Effects of Various Light Sources on the Carotenoid and Glucosinolate Contents in Chinese Cabbage (Brassica rapa L. ssp. pekinensis) (다양한 광원이 배추 내 Carotenoid와 Glucosinolate 함량에 미치는 영향)

  • BACKGROUND: Chinese cabbage biosynthesizes various phytochemicals including carotenoids and glucosinolates. Environmental stress has a major effect on the growth and yields of vegetables, and can significantly affect nutritionally important phytochemicals. Phytochemicals of plants are influenced by light, temperature, carbon dioxide, and growing conditions. The aim of this study was to investigate the effect of various light sources on carotenoid and glucosinolate contents in Chinese cabbage. METHODS AND RESULTS: [Experiment I] Set the control (field control, FC) on the ground. Using acrylic sunlight, experiments were set up transparency box (field transparency, FT), red box (field red, FR) and blue box (field blue, FB). [Experiment II] Set the control (chamber control, CC) in the greenhouse. Using plant growth chamber with artificial light, experiments were set up LED red (chamber red, CR), LED blue (chamber blue, CB), LED mixed red+blue (chamber red+blue, CRB) and fluorescent (chamber fluorescent, CF). After plant growth, Chinese cabbage was harvested at 110 days after sowing (DAS). The status of plants growth (leaf length, width, fresh weight etc.) was immediately investigated. Carotenoid and GSL contents were analyzed by HPLC. [Experiment I] Results documented that the ranges of total carotenoid contents were 25.39 ~ 58.80 mg/kg dry wt for lutein, 0.84~ 4.22 mg/kg dry wt for zeaxanthin, and 3.85~18.71 mg/kg dry wt for ${\beta}$-carotene. Lutein was the highest for the content and the largest for the variation as well. [Experiment II] Results documented that the ranges of total carotenoid contents were 24.66~137.96 for lutein, 2.51~20.65 for zeaxanthin, and 8.40~49.80 mg/kg dry wt for ${\beta}$-carotene. The total carotenoid contents of CR (156.62) and CB (115.90) were 1.6~2.3 times larger than the other treatments, and ${\beta}$-carotene content was about twice as high as that of the other treatments on the CR (38.74 mg/kg dry wt.). [Experiment I] Total GSL content was the highest in FT (19.76) that was higher 1.7 times than the lowest treatment ($11.39{\mu}mol/g\;dry\;wt$.). [Experiment II] The total content of GSL was highest in CRB (4.19) and lowest in CF ($2.88{\mu}mol/g\;dry\;wt$.). In the CRB, total GSL contents ($4.19{\mu}mol/g\;dry\;wt$.) was the highest. CONCLUSION: Total and individual carotenoid and GSL contents in Chinese cabbage show significant differences under different light sources. Red and blue lights contribute to significant carotenoids expression and antioxidant activity for nutrition and health benefits. These results concluded that the introduction of varying lights affected the synthesis of important nutrient compounds in Chinese cabbage. It is predicted that the application of good light source enhances the accumulation of functional compounds.

Chemical Composition Characteristics of Dustfall in Nakdong River Area (낙동강 하류역 강하먼지의 화학적 조성 특성)

  • Dustfall samples were collected by the modified American dust jar (bulk type) at 5 sampling sites in the Nakdong river area from lune 2002 to May 2003. Nineteen chemical species (Al, Ca, Cd, Cr, Cu, Fe, K, Mg, Mn, Na, Ni, Pb, Si, V, Zn, $Cl^-$, $NO_3^-$, $SO_4^{2-}$, and $NH_4^+$) were analyzed via the combination of ICP/AES, AAS, IC and UV. The purposes of this study were to qualitatively evaluate the chemical composition of dustfalls by examining their regional and seasonal distribution patterns. Computation of the enrichment factor showed that well-defined anthropogenic sources, particularly in Pb were found in the order Gamjeondong (industrial area), Wondong, Silla University, Samrangiin and Mulgum. The seasonal mean of soil contribution showed its highest value (16.3%) during the winter with an annual mean of 11.2%. The concentration ratio of [$SO_4^{2-}/NO_3^-$] was found to be highest (5.12) during the winter, while the lowest ratio value (3.30) was seen during the all. fall, Also regional equivalent ratios of [$SO_4^{2-}/NO_3^-$] were found in the order: Silla University (6.78), Gamjeondong (4.98), Mulgum (3.95), Wondong (3.85), and Samrangjin (2.87). Seasonal distribution of water soluble components for total dustfall were found in the order: spring (71.6%), summer (61.2%), fall (49.2%) and winter (48.6%) with a mean ratio of 57.6%. Regional contribution of sea salts of water soluble ions were found in the order: Silla university (34.5%), Gamjeondong (28.3%), Wondong (17.3%), Samrangiin (17.2%) and Mulgum (13.8%), the total mean contribution rate was 22.1%. As for the chemical composition of dustftll on the lower Nakdong river, there is a decreased influence of sea salt and artificial anthropogenic sources and increased influence of soil particle inland. Also, the total amount of deposition on the lower Nakdong river has decreased, with the river's surface serving as a confounding factor in resuspending dusts.

