염 유발 생쥐에서 피부손상 완화 효과)

  • This study aims to evaluate the anti-inflammatory effect of Coptidis Rhizoma (CR) extract for atopic dermatitis through maintaining skin barrier and regulating Th2 cell differentiation. We divided NC/Nga mice into 3 groups as follows; atopy-like dermatitis induced group with CR treatment (CT, n=10), no treatment group(Ctrl), atopy-like dermatitis elicited group(AE). Atopy-like dermatitis was induced to NC/Nga mice by sensitizing with dermatophagoides farinae(DfE) on 7, 8, 9, 11, 12, and 13th week. After inducing atopic dermatitis, CR extract was administered 20 mg/kg daily for the experimental duration to the CT group. We measured the integrity of lipid layers in the epidermis and Th2 differentiation through immunohistochemical staining against filaggrin, loricrin, IL-4, and IL-13. We also measured the distribution of subcutaneous collagen fibers by the Masson's trichrome staining. Administration of CR significantly inhibited the reduction of lipid layers in the skin that caused atopy. The expression of IL-4, IL-13, each of which is a cytokine secreted by T helper type 2 (Th2) cells, was markedly suppressed in the CT group as compared with AE group (p<0.05). CR treatment also decreased the expression of iNOS, $p-I{\kappa}B$. Atopic dermatitis induced dermatological damage to skin, such as hyperplasia of epithelium, and capillary proliferation was significantly reduced by CR administration. CR effectively inhibited the thinning of the skin barrier and inflammatory responses in atopic dermatitis-induced mice. In particular, it showed anti-inflammatory effects by reducing the expression of IL-4 and IL-13, Th2 cell cytokines, which play a crucial role in development of atopic dermatitis. Therefore, CR can be a good candidate to ameliorate and treat atopic dermatitis.

Factors associated with Atopic Dermatitis in Korean Adults: The Korean National Health and Nutrition Survey 2008 (우리나라 성인의 아토피피부염 관련요인: 2008년도 국민건강영양조사 자료를 중심으로)

  • Purpose: The purpose of this study was to investigate the factors associated with atopic dermatitis in Korean adults. Methods: The data were obtained from the Korean National Health and Nutrition Survey on 2008. Subjects of this study were 7,280 adults aged older than 18 years. The data were analyzed for demographic characteristics, health status, alcohol drinking, smoking, hypertension, hyperlipidemia, asthma, chronic sinusitis, rhinitis, stress, depression, and suicidal ideation between atopy group and non-atopy group. Results: Among 7,280 subjects, 2.9% of Korean adults had a diagnosis of atopic dermatitis from doctor. Multiple logistic regression identified that the presence of atopic dermatitis was associated with marital status (OR=2.66, CI 1.84~3.86, p<.001), rhinitis (OR=1.52, CI 1.04~2.22, p = .030), and depression (OR=1.89, CI 1.04~3.41, p = .036). Conclusion: Based on these results, marital status, rhinitis, and depression should be considered in addressing individuals with atopic dermatitis. And the other factors contributing to atopic dermatitis need to be specifically identified.

Assessment of the Environmental Conditions in Patient's Houses with Allergy by Use of a Fungal Index - A Case Study (곰팡이 센서(Fungal detector)를 이용한 알러지 환자 가정의 실내 환경 평가 - 사례연구)

  • The indoor environmental condition was assessed in houses with allergy (asthma and atopy) patients by use of a fungal detector. The fungal index was calculated from the growth rate of the sensor fungi in a fungal detector encapsulating the spores, Alternaria alternata S-78, Eurotium herbariorum J-183 and Aspergillus penicillioides K-712. Fungal indices were higher in asthma patient's houses than in control houses and Eurotium herbariorum showed the highest growth response among the sensor fungi. Dust mites allergen, Der f1, was also significantly high in allergy patient's houses where fungal indices above 10 were detected. A correlation was observed between the fungal indices and dust mite allergen proliferations in examined houses. Therefore, the fungal index can be a useful tool as an indirect indication for detecting chronic dampness that brings both contaminations by fungi and dust mite.

