Four Endophytic Ascomycetes New to Korea: Cladosporium anthropophilum, C. pseudocladosporioides, Daldinia eschscholtzii, and Nigrospora chinensis

  • Ascomycota is the largest phylum of the Fungi, including approximately 6,600 genera. They are often isolated from soils, indoor air, and freshwater environments, but also from plants as pathogens or endophytes. In this study, four species of Ascomycota (two of Cladosporium and one of each Daldinia and Nigrospora) were collected from the leaves of four woody plants (Camellia japonica, Ginkgo biloba, Quercus sp., Vitis vinifera). Their cultural characteristics were investigated on five different media (PDA, V8A, CMA, MEA, CZA) at 3 days after incubation at $25^{\circ}C$ in darkness. BLASTn search and phylogenetic analysis were performed using the internal transcribed spacer (ITS) rDNA sequences, in addition to tef1 gene sequences for Cladosporium species. Based on the cultural, morphological, and phylogenetic data, the isolates were identified as Cladosporium anthropophilum, Cladosporium pseudocladosporioides, Daldinia eschscholtzii, and Nigrospora chinensis. Previously, some members of Cladosporium and Nigrospora have been recorded as endophytes inhabiting the leaves and stems of various plants, whereas Daldinia eschscholtzii is a wood-inhabiting endophyte or wood-decaying fungus. To our knowledge, this is the first report of these four ascomycetes in Korea.

  • Fungi are ubiquitous and indispensable components of nearly all ecosystems on earth, including freshwater environments. A survey of fungal diversity in freshwater environments of Korea led to the identification of four unrecorded Pezizomycotina (Ascomycota) species in 2016 and 2017, based on morphology and molecular phylogeny; these included Cladorrhinum australe, Curvularia muehlenbeckiae, Curvularia pseudobrachyspora (Dothideomycetes), and Diaporthe longicolla (Sordariomycetes).

Diversity of Microorganisms in Decaying Maize Stalks Revealed by a Molecular Method

  • Microbial diversity in decaying maize stalk was characterized by constructing and analyzing rRNA gene clone library. Total 47 OTUs were obtained from 82 bacterial clones, including Proteobacteria (64.6%), Actinobacteria (30.5%), Bacteroidetes (2.4%) and Firmicutes (2.4%). Most proteobacterial clones were members of Rhizobium, Pseudomonas and Stenotrophomonas. Eighty-four percent of Actinobacteria was related to Microbacterium. Only 14 OTUs were identified from 124 fungal clones, including Ascomycota (88%) and Basidiomycota (12%). Sixty percent of Ascomycota were members of Eupenicillium and Paecilomyces but all Basidiomycota were close to Kurtzmanomyces nectairei.

Seven Unrecorded Species of Ascomycota Isolated from the Rhizosphere Soils of Apple and Pear Trees in Korea

  • Soil is a rich source of saprobic and pathogenic fungi in crop cultivation areas. Compared with that of the fruit trees, scarce information is available regarding the fungi present in orchard soils in Korea. We sampled rhizosphere soils of apple and pear trees from several orchards in Cheongju, Anseong, and Cheonan, Korea. During the processing of soil fungi, seven unrecorded species of the phylum Ascomycota were isolated. These included Acrocalymma walkeri, Clonostachys krabiensis, Coniella vitis, Cosmospora diminuta, Lasiobolidium spirale, Penicillium vallebormidaense, and Pseudothielavia arxii. All the species were identified and described based on morphological characteristics and molecular analysis of the ITS1-5.8S-ITS2 region and the large subunit of nuclear ribosomal RNA gene and partial β-tubulin gene (BenA) sequences. Descriptions and illustrations of the morphological characteristics are provided.

