본 연구는 솔잎착즙액이 고지방식이로 이상지질혈증을 유도한 흰쥐의 지질대사에 미치는 영향을 분석하기 위해 실험동물을 정상식이 (ND)군, 고지방식이군으로 나누어 각각의 식이를 4주간 급여한 후 고지방식이군을 설기떡 (SD)군 및 솔잎착즙액설기떡 (PSD)군으로 분류하여 6주간 사육하고, 혈장과 간 및 분변의 지질관련 지표를 분석하였다. PSD군은 설기떡 섭취에 의해 유의하게 높아진 체중증가량을 유의하게 낮추었으며 (p < 0.05), 설기떡 섭취에 의해 유의하게 높아진 혈장 총 콜레스테롤 및 LDL-콜레스테롤을 낮추는 경향을 나타내었다 (p > 0.05). 간 조직과 분변의 지질대사 관련 지표를 분석한 결과 PSD군은 설기떡 섭취에 의해 ND군보다 유의하게 높아진 간 조직의 중성지방 및 총 콜레스테롤과 간 조직의 SREBP2 유전자 발현을 유의하게 낮추는 효과를 나타내었다 (p < 0.05). 또한 PSD군은 SD군보다 분변으로 배출되는 중성지방 함량을 14.8% 높였으며 (p > 0.05), 혈장과 간의 지질과산화를 SD군보다 각각 7.7%, 20.2% 낮추는 효과를 나타내었다 (p > 0.05). 혈장과 적혈구의 항산화활성 관련 지표를 분석한 결과 PSD군은 설기떡 섭취에 의해 낮아진 혈장 ORAC을 높이는 경향을 나타내었으며, 설기떡 섭취에 의해 유의하게 낮아진 간조직의 Mn-SOD 및 CAT 효소활성을 SD군보다 각각 21.5%, 19.6% 유의하게 높이는 효과를 나타내었다. 이상의 결과에서, 고지방식이로 이상지질혈증을 유도한 흰쥐에서 솔잎착즙액설기떡의 섭취는 체중증가억제, 혈장 콜레스테롤 감소, 간 내 콜레스테롤 생합성 억제, 간 조직의 지질수준 감소, 분변으로의 지질배출 증가, 간 조직의 지질과산화 감소, 혈장과 간 조직의 항산화 활성 증가 효능을 나타내는 것으로 확인되었다. 이에 본 연구에서 개발된 솔잎착즙액을 첨가한 설기떡은 지질 감소 효능을 가진 기능성 떡으로써 활용될 수 있을 것으로 기대된다.
본 연구에서는 LPS로 자극된 RAW 264.7 세포에서 생물전환 루모라고사리 추출물(B-RAE)의 항염증 효과 및 작용기전을 연구하였다. B-RAE의 총 폴리페놀과 총 플라보노이드 함량을 측정한 결과 379.26±7.77 mg/g과 50.85±3.08 mg/g으로 각각 나타났다. B-RAE의 항산화효과를 측정한 결과 DPPH, ABTS, superoxide anion radical을 농도의존적으로 소거하는 것으로 확인되었다. 또한, B-RAE는 세포생존에 영향을 미치지 않으면서 NO 생성을 처리 농도의존적으로 저해하였다. 전염증성 사이토카인(TNF-α, IL-1β, IL-6) 발현에 미치는 영향을 정량적 실시간 PCR로 측정한 결과 전염증성 사이토카인의 mRNA발현량을 LPS 처리군과 비교하여 B-RAE 처리 농도 의존적으로 유의성 있게 감소시키는 것으로 나타났다. 염증 관련 단백질(iNOS, COX-2)의 발현 및 전사인자인 NF-κB와 MAPK 신호경로 단백질의 인산화에 미치는 영향을 Western blot분석으로 평가하였다. 그 결과 LPS 처리에 의하여 증가된 iNOS와 COX-2의 발현을 유의적으로 억제하였다. 또한, LPS 처리에 의하여 증가된 NF-κB와 IκB의 인산화가 B-RAE 처리에 의하여 감소되는 것으로 나타났다. MAPK 신호경로 단백질의 인산화에 미치는 영향을 측정한 결과 ERK와 p38 MAPK 단백질의 인산화는 농도의존적으로 감소하는 것으로 나타났으나 JNK의 인산화는 증가하는 것으로 나타났다. 따라서 이러한 B-RAE의 항염증 효과는 높은 항산화 활성, iNOS와 COX-2 발현 억제를 통한 NO 생성 억제, NF-κB경로 저해, MAPK 신호경로 조절 및 전염증성 사이토카인 발현 저해에 의해 가능하다는 것을 제시한다.
