약용 식물 첨가 된장의 숙성 중 생리기능성의 변화 (Changes of Physiological Functionalities during the Fermentation of Medicinal Herbs Doenjang)

  • 재래식 된장의 품질을 고급화하기 위해 재래식 된장과 각종 한약제와 약초류를 달리 첨가하여 제조한 두 종류의 약용 된장들을 6개월간 숙성시키면서 성인병에 관련된 몇 가지 생리 기능성의 변화를 조사하였다 약용 된장들의 고혈압을 예방하는 엔지오텐신 전환효소(ACE) 저해활성은 숙성기간이 길어짐에 따라 급격히 감소하여 숙성 6개월 후에 39.0%∼51.7%을 보였고 혈전용해활성은 숙성 6개월후에 10.4∼11.3 U를 보였다. 약용 된장들의 전자공여능은 제조직후 95%∼97%을 보였고 숙성 기간이 길어짐에 따라 큰 변화가 없었으며 SOD 유사활성은 숙성 6개월 후에 모두 10% 내외를 보였다. 약용 된장(I)의 tyrosinase 저해활성은 숙성 기간이 길어짐에 따라 증가하여 숙성 6개월 후에 상온물 추출물에서 97.6%의 높은 활성을 보였다. 6개월 숙성된약용 된장들의 조단백질과 총당 함량은 각각 14%와 33%로 재래식 된장보다 높았다.

  • 윌름씨 종양은 주로 1-5세에 복부에 생기는 종양으로 약 25% 이상에서 고혈압을 동반하지만 심각한 고혈압에 의하여 심부전을 보이는 경우는 매우 드물다. 저자들은 3세된 여자 아이가 윌름씨 종양으로 인한 고레닌 혈증으로 다음, 다뇨, 저나트륨성 고혈압과 심부전을 보였으나 수술적으로 종양을 제거한 후 증상 호전을 보였기에 문헌 고찰과 함께 보고하는 바이다.

Expression and Purification of an ACE-Inhibitory Peptide Multimer from Synthetic DNA in Escherichia coli

  • An angiotensin I-converting enzyme (EC 3.4.15.1) (ACE), which can convert inactive angiotensin I into angiotensin II, a vasoconstrictor, is one of the key enzymes in controlling hypertension. It is suggested that the inhibition of ACE prevents hypertension, and many inhibitory peptides have already been reported. In the current study, oligonucleotides encoding ACE inhibitory peptides (IY, VKY) were chemically synthesized and designed to be multimerised due to isoschizomer sites (BamHI, BglII). The cloned gene named AP3 was multimerised up to 6 times in pBluescript and expressed in BL2l containing pGEX-KG. The fusion protein (GST-AP3) was easily purified with a high recovery by an affinity resin, yielding 38 mg of synthetic AP3 from a 1-1 culture. The digestion of AP3 by chymotrypsin exhibited an $IC_50$ value of $18.53{\mu}M$. In conclusion, the present experiment indicated that AP3 could be used as a dietary antihypertensive drug, since the potent ACE inhibitory activity of AP3 could be activated by chymotrypsin in human intestine.

  • $(R)-\beta-acetylmercaptoisobutyric$ acid (RAM), a chiral compound, is an important intermediate for the chemical synthesis of various antihypertensive and congestive heart failure drugs. Microorganisms capable of converting $(R,S)-\beta-acetylmercaptoisobutyric$ acid ((R,S)-ester) to RAM were screened from soil microorganisms. A strain of Pseudomonas sp. 1001 screened from a soil sample was selected to be the best. Cells showed an activity of 540 U/mL from culture broth and the enzyme was thermostable up to $70^{\circ}C$. This strain could produce RAM asymmetrically from (R,S)-ester.

