Anti-tumor activities of Naesosungma-tang (내소승마탕(內消升麻湯)의 항암효과(抗癌效果) 연구(硏究))

  • This study was carried out to evaluate antioxidative, cytotoxic and chemosensitizing effects for anti-tumor activities of Naesosungma-tang (NST). The results were as follows ; 1. The oxidative effects were measured by DPPH radical scavenging activity. NST water extract was showed more effective than ethanol extract and also various solvent fractions from NST showed effective in the following order : butanol fraction > ethyl acetate fraction > hexane fraction > aqueous fraction. 2. NST water extract was showed cytotoxic effects on the human cancer cells Calu-6 cell and SNU-601 cell, but not on MCF-7 cell. 3. The viability of cells was measured in 4 kinds of solvent fractions from water extract of NST by MTT assay. The results were as followings : 1) On the NIH3T3 cell, $IC_{50}$ value was $200{\sim}300\;{\mu}g/m{\ell}$ in three fractions except aqueous fraction. 2) On the Caco-2 cell, effective only in the butanol fraction. 3) On the Calu-6 cell, most effective in hexane fraction($IC_{50}:80\;{\mu}g/m{\ell}$) and effective in ethyl acetate and butanol fraction($IC_{50}:100{\sim}120\;{\mu}g/m{\ell}$). 4) On the MCF-7 cell, $IC_{50}$ value was similar in three fractions except aqueous fraction. 5) On the SNU-601 cell, $IC_{50}$ value was within $100\;{\mu}g/m{\ell}$ in three fractions except aqueous fraction. 6) On the HCT-1l6 cell, $IC_{50}$ value was $150{\sim}200\;{\mu}g/m{\ell}$ in three fractions except aqueous fraction. 7) On the AML-2/WT cell, not effective in four kinds of solvent fractions. 4. In combined effects of NST and vincristine on AML-2/D100 cell, water and ethanol extracts of NST and hexane fraction of water extracts of NST more effectively inhibited proliferation of AML-2/D100 cell. These results Suggest that NST has antioxidative and cytotoxic effects against Caco-2, Calu-6, MCF-7, SNU-601, HCT116 and NIH3T3 cell especially Calu-6 and SNU-601 cell, and also chemosensitizing effects against AML-2/D100 cell in combined vincristine.

Antioxidant Component and Activity of Dropwort (Oenanthe javanica) Ethanol Extracts (미나리 에탄올 추출물의 항산화성분과 항산화활성)

  • This study was conducted to investigate the antioxidant compounds and antioxidant activity on the dropwort (Oenanthe javanica) and its solvent fraction. The dropwort was extracted with 70% (v/v) ethanol, and then partitioned using the solvents of hexane, chloroform, ethyl acetate, butanol, and aqueous. The ethyl acetate fraction contained the highest phenolic and flavonoid of 240.61 mg GA eq/g and 105.57 mg catechin eq/g, followed by ethanol extract of 37.50 mg GA eq/g and 26.50 mg catechin eq/g, respectively. The DPPH radical scavenging activity ($IC_{50}$) on the solvent fractions increased in the order of ethyl acetate, butanol, ethanol extract, chloroform, aqueous, and hexane with 0.08, 0.58, 1.07, 2.43, 2.47, and 3.31 mg/mL, respectively. The ABTS radical scavenging activity was the highest value of 382 mg AA eq/g in ethyl acetate fraction. Reducing power and chelating effect on the ethanol extracts and its solvent fraction were in range of 0.23~0.75 and 0~32.01%, respectively. Hydroxyl radical scavenging activity ($IC_{50}$) was the lowest value of $26.71\;{\mu}g$/mL in ethyl acetate fraction.

Cytotoxicity of Extracts and Fractions from Echinacea pupurea L. on Human Cancer Cells (Echinacea purpurea L. 추출물 및 분획물의 암세포 독성)

  • The cytotoxic effects of water and ethanol extracts of Echinacea purpurea (L.) (EP) and chloroform, ethyl acetate, butanol and aqueous fractions from each extract of EP were examined. Every extract and fraction of EP inhibited the growth of human hepatocarcinoma, human gastric cancer cell, human breast cancer cells and human lung carcunoma in concentration-dependent manners over a concentration range of $0.05{\sim}1.0\;mg/ml$. Most extracts and fractions with the concentraction of 1 mg/ml showed strong inhibition of more than 70% for every cancer cell. Only aqueous fractions of each extract showed very weak inhibitons of 12 to 25% on the growth of human normal lung cell with the concentration of 1 mg/ml. Overall selectivity of the extrats and fractions on the four human cancer cell lines was over 2.5. These results indicate that EP has a very potent selective toxicity for cancer cells.

Excess Volumes of Aqueous Solutions of Nonionic Amphiphile 2-(2-Hexyloxyethoxy)ethanol (비이온 양친매성 분자 2-(2-Hexyloxyethoxy)ethanol 수용액의 과잉 부피)

  • The densities of aqueous solutions of the amphiphile 2-(2-hexyloxyethoxy)ethanol (C6E2) were measured at 279.15 K and 282.15 K by vibrating-tube densitometry. Then using the density data of the binary C6E2 (1)/water(2) system, the excess volumes and partial molar volumes were determined at various compositions. Excess volume VE exhibits negative deviation for the whole region of composition, which implies relatively stronger attraction between molecules. At the C6E2 mole fraction of around 0.45, VE was at its minimum. Partial molar volume ${\bar{V}}_1$ increases monotonously with the mole fraction x1(=x) and ${\bar{V}}_2$ decreases with x. Any particular point in ${\bar{V}}_1$ and ${\bar{V}}_2$, which may point to molecular association, was unobserved.

