  • The anti-diarrhea and spasmolytic activities of Soonkijangquebo (SKJQB), a Korean herbal anti-diarrhea formulation, were subjected to pharmacological evaluation. SKJQB, at a dose of 50\ulcorner200 mg/kg, inhibited castor oil-induced diarrhea in mice. The median effective dose (ED50) of the anti-diarrhea effect was 93 mg/kg.(omitted)

  • Objectives: Presently Mume Fructus (MF) undergoes fumigation, which produces benzo[a]pyrene. As a primary analysis with the aims to minimize the production of benzo[a]pyrene and to suggest standards for processing the MF, the steaming method was chosen among the various processing methods, and reviewed through a series of experiments.Methods: Methods:Pitted and un-pitted MF were steamed and processed into samples. After testing level of benzo[a]pyrene, the samples were analyzed for amount of polyphenol and flavonoids. Scavenging activities of the samples for the DPPH and ABTS radicals were tested. In order to measure anti-inflammatory effects of the samples, cell survival rate was investigated using CCK-8 Assay. Also, water extracts of dried and steamed MF were administered to the RAW 264.7 cells to compare expressions of NO, PGE2, IL-1β, and TNF-α. In addition, anti-diarrhea effects of the herbal medicine were tested on animal models with diarrhea induced by MgSO4 and Castor oil.Results: Regardless of pitting, processed MF contained no benzo[a]pyrene. Anti-oxidation effect increased in relation to the frequency of steaming process. However, extracts of dried and steamed MF suppressed different kinds of inflammation factors, and extract of dried MF showed superior anti-diarrhea effect than extract of steamed MF.Conclusions: It is suggested that steaming method of MF is recommended for processing the herbal medicine without the production of benzo[a]pyrene. But regarding that dried and steamed MF showed differences in their anti-oxidative, anti-inflammatory, and anti-diarrhea effects, it is recommended to perform further researches on different efficacies of MF according to their processing methods.

Experimental Studies on the Anti-diarrhea Effects of Jisa-tang (지사탕(止瀉湯)의 항사하작용(抗瀉下作用)에 관(關)한 연구(硏究))

  • Objective : The aim of this experimental study was to investigate the anti-diarrhea effects of Jisa-tang using mice and guinea pigs. Methods : We feed Jisa-tang to mice and guinea pigs to investigate its effects for anti-diarrhea action. We observed its actions on gastrointestinal smooth muscles, on the transportability of small and large intestines, on diarrhea induced by castor oil and magnesium sulfate, and on enteropooling by castor oil and prostaglandin $E_2(PGE_2)$. Results : 1. Jisa-tang showed alleviation, depending on the density, only on the contraction of mice's gastrointestinal smooth muscles induced by histamine. 2. The transportability of the small intestine was not significantly constrained by Jisa-tang. However, the enhancement of pyridostigmine-induced transportability of he small intestine was significantly constrained in the group administered 900mg/kg of Jisa-tang (p<0.05). 3. The transportability of large intestine was significantly constrained in the group administered 1,800mg/kg of Jisa-tang. 4. Jisa-tang showed significant anti-diarrhea effects on diarrhea induced by castor oil and by $MgSO_4$ in the group administered 1,800mg/kg of Jisa-tang. 5. Significant reduction of effects of enteropooling induced by caster oil and by prostaglandin $E_2$ were observed only in the group administered 1,800mg/kg of Jisa-tang. Conclusions : We conclude that Jisa-tang has advantageous effects on drug-induced diarrhea and will contribute to the development of diarrhea treatment through further related studies.

Experimental Studies on the Anti-cathartic Effects of Bojangjisa-tang (보장지사탕의 항사하작용에 관한 연구)

  • Objectives : The aim of this experimental study was to investigate the effects of Bojangjisa-tang on anticatharsis by using mice and Guinea pigs. Methods : We observed the actions on gastrointestinal smooth muscles, on the transportability of small and large intestines and on diarrhea induced by castor oil, philocarpine, and barium chloride. Results : Bojangjisa-tang showed alleviation of the contraction of gastrointestinal smooth muscles induced by acetylcholine chloride and barium chloride. The transportability of the large intestine was constrained by Bojangjisa-tang, which also showed substantial anti-cathartic effects on diarrhea induced by castor oil, philocarpine and barium chloride. Conclusion : These results demostrate that Bojangjisa-tang has advantageous effects on drug-induced diarrhea and will contribute to the development of diarrhea treatment through further related studies.

