Catechin-7-O-$\beta$-D-apiofuranoside: An Anti-inflammatory constituent from alnus japonica bark

  • Alnus japonica (Betulaceae) has been traditionally used for purifying blood, and curing feces containing blood, enteritis, diarrhea, alcoholism and cut wounds. In the current work, activity guided isolation of the butanol fraction of the Alnus japonica bark led to the isolation of catechin-7-O-${\beta}$-D-apiofuranoside. Anti-inflammatory activity was determined with carrageenan-induced paw edema in mice as an acute inflammation, complete Freund's adjuvant-induced arthritis as a chronic inflammation. Carrageenan- induced paw edema in mice was significantly inhibited at 0.5, 1, 2, and 3 hr after carrageenan injection by administration of the flavonoid glycoside at the dose of 150mg/kg. (omitted)

The Root from Heracleum moellendorffii Exerts Anti-Inflammatory Activity via the Inhibition of NF-κB and MAPK Signaling Activation in LPS-Stimulated RAW264.7 Cells

  • Although the roots of Heracleum moellendorffii (HM-R) have been long treated for inflammatory human diseases, scientific evidence for the anti-inflammatory activity of HM-R is not sufficient. In this study, we investigated anti-inflammatory activity and mechanism of action of HM-R in LPS-stimulated RAW264.7 cells. HM-R blocked LPS-induced NO and PGE2 production, but not HM-L. HM-R inhibited LPS-induced overexpression of iNOS, COX-2, $IL-1{\beta}$ and IL-6 in RAW264.7 cells. HM-R inhibited LPS-induced $NF-{\kappa}B$ signaling activation through blocking $I{\kappa}B-{\alpha}$ degradation and p65 nuclear accumulation. In addition, HM-R inhibited MAPK signaling activation by attenuating the phosphorylation of ERK1/2, p38 and JNK. Furthermore, HM-R inhibited attenuated LPS-mediated overexpression of the osteoclast-specific factors such as NFATc1, cathepsin K, MCP-1 and TRAP. These results indicate that HM-R may exert anti-inflammatory activity by inhibiting $NF-{\kappa}B$ and MAPK signaling activation. From these findings, HM-R has potential to be a candidate for the development of chemopreventive or therapeutic agents for the inflammation and inflammatory diseases.

Antioxidative Activity and Anti-inflammatory Effects on the Murine Macrophages of Methanol Extracts of Amphibians

  • Oxidative stress has been reported to be one of causes of neuritis. This study examined antioxidative activities of methanol extracts of six amphibian species known to be medicinal animals (Rana catesbeiana, R. coreana, R. rugosa, R. dybowskii, R. nigromaculata, and Hyla japonica) and investigated their effects of inhibiting nitric oxide (NO) production and cytotoxicity on the murine macrophage RAW264.7 cells. As inflammation is closely associated with reactive oxygen species, assays on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, xanthine oxidase inhibitory activity, superoxide anion radical scavenging activity and NO scavenging activity of the extracts of the six species were performed to investigate their antioxidative activity. The results obtained were as follows; All extracts showed antioxidative activity, and the activity of R. dybowskii was the highest in comparison among those. Anti-inflammatory effects of the extracts were also examined, the five extracts except that of R. rugosa did not show cytotoxicity for RAW264.7 cells at the maximal concentration ($1,000{\mu}g\;mL^{-1}$). Selectivity index, meaning NO scavenging activity compared to cytotoxicity, showed the highest level in the extract of R. dybowskii. These results will be very useful basic data for future studies on prevention and treatment of human diseases to understand the biological roles of amphibian extracts throughout the antioxidative or anti-inflammatory pathways.

益氣化瘀湯加味方이 고지방 식이로 유발된 생쥐의 항고지혈 및 항산화에 미치는 효과 (Anti-hyperlipidemic and Anti-oxidant effects of Kgihwaetang-gamibang in High Cholesterol Diet-induced Mice Model)

  • This study was conducted to investigate the anti-hyperlipidemic and anti-oxidative activities of Ikgihwaetang-gamibang (IK). We observed the effect of IK on lipid factor, inflammation factor, body weight and organ weight, and liver after administering IK to hyperlipidemia mice triggered by high fat diet. Additionally, we used Raw 264.7 cells to investigate the changes in anti-oxidative activity and the effects of anti-inflammation. Administration of IK resulted in reduction of total cholesterol, LDL cholesterol, TG, and glucose in blood serum while a clear increase in HDL cholesterol. The production of IL-1β and IL-6, and ROS and NO also decreased. The results showed that IK has anti-hyperlipidemic and anti-oxidative effects, and suggested its potential use in prevention and treatment of hyperlipidemia and its associated diseases.

Propionibacterium acnes에 의해 유도된 염증에 대한 Chitosan-phytochemical Conjugates의 항염증 효과 (Anti-inflammatory Effects of Chitosan-phytochemical Conjugates against Propionibacterium acnes-induced Inflammation)

  • Propionibacterium acnes infection in skin tissue often causes acne vulgaris, commonly characterized by inflammatory papules, pustules, and nodules. Chitosan and its derivatives possess strong anti-inflammatory effects. In this study, the anti-inflammatory activity of chitosan-phytochemical conjugates on P. acnes-infected human skin keratinocytes (HaCaT) was evaluated. We designed a model of P. acnes-induced inflammation in viable HaCaT cells. Nitric oxide (NO), an inflammatory marker, was successfully elevated by P. acnes infection in HaCaT cells in a dose-dependent manner. Furthermore, the levels of NO were reduced by treatment with chitosan-phytochemical conjugates (chitosan-caffeic acid, -ferulic acid and -sinapic acid) in a dose-dependent manner. Among these conjugates, chitosan-caffeic acid exhibited the strongest NO suppression in HaCaT cells infected with P. acnes. The results obtained in this study suggest that chitosan-phytochemical conjugates could be used as a potential therapeutic agent against acne vulgaris.