오징어(Todarodes pacificus) 껍질로부터 Angiotensin I 전환효소 저해 펩티드의 분리 정제 (Purification of Angiotensin I-Converting Enzyme Inhibitory Peptide from Squid Todarodes pacificus Skin)

  • In this study, an angiotensin I-converting enzyme (ACE) inhibitor from squid skin was purified and characterized. Squid (Todarodes pacificus) skin protein isolates were hydrolyzed using six commercial proteases: alcalase, ${\alpha}$-chymotrypsin, neutrase, papain, pepsin, and trypsin. The peptic hydrolysate had the highest ACE inhibitory activity. The ACE inhibitory peptide was purified using Sephadex G-25 column chromatography and reverse phase high-performance liquid chromatography (HPLC) with a $C_{18}$ column. The purified ACE inhibitory peptide was identified and sequenced, and found to consist of seven amino acid residues: Ser-Ala-Gly-Ser-Leu-Val-Pro (657Da). The $IC_{50}$ value of the purified ACE inhibitory peptide was 766.2 ${\mu}M$, and Lineweaver-Burk plots suggested that the purified peptide acts as a noncompetitive ACE inhibitor. These results suggest that the ACE inhibitory peptide purified from the peptic hydrolysate of squid skin may be of benefit in developing antihypertensive drugs and functional foods.

THE EFFECT OF GINSENG ON BLOOD PRESSURE IN SPONTANEOUSLY HYPERTENSIVE RAT AND ESSENTIAL HYPERTENSION

  • To investigate the effect of ginseng on blood pressure (B.P.) in spontaneously hypertensive rat (SHR) and essential hypertension ginseng extract was given per se daily in 58 SHR and 35 essential hypertensive patients. SHR were divided into 5 groups according to the dosage of ginseng. In essential hypertension 1,000mg of ginseng extract was given. The B.P. in SHR was measured by tail cuff method. In essential hypertension side effect and changes in various laboratory examinations were evaluated. In SHR ginseng appeared to have hypertensive effect when it is given in small amount(10mg/kg). However, when it is given 60mg/kg/day or more difinite B.P. lowering effect was observed. The hypotensive effect was dosedependant and it lasted for 37days of observation. In essential hypertension in 12 $(80\%)$ among 15 patients hypotensive effect was seen with ginseng administration along and the effect lasted for 12 weeks. In the rest of hypertensive patients it is required addition of diuretics of other antihypertensive drugs to decrease B.P., no appreciable side effect was seen. In laboratory examinations no significant changes were seen except for serum cholesterol, ${\alpha}-and\;{\beta}-lipoprotein,$ and hematocrit. There was some evidence of relationship between plasma renin activity (PRA) and ginseng in hypotensive action.

Screening for Angiotensin 1-Converting Enzyme Inhibitory Activity of Ecklonia cava

  • Seven brown algal species (Ecklonia cava, Ishige okamurae, Sargassum fulvellum, Sargassum horneri, Sargassum coreanum, Sargassum thunbergii and Scytosiphon lomentaria) were hydrolyzed using five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase) and screened for angiotensin 1-converting enzyme (ACE) inhibitory activities. Most algal species examined showed good ACE inhibitory activities after the enzymatic hydrolysis. However, E. cava was the most potent ACE inhibitor of the seven species. Flavourzyme digest of E. cava exhibited an $IC_{50}$ of around $0.3\;{\mu}g/mL$ for ACE; captopril has an $IC_{50}$ of $\~0.05\;{\mu}g/mL$. The Flavourzyme digest was separated to three fractions by an ultrafiltration membrane (5, 10, 30 kDa MWCO) system according to the molecular weights. The active components were mainly concentrated in >30 kD fraction which are composed of the highest protein content $(27\%)$ and phenolic content (261 mg/100 mL) compared to the other two smaller molecular weight fractions. Therefore, the active compounds appear to be relatively high molecular weight complex molecules associated with protein (glycoprotein) and polyphenols. Therefore, E. cave is a potential source of antihypertensive compound.

  • As the average span of human life is continuously increasing, especially the old aged groups, are suffering from various chronic and critical diseases e.g. cardiovascular diseases, coronary heart disease, diabetes, atherosclerosis and alzheimer's etc. However, effective and safe treatment methods have not yet been investigated threathening old aged groups. This research was planned to isolate compounds with the therapeutic potential for the above mentioned chronic diseases from the mulberry leaves. Biological screenings were carried out for the following categories; anti-atherogenic, anti-diabetic and antihypertensive effects. The results were as follows; Mulberry leaves, 20% $\alpha$-treated Gaelyrangppong showed significant 81% of blood glucose lowering effects in alloxan-induced hyperglycemic mice. Particularly, butanol-soluble fractions of mulberry leaves showed the more significant hypoglycemic activity than other fractions in alloxan induced hyperhlycemic mice. Also in the group given 1g/kg doses of extract of mulberry leaves, total cholesterol level was decreased significantly by as much as 49% in hyperlipidemia-induced rats. In Mulberry leaves post-treated group, the atheroscelosis index, HDL-cholesterol/Total-cholesterol, was increased significantly by as much as 91%.

