The deer antler has been used as a major drug in oriental medicine for a long time. Recently, the demand for easy-to-take health functional foods is increasing due to economic development and changes in diet. As part of research on the development of functional materials for antlers, lactic acid fermentation of antler extract was performed. It was intended to develop a functional material with enhanced total polyphenol and flavonoid content and enhanced antioxidant activity. Lactic acid bacteria fermentation was performed by adding 4 types of lactic acid bacteria starter products, B. longum, Lb. Plantarum, Lb. acidophilus and mixture of 8 types of lactic acid bacteria to the antler water extract substrate, respectively. During the fermentation of lactic acid bacteria, the number of proliferation, total polyphenol and total flavonoid content, DPPH radical scavenging and antioxidant activity were quantified and evaluated. As a result of adding these four types of lactic acid bacteria to the antler water extract substrate, the number of lactic acid bacteria measured was 2.04~5.00×107. Meanwhile, a protease (Baciullus amyloliquefaciens culture: Maxazyme NNP DS) was added to the antler extract to decompose the peptide bonds of the contained proteins. Then, these four types of lactic acid bacteria were added and the number of lactic acid bacteria increased to 2.84×107 ~ 2.21×108 as the result of culture. The total polyphenol contents were 4.82~6.26 ㎍/mL in the lactic acid bacteria fermentation extracts, and after the reaction of protease enzyme and lactic fermentation, increased to 14.27~20.58 ㎍/mL. The total flavonoid contents were 1.52~2.21 ㎍/ml in the lactic acid bacteria fermentation extracts, and after the protease reaction and fermentation, increased to 5.59 ~ 8.11 mg/mL. DPPH radical scavenging activities of lactic acid bacteria fermentation extracts was 17.03~22.75%, but after the protease reaction and fermentation, remarkably increased to 32.82~42.90%.
본 연구에서는 율피 추출물을 이용하여 화장품 소재로서의 기능성을 조사하고자 효소종류별 및 가압추출 시간별에 따른 항산화 활성과 피부 미백 및 주름개선 효과를 실시하였다. 효소 종류별 및 가압추출 시간에 따른 율피 추출물의 추출수율은 12.42-29.80%로, C30m 추출물이 29.80%로 가장 높게 나타났다. 이화학적 특성으로 총 폴리페놀, 총 플라보노이드 및 총 탄닌 함량은 모두 C30m 추출물($1,000{\mu}g/mL$)에서 각각 22.69 g/100 g, 12.13 g/100 g 및 21.66 g/100 g으로 높은 함량을 나타내었으며, 총 당 함량은 H120m 추출물($1,000{\mu}g/mL$)에서 61.07 g/100 g으로 높은 함량을 나타내었다. 항산화 활성으로 DPPH radical 소거활성, ABTS radical 소거활성, superoxide radical 소거활성 및 FRAP 활성 모두 C30m 추출물($1,000{\mu}g/mL$)에서 각각 89.20%, 81.96%, 67.63% 및 $1,324.79{\mu}M$로 높은 항산화 활성을 나타내었으며, 모든 효소 처리구간에서 가압추출 시간이 증가함에 따라 항산화 활성이 감소하였다. 피부 미백 및 주름개선 효과를 확인하기 위해 tyrosinase 및 elastase 저해활성을 측정한 결과 C30m 추출물($1,000{\mu}g/mL$)에서 각각 61.32% 및 61.06%로 높은 저해활성을 나타내었으며, HE 추출물에서 가장 낮은 저해활성을 나타내었다. 이는 효소처리 및 가압추출에 의해 율피의 생리활성물질이 많이 용출된 것으로 판단되며, 율피의 새로운 추출공정 확립을 통해 항산화, 피부 미백 및 주름개선에 효과가 있는 항노화 기능성 소재로 화장품 제조용 원료 로 적합함을 확인하였다.
The diatom Melosira nummuloides is a microalga that is widely distributed in freshwater and seawater is used is used in the production of silicon and fucoxanthin. The objective of this experimental study was to determine the effects of diatom powder on the physiology of olive flounder Paralichthys olivaceus. In four feeding groups consuming 0%, 1%, 2% and 3% diatom powder. After 8 weeks of feeding, we investigated P. olivaceus growth rate, feed efficiency rate, survival rate, anti-oxidant enzyme rate, non-specific immune activity and immune gene expression. The rates of growth rate, feed efficiency rate and survival were significantly higher for olive flounder in all diatom groups than in the control. The results for anti-oxidant enzyme, superoxide dismutase and catalase showed no significance, but glutathione was significant, depending on the concentration of diatom addition. The galectin and lysozymes of immune genes were increased in the control group. Galectin and lysozymes were thought to have increased due to infections by from pathogens during the experiment period. These results suggest that the addition of diatoms to olive flounder diets is effective in enhancing growth rate and innate immunity.