Growth and Contents of Anthocyanins and Ascorbic Acid in Lettuce as Affected by Supplemental UV-A LED Irradiation with Different Light Quality and Photoperiod (상이한 광질 및 광주기 하에서 UV-A LED 부가 조사가 상추의 생장, 안토시아닌 및 아스코르빈산 함량에 미치는 영향)

  • The growth and contents of anthocyanins and ascorbic acid in lettuce(Lactuca sativa L., 'Jeokchima') as affected by supplemental UV-A LED irradiation under different light quality and photoperiod conditions were analyzed in this study. Five light qualities, namely B (blue LED), R (red LED), BUV (blue LED+UV-A LED), RUV (red LED+UV-A LED) and Control (white fluorescent lamps) with photoperiods of 12/12 hours (day/night), 16/8 hours, or 20/4 hours were provided to investigate the effects of light quality and photoperiod on the growth and accumulation of anthocyanins and ascorbic acid in lettuce leaves. As measured 28 days after transplanting, the number of leaves, leaf length, leaf width, leaf area, shoot fresh weight and dry weight of lettuce were significantly affected by light quality and photoperiod. The number of leaves, leaf length, leaf width, leaf area, shoot fresh weight and dry weight of lettuce grown under R treatment increased with increasing light period. By contrast, leaf development was inhibited, but chlorophyll content increased, under B treatment. Supplemental UV-A irradiation significantly decreased leaf length, leaf width, leaf area and shoot fresh weight. Anthocyanins in lettuce increased significantly with decreasing dark period under B treatment. A synergistic effect of supplemental UV-A LED irradiation on anthocyanins accumulation was found for lettuce leaves grown under R treatment but not B treatment. Ascorbic acid in lettuce was greatly affected by photoperiod. Ascorbic acid content at BUV and RUV treatments increased by 20-30% compared to without UV-A LED irradiation. From these results, it was concluded that growth and contents of anthocyanins and ascorbic acid in lettuce are significantly affected by supplemental UV-A LED irradiation. The results obtained in this study will be informative for efforts to improve the nutritional value of leafy vegetables grown in plant factories.

Leaf Shape Index, Growth, and Phytochemicals in Two Leaf Lettuce Cultivars Grown under Monochromatic Light-emitting Diodes (단색 발광다이오드에서 자란 축면상추 두 품종의 엽형, 생장 및 기능성 물질)

