목련과 식물의 항균 및 항산화활성 (Studies on Antimicrobial and Antioxidative Activities of Extracts from Magnoliaceae)

  • 목련과 식물의 활용도 제고를 위해 항산화제, 식품첨가제 개발에 필요한 항균 및 항산화 활성을 검정하였다. 우선 목련과 총 11종 식물의 수피, 잎, 목부, 꽃 및 열매의 에탄올 조추출물을 1,000 ${\mu}g/m{\ell}$로 조제하여 항진균 활성을 조사하였다. 그 결과 일본목련 수피가 백색부후균인 Trametes versicolor를 제외한 모든 공시균주에 대하여 50% 이상의 균사생장억제율을 나타내어 항균활성이 가장 높은 것으로 나타났다. 그 다음이 일본목련 목부로 탄저병균인 Glomerella cingulata와 목재부후균인 T. versicolor, Tyromyces palustris를 제외한 공시균주에 대하여 50% 이상의 균사생장억제율을 나타내었다. 수종별로는 일본목련이 가장 항균활성이 우수하여 수피, 잎, 목부 모두 항균활성이 있는 것으로 나타났으며 그 다음이 목련, 함박꽃나무 순이었다. 부위별로는 수피가 가장 활성이 우수하였으며 백목련, 자목련의 경우에는 다른 부위에 비해 꽃의 항균활성이 우수하였다. 또한, 프리라 디칼소거능을 측정하여 항산화활성을 검정한 결과에서는 100 ${\mu}g/m{\ell}$의 농도에서 태산목 수피와 잎이 70% 이상의 소거능을, 그리고 초령목 수피와 잎, 흑오미자 줄기 및 남오미자 줄기가 90% 이상의 소거능을 나타내었다. 특히, 흑오미자 줄기 및 남오미자 줄기는 10 ${\mu}g/m{\ell}$의 농도에서 각각 41.2%와 44.4%의 소거능을 나타내어 합성 항산화제인 butylated hydroxyanisole 및 천연항산화제인 ${\alpha}$-tocopherol과 동등한 항산화활성이 있는 것으로 밝혀졌다. 금후 흑오미자 및 남오미자 줄기와 같이 항산화활성 우수 시료는 활성물질 분리 및 구조 동정, 안전성 검정 등을 통하여 식품보존제 및 화장품 소재로 개발이 가능할 것으로 사료되었다.

  • 6-(4-Iodophenyl)amino-7-chloro-5,8-quinolinedione (RCK9) was evaluated for antifungal activities. The MIC values of RCK9 were determined against A. flavus, c. albicans, C. neoformans and F. oxysporium. The RCK9 showed generally potent antifungal activities against the tested fungi. Acute oral toxicity studies of RCK9 were carried out in ICR mice of both sexes. These acute oral toxicities of RCK9 had been evaluated. RCK9 were low and LD50 values were over 2,850 mg/kg in ICR mice. The genotoxicities of RCK9 had been evaluated. RCK9 was negative in Ames test with Salmonella typhimurium and chromosomal aberration test in CHL cells. The clastogenicity was tested on the RCK9 with in vivo mouse micronucleus assay. RCK9 did not show any clastogenic effect in mouse peripheral blood and was negative in mouse micronucleus assay. The results indicate that RCK9 has no genotoxic potential under these experimental conditions.

Characterization of Chitinolytic and Antifungal Activities in Marine-Derived Trichoderma bissettii Strains

  • Trichoderma fungi have been intensively studied for mycoparasitism, and the latter is closely related to their cell-wall degrading enzymes including chitinase. Here, we studied marine-derived Trichoderma spp., isolated from distinct sources and locations, for chitinolytic and antifungal activity. Based on morphological and phylogenetic analyses, two strains designated GJ-Sp1 and TOP-Co8 (isolated from a marine sponge and a marine alga, respectively) were identified as Trichoderma bissettii. This species has recently been identified as a closely related species to Trichoderma longibrachiatum. The extracellular crude enzymes of GJ-Sp1 and TOP-Co8 showed activities of chitobiosidase and b-N-acetylglucosaminidase (exochitinase) and chitotriosidase (endochitinase). The optimum chitinolytic activity of the crude enzymes was observed at 50 ℃, pH 5.0, 0-0.5% NaCl concentrations, and the activities were stable at temperatures ranging from 10 to 40 ℃ for 2 h. Moreover, the crude enzymes showed inhibitory activity against hyphal growth of two filamentous fungi Aspergillus flavus and Aspergillus niger. To the best of our knowledge, this is the first report of the chitinolytic and antifungal activity of T. bissettii.

