The Role of HS-1200 Induced Autophagy in Oral Cancer Cells

  • Bile acids and synthetic bile acid derivatives induce apoptosis in various kinds of cancer cells and thus have anticancer properties. Recently, it has been suggested that autophagy may play an important role in cancer therapy. However, few data are available regarding the role of autophagy in oral cancers and there have been no reports of autophagic cell death in OSCCs (oral squamous cell carcinoma cells) induced by HS-1200, a synthetic bile acid derivative. We thus examine whether HS-1200 modulates autophagy in OSCCs. Our findings indicate that HS-1200 has anticancer effects in OSCCs, and we observed in these cells that autophagic vacuoles were visible by monodansylcadaverine (MDC)and acridine orange staining. When we analyzed HS-1200-treated OSCC cells for the presence of biochemical markers, we observed that this treatment directly affects the conversion of LC-3II, degradation of p62/SQSTM1 and full-length beclin-1, cleavage of ATG5-12 and the activation of caspase. An autophagy inhibitor suppressed HS-1200-induced cell death in OSCCs, confirming that autophagy acts as a pro-death signal in these cells. Furthermore, HS-1200 shows anticancer activity against OSCCs via both autophagy and apoptosis. Our current findings suggest that HS-1200 may potentially contribute to oral cancer treatment and thus provide useful information for the future development of a new therapeutic agent.

Gallic Acid Enhancement of Gold Nanoparticle Anticancer Activity in Cervical Cancer Cells

  • Cervical cancer (CxCa) is the most common cancer in women and a prominent cause of cancer mortality worldwide. The primary cause of CxCa is human papillomavirus (HPV). Radiation therapy and chemotherapy have been used as standard treatments, but they have undesirable side effects for patients. It was reported that gallic acid has antioxidant, antimicrobial, and anticancer activities. Gold nanoparticles are currently being used in medicine as biosensors and drug delivery agents. This study aimed to develop a drug delivery agent using gold nanoparticles conjugated with gallic acid. The study was performed in uninfected (C33A) cervical cancer cells, cervical cancer cells infected with HPV type 16 (CaSki) or 18 (HeLa), and normal Vero kidney cells. The results showed that GA inhibited the proliferation of cancer cells by inducing apoptosis. To enhance the efficacy of this anticancer activity, 15-nm spherical gold nanoparticles (GNPs) were used to deliver GA to cancer cells. The GNPs-GA complex had a reduced ability compared to unmodified GA to inhibit the growth of CxCa cells. It was interesting that high-concentration ($150{\mu}M$) GNPs-GA was not toxic to normal cells, whereas GA alone was cytotoxic. In conclusion, GNPs-GA could inhibit CxCa cell proliferation less efficiently than GA, but it was not cytotoxic to normal cells. Thus, gold nanoparticles have the potential to be used as phytochemical delivery agents for alternative cancer treatment to reduce the side effects of radiotherapy and chemotherapy.

Induction of G1 arrest and apoptosis mediated by a novel nucleoside analog, LJ-331 in human leukemia HL-60 cells

  • In a continuous effort to develop novel anticancer agents we newly synthesized and evaluated the antitumor activity of nucleoside analogues. One analogue, 4 - [2-Chlor-6- (3-iodo- benzy lamino) -purin -9-yl]- 2,3-dihydroxy-cyclopentanecarbo xylic acid methylamide (LJ-331), has been shown to exert a potent inhibition of human cancer cell growth in vitro including human lung (A549), stomach (SNU-638) and leukemia (HL-60) cancer cells. Following mechanism of action study revealed that LJ - 331induces cell cycle arrest at the G1 phase in HL-60 cells and evokes apoptotic phenomena such as an increase in DNA ladder intensity and chromatin condensation by a dose- and time-dependent manner. LJ-331 also activated the caspase-3 activity in HL-60. This result suggests that the growth inhibition of human cancer cells by LJ-331 might be related to the cell cycle arrest and induction of apoptosis.

