본 연구는 엘크 사슴의 사육방식에 따른 사료섭취량, 체중 변화 및 녹용생산성에 미치는 영향을 확인하기 위하여 실시하였다. 엘크 사슴 12두를 공시하여 사사사육과 방목사육 형태로 사육하였으며, 처리구 간에 섭취량은 유사하게 나타났다. 일당증체량과 녹용생산성의 경우 방목사육 형태가 높게 나타났는데, 일당증체량은 방목사육 시 약 39% 높은 경향을 보였는데 이는 발육 개선효과가 있을 것으로 생각된다. 녹용 생산성 역시 방목사육 시에 약 19% 높은 경향을 보였다. 그러나 녹용 생산성의 경우 개체 간 편차가 심하여 통계적 유의성은 인정되지 않았다. 본 시험에서 방목사육 시에 일당증체량과 녹용 생산성이 높게 나타난 것은 방목에 따른 조사료원의 영양적 성분이 높기 때문으로 생각된다. 이러한 결과는 사슴 방목의 효과에 대한 기초자료로서 충분이 활용될 수 있을 것이며, 앞으로 계절과 방목지의 초종에 따른 녹용의 생산성과 안정성에 대한 추가적인 연구가 수행되어야 할 것으로 생각된다.
In the present study, the anti-inflammatory effect of Jungcheonhwadamgangki-tang plus antler water extract was tested in formalin-injected chronic inflammation model in mouse hind paw. The test articles were dosed once a day for 10 days, and changes on the body weight, paw weights were observed with histopathology of induced paw dorsum pedis. In addition, histomorphometry was also monitored at sacrifice. 15 mg/kg/10 mL of dexamethasone (DEXA) and diclofenac (DICLO) intraperitoneally dosed groups were used as reference groups. A significantly decrease of both absolute and relative paw weights were observed in all dosing groups including DEXA and DICLO groups compared to that of control, and a significantly decrease of the differences between intact and induced paw weights were also observed Jungcheonhwadamgangki-tang plus antler dosing groups compared to that of control. These histological signs-hypertrophy of paw dorsum pedis tissues were detected results from edematous changes on the cutaneous and subcutaneous tissues with severe infiltration of chronic inflammatory cells-were dramatically decreased in all dosing groups including DEXA and DICLO dosing groups compared to that of control. Especially, dose dependently decreases were detected in Jungcheonhwadamgangki-tang plus antler dosing groups compared to that of control. Base on these aforementioned results, it is concluded that Jungcheonhwadam- gangki-tang plus antler have clear anti-inflammatory effect on the chronic inflammation induced by formalin injection.
Deer horn has been used in oriental medicine and folk remedy. It has had much effects on 'health and tonic efficacy' But this word, 'health and tonic efficacy', is very obscure in modern sense This report attempts to investigate their scientific and objective effects among many papers. The biochemical components of antler are composed of pentoses, hexoses, acid mucopolysaccharides, free and ester fatty acids, glycolipids, phospholipids, prostaglandins, sterols, protein and peptides, free amino acids, minerals, hormones etc. Deer horn extract especially increases body weight gain and feed efficiency, and it stimulates the ability of spermatogenesis of animal. However, a few papers have reported concerning the toxicity and side effects of antler.
Recently, interest in food-derived bioactive peptides as promising ingredients for the prevention and improvement of hypertension is increasing. The purpose of this study was to determine the structure and antihypertensive effect of an antioxidant peptide purified from velvet antler in a previous study and evaluate its potential as a various bioactive peptide. Molecular weight (MW) and amino acid sequences of the purified peptide were determined by quadrupole time-of-flight electrospray ionization mass spectroscopy. The angiotensin I-converting enzyme (ACE) inhibition activity of the purified peptide was assessed by enzyme reaction methods and in silico molecular docking analysis to determine the interaction between the purified peptide and ACE. Also, antihypertensive effect of the purified peptide in spontaneously hypertensive rats (SHRs) was investigated. The purified antioxidant peptide was identified to be a pentapeptide Asp-Asn-Arg-Tyr-Tyr with a MW of 730.31 Da. This pentapeptide showed potent inhibition activity against ACE (IC50 value, 3.72 μM). Molecular docking studies revealed a good and stable binding affinity between purified peptide and ACE and indicated that the purified peptide could interact with HOH2570, ARG522, ARG124, GLU143, HIS387, TRP357, and GLU403 residues of ACE. Furthermore, oral administration of the pentapeptide significantly reduced blood pressure in SHRs. The pentapeptide derived from enzymatic hydrolysate of velvet antler is an excellent ACE inhibitor. It might be effectively applied as an animal-based functional food ingredient.
