마우스 대식세포에서 혈갈(血竭)(Draconis Resina)의 항산화 및 항염증 효과 (Anti-oxidative and Anti-inflammatory Effects of Draconis Resina in Mouse Macrophage Cells)

  • Objectives : Draconis Resina (DR) has been used as a traditional Korean herbal medicine since ancient times, and today it is used as a medication for wounds, tumors, diarrhea, rheumatism, in the itching of insect bites and with other conditions in the folk medicine. The aim of this study was to determine whether fractionated extracts of DR inhibit free radical generation, intracellular oxidation, production of nitrite, an index of NO, PGE2, iNOS, COX-2 and proinflammatory cytokines in lipopolysaccharide (LPS)-treated RAW 264.7 macrophages, Methods : DR extract prepared with methanol, and then fractionated with hexane, dichloromethane, ethylacetate, n-butanol and water. Inhibitory effect of DR onto free radical generation was determined by measuring DPPH, superoxide anions and nitric oxide scavenging activities in vitro. Cytotoxic activity of extracts on RAW 264.7 cells was measured using 5-(3-caroboxymeth-oxyphenyl)-2H-tetra-zolium inner salt (MTS) assay. Intracelluar oxidation was analysed by DCF-DA assay. The nitric oxide (NO) production was measured by Griess reagent system. The levels of iNOS and COX-2 expression were confirmed by western blot. And proinflammatory cytokines were measured by ELISA kit. Results : Our results indicated that fractionated extracts, especially dichloromethane and ethyl acetate extracts, significantly inhibited free radical generation, the LPS-induced H202, NO, PGE2 production and iNOS, COX-2 expression accompanied by an attenuation of TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 formation in macrophages. Conclusions : Our results indicate that dichloromethane and ethyl acetate extracts of DR have potential as an agent of chronic inflammatory diseases.

택사 열수 및 에탄올 추출물의 항산화활성과 human LDL 산화억제 및 ACE 저해효과에 미치는 영향 (Screening of Antioxidative, Anti-atherosclerotic Effect of Alisma Rhizome Extracts)

  • Objectives : The study was conducted to evaluate antioxidative, anti-atherosclerotic and anti-hypertensive effects of natural remedies. Alisma Rhizome (AR) has been used for a long time in Asia in folk remedies for treatment of hypertension and stroke and has been used in Korean traditional medicine for the treatment of glycosuria, gonorrhea, hypercholesterolemia, hypertension, and jaundice and its diuretic effect. These pharmacological effects of AR might come from antioxidant properties of phytochemicals in these materials. Methods : In this study, the antioxidant activity of extract from AR was studied with in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$ induced human LDL oxidation and on ACE. Results : The AR extracts were found to have a potent scavenging activity, as well as an inhibitory effect on LDL oxidation and on ACE against all of the reactive species tested, with the water extract showing particularly strong antioxidant activities. Conculsions : The AR extracts have antioxidative, anti-atherosclerotic and anti-hypertensive effects in an in vitro system, which can be used for developing pharmaceutical drugs against oxidative stress and atherosclerosis.

발효 이고들빼기(Youngia denticulata Houtt Kitamura) 에탄올 추출물이 알코올 투여로 손상시킨 흰쥐의 간독성 해소 효과 (Hepatotoxicity Reducing Effect of Ethanol Extracts from Fermented Youngia denticulata Houtt Kitamura in Ethanol-treated Rats)

