Screening of Genes Which are Able to Affect Kalanchoe Vegetative Reproduction (Kalanchoe 식물의 영양 번식에 영향을 줄 수 있는 유전자들의 선발)

  • The genus Bryophyllum is best known for many of its species having the ability to produce plantlets on their leaves. This phenomenon is also known as vegetative reproduction. Differential expressed gene (DEG) detecting technique was applied in order to survey the genes involved in the process of asexual reproduction for plantlet formation. Based on homology search using the NCBI database after screening of genes, 38 genes were identified from a total of 69 DEGs. Most of these DEGs were related to cell division, to intercellular signal transduction, and to hormone (cytokinin and ethylene) signaling.

Identification of the Imperfect Stage of Mycosphaerella nawae Causing Circular Leaf Spot of Persimmon in Korea (감나무 둥근무늬낙엽병균 Mycosphaerella nawae의 불완전 세대 동정)

  • Asexual spores of Mycosphaerella nawae were profusely produced on PDA after a prolonged incubation at $25^{\circ}C$ for 90 days. When persimmon trees were artificially inoculated by the conidial suspension, typical symptoms of circular leaf spot of persimmon appeared on the leaves two month later. The imperfect stag of the fungus was identified as Ramularia sp. based on following morphological characteristics examined under a light microscope and a scanning electron microscope. Conidia were mostly ellipsoid, but occasionally cylindrical, elongated oval, taro, peanut or gourd shapes and measured as 12.2~32.6$\times$6.1~10.2 ${\mu}{\textrm}{m}$. erect, hyaline, colorless-light brown. Conidia were formed solitarily or in chains on a medium and infected leaves. Conidiophore was erect, hyaline, colorless-light brown. and the size was 20.4~102.0$\times$3.1~10.2 ${\mu}{\textrm}{m}$, respectively. In this paper, we firstly demonstratrated that asexual spores of M. nawae induced persimmon circular leaf spot in nature as well as sexual spores of the fungus. Therefore, it is hypothesized that the imperfect stage of the fungus plays an important role in nature for epidemics as secondary inoculum.

Isolation and Characterization of NSD mutants in Aspergillus nidulans (Aspergillus nidulans에서 NSD 돌연변이주의 분리 및 분석)

  • Several mutants which never underwent to sexual development(NSD) of Aspergillus nidulans were analyzed genetically and physiologically. They were divided into two groups according to their characterisitics of asexual development after release from aeration block. The mutants in first group proceeded asexual development immediately after removal of aeration block, while those in second group did 10 hours or more later. The NSD mutants were separated into 4 complementation groups, nsdA, nsdB, nsdC and nsdD. The nsdA and nsdD genes were linked to AcrA1 on linkage groupⅡ and pabaA1 on linkage group I, respectively. The mutant alleles were all recessive to wild type allele in heterokayon state. The mutants did not developed cleistothecia on any of carbon sources, except NSD208 which developed cleistothecia on lactose.

A Nucleolar Protein, MoRRP8 Is Required for Development and Pathogenicity in the Rice Blast Fungus

  • The nucleolus is the largest, membrane-less organelle within the nucleus of eukaryotic cell that plays a critical role in rRNA transcription and assembly of ribosomes. Recently, the nucleolus has been shown to be implicated in an array of processes including the formation of signal recognition particles and response to cellular stress. Such diverse functions of nucleolus are mediated by nucleolar proteins. In this study, we characterized a gene coding a putative protein containing a nucleolar localization sequence (NoLS) in the rice blast fungus, Magnaporthe oryzae. Phylogenetic and domain analysis suggested that the protein is orthologous to Rrp8 in Saccharomyces cerevisiae. MoRRP8-GFP (translational fusion of MoRRP8 with green fluorescence protein) co-localizes with a nucleolar marker protein, MoNOP1 fused to red fluorescence protein (RFP), indicating that MoRRP8 is a nucleolar protein. Deletion of the MoRRP8 gene caused a reduction in vegetative growth and impinged largely on asexual sporulation. Although the asexual spores of DMorrp8 were morphologically indistinguishable from those of wild-type, they showed delay in germination and reduction in appressorium formation. Our pathogenicity assay revealed that the MoRRP8 is required for full virulence and growth within host plants. Taken together, these results suggest that nucleolar processes mediated by MoRRP8 is pivotal for fungal development and pathogenesis.