  • As an artificial light source, light-emitting diode (LED) with a short wavelength range can be used in closed-type plant production systems. Among various wavelength ranges in visible light, individual light spectra induce distinguishing influences on plant growth and development. In this study, we determined the effects of monochromatic LEDs on leaf shape index, growth and the accumulation of phytochemicals in a red leaf lettuce (Lactuca sativa L. 'Sunmang') and a green leaf lettuce (Lactuca sativa L. 'Grand rapid TBR'). Lettuce seedlings grown under normal growing conditions ($20^{\circ}C$, fluorescent lamp + high pressure sodium lamp, $130{\pm}5{\mu}mmol{\cdot}m^{-2}{\cdot}s^{-1}$, 12 hours photoperiod) for 18 days were transferred into incubators at $20^{\circ}C$ equipped with various monochromatic LEDs (blue LED, 456 nm; green LED, 518 nm; red LED, 654 nm; white LED, 456 nm + 558 nm) under the same light intensity and photoperiod ($130{\pm}7{\mu}mmol{\cdot}m^{-2}{\cdot}s^{-1}$, 12 hours photoperiod). Leaf length, leaf width, leaf area, fresh and dry weights of shoots and roots, shoot/root ratio, SPAD value, total phenolic concentration, antioxidant capacity, and the expression of a key gene involved in the biosynthesis of phenolic compounds, phenylalanine ammonia-lyase (PAL), were measured at 9 and 23 days after transplanting. The leaf shape indexes of both lettuce cultivars subjected to blue or white LEDs were similar with those of control during whole growth stage. However, red and green LEDs induced significantly higher leaf shape index than the other treatments. The green LED had a negative impact on the lettuce growth. Most of growth characteristics such as fresh and dry weights of shoots and leaf area were the highest in both cultivars subjected to red LED treatment. In case of red leaf lettuce plants, shoot fresh weight under red LED was 3.8 times higher than that under green LED at 23 days after transplanting. In contrast, the accumulation of chlorophyll, phenolics including antioxidants in lettuce plants showed an opposite trend compared with growth. SPAD value, total phenolic concentration, and antioxidant capacity of lettuce grown under blue LED were significantly higher than those under other LED treatments. In addition, PAL gene was remarkably activated by blue LED at 9 days after transplanting. Thus, this study suggested that the light quality using LEDs is a crucial factor for morphology, growth, and phytochemicals of two lettuce cultivars.

Effect of Silicate-Coated Rice Seed on Healthy Seedling Development and Bakanae Disease Reduction when Raising Rice in Seed Boxes (벼 상자육묘에서 규산코팅볍씨의 건묘육성과 벼키다리병 경감효과)

  • We investigated the effect of silicate coating of rice seeds on bakanae disease incidence and the quality of seedlings raised in seedling boxes and transplanted into pots. The silicate-coated rice seed (SCS) was prepared as follows. Naturally infested rice seeds not previously subjected to any fungicidal treatment were dressed with a mixture of 25% silicic acid at pH 11 and 300-mesh zeolite powder at a ratio of 50 g dry seed - 9 mL silicic acid - 25 g zeolite powder. The following nursery conditions were provided : Early sowing, dense seeding in a glass house with mulching overnight and no artificial heating, which were the ideal conditions for determining the effect on the seed. The nursery plants were evaluated for Gibberella. fujikuroi infection or to determine the recovery to normal growth of infected nursery plants in the Wagner pot. Seedlings emerged 2-3 days earlier for the SCS than they did for the non-SCS control, while damping-off and bakanae disease incidence were remarkably reduced. Specifically, bakanae disease incidence in the SCS was limited to only 7.8% for 80 days after sowing, as compared to 91.6% of the non-SCS control. For the 45-days-old SCS nursery seedlings, the fresh weight was increased by 11% and was two times heavier, with only mild damage compared to that observed for non-SCS. Even after transplanting, SCS treatment contributed to a lower incidence of further infections and possibly to recovery of the seedlings to normal growth as compared to that observed in symptomatic plants in the pot. The active pathogenic macro-conidia and micro-conidia were considerably lower in the soil, root, and seedling sheath base of the SCS. In particular, the underdeveloped macro-conidia with straight oblong shape without intact septum were isolated in the SCS ; this phenotype is likely to be at a comparative etiological disadvantage when compared to that of typical active macro-conidia, which are slightly sickle-shaped with 3-7 intact septa. A active intact conidia with high inoculum potential were rarely observed in the tissue of the seedlings treated only in the SCS. We propose that promising result was likely achieved via inhibition of the development of intact pathogenic conidia, in concert with the aerobic, acidic conditions induced by the physiochemical characteristics associated with the air porosity of zeolite, alkalinity of silicate and the seed husk as a carbon source. In addition, the resistance of the healthy plants to pathogenic conidia was also important factor.

