형질전환된 황기 모상근으로부터 Astragalosides의 생산을 위한 연구 (High-Yield Production of Astragalosides from Transgenic Hairy Root Cultures of Astragalus membranaceus)

  • 황기의 형질전환된 모상근으로부터 약리물질인 astragalosides I, II, 그리고 III을 효율적으로 생산하기 위하여 다양한 물리화학적 조건들 즉, 배지, 초기 당농도, 교반속도, 초기 접종농도, 광조사, 그리고 chitosan 처리에 한 elicitation 효과를 조사 하였다. 선발된 AG-04 clone의 성장과 astragalosides의 함량은 4종의 배지들 중 SH배지에서 가장 좋았으며, 배지에 첨가하는 초기 당 농도는 3%와 6%에서 각기 건물중과 astragalosides의 함량이 가장 높게 나타났다. 또한, 초기 접종량은 50 ml 배지에 500 mg(FRW)씩을 주입하는게 가장 효과적이었으며, 18시간 광조사가 되는 Shaking incubator에서 90 rpm과 120으로 교반시켰을 때 모상근의 성장과 astargalosides의 함량이 높게 나타났다. 배양 2주 후 chitosan의 처리는 모상근의 성장에는 크게 영향을 미치지 않았으나, 100 mg/L 처리구에서 대조구의 약 2.1배의 astragalosides 함량 증가를 가져왔다. 이와같은 연구결과는 황기의 대량배양을 통한 saponins 생산 연구에 효율적으로 활용될 수 있을 것으로 본다.

Inhibitory Effect of Astragaloside I and IV on Passive Cutaneous Anaphylaxis Reaction and Scratching Behaviors in Mice

  • To evaluate the antiallergic effect of the dried root of Astragalus membranaceus Bunge (AM) (Leguminosae), which inhibited the mouse passive cutaneous anaphylaxis (PCA) reaction in a preliminary experiment, its main constituents, astragalosides I and IV, were isolated and their antiallergic effects were investigated. Astragalosides I and IV inhibited the PCA reaction induced by the IgE-antigen complex, and the scratching behaviors induced by compound 48/80. These constituents reduced the protein expressions of $TNF-{\alpha}$ and IL-4 in IgE-induced RBL-2H3 cells. These findings suggest that astragalosides I and IV as well as AM can improve IgE-induced anaphylaxis and scratching behaviors.

재배 황기의 Astragalosides 함량의 변이 (Variation of Astragalosides Contents in Cultivated Astragalus membranaceus)

  • This study was conducted to determine the contents of astragalosides in Astragalus membranaceus. The total astragaloside content ($1701{\mu}g\;g^{-1}$) of 2 year-cultivated astragalus was the highest among the variously cultivated astragalus plants. Upon an increase in cultivation time, the average value of the total astragaloside content decreased (from $1650{\mu}g\;g^{-1}$ to $645{\mu}g\;g^{-1}$). Especially, the content of astragaloside decreased rapidly. Comparison between the top and subterranean parts of harvested astragalus plants showed that as the cultivation time increased, the astragaloside content of the top part decreased. On the other hand, the subterranean part demonstrated the opposite pattern. The astragaloside content of the top root increased compared to that in the lateral root (from $1.9{\mu}g\;g^{-1}$ to $33.4{\mu}g\;g^{-1}$). Further, the content of astragaloside increased in the top root as the cultivation time increased.

Effects of compound traditional Astragalus and Salvia Miltiorrhiza extract on acute and chronic hepatic injury

  • Previous reports showed that Compound Astragalus and Salvia miltiorrhiza extract (CASE), which was mainly composed of astragalosides, astragalus polysaccharide and salvianolic acids, inhibited hepatic fibrosis by mediating transforming growth factor-${\beta}$ (TGF-${\beta}$)/Smad signaling. Our aim was to examine the effects of CASE on D-galactosamine (D-GalN) treated liver injury in mice and carbon tetrachloride ($CCl_4$)-induced liver fibrosis in rats. CASE was administered to mice with D-GalN-induced liver injury and to rats with $CCl_4$-induced liver fibrosis, respectively. Liver injury was routinely evaluated by relative liver weight, serum levels of ALT, AST, hyaluronic acid (HA), hepatic malondialdehyde (MDA) content, superoxide dismutase (SOD) activity, hydroxyproline (HYP) and histopathologic changes. Treatment of mice with CASE (60, 120, and 240 mg/kg, ig) significantly lowered ALT, relative liver weight, and MDA levels when compared with D-GalN treated mice. CASE (120, 240 mg/kg) significantly lowered ALT, AST, HA, HYP, and MDA levels against $CCl_4$ treated rats. Decreased SOD level was reversed with CASE treatment. Upon histopathological examination, CASE treatment had significantly inhibitory effect on the progression of hepatic fibrosis in rats. These results indicate that CASE might be effective in treatment and prevention of acute and chronic hepatic injury due to its antioxidant activity.

재배 황기의 Phenolic Compounds 함량의 변이 (Variation of Phenolic Compounds Contents in Cultivated Astragalus membranaceus)

  • This study was conducted to determine the contents of phenolic compounds and astragalosides in Astragalus membranaceus. Between the phenolic compound groups, flavonoids compounds (51.13%) had higher average concentrations than the phenolic acids groups. Among the 30 phenolic compounds, quercetin ($353.11{\mu}g\;g^{-1}$) provided the highest concentrations. According to different cultivation year, 3-year-cultivated Astragalus ($2612.57{\mu}g\;g^{-1}$) showed the highest concentrations of phenolic compounds. According to different harvest days, 6/5 harvesting Astragalus ($2615.80{\mu}g\;g^{-1}$) showed the highest concentrations of phenolic acids. Comparison between the top and subterranean parts of harvested Astragalus plants cultivated for 2 years, 3 years, and 5 years showed big difference of total phenolic compounds in concentrations. Further, the top part had higher amounts of the total phenolic compounds than the subterranean part among all Astragalus. This tendency was similar to those of the top root and lateral root. The concentration of the phenolic compounds in the top root was higher than that of the lateral root.
