Purpose: Gleditsiae spina (GS) has been used to treat patients with several diseases such as carbuncle, swelling and parasites. Recently GS is known to have anticancer activity in abdominal solid tumor, but the effects of GS on breast cancers is not clarified. For these reasons, we investigated effects of Gleditsiae spina (GS) on gene expression of human breast cancer cells. Methods: We investigated the effects of GS on proliferation of breast cancer cell line, MDA-MB-231. In addition, the genetic profile for the effect of GS on breast cancer cells was measured using microarray technique, and the functional analysis on these genes was conducted. Results: Total 1,434 genes were up-regulated and 2,483 genes down-regulated in the cells treated with GS. Genes induced or suppressed by GS were all mainly concerned with metabolic process, regulation of biological process and protein binding. The network of total protein interactions was measured using cytoscape program, and some key molecules that can be used for elucidation of therapeutical mechanism of medicine in future were identified. Conclusion: These results suggest possibility of GS as anti-cancer drug for breast cancer, and also suggest that related mechanisms are involved in regulation of intra-cellular metabolism in breast cancer cells.

Sarcopenia, the age-related muscle atrophy, is a serious concern as it is associated with frailty, reduced physical functions, and increased mortality risk. Protein supplementation is essential for preserving muscle mass, and horse meat can be an excellent source of proteins. Since sarcopenia occurs under conditions of oxidative stress, this study aimed to investigate the potential anti-muscle atrophy effect of horse meat hydrolysate using C2C12 cells. A horse meat hydrolysate less than 3 kDa (A4<3kDa) significantly increased the viability of C2C12 myoblasts against H2O2-induced cytotoxicity. Exposure of C2C12 myoblasts to lipopolysaccharide led to an elevation of cellular reactive oxygen species levels and mRNA expression of proinflammatory cytokines, including tumor necrosis factor-α and interleukin 6, and these effects were attenuated by A4<3kDa treatment. Additionally, A4<3kDa activated protein synthesis-related proteins through the protein kinase B/mechanistic target of rapamycin pathway, while decreasing the expression of activity and degradation-related proteins, such as Forkhead box O3, muscle RING finger protein-1, and Atrogin-1 in dexamethasone-treated C2C12 myotubes. Therefore, the natural material A4<3kDa has the potential of protecting against muscle atrophy, while further in vivo study is needed.
Objectives : While treatments for cancer are advancing, the development of effective treatments for cancer metastasis, the main cause of cancer patient death, remains insufficient. Recent studies on Dichroae Radix have revealed that its active ingredients have the potential to inhibit cancer metastasis. This study aimed to investigate the cancer metastasis inhibitory effect of Dichroae Radix using network pharmacological analysis. Methods : The active compounds of Dichroae Radix have been identified using Traditional Chinese Medicine System Pharmacology Database and Analysis Platform. The UniProt database was used to collect each of information of all target proteins associated with the active compounds. To find the bio-metabolic processes associated with each target, the DAVID6.8 Gene Functional classifier tool was used. Compound-Target and Target-Pathway networks were analyzed via Cytoscape 3.40. Results : In total, 25 active compounds and their 62 non-redundant targets were selected through the TCMSP database and analysis platform. The target genes underwent gene ontology and pathway enrichment analysis. The gene list applied to the gene ontology analysis revealed associations with various biological processes, including signal transduction, chemical synaptic transmission, G-protein-coupled receptor signaling pathways, response to xenobiotic stimulus, and response to drugs, among others. A total of eleven genes, including HSP90AB1, CALM1, F2, AR, PAKACA, PTGS2, NOS2, RXRA, ESR1, ESR2, and NCOA1, were found to be associated with biological pathways related to cancer metastasis. Furthermore, nineteen of the active compounds from Dichroae Radix were confirmed to interact with these genes. Conclusions : The results provide valuable insights into the mechanism of action and molecular targets of Dichroae Radix. Notably, Berberine, the main active ingredient of Dichroae Radix, plays a significant role in degrading AR proteins in advanced prostate cancer. Further studies and validations can provide crucial data to advance cancer metastasis prevention and treatment strategies.