Analysing the effect of impervious cover management techniques on the reduction of runoff and pollutant loads (불투수면 저감기법의 유출량 및 오염부하량 저감 효과 분석)

  • Impervious covers(IC) are artificial structures, such as driveways, sidewalks, building's roofs, and parking lots, through which water cannot infiltrate into the soil. IC is an environmental concern because the pavement materials seal the soil surface, decreasing rainwater infiltration and natural groundwater recharge, and consequently disturb the hydrological cycle in a watershed. Increase of IC in a watershed can cause more frequent flooding, higher flood peaks, groundwater drawdown, dry river, and decline of water quality and ecosystem health. There has been an increased public interest in the institutional adoption of LID(Low Impact Development) and GI(Green Infrastructure) techniques to address the adverse impact of IC. The objectives of this study were to construct the modeling site for a samll urban watershed with the Storm Water Management Model(SWMM), and to evaluate the effect of various LID techniques on the control of rainfall runoff processes and non-point pollutant load. The model was calibrated and validated using the field data collected during two flood events on July 17 and August 11, 2009, respectively, and applied to a complex area, where is consist of apartments, school, roads, park, etc. The LID techniques applied to the impervious area were decentralized rainwater management measures such as pervious cover and green roof. The results showed that the increase of perviousness land cover through LID applications decreases the runoff volume and pollutants loading during flood events. In particular, applications of pervious pavement for parking lots and sidewalk, green roof, and their combinations reduced the total volume of runoff by 15~61 % and non-point pollutant loads by TSS 22~72 %, BOD 23~71 %, COD 22~71 %, TN 15~79 %, TP 9~64 % in the study site.

Distribution and Pollution Assessment of Trace Metals in Core Sediments from the Artificial Lake Shihwa, Korea (시화호 코어 퇴적물 내 미량금속 분포 특성 및 오염 평가)

  • Metal concentrations in creek water, sewer outlets and core sediments were analyzed to identify the potential origin of metal pollution and to evaluate the extent of metal pollution and potential toxicity of Lake Shihwa. Mean concentrations for dissolved metals in creek water and sewer outlets were 1.6~136 times higher than those in the surface seawater of Lake Shihwa. Metal concentrations in creek water from an industrial region were also higher than those from municipal and agricultural regions, indicating that the potential source of metal pollution in the study area might be mainly due to industrial activities. The vertical profiles of metals in core sediments showed an increasing trend toward the upper sediments. Extremely higher concentrations of metals were observed in the vicinity of Banweol industrial complex. The results of a geo-accumulation index indicated that Cu, Zn and Cd were highly polluted. By comparing the sediment quality guidelines such as TEL and PEL, six metals such as Cr, Ni, Cu, Zn, Cd and Pb levels in core sediments nearby industrial complex exceeded the PEL value. Mean PEL quotient (mPELQ) was used to integrate the estimate of potential toxicity for measured metals in the present study. Mean PELQs in core sediments from Lake Shihwa ranged from 0.2~2.3, indicating that benthic organisms nearby the industrial complex may have been adversely affected.

Effect of the Level of Carbohydrates on Bio-hydrogenation and CLA Production by Rumen Bacteria When Incubated with Soybean Oil or Flaxseed Oil In vitro (Soybean Oil 및 Flaxseed Oil 첨가 배양시 탄수화물 첨가수준에 의한 반추미생물의 Bio-hydrogenation과 CLA 생성에 미치는 효과)

  • An in vitro study was conducted to examine the effect of addition level of carbohydrates on fermentation characteristics, and bio-hydrogenation of unsaturated fatty acids by mixed rumen bacteria when incubated with soybean oil or flaxseed oil. Four levels(0%, 0.3%, 0.6% and 0.9%, w/v) of the mixed carbohydrates(glucose, cellobiose, soluble starch, 1:1:1, in weight basis) and oil sources(soybean or flaxseed oil, 60mg in 150ml culture solution) were added to the mixed solution of strained rumen fluid with artificial saliva(1:4, v/v), and incubated anaerobically for 12 hours at 39℃. pH and ammonia-N concentration were lower by increasing the substrate levels at all incubation periods(P<0.05~P<0.001). The propionate proportion increased(P<0.001), but acetic acid and butyric acid decreased(P<0.001) with the substrate level at 6 and 12 h incubations. Oil sources did not influence the proportions of individual VFA. At the end of incubation, the proportions of C18:0(P<0.01), C18:1(P<0.001) and trans-11C-18:1(P<0.01) were reduced but those of C18:2(P<0.001) and C18:3(P<0.01) were enhanced by the addition of flaxseed oil compared to addition of soybean oil. The proportions of C18:0 and total CLA were reduced(P<0.01) but those of trans-11-C18: (P<0.05) and C18:2(P<0.01) were increased with the substrate level when incubated with soybean oil or flaxseed oil. There were interactions(P<0.05) in the proportions of C18:1, C18:2 and C18:3(P<0.01) between oil source and substrate level. The proportions of cis-9, trans-11-CLA and trans-10, cis-12-CLA tended to reduce with substrate level, although there was no significant difference between treatments.

