The Effects of Chicken Leg Bone Extract on Antioxidative Properties under Different Heating Condition

  • The aim of this study was to extract chicken leg bone, which is a by-product of industrial poultry processing, using different heating temperatures (80, 90 and $100^{\circ}C$) and durations (5, 10 and 15 min). The pH value, soluble protein content, peptide content and antioxidative properties, including superoxide anion scavenging ability, 1, 1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging ability, reducing capacity and inhibitory activity of linoleic acid peroxidation, were measured. The results showed no significant differences (p>0.05) in pH value among all treatments. Decreased soluble protein content and peptide content were observed in chicken leg bone extract obtained under higher heating temperatures (90 or $100^{\circ}C$) and longer heating durations (10 or 15 min). In antioxidative properties, the extracts which were heated at 90 or $100^{\circ}C$ for 15 min exhibited significantly higher superoxide anion scavenging ability, DPPH free radical scavenging ability, reducing capacity and inhibitory activity of linoleic acid peroxidation (p<0.05).

  • 백연수 잎으로부터 얻어진 50% 에탄올 추출물의 항산화능을 조사하였다. 백연수 잎 추출물의 수율은 $18.3{\pm}0.40%$, 총 폴리페놀 함량은 $121.3{\pm}11.04{\mu}g$ gallic acid equivalents/mg, 총 플라보노이드 함량은 $105.2{\pm}2.77{\mu}g$ quercetin equivalents/mg으로 나타났다. 백연수 잎 추출물은 유리라디칼 소거활성, 양이온라디칼 소거활성, 아질산염 소거활성, 환원력 및 지질과산화 억제 활성의 항산화능을 가지고 있었다. 백연수 잎 추출물의 모든 항산화 활성은 추출물의 농도에 비례하여 증가하였으며, 유리라디칼 소거활성, 양이온라디칼 소거활성, 아질산염 소거활성 및 환원력의 $EC_{50}$은 각각 133.51, 667.22, 2.58 및 $250.00{\mu}g/mL$이었다. 백연수 잎의 우수한 항산화 활성은 높은 폴리페놀과 플라보노이드 함량에 기인한 것으로 여겨진다.

대두 사포닌이 Aflatoxin B1으로 유도된 세포돌연변이에 미치는 영향에 관한 연구 (Effect of Soybean Saponins on Aflatoxin B1-induced Mutagenicity)

  • Free radicals formed during the metabolism of environmental chemicals are known to induce mutagenicity, while different types of antioxidants suppress this event. The purpose of this study was to determine the antioxidative and antimutagenic effects of soybean saponins, and to examine the relationship between these two effects for the elucidation of mechanisms involved in the anticarcinogenicity of soybean saponins. Also, antioxidative and antimutagenic effects of soybean saponins were compared with those of kinown antioxidants. For the measurement of antioxidative capacity, soybean saponins, L-ascorbic acid, $\alpha$-tocophoerol, and BHT at concentrations between 005 and 1.0mg/ml were tested for their ability to donate hydrogens and to reduce the formation of thiobarbituric substances(TBARS). Antimutagenic activity was examined using the Ames salmonella test system at concentrations of 600, 900 or 1200ug/ml. Study results showed soybean saponins and all of the other antioxidants tested possessed dose-dependent antioxidative activities. The ability of hydrogen-donation to DPPH was in the order of L-ascorbic acid>$\alpha$-tocopherol=>BHT>soybean saponins. TBARS formation was also inhibited by these compounds, in the order of BHT>$\alpha$-tocopherol=L-ascorbic acid>soybean saponins. Soybean saponins and other antioxidants also showed antimutagenicity in a dose-dependent manner. Especially, soybean saponins and BHT were excellent antioxidants compounds, inhibiting near 80% of the mutagenic effects at a concentration of 1200ug/ml. The correlation coefficients between antioxidative capacity and antimutagenicity for each compund was statistically significant at p<0.05. These results indicate that soybean saponins possess antioxidative and antimutagenic capacities. Also, antimutagenicity of saponins and other antioxidats is partly due to their antioxidative activities.