본 연구는 육경 조직판 이식이 사슴뿔의 성장에 미치는 영향을 구명하기 위하여 육경 조직판을 육경과 전두골 정중선 부위에 각각 이식 하였을 때 조직판의 발달, 조직판을 제공한 부위의 녹용 생산량 및 이식한 조직판의 발육성적을 조사하였으며, 그 결과를 요약하면 다음과 같다. 사슴뿔의 조직판 이식은 육경과 전두골 정중선 중앙과 좌우 양 뿔의 정수리에 이식 시 이식 성공률은 12두 중 5두로 42%이었다. 사슴뿔의 발생 정규부위가 아닌 사슴의 전두골에서 조직판 이식으로 추가적인 사슴뿔 발생을 유도할 수가 있었다. 레드디어의 경우 대조구와 조직판을 제공한 쪽의 녹용 총생산량은 각각 716, 1,071g으로 이식을 하기 위하여 절단된 쪽의 녹용 생산량이 더 많았으며, 가짓수도 조직판을 제공한 쪽이 2.3개로 대조구 1개보다 많았다. 엘크의 경우 1두에서 이마에 조직판이 이식된 뿔에서 길이 17cm, 무게 302g 녹용을 생산하였으며, 레드디어는 다음해 낙각 된 녹각에서 길이 26cm에 무게 122g을 생산하였다. 조직판 이식시 이식부위의 피부는 진피까지 제거하여야만 두개골에 이식한 조직판이 견고하게 부착되었다.이상의 결과에서 조직판 이식으로 사슴뿔 발생의 정규부위가 아닌 전두골에서 부가적인 사슴뿔을 생산할 수 있다고 사료된다.
Objective: Molecular cloning and bioinformatics analysis of annexin A2 (ANXA2) gene in sika deer antler tip were conducted. The role of ANXA2 gene in the growth and development of the antler were analyzed initially. Methods: The reverse transcriptase polymerase chain reaction (RT-PCR) was used to clone the cDNA sequence of the ANXA2 gene from antler tip of sika deer (Cervus Nippon hortulorum) and the bioinformatics methods were applied to analyze the amino acid sequence of Anxa2 protein. The mRNA expression levels of the ANXA2 gene in different growth stages were examined by real time reverse transcriptase polymerase chain reaction (real time RT-PCR). Results: The nucleotide sequence analysis revealed an open reading frame of 1,020 bp encoding 339 amino acids long protein of calculated molecular weight 38.6 kDa and isoelectric point 6.09. Homologous sequence alignment and phylogenetic analysis indicated that the Anxa2 mature protein of sika deer had the closest genetic distance with Cervus elaphus and Bos mutus. Real time RT-PCR results showed that the gene had differential expression levels in different growth stages, and the expression level of the ANXA2 gene was the highest at metaphase (rapid growing period). Conclusion: ANXA2 gene may promote the cell proliferation, and the finding suggested Anxa2 as an important candidate for regulating the growth and development of deer antler.
우리나라와 중국에서 인삼과 함께 고귀한 한방 강장제로 사용되고 있는 녹용의 효능에 대하여 알고자 본 실험은 녹용분말을 급여하였을 때 흰쥐의 발육에 미치는 영향에 대하여 고찰하고자 실시하였다. Sprague-Dawley종 5주령의 어린쥐와 10주령의 큰쥐를 각각 12두씩 처리구별로 임의 배치하였고, 녹용분말을 급여하지 않는 대조구, 녹용분말 I구 (권장량) 그리고 녹용분말 II구 (권장량 3배)로 구분하여 10주간 사육하였으며 실험쥐의 성장발육, 사료섭취량, 사료요구율 및 장기발육에 대하여 조사하였다. 그리고 본 실험의 녹용분말 권장량은 몸무게가 70 kg (대사체중: 24.2 kg)인 성인에게 권장된 녹용분말 7.5 g/day를 기준으로 하여 실험쥐의 대사체중 (Wkg0.75)를 적용하여 계산하였다. 실험쥐들에 대한 성장발육은 어린쥐의 녹용분말 급여구에서 실험 종료시 체중이 다소 좋게 나타났지만 처리구간에 현격한 차이는 발견되지 않았다. 그러나 큰쥐의 경우는 녹용분말을 급여 I구와 II구 모두에서 녹용분말을 급여하지 않은 실험쥐 보다 전 실험 기간동안 좋은 성장을 보였으며, 실험 종료시 대조구, 녹용분말 I구와 II구의 체중은 각각 어린쥐가 470${\pm}$39.00g, 478${\pm}$30.33g과 475${\pm}$22.72g 그리고 큰쥐가 485${\pm}$38.50g, 521${\pm}$38.67g과 508${\pm}$34.44g으로 어린쥐 및 큰쥐 모두 녹용분말 I구의 체중이 가장 높았다. 