Effect of CRATAEGII FRUCTUS(山査) on Antioxidant Capacity in D-galactose induced Aging Rats (산사(山査)가 노화유발(老化誘發) 흰쥐의 항산화능(抗酸化能)에 미치는 영향(影響))

  • Objectives : CRATAEGII FRUCTUS(山査) is known as the substance which delays aging by the antioxidant effects. The purpose of this study is to investigate the effects of CRATAEGII FRUCTUS(山査) on antioxidant enzyme activities such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase (GSH-px) in rat erythrocytes and blood plasma. Methods : Sprague-Dawley rats were divided into 3 groups, Normal group (supplied enough water and feeds only, Normal Group), D-galatose administered group(injected D-galatose 50mg/kg, 1time/day for 6 weeks, Control Group) and CRATAEGII FRUCTUS(山査) administered group (D-galactose 50mg/kg and CRATAEGII FRUCTUS(山査) extracts 85.0mg/200g 1time/day for 6 weeks, SS Group). Rats were sacrificed and TBARS, SOD, CAT, GSH-px, neutral fat and cholesterol were measured in rat erythrocytes and blood plasma. Results : There was no significant difference in the level of TBARS in the blood plasma between each experimental group. Red blood cell SOD activities were significantly different in each group. They were significantly decreased in the Control group compared to those of Normal and there was an increasing phenomenon in the SS group compared to the Control group. There was a significant difference in the activities of the red blood cell - glutathione peroxides in each group. There was a significant increase in the SS group compared to the Control group. Red blood cell catalase activities was no significant difference in each group. Plasma total lipid concentration was significantly different in each group. It was significantly increased in the Control group compared with the Normal group and it was decreasing in the SS group compared to the control group. Plasma triglyceride was not significantly different in each group. Plasma total cholesterol and Plasma HDL -cholesterol concentrations in the blood plasma were not significantly different in each group. Conclusions : According to the above results, it is considered that CRATAEGII FRUCTUS(山査) is effective in inhibiting lipid peroxides and increasing the activities of antioxidative(anti aging) enzyme in D-galactose induced aging rat.

Protection Effects of Allylmercaptan, Metabolite of Garlic on Endothelial Cell Injury Induced by Oxidized Low Density Lipoprotein (산화된 low density lipoprotein (LDL)에 의해 유도된 내피세포의 손상에 대한 마늘 대사산물인 allylmercaptan의 보호 효과)

  • Oxidation of low density lipoprotein (LDL) has been recognized as an important role in the initiation and progression of atherosclerosis. In this study, effects of allylmercaptan, a major metabolite compound of garlic, was studied on endothelial cell injury induced by oxidized low density lipoprotein (ox-LDL). The antioxidative activity of allylmercaptan was investigated by monitoring a thiobarbituric acid substance (TBARS). Allylmercaptan inhibited LDL oxidation induced by $Cu^{2+}$ at concentrations of 0.1, 1 and 10 mM in a dose dependent manner. Lactate dehydrogenase (LDH) release, as an index of cell injury, and intracellular glutathione levels were determined. Pulmonary artery endothelial cells were preincubated with allylmercaptan at $37^{\circ}C$ and 5% $CO_2$ for 24 hr, washed, and then exposed to 0.1 mg/ml oxidized LDL for 24 hr. Preincubation of endothelial cells with allylmercaptan significantly prevented the LDH release and depletion of GSH. Peroxides were measured directly in 24 well plates using a fluorometric assay. Allylmercaptan inhibited release of peroxides induced by ox-LDL in pulmonary artery endothelial cells. In a free system, allylmercaptan was shown to scavenge hydrogen peroxide. The data indicate that allylmercaptan can protect pulmonary artery endothelial cells from injury caused by oxidized LDL, and suggest that allylmercaptan may be useful for the prevention of atherosclerosis.

