양파 열수추출물의 항산화능 및 인공소화후의 항돌연변이 효과 (Antioxidant and Antimutagenic Activities of Hot Water Extract from White and Yellow Onions after Simulated Gastric Digestion)

  • 양파 열수 추출물의 항산화능 및 인공소화 전과후의 항돌연변이 효과를 BHT와 ascorbic acid와 비교하여 조사하였다. 총 페놀 및 플라보노이드의 함량은 노란색 양파 추출물이 횐색 양파 추출물보다 많았으며, 양파 추출물의 과산화수소 소거능은 농도가 증가함에 따라 비례적으로 증가하였다. $\beta-carotene-linoleate\;system$에서 항산화능과 reducing power는 증가되었으나, BHT와 ascorbic acid보다는 낮았다. 양파열수 추출물의 인공소화 후에 IQ와 MNNG로 처리된 Salmonella typhimurium TA80과 TA100에 대하여 항돌연변이 효과가 나타났으며, 노란색 양파가 횐색 양파보다 높게 나타났다. 결론적으로 양파 추출물을 소화시키면 항산화 및 항돌연변이의 효과는 양파의 페놀과 플라보노이드의 성분 변화에 의한 것으로 판단된다.

  • The fruiting bodies of Paecilomyces japonica been used for an anticancer and immuno-stimulating agent as an oriental medicine, Antimutagenecity test with SOS chromotest and antioxidant test with NBT method were carried out using the concentrated culture broth, the extract of mycelia, and that of fruiting bodies. Among the sample extracts tested, the extract of fruiting bodies was most effective to antimutagenecity against the mutagens tested such as MNNG, ethidium bromide(EtBr), 2-aminofluorene(AF) and nitrofluorene(NF). Antimutagenic activity of all the extracts were very effective against mutagen, MNNG. When the extracts were added to certain concentration, antimutagenic activity was enhanced against mutagen, MNNG and NF. Antioxidant activity of the extract from fruiting bodies was highest. However, its activity was very low, compared to ascorbic acid.

토사자(Cuscutae Semen) 에탄올 추출물의 항산화 효과 및 항돌연변이 활성 (Antioxidative and Antimutagenic Activity of Ethanol Extracts from Cuscutae Semen)

  • 본 연구에서는 토사자 에탄올 추출물의 항산화 효과와 항돌연변이 활성을 분석하였다. 토사자 에탄올 추출물은 100 ppm, 200 ppm의 농도에서 BHT보다 높은 항산화 활성을 나타내었다. 토사자 추출물은 본 연구에서 사용한 농도에서는 돌연변이를 유발하지 않았으며, 직접돌연변이원인 4-NQO에 의해 유발된 돌연변이를 90% 이상 억제시키는 효과를 보여주어 우수한 항돌연변이능을 가지고 있는 것으로 나타났다. 또한 토사자 에탄올 추출물은 폴리페놀 화합물을 다량 함유하고 있어 이들 항산화 물질이 토사자의 항산화 효과와 항돌연변이 활성에 영향을 주었을 것으로 사료되었다. 본 연구 결과 토사자 에탄올 추출물은 항산화 효과와 함께 돌연변이 유발을 억제하는 항돌연변이 활성을 가지는 것으로 나타나 천연항산화제 및 고부가가치의 기능성식품 신소재로서 이용될 가능성을 보여주었다.

  • Against the mutagen MNNG and NOPD with SOS chromotest, the antigenotoxic activity of MeOH-soluble extract was much more effective than that of the water-soluble one. When the extract was added to the certain concentration, the antigenotoxic acivity was enhanced. Against the mutagen NOPD with Ames test, the antimutagenic activity of MeOH-soluble extract was better than that of the water-soluble one. The 60.4% of the inhibition ratio for the revertant colony-forming unit was shown at 5 mg of MeOH-soluble extract per plate. Antimutagenecity test with SOS chromotest and Ames test were performed using water-soluble and MeOH-soluble extracts from of Gleditsia sinensis. The antioxidant activity of MeOH-soluble extract with the NBT method was higher than that of the water-soluble one.

