Pharmacophore Development for Anti-Lung Cancer Drugs

  • Lung cancer is one particular type of cancer that is deadly and relatively common than any other. Treatment is with chemotherapy, radiation therapy and surgery depending on the type and stage of the disease. Focusing on drugs used for chemotherapy and their associated side effects, there is a need to design and develop new anti-lung cancer drugs with minimal side effects and improved efficacy. The pharmacophore model appears to be a very helpful tool serving in the designing and development of new lead compounds. In this paper, pharmacophore analysis of 10 novel anti-lung cancer compounds was validated for the first time. Using LigandScout the pharmacophore features were predicted and 3D pharmacophores were extracted via VMD software. A training set data was collected from literature and the proposed model was applied to the training set whereby validating and verifying similar activity as that of the most active compounds was achieved. Therefore pharmacophore develoipment could be recommended for further studies.

4'-O-β-D-Glucosyl-5-O-Methylvisamminol Attenuates Pro-Inflammatory Responses and Protects against Oxidative Damages

  • We attempted to examine anti-inflammatory and anti-oxidant effects of 4'-O-${\beta}$-D-glucosyl-5-O-methylvisamminol (GOMV), the first epigenetic inhibitor of histone phosphorylation at Ser10. While GOMV did not affect the viability of murine macrophage RAW 264.7 cells, it significantly suppressed lipopolysaccharide (LPS)-induced generation of prostaglandin $E_2$ ($PGE_2$) and nitric oxide (NO) through transcriptional inhibition of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS). GOMV also scavenged free radicals in vitro, increased NF-E2-related factor 2 (NRF2), and activated antioxidant response element (ARE), thereby resulting in the induction of phase II cytoprotective enzymes in human keratinocyte HaCaT cells. Finally, GOMV significantly protected HaCaT cells against 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced oxidative intracellular damages. Together, our results illustrate that GOMV possesses anti-inflammatory and anti-oxidant activity.

A comparative study on the phytochemical and anti-oxidant activity differences in HongJam prepared with various silkworm varieties

  • The aim of this study was to elucidate the difference in the amount of phytochemicals and anti-oxidant activities of the freeze-dried 5th instar 3rd day silkworm powder (FDSP) and 7 HongJams (HJs). Three primary colors of light of 8 silkworm powders revealed why each sample had unique color. The visual and absorption spectrum analyses of 80% methanol (MeOH) extracts of 8 silkworm powders revealed that white jade (WJ)-FDSP extracts showing strong forest-green colors had different absorption spectrum from 7 HJs. Among 7 HJs, the absorption spectrum of light green silkworm (LGS)-HJ extract was different from those of other HJ extracts. The amounts of total flavonoids and phenolic compounds of WJ-FDSP were 423.0±9.21% and 252.3±12.86%, respectively, more than those of WJ-HJ. Among 7 HJs, the amounts of total flavonoids and phenolic compounds in LGS-and red silkorm (RS)-HJ extracts were significantly more than those of WJ-HJ extract. Consistent with the amounts of phytochemicals, radical scavenging activity and reducing ability of WJ-FDSP extract were significantly higher that other HJ extracts. Among 7 silkworm powder extracts, LGS-HJ extract showed significantly higher radical scavenging activity and reducing ability than those of WJ-HJ extract. But, RS-HJ extract only showed significantly higher radical scavenging activity than that of WJ-HJ extract. Taken together, the amounts of phytochemicals in 8 silkworm powders contributed on their colors and anti-oxidant activities.

  • This paper analyzes high efficiency anti-vibration boring bars which increase stability against chatter vibration in boring operations. Structural analysis and mathematical modeling with considering dynamic properties for three types of existing boring bars are performed to search for optimal design parameters. The purpose of this paper is to find out design parameters for high efficiency anti-vibration boring bar.

Marine organisms harbour a diverse range of unique compounds with applications in a multitude of disease indications.   • Immunological imbalance eventually results in the development of various diseases. A typical example is an imbalance of cytokines with immunomodulatory abilities. In this paper, we propose a two-variable delay model to anti-pro-inflammatory cytokine therapy for autoimmune diseases, which are caused by an imbalance between the pro and anti-inflammatory cytokines. The interaction between pro- and anti-inflammatory cytokines were modeled mathematically to investigate the relevance of cytokines in disease processes. The delay time was estimated to maintain the stability of a biologically important steady state. In particular, the effects of delay with anti-pro-inflammatory cytokines therapy in autoinflammatory diseases were studied.

  • As the growth of the domestic defense industry is remarkable regarding technology level and export size, technology protection is necessary. Particularly, there is a need to apply anti-tamper measures to prevent critical technologies from illegally being taken out of weapon systems. However, there is no security protection strategy and system built yet in ROK. Precedent studies discussed the trend analysis and technical research for specific protective techniques, and the application of anti-tamper using limited procedures was provided. Recently, methods of how to select the technology for protection were studied based on risk management. Nonetheless, these studies cannot be associated with the acquisition process for the whole life-cycle, having difficulty with actual development and evaluation of the weapon systems. The objective of our study is to derive the system requirements of the weapon system for which anti-tamper measures have been determined to apply. Specifically, requirements items suitable for the development of anti-tamper weapon systems were derived based on ISO/IEC 19790, the CMVP standard for the development and verification of cryptographic modules. Also, its utilization in technical reviews and test & evaluations was presented. The usefulness of the research results was confirmed through inductive inference and comparative evaluation. The result can be expected to play a role in initiating extensive activities needed for technology protection of the weapon systems.

  • A practical recursive WLS (weighted least squares) algorithm is proposed to estimate relative range using LOS (line-of-sight) information for ASM (anti-ship missile) application. Apart from the previous approaches based on the EKF (extended Kalman filter), to ensure good convergence properties in long range engagement situations, the proposed scheme utilizes LOS rate measurements instead of conventionally used LOS angle measurements. The estimation error property for the proposed filter is investigated and a simple error compensator is devised to enhance its estimation error performances. Simulation results indicate that the proposed filter produces very accurate range estimates with extremely small computations.

A detailed design of Tail-Fan system was done and structural analysis also was done. After finishing detailed design, Detailed drawings were generated for manufacture. Through detailed design and manufacture, required techniques were achieved for helicopter development.   • Human immunodeficiency virus (HIV), the causative agent of AIDS, still continues to spread rapidly in the world population, especially in Africa and Southeast Asia. At present, two kinds of therapeutic approaches are used for treatment of AIDS. One is to target HIV reverse transcriptase, which is responsible for the viral genome transcription. The other is to inhibit HIV pretense PR, which is essential for the processing of viral proteins. Drug combinations based on these approaches can reduce the blood virus to an undetectable level. However, a small amount of virus may lurk inside the immune cells in a dormant state. Another major obstacle of long-term treatment of the disease is remarkable mutation in HIV. Most of the clinical chemotherapeutic agents have one or more of these problems. High cost and harmful side-effects further reduced the desirability of these drugs. In the course our studies on development of anti-HIV agents from natural products, we investigated various crude drugs for their inhibitory activity against HIV-induced cytopathic effects (CPE) in culture cells, HIV-pretense (PR), HIV-reverse transcriptase (RT) including ribonuclease H (RNase H), and HIV integrase (INT). In the present paper, some inhibitory substances relating to the development of anti-HIV agents are reported.