The Antitermitic and Antifungal Activities and Composition of Vinegar from Durian Wood (Durio sp.)

  • Chemical characterization of vinegars obtained from Durian wood (Durio sp.) and their termicidal activity against Coptotermes curvignathus and antifungal activity against Schizophyllum commune were evaluated. The process of pyrolysis produced wood vinegars at three distinct temperature: 350℃, 400℃, and 450℃. To determine their effectiveness against fungal growth, the vinegars were tested using a Petri dish with 1.0%, 2.0%, 3.0%, and 4.0% (v/v) against S. commune. In the experiment, termicidal activities were evaluated using a no-choice test for C. curvignathus with 3.0%, 6.0%, 9.0%, and 12.0% (v/v). The wood vinegar exhibited antitermitic activity to C. curvignathus workers in the no-choice experiment; For vinegar produced at 450℃, a 6% concentration was required to achieve 100% mortality against C. curvignathus. In addition, a 12% vinegar produced at 450℃ resulted in the lowest mass loss of treated filter paper, which was 20.00%. Furthermore, all the wood vinegars exhibited antifungal activities against S. commune at concentration of 2.0%. The dominant chemical components of wood vinegar produced at temperature of 350℃, 400℃, and 450℃ were 2-methoxy-phenol, 4-ethyl-2-methoxy-phenol, 4-ethyl-2-methoxy-phenol, 3.5-dimethoxy-4-hydroxytoluene, and creosol.

곤충의 식물병원성 진균에 대한 항균활성 (Antifungal Activities of Insect Against the Plant Pathogenic Fungi)

  • 본 연구는 식물병원성곰팡이에 대한 곤충 혈액 및 유충체의 항곰팡이 활성을 조사하였다. 집누에, 장수풍뎅이, 흰점박이 꽃무지 모두 곰팡이 저해활성을 나타냈으며, 집누에의 계통간 비교에서는 1087계통의 활성이 가장 높았다. 또한, 혈액보다는 유충체를 이용한 경우가 더 강한 항곰팡이 활성을 나타냈다. 곤충의 항곰팡이 활성은 곰팡이균주에 따라 다르며, 본 연구에 사용된 3종은 A. panax, C. gloeosporioides 및 P. oryzae 균에 항곰팡이 활성을 가진다.

Biocontrol Activity of Volatile-Producing Bacillus megaterium and Pseudomonas protegens Against Aspergillus and Penicillium spp. Predominant in Stored Rice Grains: Study II

  • In our previous studies, Bacillus megaterium KU143, Microbacterium testaceum KU313, and Pseudomonas protegens AS15 have been shown to be antagonistic to Aspergillus flavus in stored rice grains. In this study, the biocontrol activities of these strains were evaluated against Aspergillus candidus, Aspergillus fumigatus, Penicillium fellutanum, and Penicillium islandicum, which are predominant in stored rice grains. In vitro and in vivo antifungal activities of the bacterial strains were evaluated against the fungi on media and rice grains, respectively. The antifungal activities of the volatiles produced by the strains against fungal development and population were also tested using I-plates. In in vitro tests, the strains produced secondary metabolites capable of reducing conidial germination, germ-tube elongation, and mycelial growth of all the tested fungi. In in vivo tests, the strains significantly inhibited the fungal growth in rice grains. Additionally, in I-plate tests, strains KU143 and AS15 produced volatiles that significantly inhibited not only mycelial growth, sporulation, and conidial germination of the fungi on media but also fungal populations on rice grains. GC-MS analysis of the volatiles by strains KU143 and AS15 identified 12 and 17 compounds, respectively. Among these, the antifungal compound, 5-methyl-2-phenyl-1H-indole, was produced by strain KU143 and the antimicrobial compounds, 2-butyl 1-octanal, dimethyl disulfide, 2-isopropyl-5-methyl-1-heptanol, and 4-trifluoroacetoxyhexadecane, were produced by strain AS15. These results suggest that the tested strains producing extracellular metabolites and/or volatiles may have a broad spectrum of antifungal activities against the grain fungi. In particular, B. megaterium KU143 and P. protegens AS15 may be potential biocontrol agents against Aspergillus and Penicillium spp. during rice grain storage.