Ophiostomatoid Fungi in Pine Wilt Disease and Oak Wilt Disease in Korea

  • Pinewood nematode (PWN, Bursaphelenchus xylophilus) is a serious pathogenic worm that quickly dry pine trees to death. Recently, PWN has been devastating huge amounts of conifer trees in Korea. As a first step to explore the association and ecological roles of fungi in PWN life cycle in Korea, in this study we first isolated and indentified fungi from PWN-infested Korean pine and Japanese black pine wood sampled in Jinju, Sacheon, Pocheon, Chuncheon, Gwangju, and Hoengseong in Korea. A total of 144 fungal isolates were obtained from Japanese black pine wood and 264 fungal isolates from Korean pine wood. Their morphology and nucleotide sequences of the ITS rDNA and ♌-tubulin gene were examined for species identification. Ophiostoma ips, Botrytis anthophila, Penicillium sp., Hypocrea lixii, Trichoderma atroviride, O. galeiforme, Fusarium proliferatum were identified from Japanese black pine wood. Leptographium koreanum, L. pini-densiflorae, Ophiostoma ips, Penicillium raistrick, Trichoderma sp. were isolated from Korean pine wood. O. ips and L. koreanum were the major species on the two different PWN-infected pine tree. The cultivation of PWN on fungal mat of the identified species did some enhance PWN reproduction. The ambrosia beetle, Platypus koryoensis, is a serious pest of oak trees in Korea. In this study we investigated filamentous fungi present in the body of the beetle. Fourteen genera of filamentous fungi belonging to Ascomycota and Basidiomycota were isolated. All the obtained genera were isolated in the mitosporic state. The identified fungi were classified in 11 distinct orders including the Ascomycota (Eurotiales, Hypocreales, Microascales, Ophiostomatales, Pleosporales, and Sordiales) and Basidiomycota (Agaricales, Corticiales, Polyporales, and Russulales Xylariales). Within Ascomycota, 13 species were found. Meanwhile five species were found within Basidiomycota. The results showed the presence of diverse fungi in P. koryoensis. Among the isolated fungi, some were able to produce wood degrading enzymes. Further fungal isolation was performed with P. koryoensis infested Quercus mongolica trees sampled at Kumdan mountain in Hanam-Si, Gyeonggi province from June of 2009 to June of 2010. Penicillin spp. and Trichoderma spp. were the major species of mold fungi group. Pichia guilliermondii was the major species of mold yeast group. Raffaelea quercus-mongolicae was also isolated, but its isolation frequency was not high. Other species identified were Ambrosiella xylebori, Fusarium solani, Cryphonectria nitschke, Chaetomium globosum, and Gliocladium viride, Candida kashinagacola, C. maritima, C. vanderkliftii, Saccharomycopsis crataegensis.

반추위 곰팡이 다양성 조사 : 메타분석 (Diversity Census of Fungi in the Ruminal Microbiome: A meta-analysis)

  • 본 연구의 목적은 곰팡이 28S rDNA 염기서열의 메타분석을 통하여 반추위 곰팡이의 다양성을 조사하는데 있다. 'rumen'과 'ruminal'이 반추위 곰팡이 유래 염기서열들을 회수하기 위한 검색어로 사용되었다. 2016년 9월부로 모든 28S rDNA 염기서열이 보관되어 있는 Ribosomal Database Project(RDP, http://rdp.cme.msu.edu) 데이터베이스에서 반추위 곰팡이 유래 28S rDNA 유전자 염기서열(n=165)을 획득하였다. 총 165개의 염기서열은 분류학상의 '문(phylum)'인 Ascomycota, Neocallimastigomycota 및 Basidiomycota로 분류되었고, 165개의 염기서열 중에서 각각 109개, 48개, 8개의 염기서열을 차지하였다. Ascomycota 염기서열은 식물병원성곰팡이나 마이코톡신을 생성하는 곰팡이를 포함하고 있는 '속(genus)' Pseudonectria, Magnaporthe, Alternaria, Cochliobolus, Cladosporium 및 Davidiella로 분류되었다. 또한, Basidiomycota 염기서열은 식물병원성곰팡이를 포함하고 있는 '속(genus)' Thanatephorus와 Cryptococcus로 분류되었다. 뿐만 아니라, Neocallimastigomycota 염기서열의 경우 섭취된 조사료의 주요 구조탄수화물을 분해하는 '속(genus)' Cyllamyces, Neocallimastix, Anaeromyces, Caecomyces, Orpinomyces, Piromyces로 분류되었다. 본 연구는 처음으로 28S rDNA 염기서열의 메타분석을 통해 반추위 곰팡이 다양성에 대한 정보를 통합적으로 제공하였다. 본 연구의 결과는 향후 반추위 곰팡이 연구에 대한 방향을 제공할 것이고, 새로운 분석도구 개발에 응용될 수 있을 것이다.