Functional Analysis of Aspergillus nidulans Genes Selected by Proteomic Analysis under Conditions Inducing Asexual Development (Aspergillus nidulans 무성분화 촉진 조건의 단백체 및 해당 유전자 기능분석)

  • Despite the significance of external environmental factors in differentiation, putative factors involved in differentiation of Aspergillus nidulans have not yet been fully understood. A sporulation-specific proteome analysis of A. nidulans in the present study revealed that the expression levels of more than 2,400 proteins were affected under conditions inducing sporulation (0.6 M KCl) compared with normal conditions. Among the proteins with predicted functions, two targets, AN1342 and AN9419, were functionally analyzed using targeted deletion strains and phenotypic observations. For AN1342, because the deletion of the corresponding open reading frame caused a reduction in stalk length during asexual development and in pigment production in liquid culture, the gene was designated as sspA ($\underline{s}hort$ $\underline{s}talk$ & $\underline{p}igment$). Deletion of the AN9419 gene, which is predicted to encode alanyl-tRNA synthetase, led to severe growth defects due to alanine auxotrophy and abolishment of asexual reproduction and thus, the gene was designated as alaA.

Effect of High CO2 Concentration on Activation of Sexual Development in Aspergillus nidulans (고농도 CO2 노출에 의한 Aspergillus nidulans의 유성생식 촉진효과)

  • Fungal development is largely affected by many environmental factors. In a model filamentous fungus Aspergillus nidulans, asexual development is promoted by exposure of light, presence of salt and non-fermentable sugars. In other hand, sexual development is largely induced by absence of light, fermentable sugars and hypoxic condition. Also, some important genes including veA and nsdD play positive roles in activating sexual development. Here, we reported that the effect of high concentration of $CO_2$ on developmental decision in A. nidulans. When wild-type $veA^+$ strain was cultured in normal condition, sexual and asexual development occurred in balanced manner. However, high concentration of $CO_2$ (~5%) strongly activated sexual development and inhibited asexual development. Furthermore, this $CO_2$ effect was controlled by the veA or nsdD gene. High $CO_2$ culture of $veA^-$ or $nsdD^-$ mutant didn't activate sexual development, suggesting that the activation of sexual development induced by high $CO_2$ cannot overcome the genetic requirement of sexual development such as veA or nsdD. Since 5% $CO_2$ is an important condition for human pathogenic fungi for surviving and adapting in human body, this developmental pattern of A. nidulans affected by $CO_2$ concentration may provide interesting clues for comparative study with human fungal pathogens including Aspergillus fumigatus.

Gene Structure and Function of fkhE, a Forkhead Gene in a Filamentous Fungus Aspergillus nidulans (Aspergillus nidulans forkhead 유전자 fkhE의 구조와 기능 분석)

  • A homothallic filamentous fungus Aspergillus nidulans has been used as the a model organism for studying growth and development for eukaryotic system. Various studies about specific transcription factors have been performed for elucidating the molecular mechanisms of growth, asexual and sexual developmental processes. Among them, the fkhE gene (AN2025.3) is located in chromosome VII and contains an ORF encoding 718 amino acid polypeptide intervening with two short introns. The cDNA sequencing revealed that at least four types of alternative splicing events were occurred when the fkhE gene was transcribed. The putative FkhE polypeptide contains a conserved forkhead domain and a bipartite nuclear localization signal at it's N-terminus and C-terminus, respectively. Deletion of fkhE resulted in impaired conidiophore formation in a solid medium. However, the sexual developmental process or cleistothecia formation was normal. Furthermore, fkhE deletion mutant produced conidiophores and conidia under the submerged culture, indicating that the fkhE gene is involved in asexual developmental process similar to the fkhF gene.

A New Record of Stephanasterias albula (Asteroidea: Forcipulatida: Asteriidae) from the East Sea, Korea

  • A sea star was collected with fishing nets at a depth of approximately 100-160 m in the East Sea and was identified as Stephanasterias albula (Stimpson, 1853) belonging to the family Asteriidae of the order Forcipulatida. This species characterized by having the capability of asexual reproduction by self-division turned out to be new to the Korean fauna. Its morphological characteristics are redescribed with illustrations. Thirty one species of sea stars including S. albula are now recorded from the East Sea of Korea.
