Correlation of Lipid Soluble Compounds of Colored Rices and its Mutagenicity, Antimutagenicity and Antioxidativity (유색미 지용성 성분들의 변이원성.항변이원성.항산화 효과에 대한 상관관계)

  • The fractions extracted with methylene chloride from colored rice seeds of 19 cultivars were prepared to examine the correlationties of both antimutagenicity and antioxidiativity with physiological functionalities. The data revealed a positive correlation of the antimutagenicity with the content of $(3.{\beta},22Z)-Acetate-stigmasta-5,22-dien-3-ol$, 24-Oxocholesterol acetate, 6(E),8(E)-Heptadiene, and Eicosane. For antioxidativity, electron donating ability to DPPH radicals exhibited a positive correlation with the content of (24R,25S)-Aplysterylacetate, however, negative correlations were found between scavenging activity toward hydroxyl radicals and the content of either Tetradecanoic acid or Methyl ester-hexadecanoic acid, $(3.{\beta},24S)-Stigmast-5-en-3-ol$, respectively. In addition, a positive correlation was detected between the inhibitory effect of the fractions and the content of $(3.{\beta},24S)-Stigmast-5-en-3-ol$, however, a negative correlation with the content of 3',3'-Dimethylspiro [acridane-9,1'-indane], $(3.{\beta})-24-Methylene-9,19-cyclolanostan-3-ol$, and some other compounds was observed, respectively.

Antioxidative and Antimutagenic Activity of Ethanolic Extracts from Giant Embroynic Rices (거대배아미 에탄올 추출물의 항산화활성 및 항변이원성)

  • 70% ethanolic extracts were prepared from the three mutant rice cultivars with giant embryo termed Shinsunchal-giant embryonic rice, Whachung-giant embryonic rice and Nampung-giant embryonic rice, and its antioxidative and antimutagenic properties were evaluated and compared. For analysing antioxidativity, various antioxidative indices, such as electron donating ability to DPPH radical, scavenging capacity to hydroxyl radicals generated by Fenton reaction, scavenging capacity to superoxide radicals generated by HPX/XOD system, inhibitory effect on autoxidation of linoleic acid and inhibitory effect on membrane lipid peroxidation derived from rabbit erythrocyte ghost, were determined. For analysing antimutagenicity, suppressive effects on mutagenesis induced by the chemical mutagen, mitomycin C, were measured using E. coli PQ 37 as a indicator cell. The results showed that for both antioxidativity and antimutagenicity the giant embryonic rices were more effective compared to the general cooking rice, Among the giant embryonic rice cultivars, Nampung-giant embryonic rice tended to be most effective, showing its scavenging activity to DPPH radical, superoxide radical and hydroxyl radical, and inhibitory activity to lipid peroxidation was 2,3-, 3,3-, 1.7-, and 2.5-fold greater than those of normal rice, respectively.

Screening of Antioxidation-related Functional Components in Brans of the Pigmented Rices (유색미 겨에 함유된 항산화 효과 관련 기능성 성분의 검정)

  • Lipid-soluble fractions extracted with methylene chloride and ethyl acetate as well as 70% ethanolic extract were made from 5 cultivars of the pigmented rice brans (DZ 78, Elwee, LK1A-2-12-1-1, LK1-3-6-12-1-1 and Killimhukmi) to determine their antioxidative activities and to measure the contents of phenolic compounds, phytic acid, together with unsaponificated compounds such as retinol, tocopherol and stigmasterol. We also attempted to characterize candidate components in charge of antioxidation and measure their relative contents in each methylene chloride and ethyl acetate fraction by using GC-MS techniques. As the results, we found a 30% higher antioxidative activity with 26-fold greater phenolic compound content of the pigmented rices as compared to those of the normal rice. Especially, in Killimhukmi, the contents of the unsaponificated compounds such as retinol, tocopherol and stigmasterol were also found to be more than 2.3-fold greater than those of normal rice. However, we could not find the overall correlations between antioxidativity of rices and their color values, or unsaponificated compounds contents. Methylene chloride fraction from Killimhukmi and LK1A-2-12-1-1, both showing strong antioxidativity, contained relatively high contents of (Z)-9-hexadecenyl ester-9-octadecanoic acid and 2-Hydroxyl-1-1(hydroxymethyl) ethyl-9-octadecanoic acid, respectively. In ethyl acetate fractions, on the contrary, 1H indene, methyl ester-tetradecanoic acid and methyl ester-hexadecanoic acid were detected as the commonly-occurring substances in the 5 cultivars of pigmented rices, implying a possible role in antioxidative action.

