A previous study reported that Ulmus macrocarpa Hance water extract (UME) can improve hyperlipid metabolism and the involvement of suppressed lipid synthesis through adenosine monophosphate-activated protein kinase (AMPK) pathway regulation was suggested. Further exploration of the lipid metabolism between liver and peripheral tissue was necessary to confirm that work, and so this study aimed to extend the possibility that UME can regulate serum lipids. After a 6-week in vivo trial of oral administration of UME to rats with induced hyperlipidemia, serum levels of triglyceride (TG) and total cholesterol (TC) were seen to decrease while HDL cholesterol increased. The UME administrations also decreased the TC and TG levels from the control in liver analysis. However, the suggestion that UME regulates the AMPK pathway to improve hyperlipid states through the suppression of hepatic lipogenesis seems to be only one part of the extract's effect. Indeed, serum concentrations of apolipoproteins A and B were returned to baseline levels of the control group in response to UME administration whilst the liver lipid content was much reduced; this cannot occur through the suggested suppression of hepatic lipogenesis alone. Therefore, a possible mechanism of UME could be that it improves blood circulation by modulating serum lipid levels through both the prior stimulation of lipid oxidation and the suppression of hepatic lipogenesis.
This study is a basic study on the development of functional substances involved in obesity prevention, lipid metabolism, and immune regulation. Male Sprague-Dawley rats were fed a high-fat diet for 10 weeks. Allium monanthum extracts (AME) were administered orally to obesity-induced rats, and their lipid-lowering, antioxidative and various types of biological effects related to the immune system were examined. Blood free fatty acid and triglyceride concentrations decreased as the dose of AME increased. Total cholesterol and LDL cholesterol concentrations in the blood decreased as the dose of AME increased. The total cholesterol concentrations in the liver of the AME-treated groups were lower than the control group. The thiobarbituric acid reactive concentrations were lower in the plasma and liver of all AME-treated groups than the control group. Plasma AST and ALT activities did not show any significant differences among the treatment groups. IL-$1{\beta}$ and IL-6 concentrations in the liver tended to decrease as the dose of AME increased. TNF-${\alpha}$ and IL-10 concentrations did now show any significant differences compared to the control group. Lower expression levels of TNF-${\alpha}$, Apo-B and Apo-E genes were found in the AME-treated groups. Taken together, these results indicate that AME may show positive effects in lipid lowering, antioxidation and anti-inflammation.
Rhizobium tropici CIAT 899 is capable of synthesizing inactive apo-glucose dehydrogenase (GDH). To become an active holo enzyme, the GDH requires a cofactor, PQQ. When R. tropici CIAT 899 was grown in a broth culture medium containing hydroxyapatite and pyrrolo quinoline quinone (PQQ), pH decreased while the concentration of soluble P increased. The solubilization of hydroxyapatite was associated with the production of gluconic acid and 2-ketogluconic acids. The organic acid production and P solubilization were greatly enhanced when the bacterium was grown with air supply. Effect of R. tropici CIAT 899 with (CI+PQQ) and without PQQ (CI) on the common bean growth was examined. Shoot and root weight, and N and P contents in CI+PQQ treatment, were significantly higher than those in control and CI treatment. Nodule weight and acetylene reducing activities were also significantly higher in CI+PQQ treatment than in other treatments.
Single nucleotide polymorphism (SNP) in adiponectin gene has been associated with insulin resistance, diabetes, and cardiovascular disease (CVD). This study was performed to investigate the association of SNP 276G${\rightarrow}$T at adiponectin gene with CVD risk factors in Korean type 2 diabetes mellitus (DM) patients. The subjects were 351 type 2 DM patients visited a DM clinic in Seoul, and the patients with known CVD were excluded. The adiponectin SNP 276G${\rightarrow}$T was analyzed and dietary intakes were assessed by a Food Frequency Questionnaire. The prevalence of G/G, G/T, and T/T genotype was 47.6%, 43.3%, and 9.1%, respectively. Male subjects with T/T genotype showed significantly lower level of adiponectin and HDL-cholesterol and significantly higher C-reactive protein (CRP) level compared to G/G and G/T genotypes. In G/G genotype, protein intake was negatively correlated to body weight, BMI, and waist circumference, and there were positive correlation between carbohydrate intake and BMI, waist-hip ratio, and ApoB/apoA-1 ratio in G/T genotype. However, in T/T genotype, there was no significant association between macronutrient intakes and anthropometric and hematological values. In conclusion, CVD risk would be high in type 2 DM patients with T/T genotype, and the association of macronutrient intakes with anthropometric and hematologic factors was different among the three adiponectin genotypes. These results may imply the need for different dietary management regime according to adiponectin genotype to lower CVD complications in Korean type 2 DM patients.