Effect of Polygonatum Odoratum on Lowering Lipid and Antioxidation (옥죽(玉竹)의 지질강하 및 항산화효과)

  • Objectives : This study was designed to examine the effects of Polygonatum odoratum EtOH ext. on lowering lipid and anti-oxidation using hyperlipidemic rat. Methods : Male rats weighting $195.21{\pm}4.93g$ were divided into 4 groups and fed high fat diet for 8 weeks. Each of 7 rats was divided into a control and sample group. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1 time/1 day) for 4 weeks. And we fed each experimental group of rats basal diet and administered an extract of Polygonatum odoratum(100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid of plasma and liver, concentration of anti-oxidative activity and tumor necrosis factor-$\alpha$($TNF-{\alpha}$). Results : 1. Concentration of plasma free fatty acid, low density lipoprotein-cholesterol showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Concentration of plasma total cholesterol, triglyceride showed a significant decrement in all Polygonatum odoratum EtOH ext. groups than that of control group. However, concentration of plasma high density lipoprotein-cholesterol was not significantly different in all the treatment groups. 2. Concentration of liver total cholesterol showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Concentration of liver triglyceride(TG) showed a significant decrement in all Polygonatum odoratum EtOH ext. groups than that of control group. 3. Concentration of plasma thiobarbituric acid reactive substance, and liver thiobarbituric acid reactive substance showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. 4. The values of glutathione peroxidase activity showed a significant increment in all Polygonatum odoratum EtOH ext. groups than that of control group. The values of superoxide dismutase activity and catalase activity showed a significant increment in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. 5. The values of plasma aspartate aminotransferase and alanine aminotransferase activities were not significantly different in all treatment groups. 6. Concentration of liver $TNF-{\alpha}$ showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Conclusions : Based on the results in this study, the Polygonatum odoratum EtOH ext. showed a positive effect in lowering lipid and anti-oxidation.

The Effects of Panax Ginseng Extract on Antioxidative Enzyme Activity in Ovariectomized Rats (인삼 추출물이 난소를 절제한 백서의 항산화효소 활성에 미치는 영향)

  • In order to observe the bioactivity of ovariectomized rats, nonovariertoized (sham) group, ovariectomized (Ovx) group, ovariectomized ginseng total saponin (GTS)-treated (Ovx+ GTS) group and ovariectomized ginseng water extract (GW)-treated (Ovx+CW) group were made. We measured AST (L-aspartate aminotransferase) and ALT (L-alanin aminotransferase) in sera, and MDA (malondialdehyde:lipid peroxidation), SOD (superoxide dismutase), catalase, total-glutathione (GSH + GSSG) and GPx (glutathione peroxidase) in liver tissue total homogenates of rat. AST activity of serum in Ovx group was 2.11 times increased, but ALT activity was not changed compared to Sham group. In AST activity, they tend to decrease significantly in each substance such as GTS and GW administered group. Lipidperoxides of each fraction in Ovx group were highly increased compared to Sham group. Extracts of ginseng-treated group markedly inhibited lipid peroxidation by 62% ∼72%. And as the result of the measurements of SOD, catalase, total-glutathione and GPx which are antioxidant enzyme, antioxidant enzymes in Ovx group much lower than in Sham group. But they were significantly increased in each substance such as GTS and GW, administered group. Based on the results, it is supposed that more produced free radicals decreased antioxidant enzyme. And it is also thought that extracts of ginseng can inhibit aging by reducing antioxidant enzyme.

$\beta$-Glucan Suppresses LPS-stimulated NO Production Through the Down-regulation of iNOS Expression and $NF{\kappa}B$ Transactivation in RAW 264.7 Macrophages

  • The antioxidant and anti-inflammatory protective effects of $\beta$-glucan from barley on RAW 264.7 murine macrophage cells induced by lipopolysaccharide (LPS) were examined. The RAW 264.7 murine macrophages were preincubated with various concentrations ($0-200\;{\mu}g/mL$) of $\beta$-glucan and stimulated with LPS to induce oxidative stress and inflammation. The $\beta$-glucan treatments were found to reduce thiobarbituric acid-reactive substance (TBARS) accumulation, and enhance glutathione levels and the activities of antioxidative enzymes, including superoxide dismutase (SOD), catalase, glutathione reductase, and glutathione peroxidase (GSH-px) in the LPS-stimulated macrophages as compared to the LPS-only treated cells. Nitric oxide (NO) production was significantly suppressed in a dose-dependent manner (p<0.05) with an $IC_{50}$ of $104\;{\mu}g/mL$. Further treatment with $\beta$-glucan at $200\;{\mu}g/mL$ suppressed NO production to 2% of the LPS-control, and suppressed the levels of inducible nitric oxide synthase (iNOS) protein and mRNA in a dose-dependent manner. The specific DNA binding activity of nuclear factor ${\kappa}B\;(NF{\kappa}B)$ was significantly suppressed by $\beta$-glucan treatment with an $IC_{50}$ of $220\;{\mu}g/mL$ in a dose-dependent manner. Finally, barley $\beta$-glucan ameliorates NO production and iNOS expression through the down-regulation of $NF{\kappa}B$ activity, which may be mediated by attenuated oxidative stress in RAW 264.7 macrophages.