A Checklist of Mushrooms of Phousabous National Protected Area (PNPA) of Lao PDR

  • Mushroom survey and collection were conducted in the Phousabous National Protected Area (PNPA), which is located at the Northeastern area 230km from the capital city of Lao-PDR, Vientiane, from April 2017 to September 2018. During this periods, mushroom specimens were collected from 8 different locations, and then they were identified and classified into 284 species, 142 genera, 56 families, 20 orders and 7 classes by morphological and molecular analyses. The mushrooms belongs to Ascomycota were classified into 10 species, 5 genera, 5 families, 4 orders and 3 classes, while those belongs to Basidiomycota were classified into 274 species, 137 genera, 51 families, 16 orders, and 4 classes, respectively. Among these mushrooms, the most species-rich families are Boletaceae (16.06%), Polyporaceae (10.12%), Russulaceae (9.94%), Amanitaceae (7.68%), Agaricaceae (4.54%), Marasmiaceae (4.36%), Cortinariaceae (3.66%), Entolomataceae (3.66%), Mycenaceae (2.79%), Xylariaceae (2.44%), Physalacriaceae (2.09%), Omphalotaceae (2.09%), Hydnangitaceae (2.09%), Lyophyllaceae (1.92%), Tricholomataceae (1.75%) and comprised 75.19% of the total specimens identified.

Sixteen New Records of Ascomycetes from Crop Field Soil in Korea

  • The present study reports 16 species of Ascomycota that were previously unknown in Korea, namely Acremonium cellulolyticus (KNU14-25), Acremonium zonatum (KNU14-15), Chaetomium madrasense (KNU14-9), Cladosporium silenes (KNU 14-18-1), Humicolopsis cephalosporioides (KNU15-3), Leptosphaerulina chartarum (KNU14-16), Paecilomyces marquandii (KNU14-8), Paecilomyces tenuis (KNU14-18-2), Paraphaeosphaeria sporulosa (KNU15-2), Penicillium rubidurum (KNU14-12), Pochonia suchlasporia (KNU15-6), Sporothrix inflata (KNU15-8), Thielavia hyrcaniae (KNU15-1), Thielavia terricola (KNU14-23-1), Xylogone sphaerospora (KNU15-7), and Zopfiella longicaudata (KNU15-5). These fungal species were isolated from soil samples collected from various regions of Korea and identified based on their morphological characteristics and rDNA internal transcribed spacer sequence data. Full descriptions and illustrations for each species are provided.

  • Seven unrecorded species in the phylum Ascomycota, Emericellopsis pallida (KNU16-167), Scedosporium aurantiacum (KNU16-190), Duddingtonia flagrans (KNU16-279), Bionectria rossmaniae (KNU16-309), Exophiala xenobiotica (KNU16-79), Pseudocercosporella fraxini (KNU16-102), and Stachybotrys sansevieriae (KNU16-141), were isolated in 2016 from field soils collected from various locations in Korea. All of the species were identified and described based on morphological characteristics and rDNA internal transcribed spacer sequence data. Morphological features of these fungi were examined on potato dextrose agar, oatmeal agar, malt extract agar, Czapek yeast extract agar, and yeast extract sucrose agar. Full descriptions and illustrations of their morphological characteristics are provided.

  • Three new records of Ascomycota species (Chaetomium acropullum, Phialemonium globosum, Phialemonium atrogriseum) from field soils in Korea are presented in this study. These newly discovered fungal isolates were isolated from field soils from various places across Gyeongnam, Korea in 2016. All the isolates were identified and described based on morphological characteristics, and rDNA internal transcribed spacer and ${\beta}$-tubulin gene sequence data. Morphological features of these fungal species were studied on different agar media: potato dextrose agar, oatmeal agar, malt extract agar, Czapek yeast extract agar, and yeast extract sucrose agar. Full description and illustrations of their morphological characters are provided. These fungal species have not officially been previously reported in Korea.