Hypoxia-inducible factor-1 alpha (HIF-1α) is a transcription factor activated under hypoxic conditions, and it plays a crucial role in cellular stress regulation. While HIF-1α activity is essential in normal tissues, its presence in the tumor microenvironment represents a significant risk factor as it can induce angiogenesis and confer resistance to anti-cancer drugs, thereby contributing to poor prognoses. Typically, HIF-1α undergoes rapid degradation in normoxic conditions via oxygen-dependent degradation mechanisms. However, certain cancer cells can express HIF-1α even under normoxia. In this study, we observed an inclination toward increased normoxic HIF-1α expression in cancer cell lines exhibiting increased HDAC6 expression, which prompted the hypothesis that HDAC6 may modulate HIF-1α stability in normoxic conditions. To prove this hypothesis, several cancer cells with relatively higher HIF-1α levels under normoxic conditions were treated with ACY-241, a selective HDAC6 inhibitor, and small interfering RNAs for HDAC6 knockdown. Our data revealed a significant reduction in HIF-1α expression upon HDAC6 inhibition. Moreover, the downregulation of HIF-1α under normoxic conditions decreased zinc finger E-box-binding homeobox 1 expression and increased E-cadherin levels in lung cancer H1975 cells, consequently suppressing cell invasion and migration. ACY-241 treatment also demonstrated an inhibitory effect on cell invasion and migration by reducing HIF-1α level. This study confirms that HDAC6 knockdown and ACY-241 treatment effectively decrease HIF-1α expression under normoxia, thereby suppressing the epithelial-mesenchymal transition. These findings highlight the potential of selective HDAC6 inhibition as an innovative therapeutic strategy for lung cancer.
Rheumatoid arthritis (RA) is a systemic inflammatory autoimmune disorder with widespread synovitis. Isoflavones, the main active component of soy, have been reported to have potent anti-inflammatory effects; the previous RA animal models showed the promising effect of soy supplementation. We aimed to evaluate the effect of soy bread on inflammatory markers and lipid profiles in RA patients. The present study was designed as a randomized controlled trial. RA patients were randomly allocated to obtain soy bread (n = 22) or placebo bread (n = 22) for 8 weeks. Fasting serum levels of lipid profile, total antioxidant capacity (TAC), tumor necrosis factor-α (TNF-α), C-reactive protein (CRP), and DAS28 were checked. Findings showed that there were no significant differences between the two groups in physical activity and dietary intake at the beginning of the study and the end of the study. There were no significant differences between the two groups in measured lipid profile markers, including high-density lipoprotein, low-density lipoprotein, total cholesterol, triglyceride, and very low-density lipoprotein, at the end of the trial. In addition, TAC and CRP also were not significant at the end of the trial between the 2 groups (0.66 and 0.12, respectively). However, the serum levels of TNF-α reduced significantly in the soy bread group at the end of the intervention (p < 0.000) and compared with the control group (p < 0.019). Soy bread consumption only decreased circulating TNF-α serum concentration. Other outcome measures were not changed following supplementation. Future long-term, well-designed studies are needed to confirm these findings.