우리 몸의 많은 기관을 비롯하여 장기들은 반복적이거나 혹은 급성 스트레스를 이겨내지 못하고 만성 스트레스로 이어질 경우 질병이 생기게 된다. 특히 강하고 지속적인 스트레스에 노출되면 뇌의 해마 수지상 세포(hippocampal dendrites)가 위축되거나 크기가 작아진다. 이렇게 스트레스로 인하여 증가된 글루코 코티코이드 호르몬은 뉴런 흥분제인 glutamate를 유도하거나 에너지 대사를 변형시켜 신경 독작용을 일으킨다. 이러한 연속적인 반응은 TNF-$\alpha$ convertase(TACE)를 활성화시켜 TNF-$\alpha$가 분비되도록 하여 전사 조절자인 NF-${\kappa}B$가 핵내로 전이되고 신경 손상을 일으키는 iNOS와 COX-2와 같은 효소를 유도한다. 이런 산화적 스트레스의 상위조절인자 TACE는 스트레스에 의한 여러 가지 염증성 질환 및 숙주방어에서 가장 중요한 조절자인 TNF-alpha를 수용체로부터 "유리(shedding)" 시키는 역할을 한다. 따라서 이런 신호 전달계를 자극하는 TACE의 발현 양과 이로 인한 지속적인 처리과정이 중요한 문제로 대두되고 있다. 특히 여러 스트레스 중에서 고정화 스트레스 및 신체적 구속 스트레스에 대한 연구는 뇌에서 산화물 생성을 증가시키지만 인삼이 뇌의 산화물질 생성에 어떤 영향을 미치는지 체계적인 연구가 진행된 바 없다. 따라서 염증을 매개하는 TNF-alpha의 생산에 중요한 역할을 하는 TACE의 발현 조절 및 TNF-alpha 신호전달을 연구함으로써 인삼의 항산화 기전을 분자 수준에서 규명할 수 있게 될 것으로 기대된다.
Objective : Reflux esophagitis (RE) is a disease that caused gastric acid reflux and inflammation due to unstable gastroesophageal sphincter, as increasing worldwide respectively. This study was conducted to evaluate the effect of Evodiae Fructus (EF) extract on chronic reflux esophagitis in rats. Methods : The EF was measured antioxidant activity, such as total polyphenol and total flavonoid contents, 1, 1-diphenyl-2-picrylhydrazyl (DPPH) and 2, 2'-azinobis-3-ethyl-enzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity. Rats were divided into 3 groups; Nor (normal group), Con (chronic acid reflux esophagitis rats treatment with water), EF (chronic acid reflux esophagitis rat treatment with EF 200 mg/kg body weight group). A surgically-induced chronic acid reflux esophagitis (CARE) model was established in SD rats, and treated with water or EF 200 mg/kg body weight for 14 consecutive days. Results : Administration of EF to rats of induction of chronic acid reflux esophagitis was found to reduce esophagus tissues injury. Reactive oxygen species (ROS) and produces peroxynitrite ($ONOO^-$) levels of esophagus tissues were significantly decreased in EF compared to Con group. As results of esophagus protein analyses, EF effectively reduce inflammatory-related factors ($NF-{\kappa}Bp65$, $p-I{\kappa}B{\alpha}$, iNOS, $TNF-{\alpha}$, IL-6), and increase anti-oxidant enzyme (Nrf2, HO-1, SOD, catalase, GPx-1/2). Conclusions : These results suggest that EF administration comfirmed that decreased esophagus tissues injury, oxidantive stress, anti-inflammation effect, and increased anti-oxidant effect. Therefore, EF was the potential to be used as a natural therapeutic drug.
