노각나무 가지의 Phenol성 성분 (Phenolic Compounds from the Twigs of Stewartia pseudocamellia)

  • Ten phenolic compounds were isolated from the twigs of Stewartia pseudocamellia. The isolated compounds were identified as 5,7,3',5'-tetrahydroxyflavanone (1), 3,5,7,3',5'-pentahydroxyflavanone (2), quercetin (3), (+)-aromadendrin (4), (+)-ampelopsin (5), myricetin (6), (+)-catechin (7), (-)-epicatechin (8), kaempferol (9), and fraxin (10) by spectroscopic analysis. Compounds 1, 2, 4, 6, 8, and 9 were isolated from this plant for the first time. The antioxidant activities of compounds 1-10 were evaluated by the DPPH and/or ORAC (oxygen radical absorbance capacity) assay. Compounds 3, 5-9 showed significant antioxidative effects on DPPH assay. Among the active compounds, 6 exhibited higher activity compared to trolox on ORAC assay.

일부 페놀성 화합물의 항산화효과 및 아질산염 소거능 (Antioxidative Activities and Nitrite-scavenging Abilities of Some Phenolic Compounds)

  • This study investigated the antioxidant and synergistic effects and nitrite scavenging ability of some phenolic compounds(catechin, rutin, quercetin and naringin), The electron donating abilities of naringin, quercetin, rutin and catechin were 6.7%, 92.8%, 87.6% and 92.21%, respectively, The antioxidant activities in O/W emulsion substrates were in order of rutin > quercetin > catechin > naringin. The antioxidant effect of rutin was stranger than that of BHT or ${\alpha}$-tocopherol. ${\alpha}$-tocopherol showed synergistic effect with catechin and quercetin, but ascorbic acid not showed effect. The nitrite scavenging abilities of catechin, quercetin, rutin and naringin were 99.9%, 98.6%, 25.5% and 0.2%, respectively. The nitrite scavenging abilities of quercetin and actechin were very potent as compared with those of BHT and ascorbic acid.

  • $\alpha$-Benzoyloxypaeoniflorin (1), a new antioxidant monoterpene $\alpha$-glycoside anomer was isolated from Paeonia suffruticosa along with known compounds, $\beta$-benzoyloxypaeoniflorin (2), paeonolide, paeoniflorin and mudanpioside H. The structure of 1 has been determined by comparing spectral data with those of $\beta$-Benzoyloxypaeoniflorin(2). Compound 1 exhibited moderately potent radical scavenging activity on DPPH radical.

  • The acetone extract of the roots of Rhodiola sachalinensis has furnished six phenolic compounds which exhibited significant scavenging effects against DPPH free radical. The structures of these compounds were identified and determined as gallic acid (1), (-)-epigallocatechin 3-O-gallate (2), kaempferol (3), kaempferol 7-O-$\alpha$ -L-rham nopyranoside (4), herbacetin 7-O-$\alpha$ -L-rhamnopyranoside, (5) and rhodiolinin (6) by physico-chemical and spectral evidences.

  • Phytochemical examination of the barks of Pinus densiflora Siebold et Zuccarini has led to the isolation of one phenylpropanoid, one lignan, one flavonoid, one flavan 3-ol and two procyanidins : 4-O-$\beta$-D-glucopyranosyl-p-coumaric acid (1), 2,3-dihydro-2-(4-methoxy)-7-hydroxy-3-hydroxymethyl-5-(3-hydroxy propyl)-benzofuran 3-O-$\alpha$-D-glucopyranoside (2), taxifolin 3'-O-$\beta$-D-glucopyranoside (3), (+)-catechin (4), procyanidin B1 (5) and epicatechin-($4{\beta}$-8)-catechin-($4{\alpha}$-8)-catechin (6). Among them, Compound 4, 5 and 6 showed potent anti-oxidative activities and these anti-oxidative activities were significantly different compared with ascorbic acid as positive control.

  • Propolis, a natural resinous compound collected from honey bees, contains many biochemical constituents and has been used for traditional medicines as early as 300 B .C. Recently, it has been reported to possess many biological activities such as antibacterial, antiviral, fungicidal, local anaesthetic, immunostimulating, antiinflammatory and free radical scavenging properties. To investigate activities of chrysin, one of propolis effective compounds for blood coagulation system was injected endotoxin (4000 EU/kg, i.v.) in rats at 1 hr after administered chrysin (20 mg/kg, p.o.). This study was resulted that chrysin has antiplatelet aggregation activity in vitro, delay of blood clotting time and prothrombin time, and reduction of fibrinogen and FDP in vivo. Chrysin has increased SOD activity, GSH content and GST activity, and decreased MDA content in liver. The result suggests that the antithrombosis effect of chrysin is suppressive activity for a blood coagulation system and antioxidative activity.

머위(Petasites japonicus) 엽병으로부터 항산화 물질의 분리 및 동정 (Isolation and Identification of Antioxidant Substances from the Stems of Butterbur (Petasites japonicus))

