Development of new food protein through chemical modification of rice bran proteins

  • Protein concentrate was produced and succinylated from rice bran to assess and improve its functional properties for the purpose of expanding the uses of rice bran proteins. The most effective solvent for the extraction of rice bran proteins was 20% aqueous ethanol at pH 9. The protein content of rice bran protein concentrate produced was 70.0% and the total protein yield was 64.3%. The extent of succinylation of free amino groups in the modified products was 72.8%. Though the modified protein products showed good functional properties including solubility, emulsion properties, and oil absorption capacity, it did not form gel. Succinylation improved solubility and emulsion and gelling properties. These improvements in functionality will enhance the value of rice bran proteins, thus enabling them to be more competitive with other food proteins.

  • $PbTiO_3$ was prepared by addition of lead-tltanium mixed aqueous solution to an ethanol solution of oxalic acid. The influence of various ripening factors was invesigated. On the basis of the results in the mono-component system we studied the foramtion of $PbTiO_3$ phase with various ripening conditions. The $PbTiO_3$ phase formation was accelerated with the ripening time temperature and ultrasonic irradiation.

Synthesis of Silver Nanoplates with Fibronectin Nanofibril Template and Their SERS Applications

  • In this work, a novel strategy is provided to prepare silver nanoplates by a fibronectin (Fn) nanofibril template. First, Fn molecules were controlled to assemble into amyloid-like nanofibrils in highly concentrated ethanol aqueous solution. The resultant nanofibrils could serve as a soft template to direct the formation of silver nanoplates. It is worth noting that the silver nanoplates are excellent surface-enhanced Raman scattering (SERS) substrate with 4-aminothiophenol (4-ATP) molecule as a test probe. This high active SERS substrate can also be used to detect drug molecule, 2-thiouracil with high sensitivity.

물푸레나무 수피의 Oleuropein과 Ligstroside (Oleuropein and Ligstroside from the bark of Fraxinus rhynchophylla)

  • The bark of Fraxinus rhynchophylla, one of ash trees grown in domestic, was collected, extracted with acetone-$H_2O$(7:3, v/v) and freeze dried to give 50g of ethylacetate soluble powder. A portion of the freeze dried powder was chromatographed on a Sephadex LH-20 and a TSK 40F column using a series of aqueous methanol and ethanol-hexane mixture as eluents. The extractives contained a large amount of coumarin derivatives in addition to a small amount of ester compounds such as ligstroside and oleuropein which have not been isolated from this tree species yet. Some spectrometric analyses such as NMR and FAB-MS including TLC were performed to identify the structures of the isolated ester compounds.

Antimicrobial Activity of Korean Medicinal Plants and Herbal Formulations

  • In this study, we screened twenty four extracts of eight medicinal plants and three extracts of the commercial product for their antimicrobial and antioxidant properties. Ethanol and ethyl acetate extracts were found active where as aqueous extracts were little or no active. Extracts of Sophora flavescens, Salvia miltlorrhiza and Glycyrrhiza uralensis showed strong activity againsttested organisms and could be the potential antimicrobial agent. The increase of antimicrobial and antioxidant activities of formulations might be due to synergic effect. The results also indicated that the activity of bamboo salt and herbal products can be enhanced by making appropriate formulations.

  • The microcapsule for thermochromism is based on the polymerization reaction between epoxy resin and amine curing agent. The preparation process of microcapsule is based on dissolving or dispersing a hydrophobic core materials[one-dye-black(OBD), bis-phenol A(BPA), cetyl alcohol] in an aqueous solution of gelatin, epoxy resin and isophorondiamine(IPDA) ; the gelatin and IPDA used as a dispersion stabilizer and an hardening agent, respectively. The structures of epoxy resin and microcapsule materials have been analyzed by FT-IR and UV/Vis spectra. The mean diameter and size distribution of microcapsule are 1.46~1.75${\mu}{\textrm}{m}$ and 1.42, respectively. The DSC thermograms of microcapsules indicated 2 kinds of endothermic peaks at 47 and 322$^{\circ}C$. This is possibly corresponding to the melting peak of core material and wall meterial. These microcapsules are applied to the fabric by printing. Complex finished fabric showed a good wear resistance on rubbing test and the print pattern to the cotton fabric showed a reversible thermochromism ; ${\mu}_{max}$ are 580 nm below 4$0^{\circ}C$ and 276.5nm above 4$0^{\circ}C$ in ethanol/water(2/8), respectively.