머루 과피와 종자 에탄올 추출물의 항산화 활성 및 항돌연변이 활성 분석 (Analysis of Antioxidative and Antimutagenic Activities of Ethanol Extracts from Pericarp and Seeds of Wild Grape (Vitis coignetiea))

  • The antioxidative activity and antimutagenic activity of the ethanol extracts from pericarp and seeds of wild grape (Vitis coignetiea) were analyzed in this study. The antioxidative activity of the extracts from wild grape was determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay. The antimutagenic activity of the extracts was evaluated on Salmonella typhimurium TA98 and TA100 by Ames test using 4-nitroquinoline 1-oxide (4-NQO) and sodium azide as mutagens. In the antioxidative activity determination, $IC_{50}$ values of the DPPH radical scavenging activity of the extracts from pericarp and seeds were 27.16 ppm and 7.61 ppm, respectively. Additionally, ABTS radical scavenging activities of pericarp and seed extract were 99.75% and 98.87% at 200 ppm, respectively. In the antimutagenic activity determination, pericarp extract at 5 mg/plate inhibited 72.6% and 74.3% of mutagenicity of S. typhimurium TA98 induced by 4-NQO and sodium azaid, respectively. Also, the mutagenicity inhibition rates of seed extract at 5 mg/plate were 77.8% and 74.5% in S. typhimurium TA100 induced by 4-NQO and sodium azaid, respectively. These results demonstrate that the ethanol extract from wild grape has remarkable antioxidant activity and antimutagenicity.

미나리와 돌미나리의 돌연변이 유발 억제작용과 항산화 효과 (Antimutagenic and Antioxidative Effects of Water Dropwort and Small Water Dropwort)

  • The purpose of this study is to research the antimutagenic and antioxidative effects of water dropwort and small water dropwort by Ames test and by measuring malondialdehyde(MDA) production. Water dropwort and small water dropwort were extracted with methanol and then further fractionated to hexane, chloroform, ethyl acetate, butanol and water, stepwise. The methanol extracts from both samples reduced the mutagenicities by aflatoxin $B_1(AFB_1)$ in Salmonella typhimurium TA 98. The production of MDA also decreased when the methanol extracts were added to the system. The hexane, chloroform and ethyl acetate fractions revealed higher antimutagenic activities against $AFB_1$ than the butanol and water fractions. Among the five fractions, the ethyl acetate fraction showed the highest level of antioxidant activity. From the results of the experiment, water dropwort and small water dropwort seem to be good antimutagenic and antioxidative sources of food.

버섯첨가된장의 생리활성 작용 (Physiological Activity in Doenjang Added with Various Mushrooms)

  • The study was carried out to evaluate the radical scavenging activity, and antimutagenic and anticancer effects in Korean soybean paste(doenjang) added with various mushrooms. Ganoderma lucidum doenjang(Gl-TD) showed significant inhibitory activity against the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, with an IC$\sub$50/ value of 245$\mu\textrm{g}$/$m\ell$. In contrast, the other doenjang varieties appeared to have weaker antioxidant activity. Four kinds of doenjang didn't have any antimutagenic activity against N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) by adding of 5mg/plate, but exhibited a strong inhibitory effect(55∼70%) against aflatoxin B$_1$(AFB$_1$). Especially Phellinus linteus doenjang(Pl-TD) inhibited more than 70% of the mutagenicity induced by AFB$_1$ in Salmonella typhimurium TA100. On the other hand, the mushroom doenjang varieties showed relatively weak activity toward MNNG in SOS chromotest, their inhibitory rate ranging from 23% to 33%. Methanol extracts of Gl-TD and Pl-TD inhibited by 91∼92% the growth of AGS gastric cancer cells in a concentration of 100$\mu\textrm{g}$/$m\ell$.

The Antimutagenic and Antioxidant Effects of Red Pepper Seed and Red Pepper Pericarp (Capsicum annuum L.)

