칡뿌리의 항균활성과 Isoflavonoid 화합물에 관한 연구 (A Study on the Screening of Antimicrobial Activity and the Isoflavonoids in Korean Arrowroot)

  • 칡뿌리를 이용한 건강식품의 개발 가능성을 탐색하고자 항균활성과 isoflavonoid 화합물을 분석한 결과는 다음과 같다. 항균활성은 칡뿌리의 methanol 추출물의 용매분획물중 에틸아세테이트 분획물에서 나타났으며 암칡보다는 숫칡에서 활성이 높고 그람 음성균에서 활성이 크게 나타났다. 그리고 ethyl-acetate분획물은 7~9% 수율로 얻어졌고 silical gel TLC에서 CH$_2$Cl$_2$ : $CH_3$OH=7 : 3 전개용매로 puerarin은 Rf치 0.60 daidzin은 Rf치 0.69를 나타내었으며 isoflavonoid중 purearin의 함량이 가장 많고 daidzin 그리고 daidzein은 소량 함유된 것으로 나타났으며 이들의 함량은 숫칡보다 암칡에서 높게 나타났다.

Identification of Novel Bioactive Hexapeptides Against Phytopathogenic Bacteria Through Rapid Screening of a Synthetic Combinatorial Library

  • Antimicrobial peptides (AMPs) are considered to be a promising alternative to conventional antibiotics for future generations. We identified four novel hexapeptides with antimicrobial activity: KCM11 (TWWRWW-$NH_2$), KCM12 (KWRWlW-$NH_2$), KCM21 (KWWWRW-$NH_2$), and KRS22 (WRWFIH-$NH_2$), through positional scanning of a synthetic peptide combinatorial library (PS-SCL). The ability of these peptides to inhibit the growth of a variety of bacteria and unicellular fungi was evaluated. KCM11 and KRS22 preferentially inhibited the normal growth of fungal strains, whereas KCM12 and KCM21 were more active against bacterial strains. Bactericidal activity was addressed in a clear zone assay against phytopathogenic bacteria, including Pectobacterium spp., Xanthomonas spp., Pseudomonas spp., etc. KCM21 showed the highest activity and was effective against a wide range of target organisms. Application of KCM21 with inoculation of Pectobacterium carotovorum subsp. carotovorum on detached cabbage leaves resulted in an immune phenotype or a significant reduction in symptom development, depending on the peptide concentration. Cytotoxicity of the four hexapeptides was evaluated in mouse and human epithelial cell lines using an MTT test. The results revealed a lack of cytotoxic effects.

Isolation, structure elucidation and physicochemical properties of novel antibiotic polypeptide, $\varepsilon-(L-\beta-Iysine)$ polypeptide from Streptomyces sp. DWGS2

  • During the screening of material which has the antimicrobial activity against aminoglycoside-resistant bacteria, A new material $\varepsilon$-(L-$\beta$-Iysine) polypeptide from a culture medium of Streptomyces sp.(DWGS2) was isolated, and the structure and the physicochemical properties of the new material were elucidated. The new material was separated by column chromatography of the culture medium using Dowex1$\times$2, Silica gel, and Sephadex LH20 etc. The chemical structure and molecular weight were determined with the data of various NMR experiments, MALDI mass, and ESI mass experiments. The antimicrobial activity of $\varepsilon$-(L-$\beta$-Iysine) polypeptide is not only better than equal to the activity of known aminoglycoside type of antibiotics(MIC=3.125 - 6.25ug/mL) but also effective against aminoglycoside-resistant bacteria and fungi. If the mechanism of antimicrobial activity against aminoglycoside- resistant bacteria is figured out, the $\varepsilon$-(L-$\beta$-Iysine) polypeptide can be utilized for the treatment of diseases caused by aminoglycoside-resistant bacteria.

국내 분리 방선균의 항균활성 특성

  • It is well known that actinomycetes would be useful for screening of biologically active compounds. Not only to isolate diverse actinomycete strains but to ferment those strains effectively would be important. Seven hundred and forty six strains were isolated from Cheju province, 216 strains were from Chungnam province, 158 strains were from the natural caves at Chungbuk and Kangwon provinces and 202 strains were from Chungwon area at Chungbuk province. All of these 1,322 strains were fermented on a small scale using two different media and tested for their antimicrobial activities against four bacterial strains and one yeast strain. As the result, 12.3% of those isolates were active against Staphylococcus aureus KCTC 1916, 7.6% were Staphylococcus aureus KCTC 1928, 3.9% were Escherichia coli KCTC 1924, 3.0% were Candida albicans KCTC 1940, and 2.2% were Salmonella typhimurium KCTC 1926. About 40% of those isolates showed antimicrobial activities at both two media but the others showed at either one. According to the genus of isolated strains, Streptomyces and Micromonospora showed activities with higher frequencies than others.

Isolation of methicillin-resistant Staphylococcus aureus from a Shih-Tzu dog with canine distemper virus infection

  • A methicillin-resistant Staphylococcus aureus (MRSA) isolate was recovered from a 9-month-old female Shih-Tzu dog with canine distemper virus infection. We performed in vitro antimicrobial susceptibility test to determine the most effective antimicrobial drug against the isolate and thus, to emphasize its potential clinical importance in animal practices. Isolate was confirmed MRSA by oxacillin agar screening test. The isolate was fully resistant to all $\beta$-lactam antibiotics and was susceptible to glycopeptides. Of the other antibiotics, mupirocin, TMP/SMZ (trimethoprim-sulfamethoxazole), and chloramphenicol showed inhibitory effect at the concentration of 4x MIC. The MICs ranged 0.25->$128{\mu}g/ml$, and MBCs ranged 0.5->$128{\mu}g/ml$. The combined TMP/SMZ with cefamandole or novobiocin showed synergistic effect, whereas the combination of novobiocin plus cefamandole or teicoplanin resulted in antagonistic effects. Although MRSA in animals so far has been reported in the geographically limited countries, at least theoretically, it could be occurred in the future more frequently through either human or animal origin. The use of this combination may be of value in this situation. As with all antimicrobial agents, inappropriate or unnecessarily prolonged therapy may contribute to the emergence of resistance strains and loss of efficacy.