  • Aspergillus oryzae로 발효시킨 이고들빼기의 에탄올 추출물이 알코올성 간독성을 유발한 SD계 흰쥐에 미치는 영향을 조사하였다. 실험군은 정상대조군(NC), 에탄올대조군(EC), 에탄올급여 및 비발효 이고들빼기 추출물 0.5% 첨가식이 급여군(ENY), 에탄올급여 및 발효 이고들빼기 추출물 0.5% 첨가식이 급여군(EFY)의 4개군(6마리/군)으로 나누어 4주간 사육하였으며, 혈청과 간 조직의 주요 지표 성분과 항산화 및 산화관련 효소활성을 측정하였다. 그 결과, EC군이 NC군에 비하여 혈중 GOT 및 GPT 활성은 각각 87.90% 및 57.02%가 높았다. 간 조직의 중성지질과 총콜레스테롤 함량은 각각 77.51% 및 18.56%, LPO 함량은 193.86%가 높았다. 반면에 GSH 함량은 29.52% 낮았다. 또, 항산화계 효소인 SOD 및 GST 활성은 각각 42.78% 및 0.94% 낮게 나타났으며, XO(O type)효소활성은 70.83% 높았다. 이로써 알코올 급여군은 전형적인 간 손상과 함께 항산화계에도 손상이 있음을 나타내었다. ENY 및 EFY군은 EC군과 비교하여 GOT 및 GPT 활성은 각각 24.73~41.99% 및 22.91~30.11%가 낮았고, 중성지질 함량은 16.65~33.33%, 총콜레스테롤 함량은 22.44~23.11%, LPO 함량은 63.34~66.13%가 감소한 반면, GSH 함량은 24.61~35.60% 증가하였다. 또, GST 활성은 유의적인 차이를 보이지 않았으나, SOD 활성은 각각 214.94% 및 261.19% 높았다. 또, XO(O type) 효소활성은 40.34~51.20% 감소하였다. EFY군은 ENY군에 비하여 GOT 및 GPT활성은 각각 22.93%, 9.34% 낮았고, 혈중 중성지질과 총콜레스테롤 및 LPO 함량은 각각 20.01%, 0.86% 및 7.59%가 낮았다. 그리고 SOD 및 GST활성은 각각 21.52% 및 0.6%가 높았고, XO(O type) 효소의 활성은 18.19% 낮았다. 이상의 결과, 발효 이고들빼기 추출물이 알코올로 유도된 간 손상 흰쥐에 대하여 간 조직 TG와 총콜레스테롤 및 LPO의 함량을 감소시키고, GOT, GPT 및 XO의 활성감소와 동시에 SOD활성을 높임으로써 간의 해독효과가 큰 것으로 판단되며, 이러한 결과는 향후 발효 이고들빼기의 성분들을 이용한 기능성식품의 개발과 이를 위한 기초자료로 활용할 수 있을 것으로 사료된다.

Effects of the Combined Extracts of Grape Pomace and Omija Fruit on Hyperglycemia and Adiposity in Type 2 Diabetic Mice

  • Grape products have been known to exert greater antioxidant and anti-obesity than anti-hyperglycemic effects in animals and humans. Omija is used as an ingredient in traditional medicine, and it is known to have an anti-hyperglycemic effect. We investigated whether the combined extracts of grape pomace and omija fruit (GE+OE) could reduce fat accumulation in adipose and hepatic tissues and provide beneficial effects against hyperglycemia and insulin resistance in type 2 diabetic mice. C57BL/KsJ-db/db mice were fed either a normal control diet or GE+OE (0.5% grape pomace extract and 0.05% omija fruit extract, w/w) for 7 weeks. GE+OE decreased plasma leptin and resistin levels while increasing adiponectin levels and reducing the total white adipose tissue weight. Furthermore, GE+OE lowered plasma free fatty acid (FFA), triglyceride, and total-cholesterol levels as well as hepatic FFA and cholesterol levels. Hepatic fatty acid synthase and glucose 6-phosphate dehydrogenase activities were decreased in the GE+OE group, whereas hepatic ${\beta}$-oxidation activity was increased. Furthermore, GE+OE supplementation not only reduced hyperglycemia and pancreatic ${\beta}$-cell failure but also lowered blood glycosylated hemoglobin and plasma insulin levels. The homeostasis model assessment of insulin resistance levels was also decreased and the decrease seems to be mediated by the lowered activities of hepatic glucose-6-phosphatase and phosphoenolpyruvate carboxykinases. The present data suggest that GE+OE may have the potential to reduce hyperglycemia, insulin resistance, and obesity in patients with type 2 diabetes.

네 가지 처방의 항산화 및 항암 효과 비교 (Comparison of Four Multi-herbal Formulas in Antioxidant and Anticancer Activities)

  • Objective : In this study, we compared the antioxidant and anticancer properties of four multi-herbal formulas which were recorded in 'Dongeuibogam': Gilgyung-tang (GGT), Mokdanpi-tang (MDPT), Samso-eum (SSE), Samchulbobi-tang (SCBBT). Methods : We checked antioxidant properties of four multi-herbal formula through total phenolic content, radical scavenging activities, protective effects on genomic DNA oxidation. To investigate anticancer effects, we conducted MTT assay and analyzed morphologic change in A549 non-small lung cancer cells. Results : Total phenolic contents of four multi-herbal formulas were in a rich order of MDPT > SSE > GGT > SCBBT. Especially, MDPT revealed the highest activity than others in all antioxidant experiments. Our results indicated that treatment of those multi-herbal formulas induced growth retardation in A549 cells and MDPT also showed the highest anticancer effect ($IC_{50}=1.374mg/ml$) among them. Conclusions : Our data suggested that MDPT would be a powerful ingredient for lung cancer treatment.

