NMR Studies of Lipid-Protein Interaction in Apolipoprotein B / Phosphatidylcholine Recombinants

  • $^{31}P${$^1H$} nuclear Overhauser effects (NOEs) have been obtained for complexes formed between apolipoprotein B (apo B) and dipalmytoylphosphatidylcholine (DPPC) vesicles. NOE measurements have been conducted with broad-band irradiation of the entire $^1H$ spectrum in order to identify the proton source of the NOE. In a unilamellar vesicle formed spontaneously upon mixing aqueous suspensions of long-chain phospholipid with small amount of short-chain lecithin, the maximum NOE occurs at the N-methyl proton resonance position of the choline moiety. With addition of cholesterol to vesicles, the position of the NOE maximum shifts further away from the choline methyl frequency. For the ternary apo B-vesicle-cholesterol complex, the position of the maximum NOE lies halfway between those in vesicles with and without cholesterol.

Daidzein Modulations of Apolipoprotein B and Fatty Acid Synthase mRNA Expression in Chick Liver Vary Depending on Dietary Protein Levels

  • This study was designed to determine the effects of daidzein (DE) on hepatic lipid metabolism in chicks fed with low protein (LP) diet based on casein. In experiment 1, the male chicks were fed with one of the three levels of dietary protein containing 10.95%, 21.9% and 43.8% protein content for 2 days. In experiment 2, the chicks were fed one of the three levels of protein with or without DE at 1,000 mg/kg diet for 2 days. Experiment 3 was conducted to compare DE (LP+DE) with estradiol (LP+E2) in chicks fed with LP diet for 7 days. Plasma lipid profiles, hepatic lipid profiles, activities of hepatic malic enzyme and isocitrate dehydrogenase (ICDH) were measured. Transcriptions of hepatic fatty acid synthase, apolipoprotein-B (APO-B), and fructose bisphosphatase mRNA were measured by RT-PCR. Increasing dietary protein levels markedly decreased the concentrations of plasma triglycerides, hepatic total lipids, hepatic TG, and the mRNA transcriptions while the increased dietary protein levels increased hepatic ICDH activities in experiment 1. In experiment 2, the effects of dietary protein levels on blood and hepatic lipid content were more prominent than those of the additional DE. Interestingly, plasma TG levels were affected by DE supplementation (p<0.05). In experiment 3, DE inhibited APO-B mRNA expressions and stimulated the accumulation of lipid in the liver through mechanisms different from E2. In this study, we demonstrate that DE has beneficial effects on blood lipid profiles, but that it inhibits APO-B mRNA transcription and aggravates the fatty liver induced by LP diet in chicks.

흰쥐에서 김치식이가 조직과 분변의 지질조성과 Apo단백 및 Thyroxine 농도에 미치는 영향 (Effects of Kimchi on Tissue and Fecal Lipid Composition and Apolipoprotein and Thyroxine Levels in Rats)

  • This study was carried out to examine whether kimchi has hypolipidemic effect and to know how it exert lipid-lowering effect in rats. Male Sprague-Dawley rats were fed with kimchi-fee diet, or 3%, or 5%, or 10% kimchi diets for 6 weeks. Plasma cholesterol level was lowered in rats fed all concentrations of kimchi diets, and plasma triglyceride(TG) level was lowered in 10% kimchi diet group compared with that of control significantly(p<0.05). Th intake of kimchi lowered VLDL-cholesterol and VLDL-TG levels, whereas increased HDL-cholesterol level significantly(p<0.05). LDL-cholesterol level was lowered only in 5% kimchi diet group and LDL-TG level was lowered in all kimchi diet groups compared with those of control significantly (p<0.05). the intake of 5% and 10% kimchi diets also lowered the levels of hepatic cholesterol, TG, total lipid, and apolipoprotein B, whereas increased the levels of fecal total fat, cholesterol, TG, and apolipoprotein A-1 significantly(p<0.05). Triiodothyronine(T$_3$) level was elevated in rats fed kimchi diet, whereas thyroxine(T$_4$) level was not affected by kimchi treatment. These observations support that the intake of kimchin in rats loweres plasma and hepatic lipid levels by increasing the excretion of TG and cholesterol through feces, by the elevation of T$_3$ level, and by the altered lipoprotein metabolism.

