Secondary metabolites in the culture filtrates of lactic acid bacteria offer varied chiral moieties, making them a valuable resource for drug design scaffolding. Our previous methodology included using a combination of anion exchange resins, Amberlite IRA-67 and Purolite A420S, to purify significant quantities of Lactobacillus plantarum LBP-K10 peptidyl compounds. However, current experimental evidence regarding the impact of native culture extracts and/or filtrates on pathogenic fungi in vivo/in vitro is insufficient. This study analyzed the antifungal properties of two different probiotic cultures: the CH2Cl2-extracted filtrate of Chinese cabbage kimchi (CH2Cl2-extracted CCKWLB and CH2Cl2-extracted CCKWOLB) and the non-extracted filtrate of Chinese cabbage kimchi (non-extracted CCKWLB and non-extracted CCKWOLB). The samples were divided into two groups: one group was inoculated with probiotics while the other group remained non-inoculated. Filtrates from both experimental groups were utilized for antifungal assays. The treatments employing CCKWLB, with an initial inoculation of Lb. plantarum LBP-K10 as a starter, demonstrated significant antifungal activity under various experimental conditions. Our study offers new perspectives on the antifungal properties of CH2Cl2-extracted kimchi filtrates, which are naturally produced by lactobacilli. The efficacy of antifungal compounds is supported by substantial evidence demonstrating their efficient uptake by cells and the antifungal properties exerted by metabolites.
Bacillus amyloliquefaciens is an important plant disease-preventing and growth-promoting microorganism. B. amyloliquefaciens WS-8 can stimulate plant growth and has strong antifungal properties. In this study, we sequenced the complete genome of B. amyloliquefaciens WS-8 by Pacific Biosciences RSII (PacBio) Single Molecule Real-Time (SMRT) sequencing. The genome consists of one chromosome (3,929,787 bp) and no additional plasmids. The main bacteriostatic substances were determined by genome, transcriptome, and mass spectrometry data. We thereby laid a theoretical foundation for the utilization of the strain. By genomic analysis, we identified 19 putative biosynthetic gene clusters for secondary metabolites, most of which are potentially involved in the biosynthesis of numerous bioactive metabolites, including difficidin, fengycin, and surfactin. Furthermore, a potential class II lanthipeptide biosynthetic gene cluster and genes that are involved in auxin biosynthesis were found. Through the analysis of transcriptome data, we found that the key bacteriostatic genes, as predicted in the genome, exhibited different levels of mRNA expression. Through metabolite isolation, purification, and exposure experiments, we found that a variety of metabolites of WS-8 exert an inhibitory effect on the necrotrophic fungus Botrytis cinerea, which causes gray mold; by mass spectrometry, we found that the main substances are mainly iturins and fengycins. Therefore, this strain has the potential to be utilized as an antifungal agent in agriculture.

Since the mass mortality of Quercus mongolica has been first reported in Gyeonggi province at 2004, the disease spread rapidly over Korean peninsula annually. Ambrosia beetle (Platypus koryoensis) was known as the insect vector of oak wilt fungus, Raffaelea quercus-mongolicae, and control methods of the disease had mainly been focused on eradication of insect vector. However, for the efficient management of the disease, combined control methods for both of the pathogenic fungus and insect vector are strongly required. As one of the efforts to suppress the pathogenic fungus, antifungal activities of Streptomyces isolated from oak forest soil were assayed in this study. Optimum culture condition for the selected isolates was also studied, As a result, Streptomyces blastmyceticus cultured in PDB (Potato Dextrose Broth) at $25^{\circ}C$ for 1 week showed the strongest antifungal activity against oak wilt fungus. Mycelial growth inhibition rates (MGIRs) of Streptomyces isolates were compared on culture media supplemented with heated and unheated culture filtrates of S. blastmyceticus. MGIRs on culture media with unheated culture filtrates were generally higher than those on culture media with heated culture filtrates. Antagonistic mechanism to get involved in the inhibition of hyphal growth and spore formation of the pathogen is due to the antifungal metabolites produced by Streptomyces. This study will provide the fundamental information in developing biocontrol agents for the environment-friendly management of oak wilt disease.
The antifungal activity of isolated lactic acid bacteria (LAB) from a locally fermented cereal, "Kunu", was tested against toxigenic Aspergillus flavus. The liquid refreshment, "Kunu", was prepared under hygienic condition using millet, sorghum, and the combination of the two grains. The antifungal potential of isolated LAB against toxigenic A. flavus was carried out using both in vitro and in vivo antifungal assays. The LAB count from prepared "Kunu" ranged from $2.80{\times}10^4CFU/mL$ to $4.10{\times}10^4CFU/mL$ and Lactobacillus plantarum, Lactobacillus delbrueckii, Lactobacillus fermentum, Pediococcus acidilactici, and Leuconostoc mesenteroides were the isolated bacteria. Inhibitory zones exhibited by LAB against toxigenic A. flavus ranged from 5.0 mm to 20.0 mm. The albino mice infected with toxigenic A. flavus showed sluggishness, decrease in body weight, distortion of hair, and presence of blood in their stool, while those treated with LAB after infection were recovered and active like those in control groups. Except for the white blood cell that was increased in the infected mice as $6.73mm^3$, the packed cell volume, hemoglobin, and red blood cell in infected animals were significantly reduced (P<0.05) to 29.28%, 10.06%, and 4.28%, respectively, when compared to the treated mice with LAB and control groups. The antifungal activity of LAB against toxigenic A. flavus can be attributed to the antimicrobial metabolites. These metabolites can be extracted and used as biopreservatives in food products to substitute the use of chemical preservatives that is not appealing to consumers due to several side effects.