Isolation and characterization of an antifungal substance from Burkholderia cepacia, an endophytic bacteria obtained from roots of cucumber.

  • In order to develop a new microbial fungicide for the control of vegetable diseases using endophytic bacteria, a total of 260 bacterial strains were isolated from fresh tissues of 5 plant species. After they were cultured in broth media, their antifungal activities were screened by in vivo bioassays against Botrytis cinerea(tomato gray mold), Pythium ultimum(cucumber damping-off), Phytopkhora infestans(tomato late blight), Colletotrichum orbiculare(cucumber anthracnose), and Blumeria graminis f. sp. hordei(barley powdery mildew). As the results of screening, 38 bacterial strains showed potent antifungal activities against at least one of 5 plant pathogens. A bacterial strain EB072 displayed potent disease control activities against 3 plant diseases. Among the bacterial strains with a potent antifungal activity against cucunlber anthracnose, three bacterial strains, EB054, EB151 and EB215, also displayed a potent in vitro antifungal activity against C. acutatum, a fungal agent causing pepper anthracnose. A bacterial strain EB215 obtained from roots of cucumber was identified as Burkholderia cepacia based on its physiological and biochemical characteristics and 165 rRNA gene sequence. An antifungal substance was isolated from the liquid cultures of B. cepacia EB215 strain by ethyl acetate partitioning, repeated silica gel column chromatography, and invitro bioassay, Its structural determination is in progress by various instrumental analyses.

  • $NaHSO_4.SiO_2$ 촉매를 이용한 microwave 및 무수조건하에서 새로운 morpholino pyrimidines의 One-pot합성이다. 합성된 모든 화합물을 이용하여 in vitro에서 임상학적인 단리된 박테리아 품종 즉 Bacillus subtilis, Bacillus cerues, Micrococcus luteus , Salmonella typhii에 대해 항균성을 살펴보았고, 균품종 즉 Aspergillus niger, Candida 6, Candida 51에 대해 항진균성을 살펴보았다. 미생물 결과에 대한 합성된 화합물의 구조적 활동성의 관련성에 대해 토론하였다.

  • Antifungal activities of the extracts from 26 medicinal plants were investigated utilizing paper-disk diffusion method and (1,3)β-glucan synthase inhibitory assay. (1,3)β-glucan synthase is considered as valuable target in the development of antifungal agents. Among the screened extracts, the ethyl acetate extract of Equisetum arvense, the ethyl acetate extract of Polygonum aviculare, the butanol extract of Crataegus pinnatifida and the n-hexane extract of Saussurea lappa showed significant antifungal activities on Candida albacans in both disk diffusion and enzyme assays.

Verlamelin, an Antifungal Compound Produced by a Mycoparasite, Acremonium strictum

  • A strain of Acremonium strictum, the mycoparasite of Botrytis cinerea, showed strong antifungal activities both in vitro and in vivo against several phytopathogenic fungi. An antifungal substance was purified from the liquid cultures of A. strictum and identified as verlamelin by instrumental analyses. Verlamelin exhibited in vitro antifungal activity against some phytopathogenic fungi such as Magnaporthe grisea, Bipolaris maydis, and Botrytis cinerea, while it was net active against all the bacteria tested. In viva, verlamelin exhibited strong protective and curative activities, particularly against barley powdery mildew. At 100 μg/ml, it inhibited the development of barley powdery mildew with control values of more than 90% in 7-day protective and 2-day curative applications. This is the first report on the production of verlamelin by Acremonium species.