The Effects of Vit C Supplementation and Nutrition Education on Nutrition Knowledge, Food Habits and Antioxidative Enzyme Activity in Male Adolescent Smokers (비타민 C 보충 및 영양 교육이 흡연 남자 청소년의 영양지식, 식행동과 체내 일부 항산화 관련 효소체계에 미치는 영향)

  • The study evaluated the effectiveness of intervention for male adolescent smokers by making an assessment in terms of changes in food habits, nutrition knowledge, plasma catalase, superoxide dismutase(SOD), glutathione peroxidase peroxidase(GSH-px) activities and thiobarbituric acid reactive substance(TBARS) after Vit C supplementation and nutrition education. The subjects, male adolescent smokers, were assigned into four groups : Control group(19 students), education(Educ.) group(19 students), Vit C supplementation (suppl) group(19 students), and Educ. + Vit C suppl. group(19 students). The Educ. group and Educ. + Vit C suppl. group received nutrition education once a week for 2 weeks. The Vit C suppl. group Educ. + Vit C suppl. group received 500 mg ascorbic acid for 35 days. All data were collected before intervention and after intervention. Nutrition knowledge of those who received education increased, and the frequency of fruit and yellow-green vegetable consumption also increased. Plasma antioxidant enzyme activities were not different except for the SOD activity in the Educ. + Vit C suppl. group, which was significantly increased. The plasma ceruloplasmin level of groups that received Vit C supplementation was reduced more than any other groups, and the specific ceruloplasmin ferroxidase activity of groups that received Vit C supplementation was elevated more than other groups. These intervention programs had an impact on food habits, nutrition knowledge, plasma antioxidant enzyme activities, and plasma TBARS in male adolescent smokers. Various nutrition education programs must be implemented for adolescent smokers, and further studied are needed regarding sorts and amount of antioxidant nutrients and supplementation duration.

Effects of cultivated wild ginseng pharmacopuncture at Gansoo (BL18) and Gokji (LI11) on lipid composition, cytokine level, liver function and antioxidative capacity in diet-induced obese rats (간유(肝兪)(BL18) 및 곡지(曲池)(LI11) 산삼약침 처치가 비만쥐의 지질구성, cytokine, 간장기능, 항산화능에 미치는 영향)

  • Objevtives & Methods : Effects of cultivated wild ginseng pharmacopuncture at BL18 and LI11 on lipid composition, cytokine level, liver function, anti-oxidative capacity and histological characters were investigated in diet-induced obese rats. Forty male Sprague-Dawley rats weighing about 400g were divided into 4 groups of control, BL18, LI11 and BL18 plus LI11 pharmacopuncture groups and raised for 4 weeks. Results : 1. Plasma ${\beta}$-lipoprotein, free fatty acids level and TNF-${\alpha}$ levels significantly decreased in the pharmacopuncture groups compared to those of no treatment group. Plasma and liver total cholesterol, triglyceride, glucose and thiobarbituric acid reactive substance (TBARS) levels were also significantly lower than those of no treatment group. There was, however, no difference in TBARS level among pharmacopuncture groups. Liver total cholesterol level of BL18 pharmacopuncture group was lower than those of the other two pharmacopuncture groups. In LDL-cholesterol level, BL18 pharmacopuncture and BL18 plus LI11 pharmacopuncture groups only had significantly lower levels than that of no treatment group. 2. There was no significant difference between cultivated wild ginseng pharmacopuncture groups and no treatment group in IL-6, alanine transaminase (ALT), aspartic acid transaminase (AST) levels. 3. Compared with \ those of no treatment group, pharmacopuncture groups had significantly higher levels of HDL-cholesterol, superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase activities. There was, however, no significant difference among pharmacopuncture groups. 4. Histological characters of heart, kidney and liver of BL18 pharmacopuncture group were similar to those of normal rats. Conclusions : These results indicate that cultivated wild ginseng pharmacopuncture at BL18 and LI11 may suppress adipose tissue mass and lipid peroxidation and activate antioxidant system.