The aim of this study was to clarify the origins of grubs which were used in the experiments, and to determine many other effects that are now written in the classical medical and herbal books. We compared the efficacy of grubs written in the herbal and medical books on the one hand, and studied the origins of grubs used in the experiments, on the other hand. The following results were obtained. There are three kinds of grub species for medicinal uses. Holorichia diomphalia Bates mainly produced in China, Cetoniidae species mainly produced in Korea, and Protaeria brevitarsis in Korean folk remedy are three species. Grubs were used in 25 experiments. Of these experiments, Holotrichia diomphalia were used 12 times, Larve of Protaeria brevitarsis 3 times, and Protaetia orientalis was used once. So, of total 25 experiments, 36%(9 experiments) did not clarify the origins of grubs used in the experiments. Currently, the grubs, which are imported from China and used in Korea, are Holotrichia diomphalia Bates from China. Current experimental study shows that grubs have efficacy for protecting liver. So the use of grubs in folk medicine is believed to be based on the scientific evidence. Efficacy of grubs, though it is not recorded in the medical books, has the effects of anti-cancer(anti-tumor), antioxidant, and anti-diabetes mellitus. The ingredients of Holotrichia are protein, peptide, fat, enzyme, mineral, and other several nutrients. Study on the grubs so far has a tendency to be done with little focus on the differen of origin of grubs. So, from now on, it needs that the origin of grubs be clarified, and the study on the efficacy from each origin be done. Considering the effects that the classical herbal books have recorded, it is necessary that more profound study proceed to prevent and treat diseases on eyes and ears, on the basis of the fact that grub has multiple ingredients to replenish the vital essence.
The objective of this study was to investigate the effects of L-carnitine on growth performance, organ weight, biochemical parameters of blood, heart and liver, and ascites susceptibility of broilers at different ages reared under a low-temperature environment. A total of 420 1-d-old male Ross 308 broilers were randomly assigned to two dietary treatments with fifteen replicates of fourteen broilers each. Treatment diets consisted of L-carnitine supplementation at levels of 0 and 100 mg/kg. At 11-d of age, low temperature stress was used to increase ascites susceptibility. Blood, heart and liver samples were collected at different ages for analysis of boichemical parameters. The results showed that, there was no significant difference in growth performance with L-carnitine supplementation, but the mortality due to ascites was significantly decreased. Dietary L-carnitine supplementation significantly reduced heart index (HI) and ascites heart index (AHI) on d 21, lung index (LUI) on d 35 and liver index (LI) on d 42. The broilers fed diets containing L-carnitine had significantly lower red blood cell counts (RBC), hemoglobin (HGB) concentration and hematocrit (HCT) on d 42. Dietary L-carnitine supplementation significantly reduced malondialdehyde (MDA) content of heart tissue on d 21 and 35, and significantly increased total superoxide dismutase (T-SOD) and Glutathione peroxidase (GSH-Px) activity of the heart on d 21 and 42. L-carnitine supplementation significantly reduced serum triglyceride (TG) content on d 28 and 35 and serum glucose (GLU) on d 35 and 42, and significantly increased serum total protein (TP) and globulin (GLO) content on d 42. L-carnitine supplementation significantly enhanced liver succinodehydrogenase (SDH), malic dehydrogenase (MDH) and $Na^+$-$K^+$-ATPase activity on d 28, and tended to reduce the lactic acid (LD) level of liver on d 35 (p = 0.06). L-carnitine supplementation significantly reduced serum uric acid (UA) content on d 28, 35 and 42. Based on the current results, it can be concluded that dietary L-carnitine supplementation reduced organ index, red blood cell counts and hematocrit, enhanced antioxidative capacity of the heart, enhanced liver enzymes activity involved in tricarboxylic acid cycle, and reduced serum glucose and triglyceride. Therefore, it is suggested that L-carnitine can potentially reduce susceptibility and mortality due to ascites.
에탄올은 사람에 대한 발암물질로 잘 알려져 있다. 또한 여러 조직이나 세포에서의 에탄올에 의한 세포증식억제효과도 잘 알려져 있다. 본 연구에서는 여러 암세포에서 에탄올에 의한 세포증식억제 효과를 조사하였는데 특히 발암원성 $ras$로 형질전환되거나 미세주입된 세포에서의 영향을 조사하였다. 에탄올은 여러 정상세포들의 증식을 억제하였다. 반면에 여러 암세포나 발암원성 Ras에 의한 세포증식은 억제하지 못 하였다. 또한 발암원성 단백질의 세포내 미세주사에 의한 DNA합성 유도도 에탄올에 의해 억제 되지 않았다. 이러한 에탄올의 세포증식억제 효과는 $N$-acetylcysteine이나 4-methylpyrazole과 같은 항산화제에 의해 제거되었다. 이러한 실험 결과는 에탄올에 의한 세포증식억제 효과는 Ras단백질의 upstream에 있거나 또는 Ras와 독립적으로 작용하며, 활성산소 형성과 밀접한 관계가 있다는 것을 알려준다.