In our previous studies, Bacillus megaterium KU143, Microbacterium testaceum KU313, and Pseudomonas protegens AS15 have been shown to be antagonistic to Aspergillus flavus in stored rice grains. In this study, the biocontrol activities of these strains were evaluated against Aspergillus candidus, Aspergillus fumigatus, Penicillium fellutanum, and Penicillium islandicum, which are predominant in stored rice grains. In vitro and in vivo antifungal activities of the bacterial strains were evaluated against the fungi on media and rice grains, respectively. The antifungal activities of the volatiles produced by the strains against fungal development and population were also tested using I-plates. In in vitro tests, the strains produced secondary metabolites capable of reducing conidial germination, germ-tube elongation, and mycelial growth of all the tested fungi. In in vivo tests, the strains significantly inhibited the fungal growth in rice grains. Additionally, in I-plate tests, strains KU143 and AS15 produced volatiles that significantly inhibited not only mycelial growth, sporulation, and conidial germination of the fungi on media but also fungal populations on rice grains. GC-MS analysis of the volatiles by strains KU143 and AS15 identified 12 and 17 compounds, respectively. Among these, the antifungal compound, 5-methyl-2-phenyl-1H-indole, was produced by strain KU143 and the antimicrobial compounds, 2-butyl 1-octanal, dimethyl disulfide, 2-isopropyl-5-methyl-1-heptanol, and 4-trifluoroacetoxyhexadecane, were produced by strain AS15. These results suggest that the tested strains producing extracellular metabolites and/or volatiles may have a broad spectrum of antifungal activities against the grain fungi. In particular, B. megaterium KU143 and P. protegens AS15 may be potential biocontrol agents against Aspergillus and Penicillium spp. during rice grain storage.
Taiwan, formerly known as Formosa, located on tropical and subtropical climate zones with abundant biological resources. According to the latest version of the Flora of Taiwan, there are 4339 species of vascular plants including 1054 endemic species. First, Taiwania (Taiwania cryptomerioides), named after its native island of Taiwan, have been isolated more than 500 secondary metabolites, including lignans, terpenoids, steroids, and flavonoids. Several of the metabolites are reported to have antibacterial, antifungal, antimite, antitermite and antitumor activities. In order to investigate plant secondary metabolic diversity toward industrial applications, we established deep transcriptome resources for non-model plants and fungi to produce terpenoid metabolites of economic importance. Second, many plants of Lauraceae have been utilized in folk medicine for their exciting bioactivities. The twigs and leaves from 27 tree species of Lauraceae grown in Taiwan were performed to evaluate potential bioactivity. The leaves of Cinnamomum osmophloeum are traditionally used in folk medicines, and many biological activities have been identified, such as antibacterial, antifungal, antitermite, antidiabetic, antihyperuricemia, antiinflammatory, and antioxidant activities. However, C. osmophloeum has nine chemotypes with various secondary metabolite profiles. In order to efficiently produce active compounds, we established the genetic markers to identify the chemotype plants. Finally, Cinnamomum kanehirae is the host of the medicinal mushroom Antrodia cinnamomea. Several in vivo and in vitro studies indicated that A. cinnamomea possesses a diverse range of biological activities. Because of the potential pharmacological application, we established the transformation system to enhance the triterpenoid contents production.

The use of synthetic pesticides in crop production, brings the possibility of a nonselective toxicity and accumulation of toxic compounds in the environment. As an alternative, bioactive compounds from actinomycetes can provide a rich source for biodegradable pesticides. A variety of biological activities such as antibacterial, antifungal, herbicidal, insecticidal and anticoccidial are associated with these microbial metabolites. Studies on the production of bioactive compounds from marine-derived actinomycetes have been very few, compared to the terrestrial actinomycetes. With their unique metabolic and physiological capabilities, the marine actinomycetes can be useful producers of novel metabolites.
The fungal isolate Isaria javanica pf185 has potential as a mycopesticide because it demonstrates insecticidal activity against the green peach aphid and antifungal activity against Colletotrichum gloeosporioides. For commercialization of this isolate, determination of the optimal and least expensive culture conditions is required; however, these data are not currently available. This study describes the conditions for optimal development of conidia and production of metabolites for the biocontrol of the fungal pathogen. The optimal culture conditions were examined using cultures on solid agar and liquid media. High growth temperature enhanced spore formation but reduced antifungal activity in both solid and liquid media. The highest spore yield was obtained in a medium containing glucose as a carbon source and yeast extract as a nitrogen source. Soybean powder and wheat bran were effective nitrogen sources that promoted spore production and antifungal activity of the isolate. These results revealed the basic, cost-effective growth media for commercial production of a biopesticide with insecticidal and antifungal properties for use in integrated pest management.