본 연구는 고욤잎, 감잎 및 뽕잎을 단독으로 사용하기보다 혼합하여 사용할 경우 고욤잎, 감잎 및 뽕잎 복합추출물이 아토피 피부염(AD) 증상을 억제하는지 효과를 밝히고, 항염 효능을 나타내는 기능성 소재로서의 이용 가능성을 알아보고자 하였다. 그 결과 육안 평가를 통해 피부의 건조 상태, 스케일링, 미란, 찰과상 및 홍반 같은 아토피 피부염 증상이 AD 모델에서 증가하였지만, 고욤잎, 감잎 및 뽕잎 각각의 추출물과 복합추출물을 투여하였을 경우 완화되는 것을 확인할 수 있었으며, 복합추출물이 고욤잎, 감잎 및 뽕잎 각각의 추출물보다 더 뛰어난 효과를 나타내었다. 피부 두께와 염증세포 및 비만세포의 침윤은 AD 모델에서 크게 증가하였지만, 고욤잎, 감잎 및 뽕잎 단독추출물과 복합추출물을 투여하였을 경우 감소하는 것을 확인할 수 있었으며, 복합추출물이 단독추출물보다 더 뛰어난 효과를 나타내었다. 혈청중의 IgE와 IL-4의 수치를 측정한 결과 AD 모델에서 많이 증가하였으나, 고욤잎, 감잎 및 뽕잎 단독추출물과 복합추출물을 투여하였을 경우 감소하는 것을 확인할 수 있었으며, 복합추출물이 단독추출물보다 더 뛰어나게 억제하는 효과를 나타내었다. HMC-1 세포에 복합추출물을 처리하였을 경우 염증성 사이토카인인 $TNF-{\alpha}$와 IL-6의 생성량이 유의적으로 감소하였으며, 또한 RAW 264.7 세포에 복합추출물을 처리하였을 경우 염증 매개 인자인 $TNF-{\alpha}$, IL-6, $PGE_2$ 및 NO 생성량이 감소하였고, 이러한 염증 매개 인자 억제 효능은 복합추출물이 단독추출물보다 더 뛰어나게 억제하는 것을 확인하였다. 이러한 결과로 미루어볼 때 고욤잎, 감잎 및 뽕잎을 혼합한 복합추출물을 사용할 때 효능이 더 우수한 것을 확인하였고, 아토피 피부염 증상 개선 및 염증관련 질환 치료를 위한 기능성 천연물 소재 및 제품 개발에 응용이 가능할 것으로 생각한다.
본 연구의 목적은 울금의 발효를 통해 쓴맛의 관능적 기호도를 향상시키면서, 발효 울금의 항산화 및 항염효능 검증을 위한 것으로, 발효 울금의 관능적 기호도 확인을 위해 맛센서 분석과 항산화 평가를 위해 총 폴리페놀 및 플라보노이드 함량 및 DPPH radical 소거능, 지표성분 검출을 위해 HPLC분석을 진행하였고, 항염증평가는 RAW 264.7 세포에서 LPS에 의해 유도되는 염증인자인 NO, $PGE_2$, iNOS, COX-2, $NF-{\kappa}B$, IL-6와 $TNF-{\alpha}$에 대한 감소효과를 측정하였다. 실험결과 Rhizopus oryzae로 울금의 발효가 진행 될수록 쓴맛이 감소함을 보였다. 총 폴리페놀, 플라보노이드 함량과 DPPH radical 소거능은 비 발효 울금보다 울금 발효 1, 3일차에 증가함을 보였다. 발효 기간별 발효 울금은 10, 50, $100{\mu}g/ml$ 모든 농도에서 세포독성이 나타내지 않았다. RAW 264.7 세포에 LPS로 유도된 염증인자인 $NF-{\kappa}B$, IL-6와 $TNF-{\alpha}$ 생성량이 발효 기간별 발효 울금의 염증인자 생성억제를 평가한 결과 $NF-{\kappa}B$와 IL-6에서 발효 울금 $100{\mu}g/ml$에서 현저히 생성량이 억제되었다. LPS로 유도된 cytokines 억제효과를 보인 발효 기간별 발효 울금($100{\mu}g/ml$)의 NO, $PGE_2$ 생성억제를 평가한 결과, 대조군과 비교해 LPS를 처리한 군에서 NO, $PGE_2$의 생성이 현저히 증가되었고, 울금 에탄올 추출물에 의해 유의성 있는 생성억제효과를 보였고 비 발효에 비해 발효 울금의 생성억제 효과를 보였다. 또한 COX-2와 iNOS의 단백질 발현 억제효과를 확인한 결과, 대조군과 비교해 LPS에 의해 증가 된 COX-2와 iNOS의 단백질발현이 발효 울금에 의해 감소됨을 확인하였다. 위 결과 발효 울금은 RAW 264.7 세포에서 LPS에 의해 유도되는 NO와 $PGE_2$의 생성억제, iNOS와 COX-2의 발현을 억제시켰으며, $NF-{\kappa}B$, IL-6와 $TNF-{\alpha}$의 분비량도 억제시켰다. 이는 발효 울금의 항산화와 항염증성을 입증하기 위한 기초자료로 활용이 가능하다고 사료된다.