나문재 분말과 추출물 첨가 절편의 항산화 활성 (Antioxidative Activity of Jeolpyun containing Suaeda glauca (Bunge) Bunge Powder and Extract)

  • In this study, the antioxidative activity of Jeolpyun containing S. glauca (Bunge) Bunge powder (2%, 4%, 6%, and 8%) and extract (2%, 4%, 6%, and 8%) was evaluated based on the total polyphenol contents, electron-donating ability, scavenging of superoxide anion radical, and decomposition of hydrogen peroxide. Total polyphenol contents of Jeolpyun containing 8% S. glauca (Bunge) Bunge powder were the highest with a value of 2,280±0.00 ㎍ GAE/mL. It was observed that total polyphenol content exhibited a significant relation with concentrations at p<0.05. The electron-donating ability of Jeolpyun containing 8% S. glauca (Bunge) Bunge extract was the highest with a value of 83.93±0.69%. The electron-donating abilities revealed significant differences between concentrations (p<0.05). The superoxide radical scavenging ability of the blank was 0.01523±0.00068 and Jeolpyun containing 6% S. glauca (Bunge) Bunge powder was the highest with a value of 0.00977±0.00842. The hydrogen peroxide decomposing ability of Jeolpyun containing 0% S. glauca (Bunge) Bunge powder was -0.00377±0.0049; 8% S. glauca (Bunge) Bunge powder revealed the highest decomposing ability of -0.0476±0.0234. Hydrogen peroxide decomposing abilities revealed significant differences between concentrations (p<0.05). Jeolpyun containing S. glauca (Bunge) Bunge powder and extract demonstrated improved antioxidative activities.

연잎 에탄올 추출물의 항산화 효과 및 아질산염 소거능 (Antioxidative Activity and Nitrite Scavenging Ability of Ethanol Extract from Nelumbo nucifera Leaves)

  • To confirm possibility of Nelumbo nucifera leaves as biofunctional material, we investigated the antioxidant activity and nitrite scavenging ability of ethanol extract from Nelumbo nucifera leaves. Nelumbo nucifera leaves were extracted with ethanol and concentrated under vacuum using rotary evaporator. Then, antioxidative activity and nitrite scavenging ability of the extract were examined in vitro. Electron -donating ability of the extract at RC50 was 90.19 ${\mu}g/mL$. After addition of 0.96 mg/mL, autooxidation of pyrogallol decreased to 66.19% by superoxide dismutase-like activity. In antioxidative activity of the extract against linoleic acid during incubation times of 24, 48, and 96 hours at $40^{\circ}C$, lipid peroxidation values significantly decreased to 72.53%, 82.00%, 84.69% with addition of 0.2 mg/mL, respectively. Total phenolic content was determined as gallic acid equivalents (GAE) and the value revealed to be $282.84\;{\pm}\;9.03$ GAE ${\mu}g/mg$ of the extract. Nitrite scavenging ability showed the most remarkable effect at pH 1.2, exhibited to 45.55% by addition of 0.2 mg/mL. These results suggest that ethanol extract from N. nucifera leaves can be used as bioactive and functional material.

Comparative Analysis on Antioxidative Ability of Muscle between Laiwu Pig and Large White

  • This study was conducted to evaluate effects of storage temperatures ($4^{\circ}C$ and $20^{\circ}C$) and pig breeds (Laiwu pig and Large White pig) on the main antioxidative enzymes (superoxide dismutase, catalase, and glutathione peroxidase) activity and lipid oxidation in porcine Longissimus dorsi muscle. Activities of antioxidative enzymes (AOE) decreased slightly during storage, regardless of storage temperatures. Muscle antioxidative enzymes activities stored at $4^{\circ}C$ were higher than that stored at $20^{\circ}C$. Laiwu pig's enzymes activities were significantly (p<0.01) higher than Large White's. The level of malondialdehyde is a direct expression of the grade of lipid oxidation in meat. In our study, the malondialdehyde contents increased after 6 days storage. However, malondialdehyde contents of Laiwu pig were significantly (p<0.01) lower than Large White's. A lower content of malondialdehyde corresponds to a lower oxidation of lipids. These results indicated the muscle antioxidative ability of Laiwu pig was higher than Large White pig. It also implied that antioxidative enzymes were involved in the essentials and deciding mechanisms of meat quality by quenching oxygen free radicals and inhibiting lipid oxidation in muscle.