인체 폐암 세포주 A549에서 Euonymus porphyreus 추출물의 항산화 및 항암활성 분석 (Antioxidant and Anticancer Activities of Euonymus porphyreus Extract in Human Lung Cancer Cells A549)

  • Euonymus porphyreus는 노박덩굴과에 속하는 식물로 동아시아 지역에 널리 분포하며, 식물학상의 특징에 대한 보고는 있으나 항산화능과 항암활성 등에 관한 연구는 아직까지 밝혀진 바가 없다. 이에 본 연구에서는 인체 폐암세포인 A549를 사용하여 E. porphyreus 에탄올 추출물(EEEP)의 항산화 및 항암활성과 그 분자적 기전에 관하여 연구하였다. 먼저 EEEP의 총 폴리페놀 화합물과 플라보노이드 함량을 측정한 결과, 각각 115.42 mg/g, 23.07 mg/g이었다. EEEP의 DPPH radical 소거활성을 측정한 결과, IC50가 11.09 ㎍/ml로 뛰어난 항산화능을 보유한 것을 확인하였다. 또한 EEEP는 농도의존적으로 인체폐암세포주인 A549의 세포 성장을 저해하였으며, 세포 주기 변화를 분석한 결과 A549 세포의 SubG1기 세포비율이 증가하는 것을 확인하였다. Annexin V 염색과 DAPI 염색으로 EEEP 처리에 의해 apoptotic 세포와 apoptotic body가 증가하는 것을 확인하였으며, 이러한 결과는 EEEP에 의해 A549 세포의 apoptosis가 유도되는 것을 시사한다. 또한 관련 단백질들의 발현변화를 분석한 결과, EEEP에 의해 Fas, p53, Bax의 발현이 증가하고 Bcl-2의 발현은 감소하였으며, caspase-8, -9와 caspase-3의 활성화를 통해 PARP가 분해되어 apoptosis가 유도되었음을 확인하였다. 이러한 결과들로부터 EEEP는 내인성 및 외인성 경로를 통한 apoptosis 유도에 의하여 A549 세포의 증식을 억제하는 항암활성을 보유하였음을 확인하였다.

Anticancer effects of Ulva compressa extracts on FaDu human hypopharangeal squamous carcinoma cells in vitro

  • Ulva compressa Linnaeus (UCL) is a green algae seaweed that performs photosynthesis and is used as a food material in some Asian regions including Korea. It is known to be the dominant species in copper ion-contaminated seas, and many studies on copper ion resistant mechanisms have been reported. UCL is known to have an excellent antioxidant effect, but limited information is available regarding its other physiological activities. In this study, we investigated the anticancer activity of 30% prethanol extracts of Ulva compressa Linnaeus (30% PeUCL) and the underlying mechanisms of its activity on human FaDu hypopharyngeal squamous carcinoma cells. The 30% PeUCL extracts suppressed FaDu cell viability without affecting normal cells (L929), as determined by MTT and viability assays. Furthermore, the 30% PeUCL extracts induced apoptosis, as determined by DAPI staining. The 30% PeUCL extracts inhibited colony formation effectively as well as wound-healing of FaDu cells, even at noncytotoxic concentrations. In addition, 30% PeUCL extracts induced apoptosis significantly through proteolytic cleavage of caspase-3, -7, and -9, and poly (ADP-ribose) polymerase, and by downregulation of Bcl-2 and upregulation of Bax in FaDu cells, as determined by Western blot analysis. Collectively, these results suggest that the inhibitory effect of 30% PeUCL extracts on the growth of oral cancer cells, colony formation and wound-healing may be mediated by caspase- and mitochondrial-dependent apoptotic pathways in human FaDu hypopharyngeal squamous carcinoma cells. Therefore, 30% PeUCL extracts can be administered as a natural chemotherapeutic drug for the treatment of human oral cancers.

