Korean ginseng (Panax ginseng C. A. Meyer) has been used as an important medicinal plant in the Orient for a long time. It has been claimed that ginseng has many beneficial bioactive effects on human health, such as antitumor, antistress, antiaging and enhancing immune functions. Red ginseng possibly have new ingredients converted during steaming and dry process from fresh ginseng. In this study, pharmacological efficacy and ingredient conversion of ginseng by 9 repetitive steaming and drying process were investigated measuring conversion efficiency of acidic-polysaccharide, phenolic compounds and inhibition of peroxide lipides. It was found that acidic-polysaccarides were increased by heat treatment. In addition, maltol of phenolic compounds, strong antioxidant, produced during the process of red ginseng by Maillard reaction. Acidic-polysaccarides and maltol were increased after the 1st and 3rd steaming and drying treatments, but they were decreased gradually after 5th, 7th, and 9th treatments. Antioxidant activity was increased as increasing treatment times of steaming and drying without significance. Effect of red ginseng extract on inhibition of peroxide was increased gradually until after the 7th treatment, but remarkably decreased after the 9th treatment.

Kujeungkupo method which means 9 repetitive steaming and drying process was used for the processes of green tea, Polygonatum odoratum, and Rehmanniae radix preparata. In this study, ingredient conversion of ginseng by 9 repetitive steaming and drying process were investigated measuring conversion efficiency of brown materials, crude lipids, crude proteins and aromatic compounds. Brown materials, as an antioxidant, in red ginseng were produced through non-enzymatic reaction by heat. Repetitive steaming and drying treatments on ginseng root contiunously increased the content of brown materials and the chromaticity. Crude lipids were degraded by heat and converted into volatile aromatic ingredients. Crude lipids were degraded and decreased by 0.52% after the 5th and 7th. Crude proteins were also decomposed and converted to amino acid. Crude proteins after the 9th treatment were decreased by more than 85% as increased times of treatments. A bicyclogermacrene as aromatic material was decreased as increased treatment times, while but a aromatic caramel was increased.
최근에 해조류 유래 기능성소재는 항종양성, 항바이러스성, 항혈액응고 및 면역력 증강 등의 다양한 생리활성기능을 갖는 것으로 알려져 있다. 특히 해조다당류를 저분자화하면 다양한 생체조절기능성이 월등히 높게 나타나고 있음이 보고되고 있다. 따라서 해조다당류를 해조류로부터 효과적으로 추출할 수 있는 추출방법의 최적화 및 해조다당류의 저분자화를 통하여 해조다당류 기능성 증진 방법에 대한 연구가 활발히 진행되고 있다. 본 총설에서는 최근 부각되고 있는 해양 미생물 유래 해조다당류 분해효소를 이용한 기능성 신소재 개발 및 산업적 응용에 대하여 논하고자 한다.
Liposome as a carrier of topotecan (TPT), a promising anticancer drug, has been reported in attempt to improve the stability and antitumor activity of TPT. However, the biodistr ibution pattern of TPT liposome in vivo and PEG-modified liposome containing TPT have not been studied systemically. In this paper, the in vitro stability and in vivo biodistribution behavior of several liposomes containing TPT with different lipid compositions and PEG-modification were studied. Compared with the 'fluid' liposome (S-Lip) composed of soybean phosphatidylcholine (SPC), the 'solid' liposome (H-Lip) composed of hydrogenated soybean phosphatidylcholine HSPC decreased the leaking efficiency of TPT from liposome and enhanced the stability of liposome in fetal bovine serum (FBS) or human blood plasma (HBP). The results of biodistribution studies in S$_{180}$ tumor-bearing mice showed that liposomal encapsulation increased the concentrations of total TPT and the ratio of lactone form in plasma. Compared with free TPT, S-Lip and H-Lip resulted in 5- and 19- fold increase in the area under the curve (AUC$_{0\rightarrow\propto}$), respectively. PEG- modified H-Lip (H-PEG) showed 3.7-fold increase in AUC$_{0\rightarrow\propto}$ compared with H-Lip, but there was no significant increase in t$_{1/2}$ and AUC$_{0\rightarrow\propto}$ for PEG-modified S-Lip (S-PEG) compared with S-Lip. Moreover, the liposomal encapsulation changed the biodistribution behavior, and H-Lip and H-PEG dramatically increased the accumulation of TPT in tumor, and the relative tumor uptake ratios were 3.4 and 4.3 compared with free drug, respectively. There was also a marked increase in the distribution of TPT in lung when the drug was encapsulated into H-Lip and H-PEG. Moreover, H-PEG decreased the accumulation of TPT in bore marrow compared with unmodified H-Lip. All these results indicated that the membrane fluidity of liposome has an important effect on in vitro stability and in vivo biodistribution pattern of liposomes containing TPT, and PEG-modified 'solid' liposome may be an efficient carrier of TPT.
연구배경: 유전자치료에 이용되는 adenovirus를 기초로 하는 벡타는 상피세포에 강한 친화력이 있고 증식하지 않는 세포에도 유전자의 이입이 가능하며, in vivo에서 다른 백타에 비해 유전자이입 효율이 높은 장점이 있다. 그러나 adenovirus에 이해 전달된 유전자는 세포의 chromosome에 삽입되지 않기 때문에 세포가 분열하면 딸세포에 형질이 전달되지 않을 뿐 아니라 adenovirus가 숙주의 면역반응을 초래하여 이입된 유전자가 제거됨에 따라 전달된 유전자의 발현기간이 짧은 단점이 있다. 포화지방산인 butyrate는 histone의 acetylation을 증가시켜 전달된 유전자의 발현을 증가시킨다고 한다. 저자들은 유전자치료시 butyrate를 병용투여 할 경우 이입한 유전자의 발현이 증가되고, 따라서 유전자치료의 효과를 증가시킬 수 있을 것이라는 가정하에 butyrate가 herpes simplex virus thymidine kinase 유전자를 이용한 유전자치료에 미치는 영향을 조사하였다. 방 법: 악성 중피종세포인 REN세포에 Ad.CMVtk를 이입한 후 세포들을 3군으로 분류하였다. 1 군은 butyrate를 처리하지 않고 7일간 배양하였으며, 2군은 이입 후 1일째에 그리고 3군은 이입 후 2일째 1.5mM/L butyrate를 함유하는 배지에서 24시간 동안 배양한 후 butyrate가 함유되지 않은 배지로 교환하여 배양하였다. 유전자 이입 후 2일, 3일, 그리고 7일에 Western blotting을 시행하여 유전자의 발현정도를 조사하였으며, butyrate 처리 유무에 따른 bystander effect에 의한 살상효과의 차이를 조사하였다. 결 과: Butyrate를 처리하지 않은 대조군은 유전자이입 후 7일째에 유전자의 발현이 없었으나 butyrate를 처리한 군은 7일째에도 유전자의 발현이 있었다. 그리고 butyrate를 처리한 경우 bystander effect에 의한 살상효과가 유의하게 증가되었다. 결 론: 이상의 결과로 butyrate는 유전자의 발현을 증가시킴에 의해 bystander effect를 증가시킴을 알 수 있었으며, 향 후 adenovirus를 이장한 유전자치료의 효과를 증가시킬 수 있는 새로운 방법이 될 수 있을 것으로 생각된다.