  • In this study, we examined the antimutagenicity of red pepper seed and red pepper pericarp ethanol extracts using the standard Ames test in the presence and absence of S9 mix. The extracts showed inhibitory effects on both the TA98 and TA100 Salmenella Typhimurium strains against the mutagenic activity of promutagen 2-aminoanthracene, and were also protective against the directly acting mutagens sodium azide and 2-nitrofluorene. The red pepper seed elicited stronger antimutagenicity than the red pepper pericarp. Both the red pepper seed and red pepper pericarp directly quenched nitric oxide to different degrees and the scavenging activities increased with increasing concentrations. Nitric oxide scavenging activity ranged from $22{\sim}77%$ in the red pepper seed, and from $36{\sim}49%$ in the red pepper pericarp. The TEAC values for red pepper seed extract were $47.89{\pm}1.64mg\;g^{-1}$ in the ABTS radical scavenging assays, while those of red pepper pericarp extract were $94.18{\pm}1.61mg\;g^{-1}$. Therefore, we conclude that red pepper seed and red pepper pericarp have antimutagenic activities as well as antioxidant activity.

식물성 식품에 존재하는 Flavonoids의 항산화 활성 (Antioxidant Activity of Flavonoids in Plant Origin Food)

  • Effective synthetic antioxidants such as butylated hydroxyanisole(BHA) and butylated hydroxytoluene(BHT) have been widely used in the food industry, but they are suspected to be toxic and carcinogenic effects. Therefore, the development of safely available natural antioxidants such as ascorbic acid, ${\alpha}$-tocopherol, ${\beta}$-carotene, flavonoids and selenium is essential. In particular, flavonoids, 2-phenyl-benzo-${\alpha}$-pyrones, are polyphenolic compounds that occur ubiquitously in food of plant origin. flavonoids occur in foods generally as O-glycosides with sugars bound usually at the C\ulcorner position. And variations in their heterocyclic ring gibes rise to flavones, flavonols, flavanones, flavanols, catechins, anthocyanidins, chalcone and isoflavones. Vegetables, fruits, and beverages are the main dietary sources of the flavonols, primarily as quercetin, kaempferol, and myricetin and the corresponding flavones, apigenin and luteolin. These flavonoids have biological activity such as antioxidant, anti-inflammatory, antithrombotic, antimutagenic, anticarcimogenic antiallergic and antimicrobial activity effects in vitro and in vivo. Flavonoids posses strong antioxidant activities acting as oxygen radicals scavenger, metal chelators and enzyme inhibitor. The antioxidant activity of flavonoids is determined by their molecular structure and more specially, by the position and degree of hydroxylation of the ring structure. All flavonoids with the 3`, 4`-dihydroxy(ortho-dihydroxy) posses marked antioxidant activity. And antioxidant activity increases with the number of hydroxyl groups substituted on the A-and B-rings. There is as yet no certainty about the effect of the presence of a double bond between C\ulcorner and C\ulcorner on the antioxidant activity of flavonoids.

갈화(葛花)의 항돌연변이(抗突然變異) 활성(活性)에 관한 연구(硏究) (A Study on Antimutagenic acitivities of from Puerariae Flos Extracts)

  • Antigenotoxicity test (SOS chromotest), antimutagenecity test (Ames test), and antioxidant test (NBT method and xanthine-xanthine oxidase method) were carried out using water-soluble and methanolic extracts from Puerariae Flos. Against the mutagens MNNG and NQO, antigenotoxic activity of methanolic extracts were much more effective than that of water-soluble ones. When the methanolic extract was added to the certain concentration $(100{\mu}{\ell}/tube)$, antigenotoxic acivity against the mutagen MNNG was enhanced. Contrary to the water-soluble extract, the methanolic extract showed high antigenotoxicity against the mutagen NQO with increment of the extract. Against the mutagen MNNG with Ames test, antimutagenic activity of the methanolic extract at $300{\mu}{\ell}/tube$ was 96% as an inhibition ratio of revertant forming CFU/plate. The antioxidant activity of water-soluble extract was comparatively higher than that of the methanolic one.