본 연구에서는 생리활성물질유도제인 elicitor를 처리한 주름조개풀의 열수, 에탄올 추출물들의 항산화 효과, tyrosinase, elastase, collagenase 및 HAase 등의 미용관련 생리활성의 변화를 살펴보았다. Elicitor 처리한 주름조개풀 추출물의 DPPH 및 PF 등의 항산화 효과는 elicitor 비처리구인 대조구에 비해 elicitor 처리구가 농도의존적으로 더 높은 항산화 활성을 나타내었다. Elicitor 처리한 주름조개풀 추출물의 미용활성은 elastase 및 collagenase 억제 효과에 의한 주름개선효과와 tyrosinase 저해에 의한 미백효과가 비처리구인 대조구에 비해 상대적으로 매우 높은 효소 억제 효과를 나타내어 elicitor 처리가 주름조개풀추출물의 미백 및 주름개선 효과의 증대에 크게 영향을 미치는 것으로 확인되었으며, 농도의존적인 양상을 나타내었다. 또한 HAase 억제에 의해 유래되는 염증 억제 효과도 비처리구인 대조구에 비해 에탄올 추출물에서 우수한 염증 억제 효과를 나타내었다. Elicitor 처리에 의해 대조구에 비해 처리구에서 생리활성 효소에 대한 억제율이 큰 폭으로 상승하는 효과를 나타내어, elicitor 처리가 유용물질의 산업화라는 전제를 충족시키기 위한 biomass 기법으로 매우 적당하다는 것을 시사해주었다. 따라서 주름조개풀 노지재배 시에 본 연구에서 사용한 elicitor를 처리하면 수확되는 주름조개풀의 단위 무게 당 유용물질의 생산량을 높일 수 있고, 생리할성도 증대시킬 수가 있어 산업화를 위한 고기능성 원료생산에 효율적인 재배기법으로 활용할 수 있을 것이라 판단되었다.
갯메꽃은 해안 사구에 분포하며 식물로 높은 환경 적응력을 가지고 있으며 갯메꽃은 항산화, 해열, 살균, 이뇨작용 등에 효과가 있는 것으로 알려져 있다. 본 연구에서는 암전이 과정에서 중요한 역할을 한다고 알려진 matrix metalloproteinases (MMPs)의 일종인 MMP-2와 MMP-9의 활성에 대한 갯메꽃의 억제효과를 methylene chloride (MC) 및 methanol (MeOH) 추출물과 조추출물, 유기용매 분획물을 시료로 하여 인체 섬유육종 HT-1080 세포를 이용하여 ELISA (Enzyme-linked immunosorbent), gelatin zymography, Wound healing assay, RT-PCR, western blot 실험을 통해 조사하였다. 갯메꽃의 MC 및 MeOH 추출물, 조추출물과 4가지 유기용매 분획물이 HT-1080 세포에서 MMP-2와 MMP-9 생성 억제 활성을 나타냄을 확인하였다. 그 중에서도 n-hexane, 85% aq.MeOH 분획물이 전반적으로 MMP-2와 MMP-9에 대한 높은 억제활성을 보였다. 이들 결과로부터 85% aq.MeOH과 n-hexane 분획물에 MMP 억제 활성이 높은 물질이 존재할 것으로 추측되어지며, 갯메꽃 추출물 및 분획물이 암침윤 및 전이를 억제하는 효과적인 항암소재로서의 가능성이 있음을 제시한다. 이에 본 연구팀은 85% aq.MeOH과 n-hexane 분획물에 있는 활성물질을 분리하기 위한 연구를 추가적으로 수행할 계획이다.
진세노사이드(인삼 사포닌)는 인삼의 대표적 약리성분 중의 하나로 생물학적 활성을 가진 배당체 화합물이다. 이들 사포닌은 가수분해 되어 저분자화 되었을 때, 항주름 및 항산화, 항암 등에 높은 약리효능효과를 나타낸다. 본 연구에서는 인삼 esculin 배지를 활용하여 ${\beta}$-glucosidase 활성을 가진 균주를 분리하였고 인삼 사포닌 전환을 미생물을 이용하여 수행하였다. 본 균주들을 16S rRNA sequencing을 통하여 동정하여 본 결과 Stenotrophomonas rhizopilae strain GFC09로 확인되였다. 균주의 최적 활성 조건을 결정하기 위해 조효소 1 mM와 인삼사포닌 $Rb_1$과 함께 배양한 후 생물학적 전환을 TLC, HPLC를 사용하여 확인하였다. 조효소에 의한 인삼 사포닌 $Rb_1$의 전환 경로는 다음과 같다. LB: RbNeobio R&D center, Gyeonggi-do 16954, Korea${\rightarrow}$Rd${\rightarrow}$FNeobio R&D center, Gyeonggi-do 16954, Korea${\rightarrow}$compound K, TSB: $Rb_1{\rightarrow}Rd{\rightarrow}F_2$. 가수분해된 생성된 물질은 NMR로 구조 동정하였다. 전환 산물의 효능 분석결과, 콜라겐 생성을 농도 의존적으로 증가시키는 것이 관찰되었다. 이에 본 연구에서는 ginsenoside $F_2$와 compound K 함유 인삼 전환 산물의 주름 개선 소재로서 활용가능성을 확인하였다.
