채취 시기에 따른 와송 추출물의 항상화 활성 연구 (Studies on the Antioxidative Ability of Methanol and Water Extracts from Orostachys japonicus A. Berger According to Harvest Times)

  • This study examined the antioxidant activity of methanol and water extracts from Orostachys japonicus A. Berger according to harvest times, by measuring electron donating ability(EDA), reducing power, superoxide dismutase(SOD)-like activity, thiobarbituric acid reactive substances(TBARS) and antioxidant activity within a linoleic acid emulsion. EDA increased proportionally with the extract concentration and the methanol extract had slightly stronger effects than the water extract. And reducing power and SOD-like activity were highest in the methanol extract. Overall, based on the data, the methanol extract of O. japonicus A. Berger harvested during $August{\sim}October$ presented the highest level of antioxidative activity and may be a good candidate as a natural antioxidant source.

흰털오가피 부위별 물추출물의 항산화활성 (Antioxidative Activity of Water Extract of Different Parts of Acanthopanax divaricatus var, albeofructus)

  • Acanthopanax species have traditionally been used as a tonic, a sedative as well as in the treatment of rheumatism, hypertension and diabetes. In the present study, oxidative stress was induced in Vero cells by incubating the cells with glucose and the cell viability was measured by MTT assay. The concentration of glucose which 50% of cell viability was 125 mM $(IC_{50})$ and the cell viability was increased to $87.6{\pm}8.8%$ by treatment of the extracts of Acanthopanax divaricatus var. albeofructus. The antioxidative activity of water extract of different parts of the Acanthopanax plant was investigated by DPPH (1,1-diphenyl-2-picrylhydrazyl) assay, xylenol orange assay, TBARS (thiobarbituric acid reactive substances) assay and enzyme (superoxide anion and catalase) assay. Each extract (leaves, root, stem and fruits) of the plant showed free radical and $H_2O_2$ scavenging activity. The extract also inhibited lipid peroxidation and recovered enzyme (superoxide anion dismutase and catalase) activity in Vero cells treated with glucose.

유용식물로부터 Human Low Density Lipoprotein(LDL)에 대한 항산화제의 탐색 (Screening of Antioxidative Compounds toward Human Low Density Lipoprotein (LDL) from Useful Plants)

  • 본 연구는 유용식물로부터 DPPH free radical scavenger및 사람의 low density lipoprotein(LDL)의 산화에 대한 항산화 효과를 조사하기 위하여 25종의 식물로부터 메탄올로 추출하여 조사하였다. 각종 유용식물 중 항산화 활성은 우수한 달맞이꽃이 가장 높았으며 다시 메타놀로 추출하여 조사한 결과 $\alpha$-tocopherol과 항산화능이 거의 비슷하였다. 메타놀 추출물을 ethylacetate로 추출한 획분이 항산화 활성이 높았으며 ethylacetate 층 중 acidic soluble 획분을 다시 분리한 후 Sepadex LH-20 column chromatography로 추출한 결과 Fraction F-2가 전자공유능이 가장 높았다. 따라서 항산화 활성이 높은Fraction, F-2의 구조를 동정한 결과 3,4-dihydroxybenzoic acid와 3-hyoxycinnamic acid로 확인되었다.

Cell Proliferation and Antioxidative Effects of Ultrasonic Coffee Extracts

  • Recently, coffee is the most popular beverage for modern people. A great number of substances are found in coffee beans and have been studied for many years such as aliphatic and aromatic compounds. However, studies on the physiological activity of coffee extracts are insufficient. This study was performed to determine the contents of caffeine and chlorogenic acid in coffee extracts according to the solvent and to investigate the physiological activity of coffee extracts. Coffee extracts were extracted by ultrasonication method with various types of solvents including distilled water, ethanol, and other organic solvents under $50^{\circ}C$ and $80^{\circ}C$. The contents of caffeine and chlorogenic acid in coffee extracts were determined by Liquid Chromatography-Mass Spectrometry (LC-MS). Also, cytotoxic and antioxidative effects of coffee extracts were evaluated with MTT and DPPH assays to analyze the physiological activity. As a result, it was confirmed that caffeine and chlorogenic acid contents were extracted in distilled water with the highest rate. Antioxidative activity was observed below 10-fold dilute of coffee extracts, however cytotoxicity was not observed. In conclusion, distilled water was the best solvent for extracting caffeine and chlorogenic acids from coffee bean with ultrasonication and these coffee extracts are less cytotoxic in human skin cell lines and have antioxidant effect.

Isolation, Identification and Determination of Antioxidant in Ginger (Zingiber officinale) Rhizome

  • The antioxidative compounds and antioxidant contents of ginger (Zingiber officinale) rhizomes were determined. Substances reextracted using ethyl acetate from crude methanol extract of fresh ginger rhizome were separated through thin layer chromatography. Ten phenolic antioxidative bands were visualized through color reactions using ferric chloride-potassium ferricyanide and 1,1-diphenyl-2-picrylbydrazyl (DPPH). The antioxidative compounds were purified through preparative TLC and high performance liquid chromatography (HPLC), among which, five antioxidants were identified as 4-, 6-, 8-. and 10-gingerols and 6-shogaol on the basis of their molecular weights determined through LC-MS. As shown in experiments using DPPH free radicals, 6-Gingerol and PT4-HP8 (unknown) were revealed to be more efficient than BHT (butylated hydroxy toluene). Contents of gingerols were determined through reverse phase HPLC. Total gingerol contents (sum of 6-,8-, and 10-gingerols) in rhizomes of different ginger varieties varied significantly. The HG55 (collected at Wanju district in Korea) and the HG52 (imported from Brazil) showed the highest gingerol contents.