Effect of Activated Carbon, Orpar or Zeolite on Leaching Loss of Fenitrothion, Triadimefon and Diniconazole in Model Green of Golf Course (골프장 모형그린에서 활성탄, Orpar또는 Zeolite의 처리가 Fenitrothion, Triadimefon, Diniconazole의 용탈에 미치는 영향)

  • Cheju island depends on a hydrogeologically vulnerable aquifer system as its principle source of drinking water. Most of golf courses are located in the area which is important for the ground water recharge, and pesticides are applied to golf courses often at relatively high rates. Therefore, turf pesticides in golf course should be applied without adversely impacting ground water. In this experiment, downward movement of pesticides was monitored in model greens of golf course, where different adsorbents were layered in 3-cm thickness at 35-cm depth, and effect of the adsorption layer on the leaching loss of pesticides was investigated. Major leachings were observed in the periods of heavy rain and very limited leaching was observed under artificial irrigation. Fenitrothion and triadimefon, which have relatively short persistence and high adsorption coefficient, were found in the leachate in low concentrations only at the first rainfall event, around 20 days after the pesticide application. However, diniconazole, which has a relatively long half-life (97 days), was detected in the leachate during the whole period of experiment and concentration was much higher than those of the other pesticides. Maximum leachate concentrations were 1.9, 10.3, and 84.5 ${\mu}l^{-1}$ for fenitrothion, triadimefon, and diniconazole, respectively. Therefore, in golf course green which allows rapid water percolation and has extremely low adsorption capacity, persistence in soil could be more important factor in determination of leaching potential of pesticides. Total quantity of pesticides leached from the model green was <0.2% for fenitrothion and triadimefon and 1.8% for diniconazole. Adsorption layers significantly reduced pesticide leaching, and active carbon and Orpar were more effective than zeolite. In the model green having adsorption layer of active carbon or Orpar, leaching loss of pesticides was reduced below 0.01% of the initial application.

Effect of Cosmetics Contained Isotonic Water Mimicked Body Fluid on Cell Activities and Skin (생체 모사수 화장품이 세포 활성과 피부에 미치는 효과)

  • Body fluid has been studied for diverse fields like Ringer's solutions, artificial joint fluids, cell growth culture media because it plays a crucial role in controlling body temperature and acts as a solvent for diverse metabolite processes in the body and delivery media of mineral, energy source, hormone, signal and drug from and to cell via blood or lymphatic vessel by osmotic pressure or active uptake. Stratum corneum containing extracellular lipids and NMF (natural moisturizing factor) absorbs atmospheric water residing outside of cells and utilize it to hydrate inside of their own. This process is related to skin barrier function. In this study, we conducted the cell viability test with Cell Bio Fluid $Sync^{TM}$, which mimicks body fluids including amino acids, peptides, and monosaccharides to strengthen skin barrier, and the clinical skin improvement test with cosmetics containing Cell Bio Fluid $Sync^{TM}$. In the cell viability test, HaCaT cell was treated with PBS for 3 hours, followed by the treatment of a cell culture medium (DMEM) and isotonic solution (PBS) and Cell Bio Fluid $Sync^{TM}$ for 3 hours each. Then, MTT assay and image analysis were conducted. In the clinical skin improvement test, twenty-one healthy women participated. Participants applied cosmetics containing Cell Bio Fluid $Sync^{TM}$ on their face for a week and evaluated the skin hydration, skin roughness, brightness and evenness. All measurements were conducted after they washed off their face and took a rest under the constant temperature ($22{\pm}2^{\circ}C$) and constant humidity conditions ($50{\pm}5%$) for 20 minutes. All the data were analyzed by SPSS (version 21) software program. Results showed that Cell Bio Fluid $Sync^{TM}$ improved both the cell viability and in vivo skin conditions such as skin hydration, roughness, brightness and evenness.
