Complete genome sequence of Marinobacter salarius HL2708#2 isolated from a lava sea water environment on Jeju Island (제주용암 해수 환경에서 분리한 Marinobacter salarius HL2708#2의 유전체 해독)

  • During screening of microbes for compounds having cosmetic benefits, we isolated Marinobacter salarius HL2708#2 from lava seawater on Jeju Island, Republic of Korea. The complete genome sequence was determined. Strain HL27080#2 features a circular chromosome of 4,304,603 bp with 57.21% G+C content and a 244,163 bp plasmid with 53.14% G+C. There were 4,180 protein coding sequences identified, along with 49 transfer RNA and 18 ribosomal RNA noncoding genes. The genome harbored genes for the utilization of alcohol, maltose/starch, and monosaccharide as sole carbon sources. Genes responsible for halophilic characteristics and heavy metal resistance could be annotated, as well as aromatic and alkane hydrocarbons. Contrary to the prior report that M. salarius is negative for nitrate and nitrite reduction, nitrate/nitrite reductase along with nitrate/nitrate transporters and nitronate monooxygenase were evident, suggesting that strain HL2708#2 may be able to denitrify extracellular nitroalkenes to ammonia.

Ignition Characteristics of Petroleum-based and Bio Aviation Fuel According to the Change of Temperature and Pressure (온도와 압력의 변화에 따른 석유계 및 바이오항공유의 점화특성 분석)

  • In this study, the ignition characteristics of petroleum-based aviation fuel (Jet A-1), bio aviation fuel (Bio-6308), and blended aviation fuel (50:50, v:v) were analyzed in accordance with change of temperature and pressure. The ignition delay time of each aviation fuel was measured by combustion research unit (CRU) and the compositions of the fuels were analyzed by GC/MS and GC/FID for qualitative and quantitative results. From the results, it was confirmed that the ignition delay times of all aviation fuels were shortened with increasing temperature and pressure. In particular, the effect of temperature was larger than the effect of pressure. Also, the ignition delay time of Jet A-1 was the longest at all measurement conditions, and it was judged that this result is because of the structurally stable characteristics of the benzyl radical generated during the oxidation reaction of the aromatic compound (about 22.48%) in Jet A-1. Also, it was confirmed that Jet A-1 had no section where the degree of shortening of ignition delay time was decreased by increasing temperature, which was because the benzyl radical inhibits the response that can affect the negative temperature coefficient (NTC). The ignition characteristics of blended aviation fuel (50:50, v:v) showed a similar tendency to those of Jet A-1, rather than to those of Bio-6308, so that the blended aviation fuel (50:50, v:v) can be applied to the existing system without any change.

  • Through the excavated excellent articles of the Tang era, we have considered the building techniques and styles of a square pavilion, an octagonal pavilion, a miniature hill, a pond, etc., could read building techniques of the scenic structures on ancient literatures including records and additionally about their poetical life at that time and inquired into the arrangement, structure, building techniques, etc. of a miniature hill, a pond, a pavilion, a flowerplant, etc., on Nakyangmyungwonki. Thus, under the research on the private landscape architecture, especially the nobility gardening, around excavated excellent articles and literatures, 'A miniature hill(假山) and a pond for landscaping views formed the center of a garden, and additionally a pavilion was built and flowerplants were set. The miniature hill of laying stones and having a carven, steeping, stratifying, looking like dyed green and birds' singing among hills and eating water on the lakeshore${\ldots}$' was expressed. The pond of digging in the ground and conducting water had its water system developed. There were several kinds of pavilions such as 廳, 堂, 館, 亭, 臺, 樓, 閣, 榭, etc. As examples of landscaping plants, there were a bamboo, a lotus flower, a peony, aromatic trees a pine, a korean spindle tree, a big cone pine, an empress tree, a wild walnut, a peach, a plum, a Japanese apricot tree, an apricot tree, a chrysanthemum, arrowroot vines, etc. Thus, the garden of the Tang era, abundant, diverse and excellent, enjoyed the prime of the period of prosperity. Due to cultural exchange, it is supposed that the period of united Shilla of the same age would meet with the period of prosperity in the developmental history of Korea landscape, based on the nobility garden system '4 different dwelling-houses every season on a record "四節游宅"'.

NMR Spectroscopy and Mass Spectrometry of Benzyl Alcohol Galactoside synthesized using β-Galactosidase (베타-갈락토시데이즈를 이용하여 합성된 Benzyl Alcohol Galactoside의 NMR Spectroscopy 및 Mass spectrometry)

  • To characterize the molecular structure of BzO-gal synthesized using Escherichia coli ${\beta}$-gal, NMR ($^1H$- and $^{13}C$-) spectroscopy and mass spectrometry of BzO-gal were conducted. $^1H$ NMR spectrum of BzO-gal showed multiple peaks corresponding to the galactosyl group, which is an evidence of galactosylation on BzOH. Five proton peaks around the aromatic region at ${\delta}_H$ 7.43 ~ 7.24 ppm and 2 peaks from ${\delta}_H$ 4.93 and 4.67 ppm were evidence of the presence of the benzyl group. Seven proton peaks at ${\delta}_H$ 4.32 ~ 3.46 ppm showed the presence of a monosaccharide and were indicative of galactosylation on BzOH. $^{13}C$ NMR spectrum also revealed the presence of 11 carbons suggestive of BzO-gal. The mass value (sodium adduct ion of BzO-gal, m/z = 293.0994) from mass spectrometry analysis of BzO-gal, and $^1H$ and $^{13}C$ NMR spectral data were in good agreement with the expecting structure of BzO-gal. We are expecting that through future study it will eventually be able to develop a new additive of low cytotoxicity.

