Immuno-stimulating, Anti-stress and Anti-thrombotic Effects of Unossified Velvet Antlers

  • Both ethanol and water extracts of unossified velvet antler were found to exhibit a significant immuno-stimulating activity as measured by carbon clearance test in mice, a remarkable anti-fatigue effect in weight-loaded forced swimming performance in mice, a significant anti-stress effect on immobilization in rats. The antler extracts also showed a weak but significant anti-thrombotic activity. These findings are indicative of adaptogenic properties of antlers and their normalizing effects during stressful condition.

구속스트레스를 가한 흰쥐에서 Cholate류의 항스트레스 작용에 관한 연구 (Anti-stress Effect of Cholic acid Derivatives in Restraint Stress Induced Rats)

  • This study was done to investigate whether cholic acid derivatives have anti-stress activity and what is a cause of this anti-stress effect. Seven cholic acid derivatives (cholic acid, taurocholic acid, ursodeoxycholic acid, tauroursodeoxychoic acid, chenodeoxy cholic acid, dehydrocholic acid, hyodeoxycholic acid) were used, silymarin and valproic acid were used as positive controls. Stress was induced by restraint immobilization technique plus water immersion (24hrs) and adrenal weight, spleen weight, adrenal ascorbic acid, serum cholesterol, lactate dehydrogenase (LDH), alkaline phosphatase (ALP), adrenal cholesterol, glucose and corticosterone levels were measured as stress indicators. Most cholic acid derivatives markedly decreased the adrenal weight, and TUDCA and DHCA increased the spleen weight. The restraint stress induced increments in serum LDH, ALP and cholesterol were attenuated by most cholic acid derivatives. Cholic acid, taurocholic acid and tauroursodeoxycholic acid only increased the content of adrenal ascorbate. While valproic acid showed an inhibitory effect against stress, silymarin did not. Our findings suggest that most cholic acid derivatives have anti-stress effect and that their anti-stress effect is, in part, related to choleretic activity.

담츱산류의 항스트레스 효능에 관한 연구 (A Study on Anti-Stress Activities of Cholic Acid Derivatives)

  • This study was done to investigate whether cholic acid derivatives have anti-stress activity in various stress models. Two cholic acid derivatives, ursodeoxycholic acid (UDCA) and tauroursodeoxycholic acid (WDCA), were used. physical, psychological, chemical and environmental stress models were performed. Adrenal weight, serum glucose levels and ALP activity were elevated in restraint stress model, but this elevation was prevented by UDCA treatment. Moreover, UDCA and TUDCA inhibited exploratory and spontaneous movements in oscillation stress model. In alcohol-induced stress model, TUDCA improved rotarod performance. UDCA and TUDCA significantly reduced the involution of lymphoid organs and the increment of WBC counts in cold stress model. These findings suggest that choric acid derivatives have antistress effects in various stress models.

[6]-Gingerol Attenuates Radiation-induced Cytotoxicity and Oxidative Stress in HepG2 Cells

  • [6]-Gingerol, a major polyphenol of ginger (Zingiber officinale), exhibits a variety of biological properties including anti-oxidant, anti-inflammatory and anti-cancer activity. However, the radioprotective effect of [6]-gingerol is still unknown. The aim of this study was to investigate the radioprotective effect of [6]-gingerol against radiation-induced cell cytotoxicity and oxidative stress in HepG2 cells. [6]-Gingerol pretreatment attenuated radiation-induced cell cytotoxicity caused by 5Gy (half lethal dose, $LD_{50}$ of HepG2 cells). The measurements of superoxide dismutase (SOD) and catalase (CAT) activity were also performed. The results showed that [6]-gingerol pretreatment reduced increasing SOD and CAT activity after exposure of IR, indicating that [6]-gingerol protected oxidative stress by regulating cellular antioxidant enzyme (SOD and CAT) activity. These findings suggest that [6]-gingerol acts as a radioprotector by attenuating cell cytotoxicity and oxidative stress.

가미천마구등음의 항스트레스 효과에 대한 실험적 연구 (The Study of KCG Extract Under the Anti-Stress Effect of Rats)

  • To determine the anti-stress effect of KCG(加味天麻鉤藤飮) extract on sprague-dawley rats. we conducted a research about the change of weight, activity, reactivity, c-fos protein, cytotoxicity against PC12 cell line and heal shock protein. 1) KCG extract siginificantly inhibited the decrease of body weight induced by stress, compared with the control group. 2) KCG extract had no siginificant effect in the activity and reactivity of rats between the control and the experimental groups. 3) KCG extract siginificantly restrained c-fos protein manifestation, compared with the control group. 4) KCG extract siginificantly restrained heat shock protein, compared with the control group. These results suggested that KCG might be usefully anti-stress effect.

정향 추출물의 항산화.항혈소판 응집효과 및 혈전 용해능 탐색 (Screening of Antioxidative, Anti-platelet Aggregation and Anti-thrombotic Effects of Clove Extracts)

  • Clove has been frequently used as anti-diabetic, anti-microbial, anti-inflammatory, anesthetic drug and remedies for stomachache by coldness. In this study, the antioxidant activity of extract from Clove was studied in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$-induced human LDL oxidation. Anti-platelet aggregation and anti-thrombotic effects of Clove extracts were studied ex vivo methods by mesuring the inhibitory effect on thrombin induced platelet aggregation and the fibrinolytic activity. The Clove extracts were found to have a potent scavenging activity, as well as an inhibitory effect on LDL oxidation in vitro. Moreover Clove extracts were exhibited remarkable inhibitory effect on platelet aggregation and fibrinolytic activity. In conclusion, the Clove extracts have anti-oxidative and anti-atherosclerotic effects in vitro and ex vivo system, which can be used for developing pharmaceutical drug against oxidative stress and atherosclerosis.