지부자(地膚子)의 신생혈관 및 염증매개 단백질 발현에 미치는 영향 (Effects of Kochiae Fructus Extracts on the Expression of Angiogeneis and Inflammation Related Proteins)

  • Hypoxia induced angiogenesis and inflammation are essential processes for metastasis and progress of solid tumors. We examined the anti-angiogenic and inflammation related activity of Kochiae Fructus (KF) extract. To investigate the roles of the KF extract, we performed MTS assay, western blots using HaCaT cells and $HepG_2$ cells. The results are as follows. The protein level of $HIF-1{\alpha}$ was reduced when induced by $CoCl_2$ in $HepG_2$ cells treated with KF extract and induced by IGF-11 in HaCaT cells treated with KF extract. KF extract reduced the mRNA level of VEGF in HaCaT cells and KF extract reduced the protein level of iNOS in HaCaT cells. These results suggest that KF extract contributes to the anti-angiogenic and anti-inflammatory activities and also we could assume that KF extract act as antioxidant or anti-inflammatory agents via reduction of $HIF-1{\alpha}$.

  • Some pharmacological activities of acanthoic acid, isolated from Acanthopanax koreanum root was investigated in animals. It is revealed that the compound had analgesic and anti-inflammatory activities without actions on central nervous system and showed inhibition of lipid peroxidation. The anti-inflammatory activity might be related to inhibition of prostaglandin E$_2$ synthesis in exudates of inflammation.

  • Certain flavonoids possess anti-inflammatory activity. Besides their antioxidative property, the cellular action mechanisms of flavonoids include an inhibition of arachidonate metabolizing enzymes such as cyclooxygenases and lipoxygenases, and a down-regulation of proinflammatory gene expression such as cyclooxygenase-2, inducible nitric oxide synthase and tumor necrosis factor-$\alpha$. In this review, the recent findings of anti-inflammatory flavonoid research since 2004 were summarized. And the cellular mechanisms on signal transduction pathways were also discussed.

고삼(苦蔘)이 알러지성 염증 반응에 미치는 영향 (Effects of Sophorae Radix (SFR) on Allergic Inflammation related Atopy Dermatitis)

  • Sophorae Radix (SFR) is known as a therapeutic drug that has been used in Oriental traditional medicine for the treatment of skin and mucosal ulcers, gastrointestinal hemorrhage, diarrhea, inflammation and arrhythmia. In the present study, we examined the effects of the aqueous extract of SFR on anti-inflammation, anti-allergic and anti-oxidant effect in various cell lines; they include mouse lung fibroblast cells (hFCs), human mast cells (HMC-1), human monocytic cells (THP-1), and RAW 264.7 cells. Treatment with SFR extract at a concentration of 250 ${\mu}g$/ml for 24h showed no significant decrease in the survival rate of the hFCs. SFR decreased the mRNA expression of IL-8, TNF-$\alpha$, and IL-6 in HMC-1 cells. SFR extract treatment significantly inhi-bited the protein expression of IL-6 and, IL-8 induced by mite in THP-1 cells and it also did MCP-1 expression. We examined the alternation of histamine release in HMC-1 cells for investigating anti-allergic effect of SFR. Histamine secretion decreased after the treatment with SFR. In addition, SFR extract treatment at a concentration of 10 ${\mu}g$/ml, 100 ${\mu}g$ /ml, and 200 ${\mu}g$/ml lowered the $\beta$-hexosaminidase to 10.3%, 21.7%, and 50.8%, respectively. IC50 of SFR extract in RBL-2H3 cells was 196.85 ${\mu}g$/ml. Both activity of NF-$\kappa$B promoter in RBL-2H3 cells significantly diminished after the dose-dependent treatment of SFR. Therefore, our results indicate that SFR has anti-inflammatory and it may be useful for treating allergic diseases such as atopic dermatitis.

Inhibitory Activity of Cordyceps bassiana Extract on LPS-induced Inflammation in RAW 264.7 Cells by Suppressing NF-κB Activation

  • Cordyceps bassiana has long been used as an oriental medicine and reported to possess diverse biological activities. The fruiting bodies of Cordyceps bassiana was extracted with ethanol and then further fractionated with n-hexane, ethyl acetate, n-butanol and water. The butanol fraction from Cordyceps bassiana (CBBF) exhibited the most effective in anti-inflammatory activity in RAW 264.7 macrophages and the roles of CBBF on the anti-inflammation cascade in LPS-stimulated RAW 264.7 cells were studied. To investigate the mechanism by which CBBF inhibits NO, iNOS and COX-2, the activation of $I{\kappa}B$ and MAPKs in LPS-activated macrophage were examined. Our present results demonstrated that CBBF inhibits NO production and iNOS expression in LPS-stimulated RAW 264.7 macrophage cells, and these effects were mediated through the inhibition of $I{\kappa}B-{\alpha}$, JNK and p38 phosphorylation. Also, CBBF suppressed activation of MAPKs including p38 and SAPK/JNK. Furthermore, CBBF significantly suppressed LPS-induced intracellular ROS generation. Its inhibition on iNOS expression, together with its antioxidant activity, may support its anti-inflammatory activity. Thus Cordyceps bassiana can be used as a useful medicinal food or drug for further studies.