Soil Carbon Storage in Upland Soils by Biochar Application in East Asia: Review and Data Analysis (바이오차를 이용한 밭 토양 탄소 저장: 동아시아 지역 연구 리뷰 및 데이터 분석)

  • BACKGROUND: Biochar is a solid material converted from agricultural biomass such as crop residues and pruning branch through pyrolysis under limited oxygen supply. Biochar consists of non-degradable carbon (C) double bonds and aromatic ring that are not readily broken down by microbial degradation in the soils. Due to the recalcitrancy of C in biochar, biochar application to the soils is of help in enhancing soil carbon sequestration in arable lands that might be a strategy of agricultural sector to mitigate climate change. METHODS AND RESULTS: Data were collected from studies on the effect of biochar application on soil C content conducted in East Asian countries including China, Japan and Korea under different experimental conditions (incubation, column, pot, and field). The magnitude of soil C storage was positively correlated (p < 0.001) with biochar application rate under field conditions, reflecting accumulation of recalcitrant black C in the biochar. However, The changes in soil C contents per C input from biochar (% per t/ha) were 6.80 in field condition, and 12.58 in laboratory condition. The magnitude of increment of soil C was lower in field than in laboratory conditions due to potential loss of C through weathering of biochar under field conditions. Biochar production condition also affected soil C increment; more C increment was found with biochar produced at a high temperature (over 450℃). CONCLUSION: This review suggests that biochar application is a potential measures of C sequestration in agricultural soils. However, as the increment of soil C biochar was affected by biochar types, further studies are necessary to find better biochar types for enhanced soil C storage.

Breeding of 'Greenbear' for New Cultivar of Gomchwi with Disease Resistant and High Yield (내병 다수성 곰취 신품종 '그린베어' 육성)

  • 'Gondalbi' (Ligularia stenocephala (Maxim.) Matsum. & Koidz.) is the most cultivated Gomchwi species because of higher yield and low in aromatic flavor and bitter taste, But 'Gondalbi' is susceptible to powdery mildew disease and leaf shriveling after harvest in Summer. To improve powdery mildew disease resistance and post-harvest leaf shriveling problem in 'Gondalbi', 'Handeari-gomchwi' resistant to powdery mildew disease and having higher yield potential used as a paternal trait donor. Powdery mildew disease resistance and post-harvest leaf shriveling as well as agronomic performance of a new variety, 'Greenbear' were tested under field and green house conditions in Pyungchang, Korea from 2007 to 2016. Expression of both maternal and paternal characteristics in 'Greenbear' including purple colored petiole ears, glossy leaf and paternal, petiole trichome, absent at the back of a leaf were confirmed. Plant size and flowering time of 'Greenbear' were similar to check line, 'Gommany', while leaf number per plant and yield were higher in 'Greenbear'. 'Greenbear' has thinner leaves (0.66mm) compared to 'Gommany (0.69 mm)', and hardness appeared slightly higher in 'Greenbear(25.1 kg/cm2)'. The resistance to powdery mildew disease of the 'Greenbear' variety was slightly lower than that of the 'Gommany' variety, but somewhat showed high resistance.

Immunostimulatory Activity of Agarwood through Activation of MAPK Signaling Pathway in RAW 264.7 Murine Macrophages (RAW 264.7 대식세포에서 MAPKs 신호 전달 경로의 활성화를 통한 침향의 면역 자극 활성)

  • Deterioration of the immune function weakens the body's resistance to various infections, leading to a series of diseases. Immunomodulatory biomaterials have been used to reduce the side effects of immunosuppressants or to enhance immunity. Agarwood is the aromatic resinous portion of Aquilaria trees that has been traditionally used as a medicinal herb for the treatment of various diseases. Although previous studies have shown that agarwood can improve the body's immunity, evidence for this claim is still lacking. In this study, the immune-enhancing effects of the agarwood methanol extracts of Aquilaria malaccensis Lamk were evaluated in a RAW 264.7 macrophage model. Based on the results, the agarwood extracts markedly enhanced phagocytosis in the absence of cytotoxicity. The agarwood extract-treated RAW 264.7 cells exhibited the typical morphology of activated macrophages, which are spindle-shaped with elongated filopodia. Agarwood extract also significantly increased the production of nitric oxide (NO), which is associated with the increased expression of inducible NO synthase. Moreover, the secretion and expression levels of cytokines, such as tumor necrosis factor-α and interleukin (IL)-1β and IL-6, were increased by agarwood treatment. Notably, these are also associated with a mitogen-activated protein kinase signaling pathway. Taken together, our findings provide scientific evidence that agarwood has potential immune-enhancing effects in vitro.