종양세포에서의 capsaicin에 의한 apoptosis 유도와 항암제의 항암효과의 증가 (Capsaicin induced apoptosis and the enhanced anticancer effect of anticancer drugs in cancer cells)

  • 목 적 : 한국인 음식에 널리 이용되고 있는 중요한 향신료인 고추에 많이 들어있는 capsaicin이 최근 많은 연구를 통해 암을 예방하고 더 나아가 치료할 수 있는 성분으로 밝혀지고 있다. 따라서 본 연구의 목적은 한국인이 많이 섭취하고 있는 식이성분인 capsaicin이 한국인에 많은 위암세포의 세포자멸사의 유도물질로 가능성을 알아보고 이를 항암치료의 일부분으로 시도하고자 하였다. 방 법 : 한국인의 위암세포주인 SNU-668 세포에 capsaicn를 처리한 후 세포 생존은 trypan blue 와 crystal violet 분석, 세포독성은 MTT 분석, 세포사 분석은 핵 응축과 DNA 분절화 실험, bcl-2와 bax의 mRNA 발현은 역전사 중합효소 연쇄반응, 세포사와 관련된 단백질 발현은 Western immunoblot analysis로 분석하였다. Capsaicin에 의해 항암제의 감수성이 증가되는 지를 알아보기 위하여 일정 농도의 capsaicin과 농도 별 각 항암제를 같이 처리하여 2일 간 배양 하고 MTT assay로 분석하였으며 이와 관련된 세포사와 관련된 단백질 발현을 분석하였다. 결 과 : capsaicin의 농도에 따라 SNU-668 위암세포의 증식을 현저히 억제하였으며, 이러한 capsaicin의 증식 억제는 DNA 단편화, 핵 응축과 caspase 활성화에 의해 세포자멸사의 유도에 의한 것으로 입증되었다. 더욱이 capsaicin에 의해 pro-apoptotic Bax에 대한 anti-apoptotic Bcl-2의 비율이 현저히 감소하고, caspase-3활성이 증가하였다. Capsaicin을 처리한 세포는 처리하지 않은 세포에 비하여 etoposide나 adriamycin에 의해 유도되는 세포사멸에 더욱 감수성을 보였다. 결 론 : 이상의 연구에서 capsaicin은 caspase-3 의존적 경로를 통하여 SNU-668 세포에서 세포자멸사를 유도하며 항암제의 감수성을 증가시키므로 위암의 효과적인 항암치료제와 항암제 민감성 유도제의 일환으로 가능성이 있다.

Ethanol Extract of Smilax glabra Induces Apoptotic Cell Death in Human YD10B Oral Squamous Cell Carcinoma Cells

  • Background: Smilax glabra has various pharmacological activities and is widely used as a herbal medicine. Although the incidence of oral cancer is low, the recurrence rate is high, and the 5-year survival rate is poor. It is necessary to search for anticancer drugs that increase the effect of cancer chemotherapy on heterogeneous oral tissues and reduce the side effects on normal cells. This study aimed to investigate the effects and mechanism of ethanol extract of Smilax glabra (EESG) as an anticancer drug for oral cancer. Methods: Smilax glabra root components extracted with 70% ethanol were used to analyze their effects on cancer cells. A 3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide assay was performed for cytotoxicity analysis. Flow cytometry was performed to determine the cell cycle phase distribution. To observe apoptotic cells, terminal deoxynucleotidyl transferase dUTP nick end labeling and γH2AX were detected by fluorescence microscope. The protein levels of cleaved PARP and caspase were analyzed using western blotting. The activation of procaspase-3 was confirmed by measuring caspase-3 activity. Results: EESG was no cytotoxic to normal gingival fibroblast but was high in YD10B oral squamous cell carcinoma (OSCC) cells. EESG treatment increased the subdiploid DNA content of YD10B cells by assessing DNA content distribution. Chromatin condensation and DNA strand breaks increased in YD10B cells treated with EESG. EESG-treated YD10B cells had high Annexin V and low propidium iodide levels, confirming that early apoptosis was induced. In addition, increased levels of γH2AX foci, a marker of DNA damage, were observed in the nuclei of EESG-treated YD10B cells. The EESG-treated YD10B cells also exhibited decreased procaspase-3 and procaspase-9 levels, increased PARP cleavage and caspase-3 activity. Conclusion: These results indicate that EESG inhibited cancer cell proliferation by inducing apoptosis in YD10B OSCC cells.