Capsanthin Inhibits both Adipogenesis in 3T3-L1 Preadipocytes and Weight Gain in High-Fat Diet-Induced Obese Mice

  • Adipogenesis in murine preadipocyte 3T3L-1 has been used as a model system to study anti-obese bioactive molecules. During adipogenesis in 3T3-L1 preadipocytes, we found that capsanthin inhibited adipogenesis ($IC_{50}$; $2.5{\mu}M$) and also showed lipolytic activity in differentiated adipocytes from the preadipocytes ($ED_{50}$; 872 nM). We identified that the pharmacological activity of capsanthin on adipogenesis in 3T3-L1 was mainly due to its adrenoceptor-${\beta}_2$-agonistic activity. In high-fat diet animal model study, capsanthin significantly enhanced spontaneous locomotive activities together with progressive weight-loss. The capsanthin-induced activation of kinetic behavior in mice was associated with the excessive production of ATP initiated by both the enhanced lipolytic activity together with accelerated oxidation of fatty acids due to the adrenoceptor ${\beta}_2$-agonistic activity of capsanthin. Capsanthin also dose-dependently increased adiponectin and p-AMPK activity in high fat diet animals, suggesting that capsanthin has both anti-obesity and insulin sensitizing activities.

음나무 수피로부터 보체계 활성화 다당의 정제 및 특성 (Purification and Characterization of Complement System Activating Polysaccharide from the Bark of Kalopanax pictus N.)

  • It was observed that the hot-water extract of the bark of Kalopanax pictus N. had the highest anti-complementary activity among the 11 kinds of forest materials. Methanol-and ethanol-soluble portions had low anti-complementary activities, but crude polysaccharide. HKP-0 had a high activity of 80%. HKP-0 contained 54.8% of total sugar and 27.9% of protein. The neutral sugars of HKP-0 consisted of mainly arabinose, galactose and glucose. HKP-4 fraction obtained by cetavlon treatment of HKP-0 showed the highest anti-complementary activity of 90%. The activity was not changed by pronase digestion bu decreased greatly by periodate oxidation. HKP-4 consisted of mainly arabinose and glucose with molar ratio of 1.0 : 22.4, HKP-4-I, an unabsorbed fraction from HKP-4 on DEAE Sepharose CL-6B column showed higher yield and activity than those of absorbed fractions. HKP-4-I was homogeneous, and its molecular weight was about 25,000. HKP-4-I contained 84.0% of neutral sugar and consisted of arabinose and glucose with molar ratio of 1.0 : 11.2. The anti-complementary activity of HKP-4-I was not decreased by the treatment of polymyxin B, and the polysaccharide activated both classical and alternative pathway in complement system. Void volume fraction obtained from HKP-4-I hydrolyzed with ${\alpha}$-amylase on Sephadex G-25 column only had a high anti-complementary activity.

멀티 유화 기술 이용 수분산성의 항산화 효능을 함유한 커큐민의 개발 (Development of Curcumin with Anti-Oxidation Effect of Water Dispersibility using Multi-Emulsification Technology)

  • Curcumin is not soluble in water. Therefore, curcumin emulsion that can dissolve well in water were prepared using multi-emulsification technology, and the antioxidant activities and physical properties of emulsion were measured. Although curcumin was not dissolved in water, it was confirmed to be well dispersed in water when prepared in an aqueous dispersion curcumin emulsion. After dissolving curcumin using water and ethanol as solvents, respectively, the DPPH and ABTS radical scavenging abilities of the filtrate and the curcumin emulsion were measured. Because it was not dissolved in water, activities were not shown. However, when curcumin was dissolved in ethanol, the activities increased as the concentration of curcumin increased. On the other hand, when the curcumin emulsion was dissolved in water, it was found to have abilities. The curcumin emulsion was nano-homogenized and the size and distribution of the emulsified spheres were measured. It was confirmed to be nano-sized as it appeared as 9.083 nm/100%. In the results of the DPPH radical and ABTS radical scavenging abilities of curcumin nano-emulsion, it was confirmed that there was no change in the antioxidant abilities. In conclusion, water-dispersible curcumin prepared using multi-emulsification technology, and it was confirmed to exhibit antioxidant activity and emulsion stability.