Mechanisms of Resorcinol Antagonism of Benzo[a]pyrene-Induced Damage to Human Keratinocytes

  • Benzo[a]pyrene (B[a]P) is a polycyclic aromatic hydrocarbon and ubiquitous environmental toxin with known harmful effects to human health. Abnormal phenotypes of keratinocytes are closely associated with their exposure to B[a]P. Resorcinol is a component of argan oil with reported anticancer activities, but its mechanism of action and potential effect on B[a]P damage to the skin is unknown. In this study, we investigated the effects of resorcinol on B[a]P-induced abnormal keratinocyte biology and its mechanisms of action in human epidermal keratinocyte cell line HaCaT. Resorcinol suppressed aryl hydrocarbon receptor (AhR) activity as evidenced by the inhibition of B[a]P-induced xenobiotic response element (XRE)-reporter activation and cytochrome P450 1A1 (CYP1A1) expression. In addition, resorcinol attenuated B[a]P-induced nuclear translocation of AhR, and production of ROS and pro-inflammatory cytokines. We also found that resorcinol increased nuclear factor (erythroid-derived 2)-like 2 (Nrf2) activity. Antioxidant response element (ARE)-reporter activity and expression of ARE-dependent genes NAD(P)H dehydrogenase [quinone] 1 (NQO1), heme oxygenase-1 (HO-1) were increased by resorcinol. Consistently, resorcinol treatment induced nuclear localization of Nrf2 as seen by Western analysis. Knockdown of Nrf2 attenuated the resorcinol effects on ARE signaling, but knockdown of AhR did not affect resorcinol activation of Nrf2. This suggests that activation of antioxidant activity by resorcinol is not mediated by AhR. These results indicate that resorcinol is protective against effects of B[a]P exposure. The mechanism of action of resorcinol is inhibition of AhR and activation of Nrf2-mediated antioxidant signaling. Our findings suggest that resorcinol may have potential as a protective agent against B[a]P-containing pollutants.

Quantitative analysis of glycerol concentration in red wine using Fourier transform infrared spectroscopy and chemometrics analysis

  • Glycerol is a non-volatile compound with no aromatic properties that contributes significantly to the quality of wine by providing sweetness and richness of taste. In addition, it is also the third most significant byproduct of alcoholic fermentation in terms of quantity after ethanol and carbon dioxide. In this study, Fourier transform infrared (FT-IR) spectroscopy was employed as a fast non-destructive method in conjugation with multivariate regression analysis to build a model for the quantitative analysis of glycerol concentration in wine samples. The samples were prepared by using three varieties of red wine samples (i.e., Shiraz, Merlot, and Barbaresco) that were adulterated with glycerol in concentration ranges from 0.1 to 15% (v·v-1), and subjected to analysis together with pure wine samples. A net analyte signal (NAS)-based methodology, called hybrid linear analysis in the literature (HLA/GO), was applied for predicting glycerol concentrations in the collected FT-IR spectral data. Calibration and validation sets were designed to evaluate the performance of the multivariate method. The obtained results exhibited a high coefficient of determination (R2) of 0.987 and a low root mean square error (RMSE) of 0.563% for the calibration set, and a R2 of 0.984 and a RMSE of 0.626% for the validation set. Further, the model was validated in terms of sensitivity, selectivity, and limits of detection and quantification, and the results confirmed that this model can be used in most applications, as well as for quality assurance.

Contamination Characteristics of Hazardous Air Pollutants in Particulate Matter in the Atmosphere of Ulsan, Korea (울산시 미세먼지의 유해대기오염물질 오염 특성)

  • Recently, long-range atmospheric transport (LRAT) from China is regarded as a major reason for elevated levels of particulate matter (PM) in Korea. However, local emissions also play an important role in PM pollution, especially in large-scale industrial cities. In this study, PM samples were collected at suburban, residential, and industrial sites in Ulsan, Korea. Polycyclic aromatic hydrocarbons (PAHs) and heavy metals were analyzed, and a potential human health risk assessment was conducted. The concentrations of PAHs and heavy metals in total suspended particles (TSP) increased during high $PM_{10}$ episodes, and backward trajectory analysis verified the influence of LRAT from China during the high episodes. Furthermore, the concentrations of PAHs and heavy metals in $PM_{2.5}$ and $PM_{10}$ at the industrial site were higher than those at the residential site. The risk assessment of PAHs and heavy metals in $PM_{2.5}$ suggested no significant health effects. The highest levels of PAHs were measured in the particle size of $0.32{\sim}0.56{\mu}m$ at the residential site, and those of heavy metals were detected in the particle size of 1.8~5.6 and $>18{\mu}m$, reflecting different major emissions sources for both groups. On the basis of this preliminary study, we are planning long-term monitoring and modeling studies to quantitatively evaluate the influence of industrial activities on the PM pollution in Ulsan.
