Effect of silk fibroin peptide derived from silkworm Bombyx mori on the anti-inflammatory effect of Tat-SOD in a mice edema model

  • We investigated whether silk fibroin peptide derived from the silkworm, Bombyx mori, could inhibit inflammation and enhance the anti-inflammatory activity of Tat-superoxide dismutase (Tat-SOD), which was previously reported to effectively penetrate various cells and tissues and exert anti-oxidative activity in a mouse model of inflammation. Inflammation was induced by topical treatment of mouse ears with 12-O-tetradecanoylphorbol-13-acetate (TPA). Histological, Western blot, and reverse transcription-polymerase chain reaction data demonstrated that silk fibroin peptide or Tat-SOD alone could suppress elevated levels of cyclooxygenase-2, interleukin-6, interleukin-1beta, and tumor necrosis factor-alpha induced by TPA. Moreover, silk fibroin peptide significantly enhanced the anti-inflammatory activity of Tat-SOD, although it had no influence on in vitro and in vivo transduction of Tat-SOD. Silk fibroin peptide exhibited anti-inflammatory activity in a mice model of inflammation. Therefore, silk fibroin peptide alone or in combination with Tat-SOD might be used as a therapeutic agent for various inflammatory diseases.

Effects of Gardeniae Fructus on the metabolic process of antioxidant and anti-inflammation by JNK and NF-kB (치자(梔子)가 JNK와 NF-kB를 통한 항산화와 항염증의 대사과정에 미치는 영향)

  • Objectives : The purpose of this study is to observe the effects of Gardeniae Fuctus(GF) on the metabolic process of antioxidant and anti-inflammation. Methods : 4-HNE was injected into PC-12 cell to cause oxidative stress-induced inflammatory response, and then a western blot was conducted to observe the expression of Nuclear Factor-kB (NF-kB) and c-Jun N-terminal kinase (JNK) protein that are important factors involved with inflammation. Results : 1. The Gardeniae Fuctus water extract 50 ㎍ and 100 ㎍ significantly suppressed the increase in JNK protein expression in PC-12 cell. 2. The Gardeniae Fuctus water extract 50 ㎍, 100 ㎍ and 200 ㎍ significantly suppressed the increase in NF-kB protein expression in PC-12 cell. Conclusion : These results suggest that the Gardeniae Fuctus water extract has antioxidative and anti-inflammatory activity through suppressing the activity of JNK and NF-kB.

Anti-inflammatory Activity of Complex Saponin Separated from Pueraria flos and Adzuki Beans Mixture Extract

  • Pueraria flos and Adzuki Beans contains several bioactive compounds, such as saponin, oleanolic acid, and flavone. Pueraria flos and Adzuki Beans has traditionally been used to treat disorders of antioxidant activity, diabetes and liver detoxication, and it has antinociceptive and anti-inflammatory properties. However, complex saponin were validity of the anti-inflammatory activity has not been scientifically investigated. In this study, to determine anti-inflammatory activity of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract on nitric oxide and prostaglandinE2 assay. The anti-inflammatory activities of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract were evaluated for inhibitory activities against lipopolysacchride induced nitric oxide and prostaglandinE2 production protein expressions in RAW264.7 cell lines. The complex saponin separated from Pueraria flos and Adzuki Beans mixture extract inhibitory activity for both tests with protein high depressions(%) values showed in the ranges of 50~100 ㎍/ml. Overall, prostaglandinE2 tests had a higher inhibitory effect on inflammation than nitricoxide tests. Theseis result suggest a potential role of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract as source of anti-inflammation agent.

