Anti-platelet Effect of the Phenolic Constituents Isolated from the Leaves of Magnolia obovata

  • In the course of our work on anti-platelet constituents from plants, eight phenolic compounds, $({\pm})-syringaresinol$ (1), 4-hydroxybenzaldehyde (2), 4-hydroxybenzoic acid (3), vanillic acid (4), 4-hydroxy cinnamic acid (5), quercetin 3-O-rhamnoside (6), rutin (7), and quercetin $3-(2^G-rhamnosylrutinoside)$ (8) were isolated from the methanol extract of the leaves of Magnolia obovata. The compounds were identified based on the spectroscopic data. Compound 2, 3, 5, 6, 7 and 8 were isolated for the first time from genus Magnolia. 1 and 6 showed same order of inhibitory potencies as acetylsalicylic acid (ASA) to rat platelet aggregation induced by all the stimulators tested. The remaining six compounds showed only mild effects.

  • 혈소판 응집은 혈관 손상의 경우 지혈 플러그 형성에 필수적이다. 그러나, 과도한 혈소판 응집은 혈전증, 죽상 동맥 경화증 및 심근 경색과 같은 순환기 장애를 일으킬 수도 있다. Scopoletin은 Scopolia 또는 Artemisia 속 식물의 뿌리에서 발견되는 성분으로, 항응고 및 항말라리아 작용을 가지는 것으로 알려져 있다. 본 연구는 collagen에 의해 유발된 혈소판 응집에 scopoletin이 미치는 영향을 조사하였다. Scopoletin은 활성화된 혈소판에서 생성되는 응집 유도 분자인 thromboxane $A_2$ ($TXA_2$) 및 세포 내 $Ca^{2+}$ 동원 ($[Ca^{2+}]_i$)의 하향 조절을 통해 항 혈소판 효과를 나타내었다. 한편, scopoletin은 세포 내 $Ca^{2+}$-길항제인 것으로 알려져 있는 cyclic adenosine monophosphate(cAMP)와 cyclic guanosine monophosphate (cGMP) 수치를 증가시켰다. 특히, scopoletin은 cGMP보다 cAMP 수준을 강력하게 증가함으로써 콜라겐에 의해 유발된 사람 혈소판 응집에서의 ${\alpha}IIb/{\beta}_3$에 대한 피브리노겐 결합을 억제하였다. 또한, scopoletin은 용량 의존적으로 collagen에 의해 증가된 adenosine trisphosphate (ATP)의 방출을 억제하였다. 이 결과는 혈소판 내 과립 분비를 통한 응집 증폭작용이 scopoletin에 의해 억제되었음을 의미한다. 따라서, 본 연구는 scopoletin이 강력한 항혈소판 효과를 가지며 혈소판-유래의 혈관 질환을 예방할 가능성이 크다는 것을 입증하였다.

창포류 추출물이 인간 전혈혈소판 응집억제에 미치는 영향 (Effects of Acori Rhizoma Extract on the in vitro Anti-platelet Activity in Human Whole Blood)

  • Objectives : Acori Rhizoma is one of the common widely used herbal medicines with diverse bioactive effects. However, little evidence has been reported about the potential anti-platelet activity of Acori Rhizoma. The present study examined the effects on platelet aggregation by Acori Rhizoma. Methods : In this study, we tested the in vitro effect of 16 kinds of Acori Rhizoma extracts by hot water or 70% ethanol on collagen-induced platelet aggregation in human whole blood using the impedance method of aggregometry. Results : Among them, 2 kinds of 70% ethanol extract and 1 kind of hot water extract showed the significant inhibiting effect on whole blood aggregation. In particular, Acorus gramineus extracts were selected as the most effective candidate. Conclusiions : The results from this experiment provide pharmacological evidence for the traditional medicine, suggesting that Acorus gramineus could be help problems of blood circulation more than Acorus tatarinowii.

Anti-platelet Activity of Tissue-cultured Mountain Ginseng Adventitious Roots in Human Whole Blood

  • Present study investigated the effects of the 70% ethanol extracts of tissue-cultured mountain ginseng (TCMG), Korean red ginseng (KRG), and Panax ginseng (PG) on agonist-induced platelet aggregation and activation in human whole blood. The $IC_{50}$ values for TCMG, KRG, and PG were 1.159, 3.695, and 4.978mg/mL for collagen-induced aggregation, 0.820, 2.030, and 4.743mg/mL for arachidonic acid-induced aggregation, and 1.070, 2.617, and 2.954 mg/mL for ADP-induced aggregation, respectively. Also, this study assessed the effects of the most active extract, TCMG, on markers of platelet activation by determining receptor expression on platelet membranes in healthy subjects, including expression of GPIIb/IIIa-like (PAC-1) and P-selectin (CD62), by flow cytometry. A significant decrease in PAC-l expression (p=0.018) was observed in the presence of TCMG. These results show that TCMG has potent anti-platelet activity.