Enhancement of Allergen-related Eosinophilic Airway Inflammation and Airway Hyperresponsiveness by House Dust Particles in Mice (생쥐에서 가정분진 입자로 인한 항원관련 호산구성 기도 염증과 기도 과민성의 증대)

  • The number of patient with allergic asthma and atopy have increased in the cities of Korea steadily. In order to elucidate the primary factor, we investigated whether the house dust particles collected from an apartment of the middle classes has promoting effects of allergen-related airway inflammation and airway hyperresponsiveness. Mice were treated with 0.1 mL of 1 mg/mL of house dust particles suspension by intratracheal instillation once weekly for 10 weeks combined with ovalalbumin (OVA) sensitization. Intratracheal instillation of house dust particles and OVA sensitization caused an increase in the level of serum L-lactate dehydrogenase (LDH), immunoglobulun-E (IgE) and histamine, and an elevation in respiratory resistance. It also enhanced infiltration of eosinophils in the bronchoalveolar lavage fluid (BALF) of mice, IgE and eotaxin expression in blood, and T helper type 2 cell derived cytokine levels such as of interleukin (IL)-4, IL-13 and IL-5 in the BALF. However, it did not influence T helper type 1 cytokine such as interferon-gamma in the BALF. These results indicate that house dust particles elevate allergen-related airway inflammation and airway hyperresponsiveness in mice and may play an important role in the aggravation of asthma and atopy in Korea.

A Study of the Effects of Systemic Skin Care as a Adjunct Treatment for Adult Atopic Dermatitis (피부 치료 시스템이 성인 아토피 피부염에 미치는 영향에 대한 조사 연구)

  • This study was conducted with 60 adult males who visited the Atomi Cooperative Clinic(Atomi Dermatology/Pediatrics/Oriental Clinic) in Seoul between May 2003 and December 2004; they were divided into three groups, each of which consisted of 20 persons, according to the level of erythma and then each group was subdivided to consist of ten according to whether they received skin treatment and care. The standard care provided to two groups involved topical steroids and oriental medicines prescribed by a dermatologist and a oriental doctor, respectively. 5th-grade topical steroid ointment was applied to the face and 3rd-grade to the limbs; a oriental medicine was administered in a lukewarm state half an hour after meals three times a day. To determine how special and systematic skin treatment and care was helpful in treating atopy, a skin treatment system was applied to the experimental group while the control group was provided with standard care alone. By using Mexameter(MX18) manufactured by ck-mpa as a measuring tool, the inflammation level was observed at the right antecubital space during each visit to the clinic. In view of the re suits, introduction of the systemic skin care for A. D to legitimate treatment provided by a medical institution is expected to be an appropriate supplementary treatment for adult patients who suffer from frequent recurrence of atopic dermatitis.

A Case Study on Atopic Dermatitis with the Treatise on Febrile Diseases (상한론 처방을 이용한 아토피성 피부염 환자 1례 보고)

  • Introduction : Atopic dermatitis is very epidermic disease in these days. Atopic dermatitis is difficult for patient to be treated because there is no specific treatment. The oriental method of atopic dermatitis is varied as patient situation and doctor's view point. The Treatise on Febrile Diseases(傷寒論) may be a hint for atopy disease. Object: Although we use various method such as acupucture, aroma therapy, a medicine for external use, we use the two decoctions in the Treatise on Febrile Diseases according to differentiation of syndromes and abdominal diagnosis. Result : After inpatient treatment in 21 days, the itching, redness, exudation, crust, lichen get better and insomnia and constipation was removed quickly. The SCORAD index is decreased from 84.2 to 25.9. Conclusion : These decoction can treat the atopic dermatitis although there is no symptom in atopy because we cure the all situation of the patient. We can cure the atopic dermatitis not only by routine remedy for various symptoms but also by diagnosis and treatment based on overall analysis of symptoms and signs.