The beneficial effects of tea catechins (TCs) are related not only to their antioxidant potential but also to the improvement of animal meat quality. In this study, we assessed the effects of dietary TC supplementation on plasma biochemical parameters, hormone responses, and glutathione redox status in goats. Forty Liuyang goats were randomly divided into four equal groups (10 animals/group) that were assigned to four experimental diets with TC supplementation at 4 levels (0, 2,000, 3,000 or 4,000 mg TC/kg DM feed). After a 60-day feeding trial, all goats were slaughtered and sampled. Dietary TC treatment had no significant effect on blood biochemical parameters, however, low-density lipoprotein cholesterol (p<0.001), triglyceride (p<0.01), plasma urea nitrogen (p<0.01), and glucose (p<0.001) decreased and total protein (p<0.01) and albumin (p<0.05) increased with the feeding time extension, and day 20 was the turning point for most of changes. Interactions were found in glutathione (p<0.001) and the ratio of reduced and oxidized glutathione (p<0.05) in whole blood between treatment and feeding time. Oxidized glutathione in blood was reduced (p<0.05) by 2,000 mg TC/kg feed supplementation, and a similar result was observed in longissimus dorsi muscle. Though plasma glutathione peroxidase (p<0.01) and glutathione reductase (p<0.05) activities were affected by treatment and feeding time interactions, and glutathione S-transferases activity increased with feeding day extension, no changed values appeared in longissimus dorsi muscle. In conclusion, dietary TC supplementation affected the concentrations of some blood metabolites and accelerated GSH depletion in the blood of goats. In terms of less high-density lipoprotein cholesterol, the highest insulin and IGF-I concentrations, the highest ratio of reduced and oxidized glutathione in plasma, the dosage of 2,000 mg TC/kg feed might be desirable for growing goats to prevent glutathione depletion and keep normal physiological metabolism.
본 연구에서는 재배조건(하우스재배, 노지재배), 수확시기(어린잎, 성잎) 열처리에 따른 영양성분 및 항산화활성의 변화를 비교 분석하여 국가표준성분표 발간을 위한 기초자료로 활용하고자 하였다. 일반성분 4종(수분, 지질, 회분, 단백질), 무기질 5종(칼슘, 철, 마그네슘, 칼륨, 나트륨), 비타민 3종(비타민 C, 엽산, 베타카로틴)의 영양성분을 분석하였고 폴리페놀 화합물과 DPPH 라디칼 제거능을 각각 측정하였다. 연구결과 일반성분의 경우 시료 간에 유의적인 차이를 나타내지 않았으나 무기질의 경우 Ca, Mg, Na은 하우스 재배가 노지재배 보다 유의적으로 높은 함량을 나타내었다. 비타민함량과 폴리페놀 함량 및 항산화 활성의 경우 수확시기보다는 재배방법과 열처리에 의해 함량변화가 큰 것으로 나타났다. 하우스보다 노지 재배된 갯기름나물의 비타민 C, 엽산, 폴리페놀 화합물과 항산화활성이 증가하였으며 열처리할 경우 비타민 C, 베타카로틴, 폴리페놀 화합물, 항산화활성이 각각 증가하였다. 비타민 C의 기존 보고된 이론과 상이한 결과가 도출되어 다양한 열처리온도와 시간을 적용한 성분 변화 연구가 필요할 것으로 생각된다. 베타카로틴과 폴리페놀 화합물의 열처리에 의한 증가는 산화가수분해 효소의 불활성화와 세포조직의 견고성 저하로 인한 활성물질의 추출 효율증가로 설명될 수 있다. 따라서 나물을 데쳐서 섭취할 경우 미생물 오염방지 등 안전성을 확보할 수 있을 뿐만 아니라 나물 세포조직이 연해져 베타카로틴과 같은 우리 몸에 유익한 성분의 소화흡수율이 증가될 것으로 생각된다.
