가자미식해에서 분리한 유산균의 프로바이오틱스 특성 (Probiotic Properties of Lactobacillus spp. Isolated from Gajami Sikhae)

  • 가자미식해에서 분리된 유산균 40종은 Lactobacillus plantarum spp., Leuconostoc mesenteroides spp., Lactobacillus brevis spp., and Weisella paramenteroides spp.로 확인됐으며, 40종 중 10종을 선별해 검사에 사용했다. 본 연구에서는 가자미식해에서 분리된 유산균을 프로바이오틱스로 활용하기 위해 산 및 인공위액 저항성, 담즙산 저항성, 자가 응집성, 공동 응집성, 세포 표면 소수성 등의 프로바이오틱 연구를 수행하였다. 분리한 유산균주는 산성 및 인공 위액에 대해 저항성을 보여 높은 생존율을 나타내었으며, L. plantarum GS11이 가장 뛰어난 저항성을 보였다. 또한 담즙산 저항성 측정 결과 모든 유산균주가 108~109 log CFU/ml의 생균수로, 100% 이상의 생존력을 보여주었다. 그리고 세포 표면 부착능을 간접적으로 측정하기 위해 응집력 평가를 한 결과, autoaggregation 능력을 46% 이상 나타내었다. 세포 표면 소수성 평가를 위해 Xylene 부착능을 측정한 결과 분리된 유산균주에서 32.2%의 소수성을 가지는 B. subtilis 보다 더 뛰어난 세포 부착률을 보여주었다. 이와같이 유산균은 프로바이오틱스로 활용될 만한 유의미한 결과를 보였으며, L. plantarum GS12와 L. plantarum GS13을 제외한 유산균에서 항균 활성이 나타났다. 따라서 가자미 식해에서 분리된 유산균은 다양한 프로바이오틱스 특성을 가진 프로바이오틱스로 활용할 수 있을 것으로 판단되어진다.

감즙 염색에 의한 레이온직물의 염색성과 기능성 (Dyeability and Functionality of Rayon Fabrics Treated with Persimmon Juice)

  • The results of the analysis of dyed rayon fabrics treated with persimmon juice by padding are as follows. Rayon fabrics were dyed repeatedly with persimmon juice by a padding mangle for the development of dyeability. The merits of padding-based dyeing were easier color reproduction (versus traditional hand dyeing) of hard to obtain colors and color fastness to light and laundering. The add on weight increased with the number of padding times, and the color of the dyed fabrics had red-yellow colors with low brightness and high chromatic colors. Properties of fabrics treated with persimmon juice were: $2^{nd}$ level of light resistance, $4^{th}-5^{th}$ perspiration fastness levels, rubbing fastness, and $3^{rd}-4^{th}$ washing fastness levels. Dyed fabrics exposed to UV light have a deep yellow-red color that increased lightness and darken when exposed to sunlight. Rayon fabrics of a treated premordant indicated strong yellow colors, especially fabrics with Fe mordant indicated greenish red-yellow colors. The direction of the weft and warp increased and improved water-repellent qualities that increased with the number of dyeing padding times and persimmon juice; however, there was less anti-crease resistance. In addition, fabrics had good antibacterial activity and deodorization.

In vitro Antibacterial and Synergistic Activity of an Ecklonia cava Extract against Anti biotic-Resistant Streptococcus parauberis