Biological Screening of a Novel Nickel (II) Tyrosine Complex

  • A newly synthesized Nickel (II) tyrosine complex was screened as potential antimicrobial agent against a number of medically important bacteria (Bacillus subtilis, Streptococcus ${\beta}$-haemolytica, Escherichia coli, Shigella dysenterae) and fungi (Aspergillus fumigatus, Candida albicans, Aspergillus niger, Aspergillus flavus, Penicillium sp.) strains. were used for antifungal activity. The antimicrobial activity was evaluated using the Agar Disc method. Moreover, the minimum inhibitory concentration of the complexes was determined against the same pathogenic bacteria and the values were found between $4{\sim}64\;{\mu}g\;ml^{-1}$. Brine shrimp bioassay was carried out for cytotoxicity measurements of the complexes. The $LC_{50}$ values were calculated after probit transformation of the resulting mortality data and found to be 6 ${\mu}g\;ml^{-1}$.

Chitinase-producing Salinivibrio bacteria isolated from salt-fermented shrimp with antimicrobial and safety assessments

  • Chitinases are glycosyl hydrolases which cleave the ${\beta}$-1,4 linkage of chitin into oligo or monomers of N-acetylglucosamine. These bacterial enzymes have been used for a wide range of applications in the food and pharmaceutical industries. In this study, we isolated two potential chitinolytic strains, BAO-01 and BAO-02, from salt-fermented shrimp, which were shown to belong to the genus Salinivibrio through genetic characterization using 16S rRNA. These isolates were gram-positive, rod-shaped, and non-spore forming. BAO-01 showed greater growth and chitinase activity than BAO-02 after the incubation at $37^{\circ}C$ for 4 days. Both strains grew on a wide range of carbon and nitrogen sources, pH values, temperatures, and salt levels. However, they showed minor biochemical differences. In addition, their antimicrobial activities against foodborne pathogens and antibiotic susceptibilities were evaluated. These Salinivibrio spp. did not show bioamine production, hemolytic activity, and mucin degradation. Therefore, the in vitro screening results suggested that these bacteria could be widely used as new candidates for chitin hydrolyzation and seafood fermentation.

In-vitro Antimicrobial Activity Phytochemical and Cytotoxicity of Methanolic Fruits Extract of Capsicum frutescent

  • Capsicum frutescen is known in Sudan, is one of the most commonly used pepper species in cooking and in Sudanese folk medicine. The present study was conducted to investigate antimicrobial (bacteria and fungi) and cytotoxicity (Brine Shrimp Lethality Test) of methanolic extract of Capsicum frutescen (fruits). The extract have been tested in the present study to investigate the in vitro potential effects against Gram positive, Gram negative bacteria and fungi. The selected organisms were Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumonia and Candida albicans using the cup plate agar diffusion method. The methanol extract of Capsicum frutescen (fruits) exhibited inhibitory effects against Escherichia coli with zone of inhibition (23 mm) and Klebsiella pneumonia with zone of inhibition (17 mm). The phytochemical screening revealed the presence of Tannins, Saponin, Alkaloids, Anthroquinoles and Terpenoids. The Cytotoxicity of methanolic extract of Capsicum frutescens was $LD_{50}$ $64.68{\mu}g/ml$. The activity and presence of compounds known to be biologically active are a validation for the use of Capsicum as a food ingredient and as a therapeutic element of traditional medicine.

Antimicrobial Activities of White, Red, and Extruded Ginsengs with Different Extraction Conditions

  • White, red, and extruded ginsengs were studied against 8 strains of food-borne pathogens and/or food spoilage microorganisms. The ginseng powders were extracted with different extractants and screened for antimicrobial activity using the disc diffusion and broth dilution techniques. The results showed that the yield of extraction was higher with increase of aqueous solution content and temperature. Preliminary screening revealed that the red ginseng extracts were most active, that has been found to be highly effective against all tested microbe except Listeria monocytogenes. Moreover, Bacillus subtilis has shown highly susceptible, which the diameters of inhibition zone values of 28 extracts were between 7 and 14 mm. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) recorded for the different crude ginseng extracts against microorganism using ranged from 6.25 to 100 mg/mL, indicated that the methanol extract of ginseng were more effective than ethanol and water extracts. The 60% methanol extract of red ginseng had the greatest effects against B. subtilis with MIC and MBC values at 6.25 mg/mL.

Detection and Identification of Bacteriocins Produced by Propionibacteria Isolated from Commercial Swiss Cheese Products

  • Wild propionibacteria isolated from different commercial swiss cheese samples were tested for antimicrobial activities. In initial screening, six of these Propionibacterium isolates showed antagonistic activity against 10 selected indicator organisms by the deferred method. In next, only two Propionibacterium strains JW6 and JW14 showed antibacterial activity in the cell-free supernatants by the modified well diffusion method. Propionibacterium strains JW6 and JW14 were finally identified as bacteriocin producers which exhibited a bactericidal effect against closely related species. The antimicrobial substances were proteins, since their activities were completely destroyed following several degradative enzyme treatments. The bacteriocins showed a narrow inhibitory spectrum of activity against two propionibacteria and two bacilli of strains tested in this study.