Effect of Herbal Extracts Mixtures on Antioxidant System in Chronic Enthanol-treated Rats

  • Disturbance of antioxidant system is very common in chronic alcoholics and herbal or natural products with antioxidant activity have been used for its treatment. This study was to investigate the effect of Vitis vinifera extract(V), Schisandra chinensis extract(S), Taraxacum officinale extract(T), Gardenia jasminoides extract(G), Angelica acutiloba extract(A) and Paeonia japonica extract(P), and their combinations on the antioxidant and ethanol oxidation system. Male Sprague-Dawley rats were subjected to Lieber-DeCarli ethanol liquid diet(ED) and were then given different herbal extract mixtures for 6 weeks including VST(V 100+S 150+T 150mg/kg/day), VSG(V 100+S 150+G 150mg/kg/day), VTG(V 100+T 150+G 150mg/kg/day), and VAP(V 100+A 150+P 150mg/kg/day). When the activity of alcohol dehydrogenase(ADH) and acetaldehyde dehydrogenase(ALDH) were compared between ED only group and herbal extracts treatment group, the differences were statistically significant. Phase I and II(glutathione-S-transferase, phenol sulfatransferase) enzyme activities were found to be significantly higher in the VAT treatment group compared to the ED group. Herbal extracts not only repressed the ethanol-induced elevation of malondialdehyde level, but also protected against ethanol-induced decrease in glutathione content, glutathione reductase, glutathione peroxidase, catalase and superoxide dismutase activities. The administration of the herbal extracts was found to be effective in eliminating lipid-peroxides induced by long-term consumption of alcohol by activating various enzyme systems and physiological active compound formation system. After a chronic consumption of alcohol, Angelica Radix protected the liver via activating the ethanol-metabolism enzyme system, and Paeoniae Radix via activating the ethanol-metabolism enzyme and the phase I, II-metabolism enzyme system. Taraxaci Herba was also effective in liver protection via activating the ethanol-metabolism enzyme system and the phase I, II-metabolism enzyme system, Gardeniae Fructus via activating the phase II-metabolism enzyme system and the anti-oxidation system enzyme, and Schisandra Fructus and a grapestone via activating the anti-oxidation system. Our data suggest that these herbal extracts may be useful as a health functional food or new drug candidate for fatty liver and hepatotoxicity induced by chronic alcohol consumption.

삽주 육성품종 뿌리의 항산화 및 항염증 효과 비교 (Comparison of Anti-oxidative and Anti-inflammatory Effect of Atractylodes Interspecific Hybrid Cultivar Roots)

  • Background: Atractylodes radix is a well-known medicinal crop having many physiological effects. This study was conducted to select useful Atractylodes japonica ${\times}$ Atractylodes macrocephala (AJM) cultivars by comparing anti-oxidative and anti-inflammatory efficacies. Methods and Results: Seven extracts from AJM cultivars were used to treat lipopolysacchride (LPS)-treated BV2 cells, and the effects on cell viability and inhibition on reactive oxygen species (ROS) and nitric oxide (NO) production were analyzed. In vitro scavenging activities of 2,2-diphenyl-1-picrylhydrazyl (DPPH) and peroxynitrite ($NOO^-$) radicals were also investigated. Contents of total phenol, atractylenolide I, and atractylenolide III in the AJM extracts were measured using high performance liquid chromatography (HPLC) or spectrophotometry. The experiments show that none of the seven extracts was cytotoxic above 89.2% at $20-250{\mu}g/m{\ell}$. Extracts of Gowon, Dawon, Sangchul, and Huchul inhibited ROS generation in a dose-dependent manner, and Sangchul extract showed the highest inhibition on ROS production. All the AJM extracts showed effective inhibitory activity after on NO release in the LPS-treated BV2 cells, and Sangchul extract showed the highest activity. Sangchul extract had the most potent scavenging activities for $NOO^-$ and had some DPPH radical scavenging effect. Sangchul extract also had the highest content at total phenol and atractylenolide I content. Atractylenolide III was not detected in the AJM extracts. Conclusions: The results suggested that Sangchul was the most useful anti-oxidative and anti-inflammatory resource among the AJM cultivars.
