Comparative Study of Bojungikgitang in Korea, Japan and China on the Anti-Inflammatory and Anti-Oxidative Effects (보중익기탕 (補中益氣湯)의 한국, 중국, 일본 처방에 대한 항염증 및 항산화 효과 비교 연구)

  • Objectives : Bojungikgitang (BJT), the Oriental medical prescription has been traditionally used about improvement of immune response and infective disease at Asian nation. In this study, we has compared about the anti-inflammatory and antioxidative effects on BJT of three countries including Korea (Korean Traditional Medicine, KTM), China (Traditional Chinese Medicine, CTM) and Japan (Japanese Traditional Medicine, JTM). Methods : We has basically using LPS-stimulated RAW 264.7 cells. The expression of these inflammatory mediators has measured using enzyme-linked immunosorbent assay (ELISA) and reverse transcription polymerase chain reaction (RT-PCR). Also, free radical scavenging assay has tested for anti-oxidative activity as well as the contents of total flavonoid and polyphenol. Results : As a result, we were founded the inhibitory effects of BJT (KTM, CTM, JTM) on LPS-induced production of NO, TNF-${\alpha}$ and IL-6 as well as the anti-oxidative activities. Especially the KTM was most effective in anti-inflammatory and anti-oxidative activities. Conclusions : These results indicate that BJT (KTM, CTM, JTM) has a good anti- inflammatory and anti-oxidative effects. But, there were degree of effects on between pharmacopoeia of the countries. Thus, further study is required that find appropriate methods for extracting as well as establish of standardized processes in order to improve the quality of BJT (KTM, CTM, JTM) as an anti-inflammatory and anti-oxidative agent for treatment of inflammatory diseases.

Quantitative Analysis of Chemical Composition and In Vitro Anti-oxidant and Anti-inflammatory Properties of Asparagus oligoclonos (방울비짜루 지표성분의 함량분석과 항산화·항염증 활성)

  • Asparagus oligoclonos is one of the endemic halophytes used folk medicine in Korea. We isolated the main compound rutin from methanol extracts of A. oligoclonos based on nuclear magnetic resonance and TOF ESI-MS data. We have investigated the quantitative analysis method of main compound using HPLC and the results exhibit that rutin content of A.oligoclonos were 1.816%. To explore anti-oxidant from A. oligoclonos ethanol extracts (AOE), we investigated the antioxidant effects of AOE on $H_2O_2$-induced oxidative stress in RAW 264.7 cell. AOE were reduced $H_2O_2$-induced oxidative stress via enhancement of cell viability, and AOE significantly decreased ROS production depending on concentration. Next, to screen for anti-inflammatory activity of AOE, we investigated the inhibitory effects of AOE in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. AOE had no effect on cell viability at a concentration of $500{\mu}g/mL$. Nitric oxide (NO) production inhibited in a dose-dependent manner. These results suggest that AOE may be a useful antioxidant and anti-inflammatory agent.

  • With the high morbidity and mortality caused by cancer, finding new and more effective anti-cancer drugs is very urgent. In current research, biotransformation plays a vital role in the research and development of cancer drugs and has obtained some achievements. In this review, we have summarized four applications as follows: to exploit novel anti-cancer drugs, to improve existing anti-cancer drugs, to broaden limited anti-cancer drug resources and to investigate correlative mechanisms. Three different groups of important anti-cancer compounds were assessed to clarify the current practical applications of biotransformation in the development of anti-cancer drugs.

Study on The Anti-HSV (Herpes Simplex Virus) Activity of Korean Traditional Prescriptions (Herb complexes) (한약 탕제분획의 항 Herpes simplex virus 활성에 관한 연구)

  • Herpes simplex viruses(HSV) are one of the most common infectious virus of man. Though chemotherapies and antibiotics against HSV have been developed in many countries, but anti-HSV agents were not satisfactory to mankind by their toxic reaction and side effects. In order to search for anti-HSV agents from Korean traditional prescriptions, we extended the number of specimens. Both methanol extract and boilling water extract of the Korean traditional prescriptions were screened to detect anti-HSV activities by MTT assay. Korean traditional prescriptions showing anti-HSV activities as methanl extracts were Paekyopsan, Chesupwilyungtang, Yongdamsagantang, and prescription 11. Four methanol extracts showing anti-HSV activities were freationated by hexane and their efficacies were tested. Hexane freationations of Paekyopsan, Chesupwilyungtang, and prescription 11 showed in anti-HSV activities both haxane and methanol fractionation.

Screening of Functional Rhizopus stolonifer for Alcohol Fermentation and Production of High Quality Korean Traditional Rice Wine

  • Different strains of mold were screened for the production of high quality Korean traditional rice wine with anti-hypertension and good acceptability. We isolated 867 nuruk mold strains and selected 24 for further study based on measurement of amylase activity. Among them, mold No. 17 showed high ethanol production upon fermentation with Saccharomyces cerevisiae as well as anti-hypertensive properties. The No. 17 strain was therefore selected as the functional mold and later identified as Rhizopus stolonifer based on molecular biological characteristics. Optimal fermentation conditions for the brewing of anti-hypertensive traditional rice wine comprised the addition of R. stolonifer No. 17 koji at a concentration of 35 sp/g and a fermentation period of 10 days at $25^{\circ}C$ using S. cerevisiae.