  • In an effort to discover alternative phytotherapeutic antimicrobial agents to combat Streptococcus parauberis, a fish pathogenic bacterium, we evaluated the antibacterial activity of seaweed extracts in vitro. A methanolic extract of Ecklonia cava exhibited strong antibacterial activity against S. parauberis isolated from olive flounder Paralichthys olivaceus. Furthermore, the n-hexane soluble (Hexane) fraction of the E. cava methanolic extract exhibited the greatest antibacterial effect on S. parauberis strains with a minimum inhibitory concentration (MIC) ranging from 256 to $1,024{\mu}g/mL$. In addition, the MIC values of oxytetracycline against antibiotic-resistant S. parauberis were markedly reduced up to 64-fold in combination with the Hexane fraction, suggesting that the antibacterial activity of the antibiotic was restored when combined with the Hexane fraction. The interaction between both antibiotics and the Hexane fraction was assessed by the fractional inhibitory concentration (FIC) index. The Hexane fraction and oxytetracycline combination against antibiotic-resistant S. parauberis strains resulted in a median ${\sum}FIC$ range of 0.502 to 0.516. Thus, the synergistic ranges of median ${\sum}FIC$ < 1 were observed for all combinations of the Hexane fraction and oxytetracycline against S. parauberis. To the best of our knowledge, this is the first report indicating the efficacy of an E. cava extract against fish pathogenic bacterium S. parauberis.

가축질병 세균 및 항생제 내성 세균에 대한 버섯 균사체 배양 추출물의 항균 활성 (Antibacterial Activities of Mushroom Liquid Culture Extracts Against Livestock Disease-Causing Bacteria and Antibiotic Resistant Bacteria)

  • 구름버섯(Coriolus versicolor), 상황버섯(Phellinus linteus) 노루궁뎅이버섯(Hericium erinaceus)의 균사체 배양액의 ethyl acetate 추출물이 돼지와 가금류의 설사 유발 세균류인 E. coli K88, E. coli K99, E. coli 987P와 Salmonella typhimurium 14058 균주에 대하여 항균활성을 보였으며, 특히 구름버섯 추출물은 가장 높은 항균활성을 보였다. 한편, kanamycin과 ampicillin에 대해 내성을 보이는 Staphylococus aureus CARM3230과 E. coli CARM1381 균주에 대해서도 세 가지 버섯 배양 추출물 모두 항균활성을 보였으며, 특히, 구름버섯 추출물은 가장 높은 항균 활성을 보였다. 이는, 이들 버섯 균사체 배양 추출물이 기존 항생제 대체용으로 가축 사료에 첨가제로 사용될 수 있다는 가능성을 보였다. 또한 이들 버섯류로부터 항생제 내성균을 억제할 수 있는 신규 항균성 물질의 탐색을 위한 기초자료로서 활용될 수 있을 것이다.

Effect of Scutellariae Radix as a Novel Antibacterial Herb on the ppk(Polyphosphate Kinase) Mutant of Salmonella typhimurium

  • The antibacterial effects of water extracts of Scutellariate Radix (a dried root of Scutellaria baicalensis GEORGI) and its major flavonoid components, Baicalin and Baicalein, on Salmonella typhimurium, a representative enteric pathogen, were studied. Through a Kriby-Bauer disc analysis, the growth-inhibition activity of Scutellariae Radix against. S. typhimurium was found to be compatible with commercial antibiotics, such as ampicillin, chloramphenicol, and streptomycin. In contrast, the growth of a nonpathogenic E. coli strain was unaffercted by Scutellariae Radix. To examine the effect of polyphosphate kinase (ppk), a putative virulence factor, on the antibacterial activity of Scutellariae Radix, the growth profile of a ppk mutant of S. typhimurium was investigated in a tryptic soy broth containing different concentrations of water extracts of Scutellariae Radix. The ppk mutant was able to grow in 6 mg/ml of water extracts of Scutellariae Radix, whereas in 6 mg/ml of water extracts of Scutellariae Radix, whereas the wild-type could not, implying that the inactivation of ppk made S. typhimurium more resistant to the antibacterial activity of Scutellariae Radix. No enhanced resistance was observed in a ppk mutant of S. typhimurium complemented with a ppk expression vector. The attenuation of the virulence by ppk inactivation was also observed in a virulence assay using BLAB/c mice. Neither Baicalin nor Baicalein exhibited any growth-inhibition activity against S. typhimurium. The water extracts of Scutellariae Radix stimulated the transcription of ppk, especially in the early growth-stage of S. typhimurium.