연구배경: 정상세포는 보호되고 종양세포에 독성을 보인다고 알려진 TNF유전자족으로 새로이 확인된 TRAIL이 폐암세포에서 보이는 아포프토시스 효과를 확인하고, 아포프토시스로부터 세포를 보호하는 전사인자 $NF-{\kappa}B$가 TRAIL에 의하여 활성화 되는 정도를 평가하여 MG132의 $NF-{\kappa}B$활성억제가 TRAIL 유도성 아포프토시스를 감작시키는지를 확인하기 위하여 본 연구를 시행하였다. 방법: A549(wt p53) 및 NCI-H1299(null p53) 폐암세포주를 사용하였다. 세포독성 검사는 MTT assay를 이용하였고 아포프토시스는 Annexin V assay와 FACS 분석을 이용하였다. $NF-{\kappa}B$ 전사활성은 luciferase reporter gene assay를 이용하였고 $I{\kappa}B{\alpha}$ 분해는 western blot을 이용하였으며, TRAIL에 의해 활성화된 $NF-{\kappa}B$와 DNA 결합은 electromobility shift assay와 anti-p65 antibody를 이용한 supershift assay로 확인하였다. 결과: 1) TRAIL 100 ng/ml 농도에서 wild-type p53인 A549 폐암세포는 34.4%, p53 null인 NCI-H1299 폐암세포는 26.4%의 세포사를 관찰하였다. 2) Luciferase reporter gene assay로서 TRAIL에 의한 $NF-{\kappa}B$의 활성이 A549 $IgG{\kappa}B-luc$세포에서 2.45배 증가하고 NCI-H1299 $IgG{\kappa}B-luc$세포에서는 1.47배 증가함을 관찰하여 TRAIL에 의하여 $NF-{\kappa}B$가 활성화됨을 확인하였다. 3) MG132의 전처치로 TRAIL에 의한 $NF-{\kappa}B$의 활성이 A549 세포와 NCI-H1299 세포에서 각각 기저수준의 0.24, 0.21배로 강력히 억제되었다. 4) TRAIL단독으로 30% 전후의 세포독성이 MG132 전처치 후 TRAIL을 투여하면 두 세포주 모두에서 80% 이상의 세포독성이 관찰되어 MG132가 TRAIL유도성 아포프토시스에 감작효과가 있음을 확인하였다. 결론: 이상의 결과로 TRAIL에 상대적인 내성을 보이는 폐암세포주에서 MG132가 $NF-{\kappa}B$ 활성억제로서 TRAIL유도성 아포프토시스를 강화시키는 효과가 있음을 확인할 수 있었다. 따라서 본 연구는 향후 폐암치료에 있어서 TRAIL유도성 아포프토시스가 이용될 수 있는 가능성을 확인한 기초자료가 된다고 생각된다.