진달래꽃 추출물의 항산화 효과 및 인체 KB cell에 대한 세포독성 (Antioxidative Activity and Cytotoxicity on Human KB cell of Extracts from Rhododendron mucronulatum Turcz. Flower)

  • 진달래꽃 추출물의 항산화 효과와 인체 비인두암 세포인 KB cell에 대한 세포독성을 조사하였다. 또한 총 폴리페놀과 총 플라보노이드 함량을 측정하고 항산화성과의 상관성을 검토하였다. 진달래꽃 메탄올 추출물을 용매 분획하여 항산화 활성을 측정한 결과, 에칠아세테이트 분획의 활성이 가장 높게 나타났다. 에칠아세테이트 분획을 silica gel column chromatography로 부분 정제하여 항산화 실험을 한결과 fraction 2와 3은 40 ppm 농도에서 90% 이상의 활성을 나타내었다. 인체 비인두암 세포주인 KB cell의 성장 저해율은 $100{\mu}g/mL$ 농도에서 클로로포름 분획이 81.2%, 헥산분획이 74.6% 였다. 총 폴리페놀과 총 플라보노이드 함량을 측정한 결과, 에칠아세테이트 분획에는 총 폴리페놀 32.70%, 총 플라보노이드 20.30% 함유하고 있었다. 총 폴리페놀 및 총 플라보노이드 함량과 항산화성과의 상관관계는 총 폴리페놀에 대하여는 $r^2=0.8869$, 총 플라보노이드에 대하여는 $r^2=0.8469$로 상관성이 인정되었다.

엉겅퀴에서 분리 정제한 Silymarin의 사람 Low Density Lipoprotein에 대한 항산화 효과 (Antioxidative Effect of Silmarin Purified from Silybum Marianum on Modificaion of Human Low Density Lipoprotein)

  • This study was performed to evaluate the antioxidant activity of silymarin against human low density lipoproteins(LDL) oxidation. Silymarin extracted from Silybum marianum was successively purified with solvent fractionation and followed by silica gel column chromatography. The active substances were separated by HPLC and the isolated active substances, silymarin were identified by IR, NMR, GC-MS as silymarin. Silymarin inhibited at the 5 $\mu$M Cu2+-mediated oxidation of human low density lipoprotein (LDL) in a dose dependent manner. Silymarin completely inhibited LDL oxidation at 50 $\mu\textrm{g}$/$m\ell$ concentration. These findings suggest that silymarin may protect LDL against oxidation in atherosclerotic lessions.

Development of Vaccinium uliginosum L. extracts for whitening & anti-wrinkle functional food

  • This study is peformed to investigate the effect of water extract from Vaccinium uliginosum L., on melanin production in B 16 melanoma cells, procollagen production and matrix metalloproteinase-1(MMP-1) inhibition in human fibroblast cells. One hundred grams of the Vaccinium uliginosum L. was extracted with 2000 mL of water($90^{\circ}C$, 16h, 2times). The water extracts were lyophilized and stored at $4^{\circ}C$ until used. Dry weight yields of extracts of Vaccinium uliginosum L. were $3\%$(w/w). Extracts from Vaccinium uliginosum L. showed scavenger activities on DPPH radical, superoxide anion radical, hydroxyl radical, hydrogen peroxide and singlet oxygen radical. And these substances inhibited release of cyiokines from human keratinocyte after UV B exposure. Therefore we confirmed that extracts from Vaccinium uliginosum L. had antioxidative effect. These substances inhibited purified tyrosinase activity and melanogenesis in B 16 melanoma cells treated/untreated IL-$1{\alpha}$. Moreover this extract stimulated procollagen production and inhibited MMP-1 production in human fibroblast cells treated/untreated IL-$1{\beta}$. Therefore we confirmed that extracts from Vaccinium uliginosum L. had whitening effect. And these substances decreased degree of wrinkle in hairless mouse skin that induced by UV B irradiation. Therefore we confirmed that extracts from Vaccinium uliginosum L. had anti-wrinkle effect. From the above results, it is possible that Vaccinium uliginosum L. may be developed to be an anti-melanogenesis agent and anti-wrinkle agent.

Cytoprotective Effect of Green Tea Extract and Quercetin against Hydrogen Peroxide-Induced Oxidative Stress

  • In this study, we evaluated the cytoprotective effects of antioxidative substances in hydrogen peroxide ($H_{2}O_{2}$) treated Mel-Ab melanocytes. Tested substances include selenium, quercetin, green tea (GT) extract, and several vitamins (ascorbic acid, Trolox, and folic acid). Of these, both quercetin and GT extract were found to have strong cytoprotective effects on $H_{2}O_{2}$­induced cell death. We also examined additive effects, but no combination of two of any of the above substances was found to act synergistically against oxidative damage in Mel-Ab cells. Nevertheless, a multi-combination of GT extract, quercetin, and folic acid appeared to prevent cellular damage in a synergistic manner, which suggests that combinations of antioxidants may be of importance, and that co-treatment with antioxidants offers a possible means of treating vitiligo, which is known to be related to melanocyte oxidative stress.

Antioxidant Effect of Monascus koji in Sausage Mixture

  • The antioxidative capacity of Monascus koji was studied in raw bologna-type sausage by measuring thiobarbituric acid reactive substances (TBARS) and peroxide values (POV) during storage at $25^{\circ}C$ for 7 days. The addition of 2% Monascus koji effectively inhibited lipid oxidation in the mixture without vitamin C and NaNO$_2$, evidenced by its prevention of an increase in TBARS and a significantly reduction in increases in POV. Monascus koji in raw minced processed meat significantly improved the shelf life of the food.
