Antioxidative Compounds in Extracts of Eleutherococcus senticosus Max. Plantlets (가시오갈피 유식물체 추출물의 항산화 활성물질)

  • Two antioxidative compounds were isolated from the methanolic extract of Eleutherococcus senticosus Max. plantlets and identified as chlorogenic acid and 1,4-di-o-caffeoyl-quinic acid on the basis of mass spectroscopy, $^1H-NMR$ and $^{13}C-NMR$ data. The DPPH free radical scavenging activities of chlorogenic acid $(RC_{50}\;:\;1.2\;{\mu}g)$ and 1,4-di-o-caffeoyl-quinic acid $(RC_{50}\;:\;0.4\;{\mu}g)$ were more effective than those of ${\alpha}-tocopherol\;(RC_{50}\;:\;12\;{\mu}g)$. Of them, 1,4-di-o-caffeoyl-quinic acid compound were isolated for the first time from this plant.

Comparison of the Physiological Activities of Extracts from Different Parts of Prunus sargentii (산벚나무 부위별 추출물의 생리활성 비교)

  • In this study, we investigated the antibacterial activity, antioxidative activity and whitening effect of 75% ethanol extracts from different parts of Prunus sargentii. The total phenolic compound content of the branch extract was 277.92 mg/g as the highest level. In the measurement of DPPH radical scavenging ability, $SC_{50}$ values of the cork layer and branch extract were 26.79 ${\mu}g/m{\ell}$ and 30.13 ${\mu}g/m{\ell}$. In nitric oxide (NO) scavenging ability, $SC_{50}$ values of the branch and leaf extract were 49.19 ${\mu}g/m{\ell}$ and 55.55 ${\mu}g/m{\ell}$. All extracts exhibited higher NO scavenging ability than ascorbic acid used as positive control. On the other hand, in antibacterial activity against Staphylococcus epidermidis and Staphylococcus aureus by disc diffusion assay, the pure bark extract showed the highest activity. Moreover, tyrosinase inhibitory activity of cork layer, pure bark and branch extracts showed higher activity than arbutin used as positive control. In the cytotoxicity measurement by MTT assay, leaf extract was exhibited Raw 264.7 cell viabilities of 44.68~61.83% as cytotoxic result in tested concentration. In conclusion, the branch extract of Prunus sargentii will be a functional materials without damage compared to other parts such as pure bark or cork layer in the plant.

  • In order to find the antioxidative components, fractionation of Cyperus amuricus (Cyperaceae) methanol extract was performed by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging effect. Three compounds, 3,4-dimethoxy benzoic acid (1), 4-hydroxybenzoic acid (2), and piceatannol (3) were isolated from the active ethylacetate soluble fraction of C. amuricus through repeated silica gel and Sephadex LH-20 column chromatography. Among them, compound 3 showed the significant antioxidative effect on DPPH free radical scavenging test. These compounds are reported for the first time from this plant.

Isolation and Identification of Antioxidative Compounds in Fermented Glasswort (Salicornia herbacea L.) Juice (함초발효액으로부터 항산화 활성 물질의 분리 및 동정)

  • The chloroform layer of fermented glasswort (Salicornia herbacea) juice was found to have higher radical-scavenging activity than the other layers by the assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) ($ABTS^+$) radicals. Two antioxidative compounds were isolated and purified from the chloroform layer by Sephadex LH-20 column chromatography using the guided assay of DPPH radical scavenging. Based on mass spectrometer and nuclear magnetic resonance, the isolated compounds were identified as cirsiumaldehyde (1) and chrysoeriol (2). This is the first study to report the presence of those compounds in fermented glasswort juice. Compound 2 showed higher radical-scavenging activity than 1.

Antioxidant Effect of Tryptophan and Arginine on Soybean oil Rancidity (Tryptophan과 Arginine의 대두유에 대한 항산화 및 상승효과)

  • This study was conducted to investigate the antioxidant and synergistic effects upon different concentration(0.02, 0.1, 1%) of tryptophan and arginine of soybean oil during incubation at $60^{\circ}C$. Peroxide value(POV), acid value(AV) and thiobarbituric acid(TBA) value of each oil were monitored. Tryptophan and arginine showed antioxidative effects in all concentration. In case of incubating antioxidative effect of tryptophan was similar to that of TBHQ that of arginine was considerably higher than that of ${\alpha}-tocopherol$, but was lower than that of tryptophan. Synergistic effects among tryptophan, arginine and some food antioxidant were shown to be available in all substrates and the best effect was shown in substrate added compound of tryptophan and ${\alpha}-tocopherol$.

Inhibitory Effects of the Compounds Isolated from Sargassum yezoense on ${\alpha}$-Glucosidase and Oxidative Stress (왜모자반 (Sargassum yezoense)에서 분리한 화합물의 ${\alpha}$-glucosidase 및 산화스트레스 억제효과)

  • We examined ethanol extracts prepared from the Korean marine algae belonging to the Sargassaceae family for their inhibitory effects on ${\alpha}$-glucosidase activity and free radicals in vitro. Among five marine algae, the extracts of Sargassum yezoense were found to possess strongly ${\alpha}$-glucosidase inhibition and free radicals scavenging activities. Two compounds were isolated via bioactivity guided isolation and tested for their effects on ${\alpha}$-glucosidase, DPPH, $ABTS^{+}$ and $Photochem^{(R)}$ analysis. Their chemical structures were elucidated by spectral analysis and direct comparison with authentic compounds; their structures were identified as sargaquinoic acid (1) and sargahydroquinoic acid (2). The inhibitory effects of compound 1 and 2 ($IC_{50}$ value:14.2 and 12.8 ${\mu}M$, respectively) on ${\alpha}$-glucosidase were more potent that of deoxynojirimycin as a positive control ($IC_{50}$ value:18.0 ${\mu}M$). All compounds displayed antioxidative activity which was measured by DPPH, $ABTS^{+}$ and $Photochem^{(R)}$ apparatus.