In this study, we investigated the effects of lowering the fat and cholesterol in the diets of 26 Korean hypercholesteolemic men($\geq$240mg/dl). They consumed 2378kcal/day with 20.9% of the energy deriving from fat, and a cholesterol intake of 282mg(118mg/1000kcal). The experimental diet consisted of 2400kcal, the same as their usual diet, but the fat content was restricted to 15%, and the cholesterol level to below 100mg/1000kcal. The subjects kept to this diet for four weeks and were asked to maintain their usual life activities during the experimental period. The dietary intake and levels of plasma lipid, lipoprotein-cholesterol and apoprotein of the subjects were analyzed before, two weeks into, and after four weeks dietary intervention. After two of the dietary intervention, there were no significant changes of plasma total cholesterol or triglyceride levels but there was some changes of phospholipid level. However, after four weeks, the levels of plasma total cholesterol, triglyceride, and phospholipid had decreased significantly: 18.2%, 32.9%, and 11.9%, respectively. And the LDL-cholesterol and VLDL-cholesterol levels also showed a marked reduction of 18.1%, and 33.0% respectively without change of HDL-cholesterol level. There were no changes in the levels of Apo-A 1, Apo-B, or Lp(a). The changes of the plasma lipid levels were significantly associated with the changes in dietary fat intake but not the cholesterol intake. In conclusion, although the responses to the dietary intervention varied among the individual subjects, the lowering of dietary fat component from 21% to 15% of energy intake seems to be an effective way to reduce plasma cholesterol and triglyceride levels without decreasing HDL-cholesterol level. It was also found that the restriction of dietary cholesterol to below 100mg/1000kcal not seem to be effective for the hypercholesterolemic patient who already consuming below 300mg/day of cholesterol.
In order to minimize the occurrence of fatty liver, this study investigated how adding a high level of dietary choline or methionine affected the lipid metabolism of Tsaiya ducks. Feeding trials were conducted with sixty Tsaiya ducks during their growing period, when they were 8-12 weeks old, and during their laying period, 10-14 weeks after the onset of laying when they were 26-30 weeks old. The ducks were randomly assigned to one of three groups: a control group (basal diet), a group in which methionine was added to the basal diet, and a group in which choline was added to the basal diet. The levels of methionine and choline added were twice as high as the levels recommended by the NRC for layer-type chickens. Experimental results indicated that adding choline to the basal diet of growing ducks significantly (p<0.05) increased their body weight, while adding methionine significantly (p<0.05)reduced their body weight. Adding either choline or methionine reduced the ducks' liver fat content in both the growing and the laying periods (p<0.05). Ducks receiving added methionine or choline in their diets displayed enhanced egg production (p<0.05). Adding choline increased serum triacylglycerol (TG) in the laying period (p<0.05). Adding either choline or methionine did not significantly (p>0.05) affect the t-globulin level. Adding methionine increased the activity of liver malic dehydrogenase in both the growing and the laying periods, and increased fatty acid synthetase in the laying period (p<0.05). While adding choline markedly (p>0.05) increased VLDL and apo B in both the growing and the laying periods, it decreased HDL and apo A in the laying period (p<0.05). This study found that adding a high level of choline to the basal diets of Tsaiya ducks in both the growing and the laying periods had beneficial effects. Furthermore, adding methionine affected the lipid metabolism of Tsaiya ducks to a lesser extent than adding choline.

The bacterial enzyme UDP-N-acetylglucosamine enolpyruvyl transferase catalyzes the first committed step of peptidoglycan biosynthesis, i.e., transfer of enolpyruvate from phosphoenolpyruvate to UDP-N-acetyl-glucosamine. We have overexpressed the enzyme from Haemophilus influenzae in Escherichia coli and crystallized it in the apo-form, as well as in a complex with UDP-N-acetylglucosamine and fosfomycin using ammonium sulfate as the precipitant. X-ray diffraction data from a crystal of the apo-form were collected to $2.8{\AA}$ resolution at 293 K. The crystal quality was improved by co-crystallization with UDP-N-acetylglucosamine and fosfomycin. X-ray data to $2.2{\AA}$ have been collected at 100 K from a flash-frozen crystal of the complex. The complex crystals belong to the orthorhombic space group I222 (or $I2_12_12_1$) with unit-cell parameters of a = 63.7, b = 124.5, and $c=126.3{\AA}$. Assuming a monomer of the recombinant enzyme in the crystallographic asymmetric unit, the calculated Matthews parameter ($V_M$) is $2.71{\AA}^3Da^{-1}$ and solvent content is 54.6%.

Particulate $matter_{2.5}$ ($PM_{2.5}$) is notorious for its strong toxic effects on the cardiovascular, skin, nervous, and reproduction systems. However, the molecular mechanism by which $PM_{2.5}$ aggravates disease progression is poorly understood, especially in a water-soluble state. In the current study, we investigated the putative physiological effects of aqueous $PM_{2.5}$ solution on lipoprotein metabolism. Collected $PM_{2.5}$ from Seoul, Korea was dissolved in water, and the water extract (final 3 and 30 ppm) was treated to human serum lipoproteins, macrophages, and dermal cells. $PM_{2.5}$ extract resulted in degradation and aggregation of high-density lipoprotein (HDL) as well as low-density lipoprotein (LDL); apoA-I in HDL aggregated and apo-B in LDL disappeared. $PM_{2.5}$ treatment (final 30 ppm) also induced cellular uptake of oxidized LDL (oxLDL) into macrophages, especially in the presence of fructose (final 50 mM). Uptake of oxLDL along with production of reactive oxygen species was accelerated by $PM_{2.5}$ solution in a dose-dependent manner. Further, $PM_{2.5}$ solution caused cellular senescence in human dermal fibroblast cells. Microinjection of $PM_{2.5}$ solution into zebrafish embryos induced severe mortality accompanied by impairment of skeletal development. In conclusion, water extract of $PM_{2.5}$ induced oxidative stress as a precursor to cardiovascular toxicity, skin cell senescence, and embryonic toxicity via aggregation and proteolytic degradation of serum lipoproteins.