Curcumin modulates the apolipoprotein B mRNA editing by coordinating the expression of cytidine deamination to uridine editosome components in primary mouse hepatocytes

  • Curcumin, an active ingredient of Curcuma longa L., can reduce the concentration of low-density lipoproteins in plasma, in different ways. We had first reported that curcumin exhibits hypocholesterolemic properties by improving the apolipoprotein B (apoB) mRNA editing in primary rat hepatocytes. However, the role of curcumin in the regulation of apoB mRNA editing is not clear. Thus, we investigated the effect of curcumin on the expression of multiple editing components of apoB mRNA cytidine deamination to uridine (C-to-U) editosome. Our results demonstrated that treatment with $50{\mu}M$ curcumin markedly increased the amount of edited apoB mRNA in primary mouse hepatocytes from 5.13%-8.05% to 27.63%-35.61%, and significantly elevated the levels of the core components apoB editing catalytic polypeptide-1 (APOBEC-1), apobec-1 complementation factor (ACF), and RNA-binding-motif-protein-47 (RBM47), as well as suppressed the level of the inhibitory component glycine-arginine-tyrosine-rich RNA binding protein. Moreover, the increased apoB RNA editing by $50{\mu}M$ curcumin was significantly reduced by siRNA-mediated APOBEC-1, ACF, and RBM47 knockdown. These findings suggest that curcumin modulates apoB mRNA editing by coordinating the multiple editing components of the edito-some in primary hepatocytes. Our data provided evidence for curcumin to be used therapeutically to prevent atherosclerosis.

Apolipoprotein E 다형성과 고지혈증 위험 유무에 따른 혈중 지질농도, 영양소 섭취, 생활습관 및 위험요인과의 관계 (Apolipoprotein E Phenotypes and the Relationship Among Lipid Levels, Nutrient Intakes, Lifestyles and Risk Factors Between Subjects with and without Hyperlipidemic Risk)

  • This study was performed to investigate Apolipoprotein E phenotypes and the relationship among lipid levels, nutrient intakes, lifestyles and risk factors between subjects with and without hyperlipidemic risk. The data were collected from 675 industrial male workers who had completed annual medical examination. Compared to the normal group, the hyperlipidemic risk group in Apo E3 and E4 had significantly higher BMI (p < 0.05) and showed significantly higher body fat (%), waist circumference and WHR in all types of Apo E (p < 0.05). In addition, the hyperlipidemic risk group had significantly higher total cholesterol, LDL-cholesterol, triglyceride and AI than the normal group in all types of Apo E (p < 0.05). Intakes of protein, calcium, phosphorus, iron, vitamin A, vitamin B1, vitamin B2, vitamin C and niacin in Apo E3 were significantly lower in the hyperlipidemic risk group than in the normal group (p < 0.05). In the logistic regression analysis, after adjustment for other factors, Apo E2 + E4, waist and WHR were the significant risk factors associated with hyperlipidemia, but protein intakes were associated with significantly lower risks of hyperlipidemia (p < 0.05). In conclusion, genetic factor (Apo E2 or Apo E4), anthropometric index and nutrient intake seem to influence hyperlidemic risk. Further studies and efforts will be needed to evaluate the independent relationships among hyperlipidemic risk factors.

