  • A wide array of natural products have been identified to possess potenital cancer chemopreventive properties. Of particular interest is Polyporus umbellatus frs.. that exhibits antimutagenic and cytotoxic effects. The crud drug "chorei" prepared from the dried fruit body of Polyporus umbellatus is used for kidney and other such disease. In the study, we are isolating the minor components of Chorei and examined their antimutagenic and cytotoxic effects.(omitted)

Melanoidin의 항산화성 및 항돌연변이원성 (Antioxidative and Antimutagenic Characteristics of Melanoidin Related Products)

  • Melanoidins, as brown colored polymers, are formed through the diversified reaction systems of Maillard type and other reactions. Melanoidins are important components in relation to food quality and also are known to have antioxidative, mutagenic and antimutagenic activities. Since these aspects have been extensively reviwed elsewhere, only the recent studies regarding to their antioxidative and antimutagenic activities are discussed in this review. Even though their mechanisms are not clearly identified, melanoidins or specific fractions isolated from their mixtures have shown varied antioxidative activities depending on the reaction systems and reaction conditions. Those activities presumely are derived from the complex functional properties of hydrogen / electron donors and metal chelating power, which are originated from their reductone structure and others. It is considered that pyrolysate and other mutagens are formed by the given conditions in some cases during browning reaction, whereas melanoidins and their fractions have antimutagenic effects on chemical and other mutagens. There are positive correlationship among the color intensity, antioxidative activity and antimutagenicity of melanoidins or their fractions. These suggest that the antimutagenicity of melanoidins could be attributed to their antioxidative properties, however, it might also be due to other factors, because the relevant responses for antimutagenicity are very complicate and not clear. Accordingly, further studies are required to determine the actual acitivities and mechanisms involved in antioxidation and (anti)mutagenicity of melanoidins by reaction systems / conditions and by the isolated fractions. And also, additional studies are needed to evaluate the applications of melanoidins and their relevant effects to food and human health.

Antimutagenic Activities of 24 Synthetic Flavones with The Salmonella Microsomal Assay

  • Twenty-four flavones were synthesized with various hydroxyl and/or methoxyl groups on A and B rings. Their antimutagenic properties were evaluated against ben:w(a)pyrene (BaP) and a pool of mutagenic urine concentrate (U) using a modified liquid incubation method of Ames test. The tester strain was Salmonella typhimurium TA98+S9 Mix. The antimutagenic activities were calculated by non linear regression analysis and the doses of flavones (in nmoles) required for a 50% reduction of induced revertants with BaP and U were defined as the inhibition doses (TEX>$ID_{508}{\;}and{\;}ID_{508}$ respectively). Seventeen flavones possessed significant antimutagenic activity against BaP. $ID_{508}$ ranged from 15.1 nmoles (F22) to 1000.6 nmoles (F13). Eighteen f1avones showed significant antimutagenic activity against U. $ID_{50U}$ ranged from 23.5 nmoles (F22) to 354.6 nmoles (F3). The 2',3',4'-trihydroxyflavone (F22, $ID_{508}=15.1$ nmoles, $ID_{50U}=23.5$ nmoles) and the 2',3',4',7-tetrahydroxyflavone (F20, $ID_{508}=37.8$ nmoles; $ID_{50U}=62.3$ nmoles) had antimutagenic activities similar to those of chlorophyllin ($ID_{508}=19.6$ nmoles and $ID_{50U}=44.2$ nmoles) and were evaluated against B(alP 7,8-dihydrodiol-9,10-epoxide. Against this last mutagen, the flavones which included three OH in B ring showed the highest activity and this property seemed independent of the substituent groups on A ring.

Bifidobacteria에 의한 항돌연변이 효과 (Antimutagenic Effects of Bifidobacteria)

  • 분리된 균주와 표준균주의 Bifidobacterium 21종류에 대하여 Salmonella typhimurium TA 98균주를 이용한 in vitro 항돌연변이능을 조사하였다. 실험에 이용된 돌연변이원으로는 Trp-P-1 (3-amino-1,4-dimethyl-$^{5}H-pyrido$ (4, 3-b) indole), benzopyrene, IQ (2-amino-3-methylimidazo [4,5-f] quinoline)와 NQO (4-nitroquinoline oxide) 등이었다. 동결건조된 균체들은 평균적으로 각각 Trp-P-1, benzopyrene, NQO, IQ에 대하여 64, 38, 29, 20%의 항돌연변이능을 나타냈다. Trp-P-1, benzopyrene, IQ에 대하여는 균주의 종류와 성장시기에 따른 항돌연변이능에 큰 차이는 없는 것으로 나타났다. NQO에 대하여는 12시간 배양세포가 5일 배양세포보다 항돌연변이능이 우수한 것으로 나타났다. 본 연구결과는 Bifidobacterium 균주들이 일반적으로 몇 종류의 잘 알려진 돌연변이원에 대한 항돌연변이능을 보유하고 있음을 보여주었다.