고추씨 기름에 대한 다류 에탄올 추출물의 항산화 효과 (Antioxidative Effect of Ethanolic Extracts of Some Tea Materials on Red Pepper Seed Oil)

  • Antioxidative effect of ethanolic extracts of various tea materials(Camellia sinensis, Cassia tora, Lyc ium chinense, Polygonatum odoratum, Schizandrae chinensis) on red pepper seed oil was investigated. Ethanolic extracts were added to red pepper seed oil at a concentration of 0.05%(w/v). Two experimental conditions were employed : 50$\pm$0.1oC for 45 days and 150$\pm$3oC for 24 hours. Oxidation of red pepper seed oil was determined by measuring peroxide value and acid value. Electron donating ability(EDA) and total phenolic contents of each extract were also determined. The result showed that the extracts possess an antioxidative activities. The effectiveness of them was in the following order: C. sinensis>C. tora>P. odoratum>S. chinensis >L. chinense. Ethanolic extracts of C. sinensis showed substantially higher EDA value and total phenol contents than other tea materials. These results indicate that the antioxidative effect was strongly related with EDA and total phenol contents.

Antioxidative Activities of Kefir

  • This study aims to evaluate the antioxidative activities of cow-milk kefir and goat-milk kefir. Antioxidative mechanisms, including radical-scavenging effects, ferrous-ion chelating ability, reducing power and antioxidant activity, were investigated herein. Kefirs demonstrated significantly greater scavenging effects upon 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide radicals, an inhibition effect upon linoleic-acid peroxidation, and more substantial reducing power, but reduced glutathione peroxidase (GSH-Px) activity than was the case for milks. There was no significant difference between milks and kefirs as regards ferrous-ion chelating ability and superoxide dismutase (SOD) activity. These findings have demonstrated that kefirs possess antioxidant activity, thereby suggesting that kefirs are potential candidates for the role of useful natural antioxidant supplements for the human diet.

Protective Effect of Kaempferol on Cultured Neuroglial Cells Damaged by Induction of Ischemia-like Condition

  • This study was performed to evaluate the cytotoxicity induced by ischemia-like condition (ILC) in cultured neuroglial cells (C6 glioma cells). The protective effect of kaempferol (KAE), flavonoid against the cytotoxicity induced by ILC induction was assessed. In addition, antioxidative effects of KAE were done by colorimetric assays. Cell viability and the antioxidative effects such as DPPH-radical scavenging activity, superoxide dismutase (SOD)-like activity and inhibitory activity of lipid peroxidation (LP) were analyzed. ILC induction decreased cell viability in a dose-dependent manner, and the $XTT_{90}$ value (low cytotoxicity value) and $XTT_{50}$ value (high cytotoxicity value) were determined during ILC induction for 15 and 40 minutes, respectively. The butylated hydroxytoluene (BHT) antioxidant significantly increased cell viability damaged by the ILC-induced cytotoxicity. In the protective effect of KAE on ILC-induced cytotoxicity, KAE protected the ILC-induced cytotoxicity by the significant increase of cell viability, and also it showed DPPH-radical scavenging ability, SOD-like ability and inhibitory ability of LP. From these results, it is suggested that ILC induction showed cytotoxicity in these cultures and the oxidative stress is involved in the ILC-induced cytotoxicity. While, KAE prevented ILC-induced cytotoxicity by antioxidative effects. In conclusion, natural products like KAE may be a putative therapeutic agent for the treatment of disease associated with oxidative stress such as ischemia.

자소자 항산화성분의 분리 (Isolation of Antioxidative Components of Perillae semen)

  • 자소자의 탈지시료로부터 FPA, SPA 및 IPA 형태의 phenol성 물질을 추출하여 이중 항산화활성이 가장 강한 FPA 추출물에 존재하는 항산화성분을 column chromatography 및 HPLC에 의하여 분리하였다. Chlorogenic acid를 표준물질로하여 측정한 탈지된 자소자에 함유되어 있는 phenol성 물질의 총함량은 0.38% 였고, 총 phenol성 물질중 FPA, SPA 및 IPA 추출물이 차지하는 비율은 71.1%, 15.8% 및 13.1%였다. 세가지 형태의 phenol성 물질의 항산화활성을 electron donating ability (EDA)와 linoleic acid를 기질로 하여 TBA값을 측정하여 비교하였을 경우 FPA 추출물이 가장 높은 항산화활성을 나타내었다. 항산화활성이 가장 높게 나타난 FPA 추출물을 silica gel column chromatography로 분획하여 항산화활성을 비교한 결과 acetone : methanol의 비가 8 : 2인 획분에서 가장 높은 항산화활성을 나타내었다. 항산화활성이 가장 높았던 획분을 HPLC에 의하여 분리한 결과 HPLC chromatogram 상에서 5가지 활성물질 획분이 분리되었으며, 이들 획분을 분취하여 항산화활성을 비교한 결과 F-I 획분이 가장 높은 항산화활성을 나타내었다.
