Isolation and Characterization of Marine Bacterium Producing Arylsulfatase

  • A bacterial strain capable of hydrolyzing sulfate ester bonds in p-nitrophenyl sulfate and agar was isolated from the Southeast coast of Korea. The isolated strain (AS6330) is aerobic, Gram-negative, rod-shaped, and motile. Octadecanoic acid was the major cellular fatty acid in the isolate. An almost complete 16S rDNA sequence of the isolate was determined and the sequence similarity of the 16S rDNA with those of known Sphingomonas spp. was found to be at most $96.4\%$, implying that the isolate was a new Sphingomonas species. The organism was grown optimally at NaCl concentration of $1.5-3.5\%$. Optimum culture conditions were determined to be $30^{\circ}C$ and pH 7.0 for 48 h fermentation using a laboratory fermentor under constant culture conditions. Partially purified arylsulfatase through Q-Sepharose and phenyl­Sepharose chromatographies catalyzed hydrolysis of sulfate ester bonds in agar, and $97\%$ of sulfates in agar were removed after 4 h reaction at $45^{\circ}C$ and pH 7.0. The arylsulfatase from the isolated bacterium might be useful for the removal of sulfate groups in agar.

DYNAMICS OF GINGIVAL CREVICULAR FLUID VOLUME AND ENZYME ACTIVITIES AFTER APPLICATION OF ORTHODONTIC FORCE (교정력에 의한 치은열구 삼출액의 양 및 효소활성의 변동)

  • The aim of this investigation was to study the effect of orthodontic force on the flow of gingival crevicular fluid and activities of arylsulfatase and brta-glucuronidase in crevicular fluid. The material consisted of 12 persons between the ages of 13 years and 22 years and all were categorized Class I, 4-4 extraction cases Crevicular fluids were sampled from distal crevis of each canine before treatment (phase 1), after bracketing (phase 2), after application of force (phase 3) and after run out of orthodontic force (phase 4). Crevicular fluid flow did not show any significant changes during the period of treatment. The activities of arylsulfatase increased significantly after setting of orthodontic appliance without application of force, but did not show any significant difference after application of force. The activities of beta-glucuronidase increased significantly after application of orthodontic force and decreased with force deminished. These indicated that beta-glucuronidase was good indicator of bone remodelling resulted from initial orthodontic force.

Transcription level of the ars-1 promoter of Neurospora crassa (Neurospora crassa ars-1 프로모터의 발현율 조사)

  • The ars gene of the Neurospora crassa encodes arylsulfatase and is expressed under sulfur limitation. An ars-1 promoter(Pars) translationally-fused to a lacZ gene was transformed into the N. crassa RLM 35-35, a his-3 inl strain and integrated into the his-S locus by a single crossover homologous recombination. $\beta$-galactosidase specific activity was measured from mycelia grown in sulfur-limited Vogel's medium. Enzyme activity reached its maximum at 14 hour after the shift to derepressing condition. When activity from homokaryon generated by microconidiation was measured, it was 17% a higher than that from heterokaryon.

Activity of Some Hepatic Enzymes in Schistosomiasis and Concomitant Alteration of Arylsulfatase B

  • The levels of arylsulfatases A and B, $\alpha$-amylase, aspartate transcarbamylase, and $\gamma$-glutamyl transpeptidase were investigated during the infection of mice with schistosoma mansoni. This infection caused a significant (p<0.001) increase in the activity of hepatic arylsulfatase B (ASB), aspartate transcarbamylases and $\gamma$-glutamyl transpeptidase. A non-significant difference occurred for $\alpha$-amylase (p<0.3) and arylsulfatase A (p>0.5) when compared to the control. The specific activity of hepatic ASB was progressively increased with the progression of the Schistosoma-infection. Moreover, the kinetic studies of hepatic ASB in Schistosoma-infection showed that a slight decrease in the value of $K_m$ and about a 40% increase in $V_{max}$ when compared to the control. In addition, the pH optimum of hepatic ASB was altered from 6 to 7 as a result of schistosomiasis. These observations suggest that there are schistosomiasis-associated changes of the catalytic and kinetic properties of hepatic ASB.
