Determination of Chromium(VI) and Copper(II) in Organic Solvent - Solution by Liquid Core Optical Fiber Spectrophotometry

  • A new hollow fiber filled with the lower refractive index liquid as core constructs a liquid core optical fiber(LCOF). The LCOF have been used as colorimetric cell to determine elements Cr and Cu in the presence of 70%- 50% ethanol or 50% dioxane aqueous solution, based on colored complex of Chromium(VI) with Diphenycarbazid(DPC) and copper with Chromaurd S(CAS) respectively. The sensitivity (1/ng/ml) of calibration curve of Cr and Cu are 0.052, 0.017 over the range 0 - 25, 0 - 24(ng/ml) respectively. The Cr in the animal gum and Nation Standard of China are determined with recoveries of 94 - 102%.

Fluorescent Silk Fibroin Nanoparticles Prepared Using a Reverse Microemulsion

  • Color dye-doped silk fibroin nanoparticles were successfully fabricated using a microemulsion method. An aqueous silk fibroin solution was prepared by dissolving cocoons (Bombyx mori) in a concentrated lithium bromide solution followed by dialysis. A color dye solution was also mixed with the aqueous silk fibroin solution. The surfactants used for the microemulsion were then removed by methanol and ethanol, yielding color dye-doped silk fibroin nanoparticles, approximately 167 nm in diameter. The secondary structure of the nanoparticles showed a $\beta$-sheet conformation, as characterized by Fourier transform infrared spectroscopy. The morphology of the nanoparticles was determined by field emission scanning electron microscopy, transmission electron microscopy and atomic force microscopy, and their size and size distribution were measured by dynamic light scattering. The color dye-doped silk fibroin nanoparticles were examined by confocal laser scanning microscopy.

  • The pH titration curves of ampicillin, amoxicillin and cephalexin in aqueous acetone, ethanol and N, N'-dimethylformamide by use of 0.02N-KOH solution showed that 80% acetone was the most effective solvent for alkali titration of them. The pH jumps of samples in 80% acetone were sharp enough to determine the contents of them by use in 80% acetone (9, 4) was coincided with each equivalent point of ampicillin, amoxicillin and cephalexin, but those of phenolphthalein, thymolphthalein, thymol blue and bgrom cresol purple were not. The color change of brom thymol blue at equivalent points was very clear except the case of amoxicillin that the determination of smaples in 80% acetone with 0.02 N-KOH solution by the aid of brom thymol blue could be available.

  • Solutions of $Cd^{2+}$, $Co^{2+}$ and $Ni^{2+}$ were mixed with the solutions of hydroxycarboxylic acids such as salicylic, lactic and mandelic acids in the presence of cation exchange resin at room temperature. The distribution ratios of the metal ions between resin and solution were measured, using radioactive metal ions as tracer. From the observed variation of the distribution ratios with the acid anion concentrations, it was concluded that $Cd^{2+}$, $Co^{2+}$ and $Ni^{2+}$ formed the one-to-one complexes with salicylate, lactate and mandelate ions in aqueous, 20 % ethanol-water and 20 % acetone-water solutions. The results of the present study indicated that the relative stabilities of the metal-acid complexes in solution increased in the order: $Cd^{2+}$ <$Co^{2+}$ <$Ni^{2+}$ complexes. Salicylate

  • Rate of solvolysis of dansyl chloride in aqueous binary mixtures of acetone, methanol and ethanol are reported. Kinetic solvent isotope effects in methanol and product selectivities in alcohol-water mixtures are also reported. Kinetic data are interpreted with the Grunwald-Winstein and Kivinen equations. The value of $k_{CH3OH}/k_{CH3OD}=1.76$ suggests that a general have catalyzed and/or an $S_AN$ pathway is operative in methanol, a less polar solvent. Rate-rate profiles for solvolysis of dansyl chloride in the aqueous binary media indicate a change in reaction channel from $S_AN$ (in less polar media) to $S_N2$ (in more polar media) mechanism.

Physiological activity of the extract from Dolwoe (Gynostemma pentaphyllum Makino) leaves tea by different ethanol concentrations (에탄올 농도에 따른 돌외 잎 차 추출물의 생리활성)

  • The objective of this study was to compare the effects of different concentration of ethanol extraction on the total phenol and flavonoid contents and physiological activities. The total phenol content of the extracts ranged from 35.54 to 71.52 mg GAE/g. An increase in the ethanol concentration of the solvent led to an increase in the phenol content, with the highest content being found in the 80 and 99.5% ethanol extract. The same trend was observed for flavonoid content. DPPH, nitric oxide, superoxide, hydroxyl radical scavenging activity and TEAC, FRAP and ORAC were measured by antioxidant assay. Radical scavenging activity of aqueous ethanol extracts was better than that of water and 99.5% ethanol extracts. TEAC and FRAP were highly dependent on ethanol concentration and ORAC showed high activity in 40 ~ 80% ethanol extract. Antioxidant activity of Dolwoe leaves tea showed different results among the assay systems. In most experiments, the activities of water and 99.5% ethanol extracts was relatively low. ${\alpha}$-Glucosidase inhibitory activity and microorganism inhibitory activity were highest in the 80% and 99.5% ethanol extracts. Therefore, it was considered that extraction with 80% ethanol was appropriate when considering the antioxidative and physiological activities of Dolwoe leaves tea. Based on these results, it can be used as a basic data for the development of food of Dolwoe leaves tea.