항생제 내성 Cutibacterium acnes에 대한 감태(Ecklonia cava) 추출물의 항균 시너지 효과 (Addictive Effects of Ecklonia cava Methanol and its Solvent-soluble Extract Against Antibiotic-resistant Cutibacterium acnes Causing Acne Vulgaris)

  • Acne vulgaris is often associated with acne-related bacteria such as Cutibacterium acnes. In this study, we investigated the antibacterial effects of the methanol extract of the algae Ecklonia cava and its solvent-soluble extract against C. acnes. Among five solvent fractions, the ethyl acetate (EtOAc) fraction exhibited the strongest antibacterial activity against C. acnes. Furthermore, the EtOAc-soluble extract exhibited the highest total phenolic contents among the five solvent fractions tested. The EtOAc subfraction 07 (Fr. 07) extract showed the highest antibacterial effect against C. acnes and isolated C. acnes along with minimum inhibitory concentration (MIC) ranging from 32 to 64 ㎍/mL. Additionally, MICs of antibiotics against antibiotic-resistant C. acnes strains were substantially reduced when antibiotics were combined with Fr. 07, suggesting that Fr. 07 restore the antibacterial activity of the antibiotics. The fractional inhibitory concentration indices clearly revealed an additive synergistic effect of Fr. 07 with antibiotics. The results of the present study suggest a potential role for E. cava in the control of infections related to acne vulgaris.

Leptospermum scoparium 추출물중 케톤체 분획물의 항균력 및 항생제와의 병용효과 (Activities of Ketonic Fraction from Leptospermum scoparium alone and Synergism in Combination with Some Antibiotics Against Various Bacterial Strains and Fungi)

  • Whole oil and ketonic fraction (KF) of Leptospermum scoparium have been tested for their antimicrobial activity and combination effect with several antibiotics against various bacterial strains and fungi by using microbiological assay methods. Antibacterial activities of KF against a number of test strains were 2-3 fold stronger than those of whole oil. MICs of the KF were $65~125{\;}{\mu\textrm{g}}/ml$ against seven gram positive bacterial strains, $65~250{\;}{\mu\textrm{g}}/ml$ against 19 methicillin resistance Staphylococcus aureus strains, and $65~50{\;}{\mu\textrm{g}}/ml$ against 14 quinolone resistance strains. However, KF showed little or no activity against gram negative bacteria. MICs of the KF were $16~250{\;}{\mu\textrm{g}}/ml$ against more than 50% of the anaerobic bacterial strains tested. KF showed the higher antibacterial activity than bacitracin against 10 strains of Bacteroids thetaiotaomicron, or three strains of Bacteroides ovatus, and the more active than ciprofloxacin against one strain of Bacteroides thetaiotaomicron and three strains of Bacteroids ovatus. The MICs of KF was 63 and $250{\;}{\mu\textrm{g}}/ml$ against Aspergillus niger and Candida albicans, respectively. Antibacterial activities of KF in combination with 19 antibiotics against 14 strains and with four antifungal agents against one fungal strain were determined by paper strip diffusion method. While most of combination showed additivity, KF showed synergism with bacitracin, exfadroxil, cephradin, and meropenem for 29~57% of the strains tested. However, ofloxacin, enoxacin, sparfloxacin showed antagonism with KF for 43~71% of the strains. KF alone and in combination with bacitracin, gentamycin, neomycin, itraconazole, fluconazole, terfinafine and ketoconazole against five bacterial strains or one fungus strain synergistic effect was demonstrated against 33% of strains examined with FIC index value below 0.5 by checkerboard study. Synergistic effect of KF with gentamicin against Staphylococcus epidermidis 329 (QRS) was found by time-kill study.

Characteristic of Antibiotic Resistance of Foodborne Pathogens Adapted to Garlic, Allium sativum L.