Comparative analysis of nitrite scavenging activity and anti-inflammation effects in the fruiting bodies of medicinal mushrooms (약용버섯류 자실체의 아질산염 소거능 및 항염증 효능 분석)

  • This study was carried out to compare the anti-inflammation effects of various fruiting body of Ganoderma species and Cordyceps militaris, Phelinus linteus extracts. We concentrated Ganoderma species and other medicinal mushrooms by extracting with ethanol. And We made it $100{\mu}g/ml$ concentration. As a result of nitrite scavenging activity, in the contrast to the positive control; Ascorbic acid was 25%, ASI 7080 of Ganoderma species was disappeared up to around 40%. And in the contrast to Ascorbic acid was 55%, ASI 7002 was 78.5% that was the highest anti-inflammation effect in the result of "No assay test". The Cordyceps militaris showed 75% and Hericium erinaceus showed 59.7% of anti-inflammation effect. As a result of the fungus yield control test of $TNF-{\alpha}$ through ELISA method to ASI 7002 of Ganoderma species that showed the highest anti-inflammation, it was reduced as same as LPS non-treatment. We extracted RNA from ASI 7002 Ganoderma species 10, 50, $100{\mu}g/ml$ concentration and LPS $10{\mu}g/ml$ of Raw 264.7 cell. And we tested the expression of iNOS, COX-2 and TNF-a that are kinds of inflammation gene after synthesizing RNA with cDNA. Finally we could find that iNOS, COX-2 and TNF-a were all controlled expression in the result of above experiment.

Effects of three local Malaysian Channa spp. fish on chronic inflammation

  • Water and chloroform/methanol extracts of the three local Malaysian snakehead fish, Channa striatus (striped snakehead), Channa micropeltes (giant snakehead) and Channa lucius (blotched snakehead) were evaluated for inhibitory activity in chronic inflammation, using cotton pellet granuloma test. Both water extracts of C. striatus and C. micropeltes showed marked inhibition of the transudative and proliferative components of chronic inflammation (42.9 and 31.2% respectively for C. striatus, 35.6 and 26.2% for C. micropeltes) when compared to those of mefenamic acid (25.1 and 21.3% respectively) and piroxicam (36.1 and 26.2% respectively). The chloroform/methanol extracts did not exhibit any anti-inflammatory effects. These results indicated that C. striatus has more anti-transudative and anti-proliferative activities than the extract of C. micropletes. C. lucius extract in contrast, did not inhibit these two components. This present study indicated the beneficial effects of the water extracts of C. striatus and C. micropeltes, but not C. lucius on chronic inflammation.

Study on the Anti-microbacterial Activity, Anti-inflammatory and Anti-allergic Effects of Several Herb-Extract (수종의 한약 복합 추출물의 항균, 항염 및 항알레르기 효과에 대한 실험적 연구)

  • Herbal mixture water extract of (Chrysanthemum morifolium, Portulaca oleracea, Sanguisorba officinalis, Sophora flavescens, Phellodendron amurense, Cnidium monnieri) which exhibit several beneficial effects including acne and skin diseases, was tested for anti-microbial activity and anti-inflammation effects. The herbal mixture extract showed antimicrobial activity against Stapylococcus epidermis, and Propionibacterium acne. The growth of Stapylococcus epidermis, and Propionibacterium acne was inhibited completely by addition of 1.0% of the extract. Also in the present study we examined the mixture extract on compound 48/80 induced allergy and LPS induced cyclooxygenase-2(COX-2) gene expression in RAW 264.7 macrophage. The results indicated the ear swelling and histamine release induced by compound 48/80 were dose-dependently reduced, ranging 28-60%, and 48-72% , respectively. Furthermore the extract inhibited the expression of LPS-induced COX-2 proteins and mRNAs without an appreciable cytotoxic effects on RAW 264.7 cells. The LPS-induced cytokine gene expression including IL-$1{\beta}$, TNF-$\alpha$, and IL-6 were dose-dependently suppressed by the mixture extract. Based on these results, it is concluded that the herbal mixture water extract can be applied to the acne and skin diseases therapy.