Antimicrobial activities of ethanolic extracts of marine resources against Propionibacterium acnes (해양 유래 한약재의 여드름균에 대한 항균 효능 연구)

  • Objectives : Biological activities of marine resources have been rarely evaluated compared with other herbal medicines. In the present study, we aimed to evaluate antimicrobial activities of aqueous and ethanolic extracts of five marine resources(Porphyra tenera, Laminariae Thallus, Sargassum, Ostreae Concha, Maliotidis Concha) against Propionibacterium acnes. Methods : Aqueous axtracts of five marine resources were prepared by decocting in tenfold tap water for 3 h. Etanolic extracts were obtained by extracting five marine resources with tenfold ethanol for 72 h at room temperature. The zone of growth inhibition and minimum inhibitory concentration (MIC) were determined against P. acnes after incubation for 48 h under anaerobic condition. Results : Ethanolic extracts of Porphyra tenera exhibited potent antimicrobial effects(MIC $62.5\;{\mu}g/m{\ell}$ against KCTC3320, MIC $31.25\;{\mu}g/m{\ell}$ against KCTC5527). However, all aqueous extracts tested had no effects on the growth inhibition of P. acnes. In addition, four ethanolic extracts except Porphyra tenera showed little inhibitory effect. Conclusions : These results indicate that ethanolic extracts of Porphyra tenera has antimicrobial activities against P. acnes and also warrant further development of Porphyra tenera extracts as a natural anti-acne agent.

Antimicrobial Activity of Fractional Extracts from Houttuynia cordata Root (어성초(Houttuynia cordata) 뿌리에서 추출한 순차분획물의 항균활성)

  • The solvent extracts of Houttuynia cordata root, which were extracted by using several solvents with different polarities, were prepared for utility as a natural preservatives. The antimicrobial activity was investigated by disc diffusion method against 22 microorganisms consisting of food borne pathogens, food poisioning microorganisms and food-related bacteria. The extraction yields were 15.7%, 3.7%, 0.13%, 0.5% and 5.9% in ethanol, chloroform ethylacetate, butanol and aqueous fractions, respectively. Antimicrobial activities were shown in ethanol, ethylacetate and butanol fraction of Houttuynia cordata root. However chloroform and aqueous fractions showed weak antimicrobial activity against the tested bacteria. Among the four fractions, ethylacetate fraction showed the strongest antimicrobial activities against microorganisms tested, such as B. megaterium, E. coli, K. pneumoniae and S. typhimurium. The polyphenolic compounds widely occuring in the traditional medicine plants have been reported to possess high antimicrobial activity. The polyphenolic compound in ethylacetate and butanol fraction were 35.9% and 16.0%, ethanol, chloroform and aqueous fraction were 5.0%, 2.3% and 1.7%, respectively. There are some relationship between antimicrobial activity and polyphenol content in natural plants. The ethylacetate fraction could be suitable for the development of a food preservative.

Citrate Complexes of Alkaline Earth Metals in Aqueous, Acetone-Water and Ethanol-Water Solutions (수용액, 물-아세톤 및 물-에탄올 혼합 용매 내에서 형성되는 알칼리토류금속의 시트르산 착물)

  • Formation of the complexes of alkaline earths with citrate ions in aqueous, acetone-water and ethanol-water solutions was studied at room temperature by the equilibrium exchange technique. This technique involved the measurements of distribution of radioactivity between cation exchange resin and solution phase after the radioactive metal ions were equilibriated with the cation exchange resin in the presence of citrate ions ($Cit^{3-}$) of varying concentrations. The pH of the solutions was controlled to 7.2-7.5, and the ionic strength of the solutions was kept at 0.10-0.12. The present study revealed that both barium and strontium ions formed the one to one citrate complexes, $[M Cit]^-$ in all solvent systems examined. It was also observed that calcium ions formed the one to one citrate complex in aqueous solution. In acetone-water and ethanol-water solutions, however, calcium ions were observed to form both one to one and one to two complexes, $[Ca Cit]^-$ and $[Ca\;Cit_2]^{4-}$. The results of the present study indicated also that relative stabilities of the complexes increased in the order; $Ba^{++}\;<\;Sr^{++}\;<\;Ca^{++}$, and in the order of increasing concentration of the organic components in the mixed solvent systems.

Oxidation of Ethanol in the Gas Phase with Alcohol Oxidase and Alcohol Dehydrogenase (Alcohol Oxidase와 Alcohol Dehydrogenase를 이용한 기상에서의 Ethanol의 산화반응)

  • The effects of reaction temperature and the level of hydration(water activity) were studied for gas phase reactions of alcohol oxidase and alcohol dehydrogenase immobilized on DEAE-cellulose and controlled pore glass(CPG). Optimum reaction temperature zone of gas phase reaction was similar to that of aqueous phase reaction. The activity of alcohol oxidase increased dramatically and the stability decreased when the water activity was increased from 0.3 to 0.8. The apparent activation energies of the gas phase reaction decreased approaching the values obtained in the aqueous phase reaction as the water activity increased. In the both cases of alcohol oxidase and alcohol dehydrogenase, the rate constants of the gas phase reaction were lower than those of aqueous phase reaction by two orders of magnitude and these results could be correlated to the vapor-liquid equilibrium data of the substrate, ethanol.