  • Antibiotic resistance of foodborne pathogens adapted to garlic (Allium sativum Linn.) was determined in order to understand the relationship between antibiotic resistance and garlic. The Gram (-) strains of Escherichia coli and Salmonella typhimurium and the Gram (+) strains of Bacillus cereus and Staphylococcus aureus were subcultured consecutively in a garlic broth, and the surviving colonies on the agar were selected as the adapted strains. Minimal inhibitory concentrations (MIC) for 15 antibiotics on the adapted strains were determined on Muller-Hinton Infusion agar. Adaptation to 1.3%(v/v) garlic juice increased MIC for vancomycin, aminoglycoside, and erythromycin on B. cereus, and for ampicillin and erythromycin on E. coli O157:H7. MIC of aminoglycosides, chloramphenicol, and vancomycin on the adapted S. aureus increased. The adapted S. typhimurium was more resistant to penicillin and vancomycin than the non-treated strain. The adapted S. typhimurium and S. aureus lost their antibiotic resistance in non-garlic stress conditions. However, the adapted B. cereus was still resistant to erythromycin and vancomycin, and the adapted E. coli was also resistant to erythromycin. Antibacterial garlic might increase the antibiotic resistance of E. coli, B. cereus, S. aureus, and S. typhimurium and this resistance can continue even without the stress of garlic. Therefore, garlic as a food seasoning could influence the resistance of such pathogens to these antibiotics temporarily or permanently.

항생제 다제내성균 Pseudomonas aeruginosa에 대한 생육저해물질 생산 방선균의 분리 및 항균활성 (Isolation and Antibacterial Activity of Actinomycetes Producing Growth Inhibition Compounds Against Multi-antibiotic Resistant Pseudomonas Aeruginosa)

  • Of the 500 Actinomycetes isolates obtained from soil, one isolate grown on maltose as the sole carbon source produced compound BHK-P19, which inhibited the growth of multiple drug resistant P. aeruginosa 0245. Ultraviolet radiation mutagenesis curtailed production of BHK-P19. Mutation of the BHK-P19 producer using N-methyl-N'-nitro-N-nitroso-guanidine obviated the antibacterial activity to P. aeruginosa 0245, but not towards P. aeruginosa 0225. The mixing of BHK-P19 and BHK-S5 culture extracts inhibited P. aeruginosa 0254, 0225 and 1113. The combined application of BHK-P19 culture extract and Schizandra chinensis Baillon extract inhibited P. aeruginosa 0254, 0225, 0826, 1113, 1378, 1731 and 2492. Use of various concentrations of BHK-P19 culture extract and ampicillin markedly increased antibacterial activity against multi-drug resistant P. aeruginose 1113.

한약재발효액의 항생제 다제내성균에 대한 항균활성 및 항산화활성 (Antibacterial Activities of Fermented Sayuksan Ingredient Extracts for Multidrug-resistant Strains)

  • Sayuksan has been widely applied to treat a variety of diseases such as acute hepatitis, gastritis, and colitis. Sayuksan consists of medicinal herbs such as Glycrrhizae uralensis Fischer, Paeonia lactiflora Pallas, Bupleurum falcatum Linne, and Poncirus trifoliata Rafinesqul. Methanol extracts (1 mg/mL) from the four kinds of medicinal herbs did not show antibiotic activities against general test strains and multi-drug resistant strains. The antibacterial activity of fermented medicinal herbs extracts with Lactobacillus spp. strain was confirmed as Gram-positive bacteria which are higher than Gram-negative bacteria. Extracts of Glycrrhizae uralensis Fischer fermented with Lb. casei KCTC 3109 displayed inhibitory diameters of 16 mm against Pseudomonas aeruginosa P01828. Superoxide dismutase (SOD)-like activity of the medicinal herb extracts was not determined, but the extract of Paeonia lactiflora Pallas fermented with six strains of Lactobacillus spp. had the highest antioxidant activity. SOD-like activity of Paeonia lactiflora Pallas extracts fermented by Lb. brevis KCTC 3498 was $41.4{\pm}0.8%$, which was the highest antioxidant activity among the fermented extracts with the other medicinal herbs.