굴 효소 가수분해물의 angiotensin converting enzyme 저해작용 (Angiotensin-Converting Enzyme Inhibitory Activity of Enzymatic Hydrolysates of Crassostrea gigas (Oyster))

  • 굴 효소 가수분해물은 굴을 alcalase, protamex, neutrase, flavourzyme, pepsin으로 각각 처리하였고, 이들의 ACE 저해활성을 측정하였다. ACE 저해활성은 가수분해 시간에 따라 증가 또는 감소하였으며, 그 중 10시간 이상 가수분해 처리한 PEH에서 가장 높은 ACE 저해활성을 나타내었다. PEH 가수분해물을 한외여과(30, 10 kDa) 하여 낮은 분자량의 분획물을 얻었다. 30 kDa와 10 kDa 이하 분자량 별 ACE 저해활성은 각각 $69.18{\pm}0.75$, $83.71{\pm}1.12$의 활성을 나타내었다. 이 결과 10 kDa 이하의 시료로부터 HPLC column (watchers 120 ODS-AP $250{\times}4.6$ ($5{\mu}m$))을 이용하여 각각의 활성 분획을 분취하여 얻은 6분획의 ACE 저해 활성은 29.56~85.85% 로 나타났다. 그 중 저해활성이 가장 높게 나타나는 B fraction의 아미노산 서열을 확인한 결과 Leu-Gln-Pro 임을 확인하였고, peptide합성하여 얻은 저해활성 농도($IC_{50}$)가 1.18 uM임을 확인하였다. 이러한 결과는 PEH를 이용하여 건강 기능성 식품의 개발에 도움이 될 수 있을 것으로 생각된다.

폴리페놀 화합물과 항생제의 상호작용 (Interaction with Polyphenols and Antibiotics)

  • 항산화, 항알러지, 항염증, 항암, 항고혈압, 항균 활성을 나타내는 폴리페놀 화합물은 고등식물에 의해 생산되는 2차 대사산물이다. 일반적으로 폴리페놀 화합물은 플라보노이드와 비 플라보노이드 화합물로 나뉘며 병원균에 대한 항균활성은 비 플라보노이드 화합물에 비해 플라보노이드 화합물이 우수하다. 항균활성을 나타내는 플라보노이드 화합물은 칼콘, 플라반-3-올(카테킨), 플라바논, 플라본, 플라보놀, 프로안토시아니딘 등이며 플라보놀 화합물은 그람 음성 세균(Fusobacterium nucleatum, Porphyromonas gingivalis, Prevotella melaninogenica, Prevotella oralis)과 그람 양성 세균(Actinomyces naeslundii, Lactobacillus acidophilus, Staphylococcus aureus)에 대해 항균활성을 나타낸다. 비 플라보노이드 화합물 중에서 항균활성을 나타내는 화합물은 커피산, 갈릭산, 페룰산 등이며 그람 양성 세균(Listeria monocytogenes, S. aureus)과 그람 음성 세균(Escherichia coli, Pseudomonas aeruginosa)에 대해 젠타마이신, 스트렙토마이신보다 우수한 항균활성을 나타낸다. 플라본 화합물인 캠퍼롤과 퀘세틴은 단독 사용보다는 리팜피신, 시플록사신과 병용하였을 떄 S. aureus와 메티실린 내성 S. aureus (MRSA)에 대해 시너지 효과를 나타내었고, 에피갈로카테킨 갈레이트(EGCG)는 베타락탐계 항생제와 병용했을 때 MRSA 에 대한 대해 시너지 효과를 나타내었다. 특히 한국 녹차에서 분리된 에피카테킨, 에피갈로카테킨, 에피갈로카테킨 갈레이트, 갈로카테킨 갈레이트 등의 차 폴리페놀류는 임상에서 분리한 MRSA에 대해 항균활성을 나타내었고 옥사실린과 병용했을 때 시너지 효과를 나타내었다.