Desmodium heterocarpon은 콩과에 속하는 덩굴식물로 주로 한국, 일본 등의 아시아 국가에 분포되어 있다. 본 연구에서는 식물에 존재하는 신규 기능성 소재 개발의 일환으로 Desmodium heterocarpon 에탄올 추출물(DHEE)의 항산화 및 항염증 생리활성을 DPPH 라디칼 소거능, ROS 소거능, NO 생성 억제능 및 관련 단백질 발현을 통해 분석하였다. 먼저 DHEE의 항산화능을 DPPH 라디칼 소거능을 통해 분석한 결과 높은 소거활성을 보여 DHEE가 매우 강한 항산화능을 보유함을 확인하였다. 또한 RAW 264.7 세포주에서 H2O2에 의해 유도된 ROS에 대한 DHEE의 소거능을 분석한 결과 농도의존적인 강한 ROS 소거능을 보였다. 뿐만 아니라 대표적인 항산화 효소 중 하나로 항산화능 보유 천연물에 의해 발현이 유도되는 HO-1 및 그 전사 인자인 Nrf2의 단백질 발현이 DHEE의 처리에 의해 유의적으로 증가됨을 보였다. 한편 DHEE가 LPS에 의해 유도된 NO 생성에 미치는 영향을 분석한 결과 농도 의존적인 NO 생성 저해능을 보였으며 이는 NO 생성 단백질인 iNOS의 발현 저해에서 기인함을 확인하였다. 이러한 결과를 통해 DHEE의 높은 항산화능과 항염증 활성을 확인하였으며 향후 잠재적인 기능성 소재로서 유용하게 활용될 수 있을 것으로 판단된다. 추후 계속적인 연구를 통해 활성 물질의 규명이 필요할 것으로 보인다.
Malus melliana (Hand.-Mazz.) Rehder (M. melliana)는 장미과에 속하는 중국 자생 식물 중 하나로 현재까지 보고된 생리활성은 전무하다. 본 연구에서는 M. melliana 에탄올 추출물(MMEE)의 항산화 및 항염증 생리활성을 DPPH 라디칼 소거능, ROS 소거능, NO 생성 저해능 및 Western blot hybridization을 통한 연관 단백질 발현분석을 통해 평가하였다. MMEE의 항산화능을 DPPH 라디칼 소거능을 통해 분석한 결과 양성 대조군으로 사용한 대표적인 항산화제인 아스코르빈산과 유사한 정도의 높은 소거활성을 보여 MMEE가 매우 강한 항산화능을 보유함을 확인하였다. 또한 RAW 264.7 세포주에서 $H_2O_2$에 의해 유도된 ROS에 대한 MMEE의 소거능을 분석한 결과, 농도의존적인 강한 ROS 소거능을 보였다. 뿐만 아니라 대표적인 항산화 효소인 HO-1 및 그 전사 인자인 Nrf2의 단백질 발현에 미치는 영향을 분석한 결과 MMEE에 의해 HO-1 및 Nrf2의 발현이 증가됨을 보였다. 한편 MMEE가 LPS에 의해 유도된 NO 생성에 미치는 영향을 분석한 결과 농도의존적인 NO 생성 저해능을 보였으며 이는 NO 생성 단백질인 iNOS의 발현 저해에서 기인함을 확인하였다. 이러한 결과를 통해 MMEE의 높은 항산화능과 항염증 활성을 확인하였으며 향후 잠재적인 기능성 소재로서 유용하게 활용될 수 있을 것으로 판단된다. 추후 계속적인 연구를 통해 활성 물질의 규명이 필요할 것으로 판단된다.

회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
