To isolate antifungal substances from Korean radish (Raphanus Sativus L.) seeds, various purification techniques such as DE52 cellulose anion exchange, SP-Sephadex C-25 cation exchange, and Sephadex G-50 gel filtration chromatographies were used. The molecular masses of two purified R. sativus antifungal proteins (RAPs) were estimated to be about 6.1 kDa (RAP-1) and 6.2 kDa (RAP-2) by SDS-PAGE, and 5.8 kDa(RAP-1) and 6.2 kDa (RAP-2 by a gel filtration chromatography, respectively. Purified proteins RAP-1 and 2 clearly exhibited different growth inhibitory activities against other microorganisms like Candida albicans and Saccharomyces cerevisiae. Although they have similar molecular masses, both RAP-1 and 2 proteins are not identical because their microbial inhibitory actions were different. Therefore, RAP-1 could be a new antifungal protein when compared with the antifungal activities of 2S albumins, Rs-AFPs, Mj-AMPs, chitinase, glucanase, permatin, and ribosome inactivating proteins, all of which are anifungal proteins of plants.
In order to use the symbiotic bacteria from ethomophatogenic nematodes as a biological control agent for agriculture, the cultural condition for maintaining phase I and antifungal activity was investigated. Symbiotic bacteria (SB) 1 stain from nematodes were selected from the three strains isolated from entomopathogenic nematodes. The growth of the SB 1 strain in NB, TSB, TY and YS medium was higher than that of the SB 2 and SB 3 strain. The packed cell volume of the SB 1 strain was reduced in NB medium which showed radical pH change. Phase I of the SB 1 strain was maintained in TSB medium after being stored for 2 weeks at $4^{\circ}C$. Culture broth with the SB 1 strain in TSB medium for 6 days and 7 days showed antifungal activities against Rhizoctonia solani KACC 40142, Botrytis cinerea Pers. KACC 40854, and Botrytis cinerea Pers. KACC 41008. Culture broth with the SB 1 strain in TSB medium containing 100 mM L-proline for 5 days showed antifungal activities against Rhizoctonia solani KACC 40142, and Botrytis cinerea Pers. KACC 40854.
Antifungal and antibacterial activities of wood vinegars were investigated in this study in order to apply for the natural fungicides and food preservatives. The antifungal activities of wood vinegars were measured as a hyphal growth inhibition rate using four plant pathogenic, four wood rot and one mushroom pathogenic fungi. Inhibition effect on the fungi growth was explicitly observed at the concentrations higher than 0.5% of unrefined wood vinegar and higher than 1.0% of refined wood vinegar, respectively, suggesting that unrefined wood vinegars showed the higher antifungal activities than that of refined wood vinegars. The highest inhibition effect on the fungi growth was shown in Libertella betulina which is a mushroom pathogenic fungus. In addition, the wood vinegars from Pinus densiflora showed the higher antifungal activities against the plant pathogenic and wood rot fungi than that from Quercus spp.. On the other hand, the antibacterial activities of wood vinegars were determined by a paper disc method using the three gram positive and five gram negative bacteria. The unrefined wood vinegars showed a prominent effect on the suppression of bacteria growth at the concentration of 10%, while the suppression of bacteria growth was not observed in the refined wood vinegars. These results also suggest that the unrefined wood vinegars showed the higher antibacterial activities than the refined wood vinegars. The antibacterial activities of wood vinegars against gram positive bacteria were higher than those of wood vinegars against gram negative bacteria. From these results, it can be inferred that the wood vinegars has the strong antimicrobial activities and can be applied as the natural fungicides and food preservatives.
Antimicrobial and antioxidative activities on heartwood extractives of domestic species were investigated to develop a natural fungicide or preservative. Six flavanones including pinostrobin, eriodictyol, naringenin, pinocembrin, taxifolin and verecundin were isolated from Prunus sargentii which has been selected due to its high antimicrobial and antioxidative activities among the tested species. According to the results of antifungal test, pinocembrin was evaluated as the highest antifungal compound among the test compounds, which showed 80% of hyphal growth inhibition rate. Antifungal activity of pinocembrin was similar to hinokitiol(${\beta}$-thujaplicin), strong antimicrobial compound isolated from Thujopsis dolabrata. Naringenin followed pinocembrin in its antifungal activity. However, verecundin did not show any antifungal activity. No compound was effective in antibacterial activities. As a result of the measurement of free radical scavenging activity, antioxidative activities of taxifolin and eriodictyol were 2 times that of ${\alpha}$-tocopherol, and antioxidative index of these compounds were even superior to that of ${\alpha}$-tocopherol. In this regard, it could inferred that high antifungal and antioxidative activities of extractives of P. sargentii were derived from pinocembrin, taxifolin and eriodictyol, respectively.
For the purpose of searching for the new antifungal agent from the plant origin, we have examined twenty seven species of medicinal plants for the antifungal activity against Candida albicans and Pityrosporum ovale. The whole MeOH extract of each plant material and the n-hexane and EtOAc fractions were evaluated individually for the antifungal activity. Among the test materials, the fractions from the root bark of Zanthoxylum schinifolium, Zanthoxylum piperitum and Citrus unshiu showed significant antifungal activities in a dose dependent manner against Pityrosporum ovale and Candida albicans, respectively.