Correlation of Antioxidant and Antimutagenic Activity with Content of Pigments and Phenolic Compounds of Colored Rice Seeds (유색미 색소체 및 페놀성화합물 함량과 항산화.항변이원성 간의 상관관계)

  • Twenty-three cultivars of colored rice were collected from inside and outside Korea to determine the contents of pigments and phenolic compounds, and also to compare their correlations with antioxidativity and antimutagenicity. The pigment content decreased in the order of LK 1-3-6-12-1-1>LK 1D 2-7-12-1-1>Elwee>Mutumanikam>IR 1544-38-2-2-1-2-2>wx 124-163-45-7-1-1-1, LK 1A-2-12-1-1. Polyphenolic content was the greatest in IR 17491-5-4-3-3 with a ratio of 0.244 g per 100 g brown rice, followed by LK 1-3-6-12-1-1>LK 1D2-12-1>Elwee>Mutumanikam 7 IR 1544-38-2-2-1-2-2, LK 1A-2-12-1-1. The pigment contents for each colored rice cultivar showed a highly positive correlation with polyphenolics in colored rice seeds. For chromaticity, a positive correlation was exhibited between the lightness and hydroxyl radical scavenging activity. In contrast, a negative correlation was observed between the redness and the inhibitory effect of lipid peroxidation.

Changes in Functional Characteristics of Maillard Reaction Products by Ozonolysis (Maillard 반응 생성물의 Ozonolysis에 따른 기능적 특성 변화)

  • It was investigated that the reduction of the intense brown color and increase of solubility, and depolymerization of polymerized melanoidins by ozonolysis affect their antioxidative and antimutagenic activities. Melanoidins was depolymerized and decolorized by ozonolysis. Ozone-treated melanoidins revealed a lower antioxidative activity and a higher antimutagenic activity than those of the untreated control. Fractions of ozone-treated melanoidins showed three peaks resemble to that of the melanoidins on Sephadex column chromatogram. Melanoidins of above MW 1,200 showed intense brown color. However, the strongest electron-donating ability was detected in the melanoidins of between MW 750 to 900. Ozone-treated and ozone-untreated melanoidins of between MW 900 to 1,000 revealed the higher antioxidative and antimutagenic activities.

Antioxidative Activity of the Extracts from Browned Oak Mushroom (Lentinus edodes) with Unmarketable Quality (시장성이 없는 품질의 갈변 표고버섯(Lentinus edodes) 추출물의 항산화 활성)

  • Physiological functionality of browned oak mushroom was evaluated with focusing its electron-donating ability to DPPH radicals, scavenging ability to superoxide radicals and hydroxyl radicals, and inhibitory activity on lipid peroxidation. The results showed that overall antioxidative activities of browned oak mushroom were superior to those of raw oak mushroom with marketable quality, implying possible involvement of resultant browning reaction products in an increment of antioxidativity. The increased radical-scavenging ability was suggested to mainly be exerted by direct quenching of both superoxide and hydroxyl radicals, not by inhibition of xanthine oxidase activity and $Fe^{2+}$ chelation, respectively. Collectively, these results indicate a possible use of unmarketable browned mushroom as a material for manufacturing various processed functional foods.