Diarylheptanoids from Bark of Alnus hirsuta Turcz (물오리나무 수피의 Diarylheptanoid)

  • This study was carried out to investigate the valuable antioxidative compound from extracts of Alnus hirsuta Turcz. which has a nationwide distribution. The bark (2 kg) was extracted with 70% aqueous acetone and fractionated, and the ethyl acetate and $H_2O$ fractions, separately, were chromatographed on a Sephadex LH-20 column to purify the mixture and to give two diarylheptanoid compounds. The isolated compounds were analyzed by NMR spectroscopy, including $^1H$, $^{13}C$ and two-dimensional NMR, and identified as oregonin (1) and 1,7-bis-(3,4-dihydroxyphenyl)-heptane-5-O-${\beta}$-D-xylopyranoside (2). The antioxidative activity was evaluated by DPPH method using two diarylheptanoids and acid-hydrolyzed oregonin derivative which indicated higher activity potential. Of those the acid-hydrolyzed oregonin derivative showed highly active potential with the value of 2.6 of $IC_{50}$.

Effect of Antioxidant Activity and Change in Quality of Chemical Composition and Polyphenol Compound during Long-term Storage (한국산 배의 저장기간에 따른 polyphenol화합물의 함량 및 성분변화와 항산화효과)

  • Changes in polyphenol, free sugar, and organic acid concentrations and their antioxidative activities in Korean pear during storage were investigated. The antioxidant activity of polyphenols was evaluated. Polyphenol concentration in different parts of pear, peel, pulp and core, reached 25.7%. pH decreased while concentrations of free sugars, including fructose, glucose, and sucrose, increased during storage. Four organic acids, citric malic, succinic, and fumaric acids, were determined. Succinic acid was the major organic acid in the pulp and decreased after 5 month of storage. The antioxidative of polyphenol fractions I, II, and III on cottonseed, linseed, and fish oils were higher than those of the control group.

Antioxidant Activity and Total Phenolic Compounds in Grain Extracts of Wheat, Barley, and Oat

  • Cereal grains are rich in phenolic compounds that give beneficial effect in human health. Although several research works have been reported on the effects of phytochemicals of plant origin, such as fruits, vegetables, few studies have examined the antioxidative effects of whole cereal grains. The objective of this study was to determine total antioxidant capacity of 80% ethanolic extracts of cereal grains by testing the ability of the extracts to inhibit UV -induced lipid peroxidation in vitro using linoleic acid in comparison to well-known antioxidant such as ascorbic acid and tannic acid. The total phenolic content of the cereal grain (80% ethanolic extracts) investigated in this study varied from 2.1 mg/g (wheat cv. Olgeurumil) to 10.4 mg/g (barely cv. Seodunchalbori). Highly positive relationship between total phenol compounds and antioxidant activity was found. When the antioxidant activities of all investigated extracts were measured with application of same quantity of phenol compounds, oat grain extracts showed similar antioxidant activity of barely cultivars. However, barely extract appeared as the most potent antioxidant activity of inhibition of UV -induced lipid peroxidation. This indicated that factors such as phenolic compound composition and their individual antioxidant activity could playa crucial role in the total antioxidant activity of cereal grains.

Phenolic Compounds from Barks of Actinidia arguta Planchon Growing in Korea and its Anti-Oxidative and Nitric Oxide Production Inhibitory Activities (국내산 다래나무 수피의 페놀성 화합물의 항산화 및 Nitric Oxide 생성 억제 활성)

  • Phytochemical examination of the barks of Actinidia arguta led to the isolation of five flavonoids. Structures of compounds were elucidated as catechin (1), (-)-epicatechin (2), quercetin (3), $quercetin-3-O-{\beta}-D-glucopyranoside$ (4), $quercetin-3-O-{\beta}-D-galactopyranoside$ (5) by comparison with previously reported spectral evidences. To investigate the anti-oxidative effect and nitric oxide (NO) production inhibitory activity of these compounds, DPPH radical scavenging activity and nitric oxide production inhibitory activity in $IFN-{\gamma}$, LPS stimulated RAW 264.7 cell were examined. The $IC_{50}s$ were determinied as follows : $1\;$IC_{50}=26.61\;{\mu}g/ml$, $2\;IC_{50}=25.30\;{\mu}g/ml$, $3\;IC_{50}=20.41\;{\mu}g/ml$, $4\;IC_{50}=18.23\;{\mu}g/ml$ , $5\;IC_{50}=30.46\;{\mu}g/ml$, $6\;IC_{50}=28.0;{\mu}g/ml$, $7\;IC_{50}=27.24\;{\mu}/ml$. These NO production inhibitory effects were significantly different compared with the positive control, L-NMMA $(IC_{50}=20.77\;{\mu}g/ml)$, respectively. Compound $1\;(IC_{50}=6.19\;{\mu}g/ml)$, $2\;(IC_{50}=8.98\;{\mu}g/ml)$, $3\;(IC_{50}=7.30\;{\mu}g/ml)$ and $4\;(IC_{50}=7.64\;{\mu}g/ml)$ also showed potent antioxidative activities similar level to ascorbic acid $(IC_{50}=9.22\;{\mu}g/ml)$. These results suggest that barks of A. arguta have a potent anti-oxidative and anti-inflammatory activity.