Whitening and Anti-wrinkle Effects of Korean Native Dendrobium moniliforme Methanol Extract (한국 자생 석곡 메탄올 추출물의 미백 및 주름개선 효과)

  • Dendrobium moniliforme (DM) is a valuable and versatile herbal medicine with the anecdotal claims of antioxidant and anti-inflammation. In the present study, we investigated the whitening and anti-wrinkling effects of DM under various conditions with B16F10 melanoma cells and human dermal fibroblasts. The DM extract inhibited melanin contents and tyrosinase activity in a dose-dependent manner, compared with untreated group. Treatment of the DM extract effectively suppressed the ${\alpha}$-MSH-stimulated melanin formation, tyrosinase activity and dendrite outgrowth. Moreover, the ${\alpha}$-MSH-induced mRNA expressions of tyrosinase-related protein-1 (TRP-1) and tyrosinase-related protein-2 (TRP-2) and protein expression of tyrosinase were significantly attenuated by DM treatment. We also investigated the DM increased the production of type I procollagen and inhibited TNF-${\alpha}$-induced mRNA expressions of MMP-1, -3 in the human dermal fibroblast. These results indicate that DM may be a great cosmeceutical ingredient for its whitening and anti-wrinkle effects.

Analysis of Research Trend on Ultraviolet Induced Skin Damage in the Korean Traditional Medicine Field (자외선으로 유도된 피부손상에 대한 한의학계의 연구동향)

  • Objective : In order to establish the scientific research methods to prove the effects of korean traditional medicine about ultraviolet induced skin damage, we investigated the research on ultraviolet induced skin damage in the korean traditional medicine field. Method : We searched the papers about ultraviolet induced skin damage published in the journals of korean traditional medicine. The searching end date was on June 29, 2015. Results : Thirty papers about ultraviolet induced skin damage were found. Twenty nine papers were experimental research, and one paper was clinical research. Sixty-three percent of papers were written the years after 2011. Fifty-seven percent of papers were published in the korean journal of oriental physiology & pathology and the journal of korean oriental medical ophthalmology & otolaryngology & dermatology. Main outcome measures of experimental research were anti-wrinkle effects, anti-oxidant capacity, anti-apoptosis effects, whitening effects and anti-inflammatory effects. Conclusion : We need more systematic research in order to prove the effects of korean traditional medicine, and use extensively.

Study on the Anti-hypertension mechanism of Prunella Vulgaris based on entity grammar systems

  • Literatures and experimental studies have shown that Prunella has an effect on anti-hypertension, however, its components are complicated, so that it is still difficult to clear the specific roles of its various components in blood pressure regulation in. So we decide to systematically study the anti-hypertension mechanism of Prunella. We integrated multiple databases and constructed molecular interaction network between the chemical constituents of Prunella Vulgaris and hypertension based on entity grammar systems model. The network has 262 nodes and 802 edges. Then we infer the interactions between chemical compositions and disease targets to clarify the anti-hypertension mechanism. Finally, we found Prunella could influence hypertension by regulating apoptosis, cell proliferation, blood vessel development and vasoconstriction, etc. Thus this study provides reference for drug development and compatibility, and also gives guidance for health care at a certain extent.

Chemical constituents of Dicentra spectabilis and their anti-inflammation effect

  • Column chromatographic separation of the MeOH extract from the roots of Dicentra spectabilis yielded fourteen compounds, menisdaurin (1), menisdaurilide (2), trans-N-p-coumaroyltyramine (3), trans-N-p-feruloyltyramine (4), 4-O-feruloylquinicacid (5), chlorogenic acid (6), 3-O-feruloylquinicacid (7), ferulic acid (8), protopine (9), Kaempferol 3,7-di-O-${\beta}-{\text\tiny{D}}$-glucopyranoside (10), kaempferol 3-O-${\beta}-{\text\tiny{D}}$-glucopyranosyl-7-O-${\alpha}-{\text\tiny{L}}$-rhamnopyranoside (11), ${\alpha}-rhamnoisorobin$ (12), astragalin (13), and nicotiflorin (14). Their structures were determined on the basis of NMR spectroscopic data. Among them, compound 1, 3-8, and 10-14 isolated from this plant were reported for the first time. The isolated compounds (1-14) were tested for nitric oxide (NO) inhibitory activity on lipopolysaccharide-stimulated RAW 264.7 cells. Compound 3, 4 and 12 significantly inhibited NO production. Moreover, Compound 3 suppressed pro-inflammatory cytokines ($TNF-{\alpha}$, $IL-1{\beta}$ and IL-6) in a dose- dependent manner. These data suggest that compound 3 possess anti-inflammatory activity and might be useful natural materials for development of anti-inflammatory agent.