Extracts from sawdust of Pinus densiflora were showed antifungal activity against Lentinula edodes. It was extracted by hot water and then successively extracted by n-hexane, ethyl acetate, and methanol. The yields of the n-hexane-soluble, ethyl acetatesoluble, methanol-soluble and methanol-insoluble fractions of water extracts were 8.2%, 10.6%, 32.0%, and 49.2%, respectively. The ethyl acetate-soluble fraction showed the greatest antifungal activity against L. edodes: 41.5% inhibition at 1,000 ppm. However, there were not significant differences of antifungal activities between n-hexane-soluble fraction and methanol-soluble fraction at a concentration of 1,000 ppm. The hot water extracts showed 23.5% of antifungal activity against L. edodes at a concentration of 1000 ppm. The four antifungal compounds were separated from ethyl acetate fraction by thin layer chromatography.
Efficacy of antifungal activity on plant pathogens by compost teas fermented under different temperatures was studied. Compost teas are recently chosen by agricultural producers for the better method of controlling plant diseases under increase of public consciousness against use of chemicals for controlling the diseases. Compost tea has been intensively studied; however, understanding of compost tea is still not well developed, and temperature influence during fermentation of compost tea on its antifungal activity has not been investigated. In this study, antifungal activities of compost teas fermented at 10, 20, 30, and $40^{\circ}C$ against selected 10 pathogens were observed. From the results, antifungal activities of compost teas at 20 and $30^{\circ}C$ of fermentation-temperatures showed the strongest while the weakest activity was observed with the compost tea at $10^{\circ}C$. Change of the activity by the fermentation-temperature apparently implied that microbes in the compost tea were strongly involved in its antifungal activity.
In this study, we evaluated antifungal and antioxidant activities of condensed liquid of volatile organic compounds generated during the drying process of Chamaecyparis obtuse. Condensed liquid of volatile organic compounds were extracted with hexane and ethyl acetate, respectively. The extracts were used for antifungal activity at range from 100 to 500 mg/L Antifungal activity of extracts increased as the extracts concentration increased, the activity against tested dermatophytes was high at 500 mg/L. Among extract solvents, antifungal activity was higher at hexane extract than the ethyl acetate. Especially, the antifungal activity against Trichophyton rubrum was higher than other tested dermatophytes. Antioxidant activity was 90% above 25 mg/L regardless of the extractive solvents, while the hexane extract showed high antioxidant activity below 25 mg/L. Major compounds of hexane extract were torreyol, alpha-cadinol, and tau-cadinol. However, major compounds of ethyl acetate extract were alpha-amorphene, alpha-cadinol, and gamma-cadinene.
Antimicrobial activities of plant extractives were investigated to develop a natural fungicide. Two stilbenoids and five flavonoids were isolated from wood extractives of Hovenia dulcis (Rhamnaceae) which had been selected due to its high antifungal activity among the tested species. The chemical structures of isolated compounds were determinded as : 5-hydroxy-7-methoxyflavone, 5,7-dihydroxyflavone (chrysin), 5,7-dihydroxyflavanone (pinocembrin), 3,5,7-trihydroxyflavanone (pinobanksin), 3,4',5,7-tetrahydroxyflavanone (aromadendrin), 3-hydroxy-5-methoxystilbene and 3,5-dihydroxystilbene (pinosylvin) on the basis of Mass and NMR spectroscopic data. According to the results of antifungal test, 3-hydroxy-5-methoxystilbene was evaluated as the strongest antifungal compound among the tested compounds and next were pinocembrin and pinosylvin, but those also had high hyphal growth inhibition activities against C. parasitica, T. versicolor, T. palustris and T. viride. However, pinobanksin, 5-hydroxy-7-methoxyflavone, chrysin and aromadendrin showed very low antifungal activity. In this regard, it could inferred that high antifungal activity of wood extractives of H. dulcis were derived from 3-hydroxy-5-methoxystilbene, pinocembrin and pinosylvin, respectively.
Objectives: The purpose of this study was to investigate the antifungal effect of bee venom (BV) and sweet bee venom (SBV) against Candida albicans (C. albicans) clinical isolates. Methods: In this study, BV and SBV were examined for antifungal activities against the Korean Collection for Type Cultures (KCTC) strain and 10 clinical isolates of C. albicans. The disk diffusion method was used to measure the antifungal activity and minimum inhibitory concentration (MIC) assays were performed by using a broth microdilution method. Also, a killing curve assay was conducted to investigate the kinetics of the anti-fungal action. Results: BV and SBV showed antifungal activity against 10 clinical isolates of C. albicans that were cultured from blood and the vagina by using disk diffusion method. The MIC values obtained for clinical isolates by using the broth microdilution method varied from $62.5{\mu}g/mL$ to $125{\mu}g/mL$ for BV and from $15.63{\mu}g/mL$ to $62.5{\mu}g/mL$ for SBV. In the killing-curve assay, SBV behaved as amphotericin B, which was used as positive control, did. The antifungal efficacy of SBV was much higher than that of BV. Conclusion: BV and SBV showed antifungal activity against C. albicans clinical strains that were isolated from blood and the vagina. Especially, SBV might be a candidate for a new antifungal agent against C. albicans clinical isolates.