  • 본 연구에서는 머위 엽병의 에탄올 추출물을 용매 극성에 따라 hexane, chloroform, ethyl acetate 및 n-butanol로 분배한 분획물에 대해 전자공여능, 지질과산화 억제능 및 lipoxygenase 저해효과 효과를 살펴봄으로써 항산화능을 측정하였다. 분획물 중 ethyl acetate 분획물의 경우 $100\;{\mu}g/mL$ 농도에서 전자공여능, 지질과산화 억제능 및 lipoxygenase 저해능에서 각각 65.41%, 37.04%, 50.67%로 가장 우수한 항산화능을 나타내었다. 이를 근거로 하여 ethyl acetate 분획물로부터 머위 엽병의 항산화 활성 성분을 규명하기 위하여 open column chromatography를 이용하여 화합물 PJ-4를 분리, 정제하였으며, 1H- 및 13C-NMR 분석을 통해 kaempferol로 동정하였다. 단리된 kaempferol의 항산화 활성을 확인하기 위하여 앞에서 언급한 세 가지 항산화 측정법을 통해 실험한 결과, 농도 의존적으로 활성이 증가하는 양상을 보였다. 특히 $100\;{\mu}g/mL$의 농도에서 전자공여능, TBARS에 의한 지질과산화 억제 및 lipoxygenase 저해실험에서 각각 65.76%, 43.47%, 58.60%로 높은 항산화 활성을 나타내었다. 이와 같은 결과를 통해 kaempferol은 머위 엽병의 항산화 활성에 있어서 기여하는 물질일 것으로 보이며, 이로써 머위엽병을 이용한 천연항산화제 개발 및 활용에 긍정적인 가능성을 제시할 것으로 사료된다.

옻나무(Rhus verniciflua) 목질부에서 분리한 화합물의 항산화활성 (Antioxidant Activity of Isolated Compounds from the Heartwoods of Rhus verniciflua)

  • 옻나무 목질부를 열수 추출하고, 얻어진 추출물을 $CHCl_3$, EtOAc, H-BuOH 및 $H_2O$로 용매 분획하였다. 이중 EtOAc와 n-BuOH분획으로부터 silica gel, ODS 및 Sephadex LH-20 column chromatography로 정제하여 4개의 화합물을 분리하였다. 각 화합물의 화학구조는 NMR 스펙트럼 데이터를 해석하여, sinapyl aldehyde (1), 2,4-dihydroxybenzaldehyde (2), 2,4-dihydroxybenzoic acid (3) 및 2,4-dihydroxyphenyl-glyoxylic acid (4)으로 동정하였다. 이 화합물들 중 2-4은 옻나무목질부에서 처음 분리된 화합물이다. 이들 화합물 중 sinapyl aldehyde (1)와 2,4-dihydroxyphenylglyoxylic acid (4)의 DPPH radical 소거작용이 $34.7{\pm}0.6%$$43.8{\pm}0.9%$로 나타나 합성항산화제인 $BHT(14.4{\pm}0.3%)$보다 높게 나타났다.

압출성형 백삼추출물의 화학적 조성 및 항산화 활성 (Chemical Compositions and Antioxidant Activity of Extract from a Extruded White Ginseng)

  • 본 연구는 압출성형 공정을 통한 인삼 시료의 성분 변화를 측정하기 위하여 일반성분 및 항산화활성을 측정하였다. 수분함량을 20%로 고정시키고, 배럴온도는 100, $140^{\ciirc}C$로 변화시켜 압출성형백삼을 제조하였다. 백삼, 홍삼, 압출성형백삼의 일반성분 실험에서는 압출성형 공정을 통한 성분의 증가를 확인할 수 없었다. 항산화활성 측정 결과 산성다당체 함량은 홍삼이 62.94 mg/mL로 높게 측정되었고 총 폴리페놀함량과 DPPH 측정에서는 $140^{\circ}C$ 압출성형백삼이 각각 14.99 mg/g, 80.21%로 높게 측정되어 압출성형 공정을 통한 항산화 물질의 증가를 확인할 수 있었다. $140^{\circ}C$ 압출성형백삼의 조성 분석을 위하여 BuOH 분획을 실시한 결과 총페놀성 화합물과 산성다당체 측정 결과가 각각 27.2 mg/g, 217.6 mg/mL로 측정되었으며 HPLC를 통해 알아본 사포닌 조성은 $140^{\circ}C$ 압출성형백삼에서 모든 성분의 증가를 확인할 수 있었고 특히 홍삼의 특유성분인 Rg3s, Rg3r의 증가로 압출성형 공정을 통한 백삼의 홍삼화 가능성을 확인할 수 있었다.

오미자의 이화학적 특성 및 항산화 활성 (Physicochemical Properties and Antioxidative Activities of Omija(Schizandra chinensis Bailon))

  • To develop physiological functionality of Omija extracted with water was evaluated on antioxidative activity. Omija, high acid material with pH 3.6, contain $57.5{\pm}1.03%$ of moisture, and $18.8{\pm}0.12%$ of crude fat. This material have $12.6{\pm}0.04%$ of carbohydrate and $11.1{\pm}0.07%$ of crude protein as well, but ash and crude protein contents were found less than 10%. 10 mineral contents were also found, too; K and Ca showed the highest level, then Al, Mg, Na and Mn were followed. In composition amino acid contents, glutamic acid took the largest portion, $131.7{\pm}1.3$ mg/100 g, aspartic acid $51.5{\pm}0.6%$, and other composition amino acid under 50%. In case of free sugar contents, 7 types were found. Most of them were glucose and fructose. Total phenolic compounds showed the highest level, $2,862.6{\pm}31.7$ mg/100 g. $197.8{\pm}14.6$ mg/100 g of flavonoid and $225.6{\pm}18.2$ mg/100 g were included. In terms of electron donating ability, radical scavenging ability activated as the amount of Omija extract increased. In particular, Omija extract in 1,000 ${\mu}g/m{\ell}$ demonstrated almost similar electron donating ability, $72.4{\pm}0.21%$, to BHT. It was also found that antioxidant activities of electron donating ability, SOD-like ability, hydroxyl radical scavenging ability and nitrite scavenging ability were highly promoted as Omija extract concentration increased. The nitrite scavenging ability was significant when the extract belonged to strong acid region and doping concentrations increased.