  • This study examined the antimicrobial activity of Myagropsis yendoi (MY). The antimicrobial activity of ethanol and aqueous MY extracts were measured using a paper disc assay. The MY ethanol extract had the best antimicrobial activity. It inhibited the growth of Candida tropicalis and Gram-positive bacteria such as Bacillus subtilis, Listeria innocua, L. monocytogenes, and Staphylococcus aureus at 4 mg/mL. The minimum inhibitory concentration (MIC) of the MY ethanol extract ranged from 0.1 to 0.5%. The MY ethanol extract inhibited the growth of B. subtilis and L. innocuaat concentrations of 0.15% and 0.1%, respectively. The antimicrobial activity of the MY ethanol extract was not affected by heating at $121^{\circ}C$ for 15 min or pH 2.10. Therefore, the main substances responsible for the antimicrobial activity of the MY ethanol extract are believed to be stable with changing heat and pH.

Apigenin Derivatives of Paulownia coreana Uyeki Leaves

  • The leaves of Paulownia coreana Uyeki were extracted with acetone-$H_2O$ (7:3, v/v), concentrated under reduced pressure and fractionated successively with n-hexane, methylene chloride and ethyl acetate, leaving residual water soluble fraction. A portion of the resulting aqueous soluble powder was chromatographed on a Sephadex LH-20 column using aqueous methanol and ethanol-hexane as washing solvents. Three apigenin derivatives were isolated and identified as apigenin-7-O-${\beta}$-D-glucpyranoside, apigenin-7-O-${\beta}$-D-glucuronopyranoside and apigenin-7-O-[${\beta}$-D-glucuronopyranosyl($1{\rightarrow}2$)-O-${\beta}$-D-glucuronopyranoside] by spectroscopic methods including NMR and FAB-MS.

Correlation of the Rates of Solvolyses of 4-Methylthiophene-2-carbonyl Chloride Using the Extended Grunwald-Winstein Equation

  • The specific rates of sovolysis of 4-methylthiophene-2-carbonyl chloride (1) have been determined in 26 pure and binary solvents at $25.0^{\circ}C$. Product selectivities are reported for solvolyses of 1 in aqueous ethanol and methanol binary mixtures. Comparison of the specific rates of solvolyses of 1 with those for p-methoxybenzoyl chloride (2) in terms of linear free energy relationships (LFER) are helpful in mechanistic considerations, as is also treatment in terms of the extended Grunwald-Winstein equation. It is proposed that the solvolyses of 1 in binary aqueous solvent mixtures proceed through an SN1 and/or ionization (I) pathway rather than through an associative $S_N2$ and/or addition-elimination (A-E) pathway.

Synthesis and Evaluation of Molecularly Imprinted Polymeric Microspheres for Chloramphenicol by Aqueous Suspension Polymerization as a High Performance Liquid Chromatography Stationary Phase

  • Molecularly imprinted microsphere for chloramphenicol (CAP) with high adsorption capacity and excellent selectivity is prepared by aqueous suspension polymerization, in which chloramphenicol is used as template molecule and ethyl acetate as porogen. The CAP-imprinted microspheres are used as high performance liquid chromatography (HPLC) stationary phase and packed into stainless steel column ($150mm{\times}4.6mm$ i.d.) for selective separation of chloramphenicol. HPLC analysis suggests that chloramphenicol can be distinguished from not only its structural analogs but also other broad-spectrum antibiotic such as erythromycin and tetracycline. In addition, the binding experiments of CAP-imprinted microspheres are carried out in ethanol/water (1:4, V:V), the results indicate that the maximum apparent static binding capacity of molecularly imprinted microspheres is up to 66.64 mg $g^{-1}$ according to scatchard model.

Adhesion of Ice Slurry in a Multi-component Aqueous Solution with Stirring and Cooling (다성분계 수용액의 교반/냉각에 의한 빙부착)

  • To resist ice adhesion on cooling wall is concerned to continuous ice formation in thermal storage system. In this study, ice slurries were formed with two ecological aqueous solution, one is ethanol+silanol and the other is propylene glycol+silanol. By freezing under stirring the solution of $300m\ell$ in a stainless steel vessel which was immersed and cooled in a temperature controlled bath, the shape of ice slurry and the strength of ice adhesion on wall was observed with measuring the temperature and stirring load variation. As the concentration is smaller and the supercooling degree is larger, the ice adhesion is easy to occur. When the stirring load is larger than$ 2.1\times10^{-5}W$, the ice adhesion occurred.

Stilbenoids of Korean Pine (Pinus koraiensis) Inner Bark

  • Pinus koraiensis inner bark was collected and extracted with 95% ethanol. The extracts were concentrated and then sequentially fractionated using n-hexane, $CH_2Cl_2$, EtOAc, and $H_2O$ to be freeze dried. A portion of EtOAc fraction (6.6 g) was chromatographed on a Sephadex LH-20 column using aqueous methanol to isolate (+)-catechin (1), (-)-epicatechin (2), and trans-pinostilbenoside (3). Resveratrol (4) and trans-pinostilbene (5) were isolated by column chromatography using EtOH-hexane mixture after purification with aqueous methanol. The structures of these stilbenosides and flavans were characterized by spectroscopic tools using NMR and MS.