  • Objectives: The aim of this experimental study is to investigate effects of Dolichoris Semen, Terminaliae Fructus and Bojanggunbi-tangon the anti-cathartic action of mice and guinea pigs to develop effective herbal medicine and prescription to cure diarrhea early and to prevent side effect of chronic diarrhea. Methods: To compare effects of Dolichoris Semen, Terminaliae Fructus and Bojanggunbi-tangon the anti-cathartic action of mice and guinea pigs, we evaluated the actions on isolated gastrointestinal smooth muscles. on the transportability of small and large intestines, onthe neostigmine-induced transportability of small intestine and on diarrhea induced by castor oil, pilocarpine, barium chloride. Results: (1) Dolichoris Semen, Terminaliae Fructus and Bojanggunbi-tang showed the strong alleviation on the contraction of mice's isolated gastrointestinal smooth muscles induced by acetylcholine chloride and barium chloride. while Terminaliae Fructus, strong, Bojanggunbi-tang, intermediate and Dolichoris Semenshowed the minor alleviation on the contraction of guinea pigs' isolated gastrointestinal smooth muscles induced by acetylcholine chloride and histamine. All effects depended on the density in each medicine. (2) The tranbportability of small intestine was contrained in the groups controlled by 600 mg/kg of Dolichoris Semen, Terminaliae Fructus and Bojanggunbi-tang, and 200 mg/kg of D olichoris Semen and Terminaliae Fructus. However, the enhancement of neostigmine-induced transportability of small intestine was constrained only in the groups controlled by 600 mg/kg of Dolichoris Semen, Terminaliae Fructus and Boianggunbi-tang. (3) The transportability of large intestine was constrained by Dolichoris Semen and Bojanggunbi-tang. They showed substantial results. which depended on the amount of each medicine. (4) Terminaliae Fructus showed substantial anti-cathartic effects on diarrhea induced by caster oil in both groups controlled by 200 and 600 mg/kg of it. However, Bojanggunbi-tangshowed the effects only in the group controlled by 600 mg/kg. in diarrhea induced by pilocarpine, the groups controlled by 600 mg/kg or Terminaliae Fructus and 200 and 600 mg/kg of Boianggnbi-tangshowed substantial results. In diarrhea induced by barium chloride. the groups controlled by 200 and 600 mg/kg of Terminaliae Fructus and 600 mg/kg of Boianggunbi-tangshowed substantial results in anti-cathartic effects. Conclusions: These results show that Terminaliae Fructus is more effective on the contraction of mice and guinea pigs' isolated gastrointestinal smooth muscles, Dolichoris Semen is more effective on the transportability of small intestine, Dolichoris Semen and Bojanggunbi-tang are more effective on the transportability of large intestine. Terminaliae Fructus and Bojanggunbi-tang show substantial results in anti-cathartic actions.

Characterization of porcine cytokine inducible SH2-containing protein gene and its association with piglet diarrhea traits

  • Objective: The cytokine inducible SH2-containing protein (CISH), which might play a role in porcine intestine immune responses, was one of the promising candidate genes for piglet anti-disease traits. An experiment was conducted to characterize the porcine CISH (pCISH) gene and to evaluate its genetic effects on pig anti-disease breeding. Methods: Both reverse transcription polymerase chain reaction (RT-PCR) and PCR were performed to obtain the sequence of pCISH gene. A pEGFP-C1-CISH vector was constructed and transfected into PK-15 cells to analysis the distribution of pCISH. The sequences of individuals were compared with each other to find the polymorphisms in pCISH gene. The association analysis was performed in Min pigs and Landrace pigs to evaluate the genetic effects on piglet diarrhea traits. Results: In the present research, the coding sequence and genomic sequence of pCISH gene was obtained. Porcine CISH was mainly localized in cytoplasm. TaqI and HaeIII PCR restriction fragment length polymorphism (RFLP) assays were established to detect single nucleotide polymorphisms (SNPs); A-1575G in promoter region and A2497C in Intron1, respectively. Association studies indicated that SNP A-1575G was significantly associated with diarrhea index of Min piglets (p<0.05) and SNP A2497C was significantly associated with the diarrhea trait of both Min pig and Landrace piglets (p<0.05). Conclusion: This study suggested that the pCISH gene might be a novel candidate gene for pig anti-disease traits, and further studies are needed to confirm the results of this preliminary research.

  • The rotavirus-induced diarrhea of human and animal neonates is a major public health concern worldwide. Until recently, no effective therapy is available to specifically inactivate the rotavirion particles within the gut. Passive immunotherapy by oral administration of chicken egg yolk antibody (IgY) has emerged of late as a fresh alternative strategy to control infectious diseases of the alimentary tract and has been applied in the treatment of diarrhea due to rotavirus infection. The purpose of this concise review is to evaluate evidence on the properties and performance of anti-rotavirus immunoglobulin Y (IgY) for prevention and treatment of rotavirus diarrhea in human and animal neonates. A survey of relevant anti-rotavirus IgY basic studies and clinical trials among neonatal animals (since 1994-2015) and humans (since 1982-2015) have been reviewed and briefly summarized. Our analysis of a number of rotavirus investigations involving animal and human clinical trials revealed that anti-rotavirus IgY significantly reduced the severity of clinical manifestation of diarrhea among IgY-treated subjects relative to a corresponding control or placebo group. The accumulated information as a whole depicts oral IgY to be a safe and efficacious option for treatment of rotavirus diarrhea in neonates. There is however a clear need for more randomized, placebo controlled and double-blind trials with bigger sample size to further solidify and confirm claims of efficacy and safety in controlling diarrhea caused by rotavirus infection especially among human infants with health issues such as low birth weights or compromised immunity in whom it is most needed.

Antibiotic Resistance of Pathogenic Escherichia coli Isolated from Piglets with Diarrhea (설사자돈 유래 대장균의 항생물질 내성에 관하여)

  • The present study was conducted to investigate the biochemical characteristics and anti-biotic resistance of Escherichia coli(E. coli) isolated from piglets with diarrhea in Kyongbuk province during the Period from February to November 1991. 368 E. coli strains were isolated from 382 piglets with diarrhea and the biochemical and cultural reaction were compared with the classification criteria of Edwards and Ewing. Tetracycline and sulfadimethoxine were found to be highly ineffective at in vitro inhibition of the E. coli of piglets origin. The majority of E. coli were susceptible to amikacin, chloramphenicol and gentamicine. 89 (89.0%) of 100 strains of E. coil were resistant to one or more drugs. The organisms resistant to 20 or 3 drugs were 54(60.6%) of 89 strains, whereas 16(17.9%) strains were found to be resistant to one drug. 55(61.8%) out of 89 drug resistance strains carried R factors($R^+$) which were transfer-able to the recipients by conjugation.