  • In this study, antioxidative effectiveness of BHA, BHT of 0.02% was compared to that of separated phenolic acid, ester form and insoluble phenolic acid were extracted from 50g mustard seed removed fat antioxidative effectiveness was assumed, measuring Peroxide value, TBA value for 5 days, storaging respective substrate and contrast tube at $45{\pm}1^{\circ}C$ for 25 days. 1. Laboratory tube was added by BHA, BHT separated phenolic acid ester form and insoluble phenolic acid extract and peroxide value of contrast tube after 25 days storage were 31.9, 13.2, 16.6, 11.2, 35.91. On the other hand at the same condition TBA of each antioxidativity matter were 0.24, 0.16, 0.19, 0.17, 027, 0.35 as a result remarkably appeared antioxidative effectiveness in meal soybean oil substrate. 2. Total phenolic contents of free phenolic acid and insoluble phenolic acid in mustard were 13.2mg/10ml, 340.5mg/10ml, 2.1mg/10ml. 3. Phenolic acid separated and identificated were catechol, methylcatechol, salicylic acid, cinnamic acid, p-hydroxybenzoic acid, syringic acid, caffeic acid, sinapic acid.

Antioxidant and Antimutagenic Activity of Solvent-Fractionated Layers of Colored Rice Bran (유색미 품종별 유기용매 추출물의 항산화성 및 항변이원성 검정)

  • Twenty-four cultivars of colored rice seeds were collected inside and outside Korea, and the antioxidant and antimutagenic activity was determined for the solvent-fractionated layers of their bran parts lipid soluble fraction, pigment containing fraction, and pigment component per se. As the serial organic solvent extraction proceeded, the overall tendency of antioxidant activities declined with increased chemical homogeneity of each fraction. This markedly showed the low antioxidativities of the pigment components from LK 1-3-6-12-1-1 and Gillimhukmi. Even all the colored rice cultivars, with considerable antimutagenic activity in 70% ethanolic extract, exhibited mutagenicity when measured with its pigment containing fraction (wx 124-163-45-7-1-1-1 and LKlB-2-1-1 being the strongest). The pigment content in each colored rice seeds decreased in the order of IR 17491-5-4-3-3>LK 1-3-6-12-1-1>LK 1D-2-12-1>RGS No.336, Elwee. In addition, a substantial difference in both chemical composition of the constituents and its amount could be found between the colored rice and cooking rice cultivars. This revealed that, compared to cooking rice, major components of organic solvent fractions from colored rice probably have long hydrocarbon chain moieties.

Antioxidant activity and inhibition activity against α-amylase and α-glucosidase of Juniperus rigida Sieb extracts (노간주나무(Juniperus rigida Sieb) 추출물의 항산화 활성 및 α-amylase와 α-glucosidase에 대한 저해활성)

  • In this study, the antioxidant effect of water and ethanol extracts from Juniperus rigida Sieb were investigated. The activities of each of the extracts were measured based on their total phenolic and flavonoid contents and using antioxidant test such as of 2,2-azinobis (3-ethyl benzothiazoline-6-sulfonic acid (ABTs) radical scavenging activities, ferric reducing antioxidant power (FRAP), angiotensin I converting enzyme (ACE) inhibition activity, antioxidant protection fator (PF), thiobarbituric acid reactive substances (TBARs) content, and ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibition activity assay. The result of the examination to measure the polyphenol content by investigating the antioxidativity of the J. rigida Sieb. extract showed 71.3 mg/g polyphenol content in the water extract, and 116.0 mg/g in the ethanol extract and a 17.7 mg/g flavonoid content in the water extract and in 76.4 mg/g in the ethanol extract. The ABTS radical cation decolorization showed 76.4% and 79.3% scavenging activities of the $500{\mu}g/mL$ water extract and ethanol extract, respectively. The FRAP showed 1.83 mM efficacy in the water extract and a lower 1.77 mM in ethanol extract. Both the water extract and the ethanol extract showed reduced ACE activities of 75.39% and 71.25% at $500{\mu}g/mL$, respectively. The antioxidant protection factor of the water and 70% ethanol extracts of J. rigida Sieb were 1.5 PF and 2.1 PF, respectively. In the TBARS inhibitory activity, the extracts showed 55.78% and 71.48% antioxidant activities at the $500{\mu}g/mL$ concentration. The results of the measurrement of the ${\alpha}$-amylase inhibitory activity indicated more than 90% of activity inhibition in the $500{\mu}g/mL$ concentration of the ethanol extract. For the ${\alpha}$-glucosidase inhibitory activity, the ethanol extract showed 70% activity inhibition at the $500{\mu}g/mL$ concentration.