독활의 항혈소판 및 항산화 효과 (Effects of Aralia continentalis Kitagawa on Antiplatelet and Antioxidative Activities.)

  • 독활 메탄올 추출물과 그 분획의 항혈전 및 항산화 효과를 확인하기 위하여 ADP를 이용하여 혈소판 응집억제효과를 탐색하고 DPPH 및 ABTS-radical 소거능을 측정하여 항산화능을 측정하였다. 그 결과 독활, 특히 EtOAc 분획은 ADP에 의해 유도된 혈소판응집과 트롬빈에 의한 혈소판 부착에 대하여 농도 의존적으로 저해효과를 나타내었으며, 높은 폴리페놀 함량과 가장 높은 라디칼 소거능을 통하여 독활의 항산화 효과를 확인하였다. 이상의 결과로 독활 EtOAc 분획의 높은 DPPH 및 ABTS radical 소거능이 항혈소판 효과에 영향을 미치는 것으로 사료된다.

  • We have previously reported that green tea catechins (GTC) displayed anti-thrombotic activity, and that this might be due to anti-platelet rather than anti-coagulation effects. In the present study, we have studied the anti-platelet activity and mechanism of gallocatechin gallate (GCG), which is a component of GTC. GCG inhibited the collagen- and U46619-induced aggregation of rabbit platelets, with IC$\^$50/ values of 63.0 and 48.3 ${\mu}$M, respectively. GCG also inhibited collagen-induced serotonin release and TxB$_2$ formation in a similar manner of platelets aggregation. (omitted)

백지 BuOH 가용분획의 항혈전 활성에 관한 연구 (Antithrombotic Effect of the BuOH Soluble Fraction of Angelica dahurica Root)

  • Several coumarins isolated from Angelica sp. were described to show inhibitory effects against human platelet aggregation. The anti-thrombotic and anti-platelet potential was evaluated, in this paper, with the BuOH soluble fraction of Angelica dahurica root. The BuOH fraction was divided into five subfractions fr. A - E and tested in the mouse model of thrombosis. Survival was enhanced to 35% with fr. A or fr. E treated (500 mg/Kg, p.o.) group of mice compared with 5% survival of the control group. However, none of the 8 coumarin glycosides obtained from fr. A, at the conc. of 0.5 mg/ml, showed inhibitory effects against rat platelet aggregation induced by ADP or collagen.

목초액의 혈소판 응집억제를 통한 혈행개선 작용에 관한 연구 (Improvement of Haemostasis Mediated by Anti-Platelet Activities by Plant Vinegar)

  • We investigated the effects of plant vinegar on platelets and blood coagulation system. Plant vinegar inhibited in vitro platelet aggregation in a concentration dependent manner, when platelets were activated by thrombin and collagen. In addition, plant vinegar showed inhibitory effects on the serotonin secretion induced by thrombin in a concentration dependent manner. However, treatment with plant vinegar to platelets did not induce any cytotoxicity, as determined by the release of lactate dehydrogenase. Plant vinegar did not change the coagulation parameters such as activated partial thromboplastin time (aPTT) and prothrombin time (PT) using rat citrated plasma. In vivo study revealed that, treatment with plant vinegar prolonged the bleeding time from mouse tail. All these results suggest that plant vinegar might improve blood hemostasis mediated via anti platelet activities.

  • Rhus verniciflua Stokes (RVS) is a widely used herbal plant with various biological properties. Our previous study using in vitro platelet aggregation in whole blood showed that ethyl acetate layer of RVS had strong anti-aggregatory activity. In this study. to investigate the anti-aggregatory activity and antioxidative effects of RVS ethyl acetate layer. the layer was subsequently fractionated by ODS columm chromatograph (50% MeOH). As a result. the fraction 3 was most inhibited the aggregation of platelet in rat whole blood induced by thrombin and all fraction of RVS was detected strong antioxidative effect. (omitted)

  • In the course of our work on anti-platelet constituents from plants, five phenolic compounds, magnolol, honokiol, obovatol, methyl caffeate, and syringin, were isolated from the methanol extracts of the barks and fruits of Magnolia obovata. The compounds were identified based on the spectroscopic data. Methyl caffeate was isolated for the first time from the genus Magnolia and it showed 3∼4-folds higher potency than ASA. The activities of obovatol and honokiol were comparable to ASA. Magnolol and syringin showed only very mild inhibitory effects to all the stimulators.