The anti-oxidant activities of processed fruits and vegetables in APAP induced oxidative stress in BALB/c mice

  • There is a strong connection between the diet rich in antioxidants and the decreased incidence of cardiovascular and cancerous diseases. Diets that are rich in anti-oxidants particularly include fruits and vegetables containing the high amounts of vitamin A-E, carotenoids, and minerals. Different processing conditions applied for vegetables and plants results in the alteration of the nutrients present in them. Therefore the rationale of our study was to compare the antioxidant effects of different processed vegetables and plants and to see that which one of them showed best anti-oxidant activity. For this purpose, we have used acetaminophen induced oxidative stress model in mice to check the effects of processed apple, pear, carrot, cabbage, broccoli and radish. Our results have shown that the administration of these samples effectively decreased the expression of parameters related with oxidative stress like ALT, AST, catalase, superoxide dismutase, GPx and 8-OHdG. Moreover they also significantly protected the mice livers from APAP induced damage as shown by histological changes. Therefore our results have demonstrated the effects of processed fruits and vegetables in mice model of oxidative stress.

Ellagic acid plays a protective role against UV-B-induced oxidative stress by up-regulating antioxidant components in human dermal fibroblasts

  • Ellagic acid (EA), an antioxidant polyphenolic constituent of plant origin, has been reported to possess diverse pharmacological properties, including anti-inflammatory, anti-tumor and immunomodulatory activities. This work aimed to clarify the skin anti-photoaging properties of EA in human dermal fibroblasts. The skin anti-photoaging activity was evaluated by analyzing the reactive oxygen species (ROS), matrix metalloproteinase-2 (MMP-2), total glutathione (GSH) and superoxide dismutase (SOD) activity levels as well as cell viability in dermal fibroblasts under UV-B irradiation. When fibroblasts were exposed to EA prior to UV-B irradiation, EA suppressed UV-B-induced ROS and proMMP-2 elevation. However, EA restored total GSH and SOD activity levels diminished in fibroblasts under UV-B irradiation. EA had an up-regulating activity on the UV-B-reduced Nrf2 levels in fibroblasts. EA, at the concentrations used, was unable to interfere with cell viabilities in both non-irradiated and irradiated fibroblasts. In human dermal fibroblasts, EA plays a defensive role against UV-B-induced oxidative stress possibly through an Nrf2-dependent pathway, indicating that this compound has potential skin antiphotoaging properties.

백작약 추출물이 항산화활성, LDL 산화 억제 및 혈전용해에 미치는 영향 (Preventive Effects of Peony Root Extracts on Oxidative Stress, Thrombosis and Atherosclerosis)

  • Objectives: There is currently increased interest in the identification of natural antioxidant compounds derived from various plants. Peony Root (PR) is used worldwide for the treatment of many types of cardiovascular disease including atherosclerosis and hypertension. It has been used in Korean traditional medicine for the treatment of glycosuria, hypertension and cancer. However, to date, no studies concerning the antioxidant properties of PR have been conducted. Therefore, this study was conducted to evaluate the in vitro scavenging activity, inhibitory effect of LDL oxidation of pro-oxidant reactive species and anti-thrombosis effect in response to treatment with PR using various screening methods including biological and non-biological oxidants. Methods: In this study, the antioxidant activity of extract from PR was studied with in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$-induced human LDL oxidation and the inhibitory effect on collagen-induced platelet aggregation. Results: The PR extracts were found to have a potent scavenging activity of oxidative stress [DPPH, superoxide anion, hydroxyl radical, nitric oxide and peroxynitrite, etc.] as well as an inhibitory effect on LDL oxidation and on platelet aggregation. Conclusions: The PR extracts have anti-oxidative and anti-atherosclerotic effects in vitro system, which can be used for developing pharmaceutical drugs against oxidative stress and atherosclerosis.

한국독활(韓國獨活)과 중국독활(中國獨活)의 항산화효능 비교 연구 (Comparative Studies on the Anti-oxidation Activities of Aralia continentalis Root and Angelica pubescens Root)

  • Objectives : The present study compared Aralia continentalis Root and Angelica pubescens Root used in Korea and China respectively concerning their anti-oxidant effect Methods : We tested the anti-oxidant effect through in vitro experiment and in vivo experiment that induced oxidative stress using ethanol. Results : 1. DPPH scavenging activity was stronger in Aralia continentalis Root than in Angelica pubescens Root 2. Superoxide anion radical scavenging activity was similar between Aralia continentalis Root and Angelica pubescens Root 3. The linoleic acid peroxidation inhibition effect was stronger in Aralia continentalis Root than in Angelica pubescens Root. 4. The phenolic component was higher in Aralia continentalis Root than in Angelica pubescens Root. 5. Both Aralia continentalis Root and Angelica pubescens Root increased the concentration of GSH and decreased SOD activity in mice, in which oxidative stress was induced, and the effect was stronger in Aralia continentalis Root. 6. Aralia continentalis Root increased GSH peroxidase activity but Angelica pubescens Root did not have such an effect. 7. Neither Aralia continentalis Root nor Angelica pubescens Root had a significant effect on catalase, ADH and ALDH in mice, in which oxidative stress was induced. Conclusions : Aralia continentalis Root has a stronger anti-oxidant effect than Angelica pubescens Root. Thus, although Aralia continentalis Root is not an original plant recorded in botanical literature, it may be usable based on the data about its effects.