Antimutagenic Effect of Polysaccharides Extracted from Soybeans Fermented with Basidiomycetes on 2-Amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx)

  • The antimutagenic activity of polysaccharides extracted from soybeans fermented with Agrocybe cylindracea (AC) or Phellinus igniarius (PI) against 2-amino-3,8-dimethylimidaxo[4,5-f]quinoxaline (MeIQx) was examined using a Salmonella/Ames test and host-mediated assay in mice. The polysaccharides from the soybeans fermented with A. Cylindracea and P. igniarius inhibited the mutagenic acitivity of the cooked food mutagen, MeIQx, by 31.2% and 35.3%, respectively. The polysaccharides also inhibited MeIQx genotoxicity in a dose-dependent manner in micel. These results suggest that the polysaccharides from soybeans fermented with A. cylindracea or P. igniarius exhibit antimutagenic properties against MeIQx in vitro and in vivo.

수세미오이(Sponge-gourd: Luffa cylindrica L.) 추출물의 항산화, 항돌연변이 및 암세포 증식 억제 효과 (Antioxidative and Antimutagenic Effects and Hyperplasia Inhibitory Activity of Cancer Cells from Luffa cylindrica Extracts)

  • Considering the dearth of information regarding the medicinal properties of Luffa cylindrica, we assessed the antioxidative, antimutagenic and hyperplasia inhibitory activity of cancer cells from Luffa cylindrica extracts by employing biological and biochemical assays. Ethanol extracts of Luffa cylindrica inhibited MDA-BSA (malondialdehyde-bovine serum albumin) conjugation reaction ($66.38{\pm}2.65$), DPPH (1,1-diphenyl-2-picryl-hydrazyl) radical production ($60.13{\pm}0.42$) and lipid peroxidation ($56.04{\pm}3.24$). In this study, Luffa cylindrica is believed to exert possible antioxidative effects. The direct and indirect antimutagenic effects of the ethanol extracts of Luffa cylindrica were examined by the Ames test using Salmonella typimurium TA98 and TA100. The inhibitory effects on indirect and direct mutagenicity shows an weak tendency, particularly in direct mutagenicity mediated by 2-nitrofluorene in Salmonella typimurium TA98 ($5.82{\pm}5.74$) and in indirect mutagenicity mediated by 2-anthramine in Salmonella typimurium TA100 ($5.76{\pm}2.15$). The ethanol extracts of Luffa cylindrica on cancer cell hyperplasia inhibitory activity via MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay exerted cytotoxic effects on Hela cells ($55.83{\pm}3.83$) and MCF-7 cells ($33.03{\pm}2.09$), which were used in this study. Based on these results, it believed that the ethanol extracts of Luffa cylindrica have antioxidative capacities as well as hyperplasia inhibitory activity of cancer cells. Furthemore, Luffa cylindrica is a candidate for the prevention and dietetic treatment of chronic diseases and for the development of functional food.

Mutagenicity and Antimutagenicity of Hydrophilic and Lipophilic Extracts of Thai Northern Purple Rice

  • Purple rice (Oryza sativa L. var. indica) cv. Kum Doisaket is cultivated in northern Thailand. This study evaluated the mutagenic and antimutagenic properties of hydrophilic and lipophilic components of purple rice using the Ames test. The seed and hull of purple rice were extracted with hexane, methanol, ethanol, and water. The methanol extracts had the highest amounts of phenolic acids and flavonoids, while the hexane extracts contained large amount of tocols and ${\gamma}$-oryzanol. None of the extracts were mutagenic in Salmonella typhimurium strains TA98 and TA100. The hexane extract of rice hull and the methanol extract of rice seed were strongly effective against aflatoxin B1- and 2-amino-3, 4 dimethylimidazo (4, 5-f) quinoline-induced mutagenesis, while aqueous extracts showed weakly antimutagenic properties. All extracts with the exception of aqueous extracts enhanced the number of revertant colonies from benzo (a) pyrene induced-mutagenesis. None of the extracts inhibited mutagenesis induced by the direct mutagens 2-(2-furyl)-3-(5-nitro-2-furyl)-acrylamide and sodium azide. The hull extracts showed more potent antimutagenicity than the seed extracts. Based on a chemical analysis, ${\gamma}$-oryzanol and ${\gamma}$-tocotrienol in the hull and cyanidin-3-glucoside and peonidin-3-glucoside in the seed are candidate antimutagens in purple rice. The antimutagenic mechanisms of purple rice might be related to either modulation of mutagen metabolizing enzymes or direct attack on electrophiles. These findings supported the use of Thai purple rice as a cancer chemopreventive agent.