Hydrophobic and Ionic Interactions in the Ester Hydrolysis by Imidazole-Containing Polymers

  • N-Methacryloyl-L-histidine and N-methacryloyl-L-histidine methyl ester were synthesized and polymerized to obtain polymeric catalysts with different functions. In the presence of each of these polymers the solvolytic reactions of p-nitrophenyl acetate (PNPA), 3-nitro-4-acetoxybenzoic acid(NABA), 3-acetoxy-N-trimethylanilinium iodide(ANTI) and 3-nitro-4-decanoyloxybenzoic acid(NDBA) were performed in 20% aqueous ethanol. For the purpose of comparison the low molecular weight analogs(LMWA's), L-histidine, L-histidine methyl ester and N-acetyl-L-histidine were also subjected to catalyze the solvolyses of above substrates. In the solvolysis of PNPA the polymeric catalysts exhibited lower activities than the LMWA's. However the ionic substrates, NABA and ANTI were solvolyzed at anomalous rate by polymeric catalyst, indicating that electrostatic effects are operative in the catalysis by polymers. Furthermore in the solvolysis of hydrophobic monomer NDBA, polymeric catalysts exhibited highly enhanced activities compared with the LMWA's implying that hydrophobic interaction can be the most important contribution to the high catalytic activity of imidazole-containing polymers.

  • A general spectroscopic method is described for studies on the complex formation between metal ions and ligands, and is applied to $Cu^{2+}$ and $Ca^{2+}$binding to glycosaminoglycans. The order of binding constants for both ions is heparin >dermatan sulfate >chondroitin sulfate. The electrostatic forces are shown to be the predominant factor in the interaction. The 2- to 3-fold higher affinity for $Cu^{2+}$ than for $Ca^{2+}$ is obtained for heparin and dermatan sulfate, but little difference for chondroitin sulfate. These results are explained as chelation of both carboxyl and sulfate groups to $Cu^{2+}$ in former cases. The difference of binding constants among glycosaminoglycans is related to proposed various biological functions of the biopolymers.

Solvolyses of N-Methyl-N-phenylcarbamoyl Chlorides with Electron Acceptor Substituents in A Queous Binary Mixtures

  • Solvolyses of N-methyl-N-phenylcarbamoyl chlorides in aqueous binary mixtures of acetone, ethanol, methanol and in water, D2O, and 50% D2O-CH3OD are investigated at 25.0 $^{\circ}C.$ The Grunwald-Winstein plots of first-ord er rate constants for N-methyl-N-phenylcarbamoyl chlorides with YCl (based on 2-adamantyl chloride) show a dispersion phenomenon. The ring parameter (I) has been shown to give considerable improvement when it is added as an hI term to the original Grunwald-Winstein and extended Grunwald-Winstein correlations for the solvolyses of N-methyl-N-phenylcarbamoyl chlorides. This study has shown that the magnitude of l, m and h values associated with a change of solvent composition is able to predict the dissociative SN2 transition state. The kinetic solvent isotope effects determined in deuterated water are consistent with the proposed mechanism of the general base catalyzed and/or a dissociative SN2 mechanism channel for N-methyl-N-phenylcarbamoyl chlorides solvolyses.

Nucleophilic Substitution Reaction of α-Methoxy-α-(trifluoromethyl)phenylacetyl Chloride in Alcohol-Water Mixtures

  • Solvolyses of α-methoxy-α-(trifluoromethyl)phenylacetyl chloride in H₂O, D₂O, CH₃OD, 50% D₂O-CH₃OD, and in aqueous binary mixtures of acetone, dioxane, ethanol and methanol are investigated at 25.0℃. The Grunwald-Winstein plots of first-order rate constants for α-methoxy- α-(trifluoromethyl)phenylacetyl chloride with $Y_{Cl}$ show a dispersion phenomenon. Solvent nucleophilicity N has been shown to give considerable im-provement when it is added as an 1N term to the original Grunwald-Winstein for the solvolyses of α-methoxy- α-(trifluoromethyl)phenylacetyl chloride. The dispersions in the Grunwald-Winstein correlations in the present studies are caused by solvent nucleophilicity. The magnitude of l and m values associated with a change of solvent composition predicts the associative $S_N2$ transition state. The kinetic solvent isotope effects determined in deuterated water and methanol are consistent with the proposed mechanism of the general base catalyzed associative $S_N2$ or $S_AN$ mechanism for the of α-methoxy- α-(trifluoromethyl)phenylacetyl chloride.