Forsythiaside, a Constituent of the Fruits of Forsythia suspense, Ameliorates Scopolamine-Induced Memory Impairment in Mice

  • Forsythiaside is a polyphenolic constituent of the fruits of Forsythia suspense Vahl which are widely used as anti-inflammatory herbal raw materials in traditional Chinese medicine. In the present study, the authors assessed the effects of forsythiaside on learning and memory impairments induced by scopolamine using a passive avoidance and the Morris water maze tests in mice. Drug-induced amnesia was induced by scopolamine treatment (1 mg/kg, i.p.). Forsythiaside (10 mg/kg, p.o) administration significantly prevented scopolamine-induced step-through latency reduction in the passive avoidance test and scopolamine-induced increased escape latency in the Morris water maze test (p<0.05). Moreover, in an ex-vivo study, forsythiaside treatment (10 mg/kg, p.o) significantly reduced the increase of thiobarbituric acid reactive substance levels induced by scopolamine (p<0.05). Taken together, the present study suggests that forsythiaside could be useful for the treatment of cognitive impairment, and that its beneficial effects are mediated, in part, by its antioxidative properties.

Hypoglycemic and Antioxidative Effects of Dietary Sea-Tangle Extracts Supplementation in Streptozotocin-Induced Diabetic Rats (Streptozotocin-유발 당뇨쥐에서 다시마 추출물 첨가식이의 항당뇨 및 항산화 효과)

  • The purpose of this study was to investigate the effect of dietary sea-tangle extracts on blood glucose levels, serum lipid levels, thiobarbituric acid reactive substance (TBARS) and glutathione enzymes in diabetic rats treated with streptozotocin (STZ) Four groups of rats (Sprague-Dawley male rats, 180 - 200g) were consisted of normal rats fed control diet (C), diabetic rats fed control diet (CD), normal rats fed sea-tangl extracts diet (E), and diabetic rats fed sea-tangle extracts diet (ED). Diabetes was induced by single injection of streptozotocin (60 mg/kg B.W.). After 7 weeks, rats were sacrificed, serum glucose, serum total cholesterol, triglyceride levels and glutathione enzymes were measured. Urine was significantly higher in CD and ED groups than those of others (p < 0.05). Levels of amylase, calcium, uric acid, hemoglobin, cholesterol and low density lipoprotein (LDL)-cholesterol were different among four groups. But high density cholesterol (HDL)-cholesterol of ED group was significantly higher (p < 0.05) than other groups (C and E group) And the weekly change of serum glucose was decreased in the 3th,4th and 5th weeks. But serum triglyceride (TG) of diabetic rats fed sea-tangle extracts diet (ED) was lower than diabetic rats fed control diet (CD). Activity of hepatic microsomal G6Pase was significantly increased CD and ED groups higher than C and E group, but kidney was decreased ED group. Hepateic glutathione S-transferase (GST) of CD and ED group were significantly lower than C and E group (p<0.05), glutathione peroxidase (GPX) of E and ED group were significantly higher than C and CD group (p<0.05), glutathione reductase (GR) activities of ED group was significantly lower than other groups, malondialdehyde (MDA) of ED was lower than E and CD group, but kidney was increased significant in ED group compared to liver. These results suggested that dietary sea-tangle extracts reduce .hepatic disorders such as oxidant than kidney. In conclusion, dietary sea-tangle extracts groups reduced blood TG and hepatic MDA levels in STZ-induced diabetic rats.

Effect of Gamisogunjung-tang on Antioxidation Activity in Rats Induced Aging by D-Galactose (가미소건중탕(加味小建中湯)이 D-galactose로 노화(老化)를 유발(誘發)시킨 백서(白鼠)의 항산화능(抗酸化能)에 미치는 영향(影響))

  • Objective : This experiment has been done to evaluate the effects of Gamisogunjung-tang(GST) on antioxidant capability and lipidic concentration in blood which are presumed to be related to aging. Method : In this study, we divided 14 weeks old SD rats into normal group, control group and GST group. Control and GST group were induced aging by D-galactose. At the same time GST group were administered extract of Gamisogunjung-tang for 6 weeks. After then we took blood, and measured the activities of SOD, GSH-px, catalase in erythrocytes and measured TBARS values, concentrations of total lipid, tryglyceride, total cholesterol, HDL-cholesterol in plasma. Results : The activities of SOD, GSH-px, catalase in erythrocytes increased significantly in GST group compared with control group. The value of TBARS and the concentration of total lipid, total cholesterol in plasma decreased significantly in GST group compared with control group. The concentration of HDL-cholesterol increased significantly in GST group compared with control group. The concentration of triglyceride were not noticeable. Conclusion : it is considered that Gamisogunjung-tang has an influence on control aging by activation the antioxidative enzyme systems in erythrocytes and decreasing the concentration of lipid in blood plasma.