연구배경 : 종양괴사인자(tumor necrosis factor ; TNF)는 다양한 생물학적 기능을 가지고 있는바, 그 중 생체 외에서 증명된 뚜렷한 항암 효과로 말미암아 최근 항암유전자요법의 중요한 대상으로 관심을 모으고 있다. 그러나 TNF의 세포독성의 기전에 대해서는 확실히 밝혀진 바가 없고 반응성 산소기가 관여할 것임을 시사하는 연구들이 있었으나, 항산화제에 의해서 TNF의 세포독성이 감소되는 지에 대해서는 상충되는 결과가 있었다. 본 연구는 TNF의 세포독성의 기전에서 반응성 산소기의 역할을 알아보기 위해 TNF 감수성 종양세포에 여러 가지 항산화제를 전처치한 후의 TNF 세포독성이 감소하는 지를 대조군과 비교하였다. 방 법 : TNF에 비교적 감수성을 보이는 세포주인 WEHI164(murine fibrosarcoma cell line)와 ME180(human cervix cancer cell line)을 배양하여 각각 3가지 항산화제로 전처치한 것과 대조군을 TNF 10ng/ml로 세포독성을 유도하고 그 정도를 MTT(dimethylthiazolyl-diphenyltetrazolium bromide) assay를 이용하여 세포독성 정도를 측정 비교하였다. 결 과 : 항산화제를 전처치하지 않은 대조군에서는 WEHI164 세포와 ME180 세포가 각각 $92.2{\pm}2.8%$와 $59.9{\pm}7.0%$의 세포독성을 보였고, TMTU를 전처지한 군은 각각 $91.4{\pm}3.7%$와 $74.6{\pm}7.0%$, PMZ을 전처치한 군은 $90.2{\pm}2.5%$와 $62.5{\pm}5.7%$, BHT을 전처치한 군은 $93.2{\pm}1.3%$와 $66.3{\pm}6.1%$로 세포독성의 유의한 감소는 없었다. 결 론 : 종양세포에 대한 TNF의 세포독성은 항산화제에 의해 감소되지 않으며 따라서 TNF의 세포독성은 반응성 산소기에 의한 것이 주된 기전은 아니라고 판단된다.
세포괴사는 세포막의 파열, HMGB1을 포함한 세포 내용물의 세포외부로의 방출 등을 수반하는 세포죽음이다. HMGB1은 핵 단백질로 전사조절자로 작용하지만 세포괴사에 의해 세포 밖으로 방출되면 염증을 유발하고 암을 촉진하는 cytokine으로 작용한다. HMGB1의 과발현은 암 발생 및 항암제 저항과 밀접한 연관성을 가지고 있지만, 그 기작에 대한 연구는 미흡한 실정이다. 본 연구에서는, HMGB1이 항암제에 의한 세포 죽음에 미치는 영향을 조사하였다. 그 결과, HMGB1은 MCF-7, MDA-MB231, MDA-MB361 세포에서 cisplatin에 의한 세포사멸을 억제하고 세포운명을 세포괴사로 바꾼다는 사실을 확인하였다. HMGB1의 세포사멸-세포괴사 전환 작용을 4-HC를 처리한 세포에서도 관찰되었다. 그러나, HMGB1은 docetaxel (DOC)에 의한 세포사멸에는 영향을 주지 않음을 확인하였다. MTS를 이용하여 항암제에 의한 세포 죽음에 미치는 영향을 조사한 결과, necrotic core가 형성된 8일째 MCF-7 MTS에서 cisplatin에 의한 세포사멸이 세포괴사로 바뀌는 반면, DOC에 의한 세포사멸은 세포괴사로 전환되지 않는 것을 확인하였다. 또한 spheroid에서 HMGB1 receptor인 RAGE의 발현이 증가함을 확인하였다. 이러한 결과를 통해, HMGB1이 alkylating agent에 의한 세포사멸을 세포괴사로 전환시킴을 알 수 있었다. 따라서, alkylating agent에 의한 항암제 효능을 나타내기 위해선, 이들 항암제의 부작용 즉 세포괴사를 억제하는 전략이 필요한 것으로 생각된다.

① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