Induction of Cell Cycle Arrest, Apoptosis, and Reducing the Expression of MCM Proteins in Human Lung Carcinoma A549 Cells by Cedrol, Isolated from Juniperus chinensis

  • Proteins related to DNA replication have been proposed as cancer biomarkers and targets for anticancer agents. Among them, minichromosome maintenance (MCM) proteins, often overexpressed in various cancer cells, are recognized both as notable biomarkers for cancer diagnosis and as targets for cancer treatment. Here, we investigated the activity of cedrol, a single compound isolated from Juniperus chinensis, in reducing the expression of MCM proteins in human lung carcinoma A549 cells. Remarkably, cedrol also strongly inhibited the expression of all other MCM protein family members in A549 cells. Moreover, cedrol treatment reduced cell viability in A549 cells, accompanied by cell cycle arrest at the G1 phase, and enhanced apoptosis. Taken together, this study broadens our understanding of how cedrol executes its anticancer activity while demonstrating that cedrol has potential application in the treatment of human lung cancer as an inhibitor of MCM proteins.

Effects of polysaccharides derived from Orostachys japonicus on induction of cell cycle arrest and apoptotic cell death in human colon cancer cells

  • Crude Orostachys japonicus polysaccharide extract (OJP) was prepared by hot steam extraction. Polysaccharides (OJPI) were separated from OJP by gel filtration chromatography and phenol-sulfuric acid assay. The average molecular weight of the OJPI was 30-50 kDa. The anti-proliferative effect of OJPI on HT-29 human colon cancer cells was investigated via morphology study, cell viability assay, apoptosis assay, cell cycle analysis, and cDNA microarray. OJPI inhibited proliferation and growth of HT29 cells and also stimulated apoptosis in a dose- and time-dependent manner. In cell cycle analysis, treatment with OJPI resulted in a marked increase of cells in the G0 (sub G1) and G2/M phases. To screen for genes involved in the induction of cell cycle arrest and apoptosis, the gene expression profiles of HT-29 cells treated with OJPI were examined by cDNA microarray, revealing that a number of genes were up- or down-regulated by OJPI. Whereas several genes involved in anti-apoptosis, cell proliferation and growth, and cell cycle regulation were down-regulated, expression levels of several genes involved in apoptosis, tumor suppression, and other signal transduction events were up-regulated. These results suggest that OJPI inhibits the growth of HT-29 human colon cancer cells by various apoptosis-aiding activities as well as apoptosis itself. Therefore, OJPI deserve further development as an effective agent exhibiting anticancer activity.

Effect of AC-264, a Novel Indole Derivative, on Apoptosis in HL-60 Cells

  • The anticancer effect and apoptotic mechanism of a novel indole derivative AC-264, a lead derived from a chemical library, were investigated in human promyelocytic leukemia HL-60 cells. HL-60 cells treated with AC-264 at various concentrations showed the morphological features of apoptosis, such as plasma membrane blebbing and cell shrinkage. AC-264 exhibited cytotoxic effect in various cancer cell lines with different degrees of potency. Especially, AC-264 was effective on increasing the population of apoptotic cells in HL-60 cells, as detected by the number of cells stained with Annexin V and PI. Furthermore, AC-264 activated caspase-3 enzyme activity and induced internucleosomal DNA fragmentation. These results indicated that AC-264 produces anti-cancer effect via apoptotic cell death by activating caspase-3 and inducing internucleosomal DNA fragmentation in HL-60 cells.