비만아에서 고지혈증과 Apolipoprotein E 다형성의 관계 (Association of apolipoprotein E polymorphisms with serum lipid profiles in obese adolescent)

  • 대 상 : Apolipoprotein E (Apo E)는 지단백 대사와 지질의 수송에 중요한 역할을 하는 물질이며 많은 연구들에 의해 Apo E의 다형성이 심혈관계 질환의 중요한 유전 인자임이 알려져 있다. 이에 저자들은 Apo E 의 다형성이 비만 청소년에서 이상 지혈증의 위험도를 증가시키는지를 알아보고자 하였으며 Apo E가 비만 청소년 중에서도 심혈관계 합병증의 위험성이 높은 군을 선별하여 적극적인 치료를 하는 기준이 될 수 있는지를 확인하기 위해 본 연구를 시행하였다. 방 법 : 14-18세의 86명의 비만아(각 연령 및 성별에 따른 체질량 지수[body mass index; BMI] 95백분위수 이상)를 대상으로 하였으며 8시간 금식 후 시행한 혈액검사를 통해 혈당, apolipoprotein (Apo) A1, Apo B, 총콜레스테롤(total cholesterol; TC), 중성지방(triglyceride; TG), 저밀도 콜레스테롤(LDL), 고밀도 콜레스테롤(HDL)을 측정하였고 polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP)를 통해 Apo E의 다형성을 검사하였다. 결 과 : 남아와 여아의 비는 1.7이었으며 86명의 평균 나이는 $16.2{\pm}1.8$세이었고 평균 BMI는 $27.4{\pm}2.5kg/m^2$이었다. 유전자 분석에 따른 아형의 빈도(allele frequency)는 ${\varepsilon}2$형이 8.1%, ${\varepsilon}3$형이 87.2%, ${\varepsilon}4$형이 4.7%이었다. Apo E의 유전자형에 따라 ${\varepsilon}2/{\varepsilon}3$${\varepsilon}2/{\varepsilon}3$는 E2군으로 ${\varepsilon}3/{\varepsilon}3$는 E3군, ${\varepsilon}3/{\varepsilon}4$${\varepsilon}4/{\varepsilon}4$는 E4군으로 분류하였고 E2군은 13명(15.1%), E3군은 65명(75.6%), E4군은 8명(9.3%)이었다. 세 군 사이에 나이, 성별, 체중, 키와 BMI는 차이가 없었다. 고지혈증에 대한 검사에서 혈당, Apo A1, TC, TG, HDL은 각 군간 차이를 보이지 않았으며 Apo B와 LDL만이 E4형에서 유의하게 높은 수치를 보였다(P<0.05). 결 론 : 저자들은 본 연구를 통해 비만아에서 Apo E의 유전자형의 빈도를 알 수 있었으며 Apo B와 LDL 농도가 Apo E의 유전자형에 따라 차이가 있음을 확인할 수 있었고 따라서 비만아 중에서도 특정 Apo E4의 유전형을 가진 경우 이상 지혈증의 위험성이 더 높은 것으로 사료된다.

12주간 라인댄스가 노인여성의 치매 및 노화관련인자에 미치는 영향 (The Effects of 12week Line Dance on dementia and Aging-related factors in Elderly Women)

  • 본 연구는 60-70대 노인여성을 대상으로 하였으며 12주간 주 3회, 매회 60분 라인댄스가 치매 및 노화관련호르몬에 미치는 변화를 알아본 결과 결론은 다음과 같다. 12주간 라인댄스 프로그램 실시 후 BDNF, DHEA-s, 에스트로겐은 라인댄스 그룹에서 증가하였으나 통제그룹과 통계적으로 유의미한 차이가 나타나지 않았다. 또한 12주간 라인댄스 프로그램 실시 후 Apolipoprotein B는 라인댄스 그룹에서는 감소하였으나 통제그룹과 통계적으로 유의미한 차이가 나타나지 않았다. 본 연구의 결과에서 그룹과 유의미한 차이는 확인 할 수는 없었지만, 라인댄스 프로그램 적용 그룹에서 치매 예방 및 노화를 지연시키는데 긍정적인 효과를 확인하였으며, 앞으로 라인댄스의 뚜렷한 효과를 확인하기 위해서는 새로운 연구대상과 높은 운동강도, 빈도, 시간 및 운동 장르를 고려한 후속 연구가 필요하다고 사료된다.