사료섭취량은 녹용분말 급여에 의하여 특별한 영향을 받지 않았으며, 사료요구율은 대조구, 녹용분말 I구와 II구에서 각각 어린쥐가 5.99, 5.47과 5.54, 그리고 큰쥐가 9.04, 7.73와 8.18로 어린쥐와 큰쥐 모두에서 녹용분말 급여구에서 사료효율이 좋았으며 권장량의 녹용분말을 급여하였을 때 가장 좋은 사료효율을 보였다. 그리고 장기 무게에 있어서는 처리구간 큰 차이를 보이지는 않았지만 녹용분말을 급여한 어린쥐와 큰쥐의 간장, 심장 신장 및 위장의 발육이 좋은 경향을 보였으며 녹용분말 급여량을 달리함에 따라 장기 발육에 미친 영향에 대한 특별한 경향은 발견하지 못하였다. 또한 간장과 신장은 혈액과 중요한 관계가 있는 장기로 녹용분말 급여가 이들 발육에 영향을 주었다는 결과는 녹용의 조혈 작용에 대한 가능성을 시사한다고 볼 수 있다. 결론적으로 녹용분말 급여가 어린쥐와 큰쥐의 성장, 사료요구율 및 장기 발육에 좋은 영향을 주었으며, 녹용 급여량에 대한 세부적 연구는 더 필요하지만 본 실험에서는 권장량을 급여하였을 때 더 바람직한 결과를 얻었다.
사슴뿔은 동물세계에서 가장 빨리 성장하는 조직이다. 따라서 성장중인 사슴뿔은 뼈 성장을 촉진하는 인자가 풍부하게 포함된 것으로 생각된다. 이들 성장인자들 중 IGF-1은 뼈를 자라게 하는 조골세포의 대사에 중요한 역할을 한다고 알려져 있어 이를 정제하고자 하였다. IGF-1의 정제는 상대라고 불리는 신선한 사슴뿔을 유안침전, DEAE-Sepharose CL-6B 이온교환수지, CM-Sepharose CL-6B 양이온교환수지, Sephadex G-50의 순차적인 방법으로 할 수 있었다. 각 과정마다 IGF-1의 거동을 HPLC, SDS-PAGE, Dot blot, 그리고 western blot으로 분석하였다. IGF-1의 정량은 ELISA기술로 재조합 인간 IGF-1을 이용하여 계산되었으며, 최종 분별 액은 두 개의 단백질을 보였으나, Western-blot에서 작은 분자량인 12 kDa으로 최종 판명할 수 있었다. 정제된 단백질은 HPLC에서 retention 시간 8분만에 검출되었으며, 총 농도는 2910 ng/ml 이고 중량은 0.291 g 이었다.
Although it has traditionally known that deer antler and medicinal herbs extract contain some functional components for health promotion, the nutritional significance remains to be elucidated. This study examined the efficacy of deer antler extract (DA) , medicinal herbs extract (MH) and their mixture (DAMH) on serum IGF-I, bone growth with growing rats in vivo and splenocyte proliferation with spleen cells in vitro. Three week-old young female rats (Sprague-Dawley) were divided into 4 groups and then fed basal diet (AIN-93G) or experimental diets containing DA, MH, DAMH, respectively, for 7 weeks. We collected blood, liver, kidney, spleen, femur and tibia from rats. There was no significant difference in weight gain, but food intake increased in DA- and MH-fed groups. There were no signs of liver and kidney damage in the DA, MH and DAMH-fed groups compared to basal diet group. In femur and tibia, wet weights: breaking forces and bone minerals (Ca, Mg and Zn) were significantly higher in the DA-fed group than in the other groups. Serum alkaline phosphatase (ALP) , bone-specific alkaline phosphatase (BALP) activities were significantly lower in the DA, MH, DAMH-fed groups than in basal diet group. Also, serum insulin-like growth factor-I (IGF-I) concentrations were significantly increased in DA-fed group compared to the other groups. Therefore DA was shown to have an activity of bone growth promotion by increasing the IGF-I, a major bone growth factor. The deer antler extract showed an enhanced immune action on the primary cultured-cells from spleen of rats, representing that splenocytes were proliferated by lipopolysaccharide (LPS), but not by concanavalin A (Con A). These results indicate that deer antler extract has beneficial effects on bone growth via IGF-I and on splenocyte activation.