일반재배 및 유기재배로 생산된 쌀의 품질 및 이화학적 특성, 기능성 및 관능평가에 대한 결과는 다음과 같다. 쌀 품질 분석 결과, 일반 쌀이 유기재배 쌀보다 6% 완전립 비율이 높고 쇄미 등 불완전립 비율이 낮았으며, 이화학적 특징인 아밀로스, 단백질, 취반미 윤기특성, 산가 등은 일반 쌀에 비해 유기재배 쌀이 약간 양호하였으나, 알칼리 붕괴도와 아미노산 Asp, Ser, Glu, Ala, Leu, Arg는 일반 쌀과 유기재배 쌀 간에 차이가 거의 없었다. 그러나 Thr, Val, Ile, Met, Phe, Lys, His 아미노산은 유기재배 쌀이 일반 쌀보다 함량이 많은 경향을 볼 수 있었다. 아미노산 성분중 일반 쌀과 유기재배 쌀의 무기성분 중 칼슘 같은 다량 성분은 차이가 없었지만, 붕소, 망간, 철, 아연 4가지 미량요소는 유기재배 쌀에서 1.1~2.7 mg/100 g 함량이 더 많았다. 기능성 조사 결과, 총 페놀화합물과 phytic acid 함량은 유기재배 쌀에서 89%와 23% 각각 더 많았고 항산화 활성도 유기재배 쌀에서 일반 쌀보다 4% 더 높게 나왔다. 관능평가 결과, 쌀 찰기, 맛, 식미감 분야에서 일반 쌀이 유기재배 쌀보다 밥맛이 우수한 것으로 조사되었다.
Cadmium, a human carcinogen, can induce toxicity in various cell lines and organs. Despite extensive research, the mechanisms of cadmium-induced cell toxicity and estrogenic potential in human are not clear. This study was performed to investigate cadmium-induced toxicity on human breast cancer cells: MCF-7 cells, an estrogen receptor (ER) positive breast cancer cells, and MDA-MB-231 cells, an ER negative breast cancer cells. MCF-7 cells was proved to be more sensitive than the other cell lines (IC50 = $50\;{\mu}M$ at MCF-7 cells and $120{\mu}M$ at MDA-MB-231). The expression of JNK and AP-1 transcription factors such as c-Jun and c-Fos dependent transcription were increased by cadmium treatment. Inhibition of ER activation by ER antagonist (tamoxifen or ICI 182,780) significantly recovered the viablity and inhibited apoptotic cell death. This suggested that cadmium-induced cell death in ER (+) cells was mediated by JNK/AP-1 pathway and this pathway was more stimulated by ER activated by cadmium. Co-treatment of antioxidants such as selenium (Se), butylated hydroxyanisole (BHA), glutathione (GSH), or N-acetyl-L-cysteine (NAC) recovered the cadmium-induced cell death in MCF-7 cells. Cadmium-induced lipid peroxidation was decreased by GSH, NAC, or BHA in MCF-7 cells. The expression of SOD protein was decreased by cadmium ($100{\mu}M$) but recovered by GSH, NAC, BHA, or Se. Our data showed that the cadmium-induced cell toxicity in human breast cancer cells could be protected by the antioxidants (Se, BHA, NAC, GSH, or NAC) and ER antagonist (tamoxifen or ICI 182,780). Therefore, toxicity of cadmium in breast cancer were mediated by oxidative stress and $ER{\alpha}$.
Background: Reactive oxygen species play critical roles in homeostasis and cell signaling. Dexmedetomidine, a specific agonist of the ${\alpha}2$-adrenoceptor, has been commonly used for sedation, and it has been reported to have a protective effect against oxidative stress. In this study, we investigated whether dexmedetomidine has a protective effect against $H_2O_2$-induced oxidative stress and the mechanism of $H_2O_2$-induced cell death in normal human fetal osteoblast (hFOB) cells. Methods: Cells were divided into three groups: control group-cells were incubated in normoxia without dexmedetomidine, hydrogen peroxide ($H_2O_2$) group-cells were exposed to $H_2O_2$ ($200{\mu}M$) for 2 h, and Dex/$H_2O_2$ group-cells were pretreated with dexmedetomidine ($5{\mu}M$) for 2 h then exposed to $H_2O_2$ ($200{\mu}M$) for 2 h. Cell viability and apoptosis were evaluated. Osteoblast maturation was determined by assaying bone nodular mineralization. Expression levels of bone-related proteins were determined by western blot. Results: Cell viability was significantly decreased in the $H_2O_2$ group compared with the control group, and this effect was improved by dexmedetomidine. The Hoechst 33342 and Annexin-V FITC/PI staining revealed that dexmedetomidine effectively decreased $H_2O_2$-induced hFOB cell apoptosis. Dexmedetomidine enhanced the mineralization of hFOB cells when compared to the $H_2O_2$ group. In western blot analysis, bone-related protein was increased in the Dex/$H_2O_2$ group. Conclusions: We demonstrated the potential therapeutic value of dexmedetomidine in $H_2O_2$-induced oxidative stress by inhibiting apoptosis and enhancing osteoblast activity. Additionally, the current investigation could be evidence to support the antioxidant potential of dexmedetomidine in vitro.

제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에

제 5 장 계약 해지 및 이용 제한

일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