Antioxidant, anti-cholinesterase, and inflammation inhibitory activities of fruiting bodies of Phallus impudicus var. impudicus L. (말뚝버섯 자실체의 항산화, 항콜린에스테라제 및 염증 저해 활성)

  • Phallus impudicus var. impudicus L. is an edible mushroom that has long been used as folk medicine in China. The aim of this study was to evaluate the antioxidant, anti-cholinesterase, and inflammation inhibitory activities of a methanol extract of fruiting bodies of P. impudicus var. impudicus L. The extract exhibited good 1,1-diphenyl-2-picrylhydrazyl scavenging activity, excellent ferrous ion chelating activity, and moderate hydroxyl radical scavenging activity compared with BHT at 2.0 mg/ml. However, the reducing power of the extract was significantly lower than that the BHT positive control. Although the inhibitory activities of methanol extract on acetylcholinesterase and butyryl cholinesterase were significantly lower than the galanthamine positive control at the concentration tested, the inhibition of acetylcholinesterase and butyryl cholinesterase was 52.83% and 55.17%, respectively, at 1.0 mg/ml. The methanol extract also demonstrated excellent inhibition of inflammation-related activities, such as production of nitric oxide in lipopolysaccharide-induced RAW 264.7 macrophage cells and acute edema induced by administration of carrageenan on the hind paw of rats. The collective results suggest that the fruiting body of P. impudicus var. impudicus L. might be a good source of antioxidant, anti-cholinesterase, and anti-inflammation compounds.

Effect of Phellinus igniarius Quel Extract on the Anti-inflammatory, Anti allergy, Anti-oxidant, Anti-wrinkle (상황버섯 추출물이 항염증 및 항알레르기, 항산화, 주름제거에 미치는 실험적 연구)

  • Objectives : This study was carried out to investigate the effects of Phellinus igniarius Quel extract on the Anti-inflammatory, Anti allergy, Anti-oxidant and Anti-wrinkle. Methods : To investigate in vitro anti-oxidant activity assay, ethanol extracts of medicinal plants tested by DPPH method. In the next experiment, to investigate anti-inflammatory test, the RAW 264.7 macrophage cells was cultured using DMEM including the 10% FBS. To study anti-allergic effect, we blended cultured Human Mast Cells(HMC-1), and then observe TNF-$\alpha$, IL-8 by ELISA Results : Phellinus igniarius Quel extract has the effects of anti-inflammation and anti-allergy, which may be due to its inhibitory potential on the macrophage activation. Furthermore, Phellinus igniarius Quel extract has the anti-wrinkle effects through the inhibitory potential on the collagnease, elastase and gelatinase activities. Conclusions : The above results suggest that Phellinus igniarius Quel extract could be applicable for improvement of several skin functions.

  • The seeds of Astragalus sinicus grows in Korea have been used for oriental traditional medicine as the remedies for inflammation. Eight flavonoids were isolated from the Seeds of Astragalus sinicus and studied its anti-inflammatory activity. Some flavonoid compounds showed significant nitrogen monoxide(NO) production inhibitory activity in IFN-${\gamma}$, LPS stimulated RAW 264.7 cell. There compounds also showed significant antioxidative in DPPH assay. (omitted)

Inhibition of Contact Dermatitis in Animal Models and Suppression of Proinflammatory Gene Expression by Topically Applied Flavonoid, Wogonin

  • Wogonin (5,7-dihydroxy-8-methoxyflavone) is a down-regulator of cyclooxygenase-2 and inducible nitric oxide synthase expression, contributing to anti-inflammatory activity in vivo. For further characterization of modulatory activity on ploinflammatory gene expression in vivo, the effect of wogonin was examined in this experiment using animal models of skin inflammation. By topical application, wogonin inhibited an edematic response as well as ploinflammatory gene expression against contact dermatitis In mice. Wogonin inhibited ear edema ($19.4-22.6\%$) at doses of $50-200\;{\mu}g$/ear and down-regulated interleukin-$1{\beta}$ induction ($23.1\%$) at $200{\mu}g$/ear in phenol-induced simple irritation. Wogonin ($2{\times}50-2{\times}200{\mu}g$/ear) also inhibited edematic response ($51.2-43.9\%$) and down-regulated ploinflammatory gene expression of cyclooxygenase-2, interleukin-$1{\beta}$, interferon-$\gamma$, intercellular adhesion molecule-1 and inducible nitric oxide synthase with some different sensitivity against picryl chloride-induced delayed hypersensitivity reaction. All these results clearly demonstrate that wogonin is a down-regulator of ploinflammatory gene expression in animal models of skin inflammation. Therefore, wogonin may have potential for a new anti-inflammatory agent against skin inflammation.