발효쪽 추출물의 생리적 기능과 염색특성(제1보) (A Study on the Physiological Effects and Dyeing Properties of the Extract of Fermented (Part I))

  • The purpose of this study was to investigate the antimicrobial activity, antimutagenic and anticancer effects and dyeing properties of the fermented indigo extract. The physiological effects of natural color extracts from colorant plants(gardenia, beet and indigo) were studied. The methanol extract of indigo showed an inhibitory effect on the growth of E. coli and Staph. aureus, and also showed a strong antimicrobial effect on Trich. mentagrophytes compared to others. The methanol extract of indigo showed antimutagenic activities against aflatoxin B1(AFB1) in the Ames test using Salmonella typhimurium TA 100. The proliferation of Clone M-3 mouse melanoma cells and A431 human epidermoid carcinoma cells was inhibited by the methanol extract of indigo. So we decided to use natural indigo for dyeing the fabrics because of those effects. Dried indigo leaves were fermented at variouss temperature and the fermented indigo was reduced by using alkaline(NaOH, Ca(OH)2) and glucose to dye the fabrics. The values of K/S fermented indigo showed the highest value when it was fermented at 3$0^{\circ}C$. The indigo fermented at 3$0^{\circ}C$ had the greatest number of total bacterial counts and we identified one of the main microorganisms as Aspergillus niger. This microorganism was responsible for the indigo fermentation and accelerated indigo fermentation. So it can be supposed to reduce the fermentation period of indigo by inoculating Aspergillus niger into the indigo leaves at 3$0^{\circ}C$.

아플라톡신에 대한 익모초의 돌연변이 억제 효과 (Desmutagenic Effect of Leonurus sibiricus L. to Aflatoxin B1 in Salmonella Mutation Assay)

  • By the 505 chromotest which utilized Escherichia bolt PQ 37, Korean medicinal plants had been screened to Investigate the antimutagenic effect to aflatoxin B1(AFBl). Ikmocho(IMC, Leonurus sibiricus L.) was extracted with hot water. The extract was not found to be mutagenic in the Salmonella mutation test with or without metabolic activation, and the extract was showed to possess the antimutagenic properties towards AFB1-induced metation. The mutagenicity of AFB1 was inhibited by methanol soluble fracstion (IMC-MS) in dose-dependent. However, water-soluble fraction exhibited comutagenic activity. The greatest inhibitory effect of IMC-MS on AFB1 mutagenicity occurred when IMC-MS was first incubated, AFB1 followed by a second incubation with the cells and 59 mixture. Also lower inhibition was occurred when S9 mixtures were first incubated, with IMC-MS followed by a second incubation with AFBI. The results of the sequential incubation study support the probability that one mechanism of inhibition could involve the formation of chemical complex between IMC-MS and AFB1 rather than deactivation of S9 enzyme.

Ames 실혐계 및 SOS Chromotest에서 Linolenic acid의 돌연변이유발 억제효과 (Inhibitory Effect of Linolenic Acid on the Mutagens-Induced Mutagenicities in Ames Assay System and SOS Chromotest)

  • To determine whether the omega 3 family, linolenic acis(LnA) is effective to inhibit carcinogens/mutagens-induced mutagenesis, we employed the Ames test using Salmonella typhimurium strain of TA100 and the SOS chromotest using Escherichia coli PQ37 strain. The inhibitory effect of LnA shown in the Ames assaying system was 95%, 78% and 73% when the mutagenicities were mediated by AFB$_{1}$, MNNG and 4-NQO, respectively. LnA shows a strong antimutagenic activity against indirect mutagen of AFB$_{1}$, whereas the same concentration of LnA exhibited weaker inhibitory effects on the direct mutagen of MNNG and 4-NQO than that of AFB$_{1}$. However. LnA reduced more than 80% of SOS responses induced by MNNG and 4-NQO when the adding concentration increased to 5%. We conclude that LnA contains in vitro antimutagenic properties and that this finding warrants further investigation both in vitro and in vivo to assess its possible chemotherapeutic potential.