this study was conducted to examine the atherogenic effect of high cholesterol diet (experimental diet) that influences changes of lipoprotein cholesterol metabolism and arterial wall. Seven NewZealand white rabbits were fed control diet, an the other 7 rabbits 2% cholesterol diet for 10 weeks. Results obtained from this study are as follows: 1) High cholesterol diet resulted in a gradual increase of plasma total cholesterol level, reaching upto 1422 mg/dl at the seventh week. 2) CETP (cholesteryl ester transfer protein) activity was significantly higher in high cholesterol group (64.9% at the 7th week) than control group (49.3% at the 7th week) during most of the experimental period except the 6th week. 3) The cholesterol supplementation induced fatty liver and a decrease of hepatic HMG-CoA reductase activities (2.1 moles vs. 0.3nmoles) compared to control group. 4) Bands of apo B-100 and apo E in plasma lipoprotein were thicker in high cholesterol-fed animals tan control animals as visualized by SDS-PAGE. 5) Oxidizability of plasma lipoproteins measured in vitro was greater in high cholesterol group tan control group, but vitamin E level higher in control group. 6) he effect of cholesterol feeding for 10 weeks also led to early fatty streaks in aortic intima. High cholesterol feeding was atherogenic to rabbits, an this seems to be mediated through elevated CETP activities that regulate plasma HDL cholesterol level and decrease an efficiency of reverse cholesterol transport in lipoprotein cholesterol metabolism. The enhanced oxidizability of plasma lipoproteins and lowered vitamin E level may also contribute to the formation of faaty streaks in aorta of cholesterol-fed rabbits.
Objective : This study was designed to examine the effects of stem bark extracts of Cornus walteri Wanger on the lipid lowering, anti-oxidative activity and concentration of proinflammatory cytokines in hyperlipidemic rat. Methods : Male rats weighing $195.21{\pm}5.85g$ fed high fat diet for 8 weeks and 40 rats(above 400 g) were divided into 4 groups. Each groups were divided into a control group and 3 experimental groups. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1 time/1 day) for 4 weeks. And we fed each experimental group of rats Basal diet and administered an extract of Cornus walteri Wanger(100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid of plasma and liver, concentration of proinflammatory cytokines, anti-oxidative activity and gene expression. Results : 1. Concentration of plasma free fatty acid, LDL-cholesterol showed a tendency to decrease in Cornus walteri Wanger ext. groups. Concentration of plasma triglyceride, total cholesterol showed a significantly decrement in all Cornus walteri Wanger ext. group than that of control group. HDL-cholesterol showed a significantly increment in the 300 mg/kg Cornus walteri Wanger ext. group. 2. Concentration of liver total cholesterol showed a tendence to decrease in Cornus walteri Wanger ext. groups. Concentration of triglyceride liver showed a significantly decrement in all Cornus walteri Wanger ext. group than that of control group. 3. Concentration of plasma and liver TBARS showed a tendence to decrease in Cornus walteri Wanger ext. groups. The values of GSH-Px, SOD and CAT activity showed a significantly increment in the 300 mg/kg Cornus walteri Wanger ext. group than that of control group. 4. The values of plasma AST and ALT activity showed no significantly different in all treatment groups. 5. Concentration of plasma $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ showed a tendency to decrease in the Cornus walteri Wanger ext. groups. However the concentration of IL-10 in the 200 and 300 mg/kg Cornus walteri Wanger ext. groups showed a significantly increment than that of control group. Concentration of liver $IL-1{\beta}$, $TNF-{\alpha}$ and IL-10 showed no significantly difference in all treatment groups. However concentration of IL-6 in the 200 and 300 mg/kg Cornus walteri Wanger ext. groups showed a significantly decrement than that of control group. 6. In the analysis of RT-PCR, gene expression of $TNF-{\alpha}$, Apo-B, Apo-E and leptin in the Cornus walteri Wanger ext. groups showed a lower expression than that of control group. 7. The ratio of $TNF-{\alpha}$, Apo-E and leptin expression per $\beta$-actin expression in the 200 and 300 mg/kg Cornus walteri Wanger ext. showed a significantly decrement than that of control group. The ratio of Apo-B expression per $\beta$-actin expression in the 300 mg/kg Cornus walteri Wanger ext. showed a significantly decrement than that of control group. Conclusions : According to above results, in lowering lipid effect, antioxidative activity and antiinflammatory effect, the Cornus walteri Wanger ext. gives positive effect.
